신광벼 유래의 벼 줄무늬잎마름병 저항성 주동 QTL qSTV11SG탐색 (Identification of a Major QTL, qSTV11SG, Associated with Resistance to Rice Stripe Virus Disease Originated from Shingwangbyeo in Rice (Oryza Sativa L.))

  • 벼 줄무늬잎마름병 저항성 유전자 및 연관 DNA 마커 탐색을 위하여 줄무늬잎마름병에 저항성인 통일형 품종인 신광 벼 이용 여교잡 집단을 육성하였다. 줄무늬잎마름병 저항성 유전자에 대한 QTL을 분석한 결과 11번 염색체에 위치하는 SSR 마커 RM6897이 탐색되었으며 전체 표현형 변이의 44.2%를 설명하였다. DNA 마커 RM6897은 여교잡 집단에서 생물검정과 유전자형이 일치하였다. 또한 자포니카 품종들에서 저항성 27품종과 감수성 23품종에 대해 구분이 가능하였다. 따라서 신광벼 유래의 줄무늬잎마름병 저항성원 및 분자마커는 자포니카 품종의 바이러스 저항성 향상에 효율적으로 활용될 것으로 기대된다.

  • 2002년은 여러 가지로 기상이 특이한 해였다. 겨울철의 이상난동, 3-4월의 고온 및 한발, 그리고 6월, 7월초중순의 가뭄, 7월하순부터 8월한달 내내 지속된 강우, 저온, 일조부족 등 여러모로 작물환경에 불리한 환경이 조성된 해였다. 이러한 기상특성과 상관하여 금년은 일반적으로 저온, 일조부족, 다우조건을 좋아하는 병해의 발생이 많았으며 벼의 경우 세균성알마름병, 이삭누룩병의 발생이 증가한 반면 도열병 등 주요병해는 질소비료의 시용량이 감소함에 따라 발생이 감소하였다. 채소, 과수류, 서류 병해의 발생은 생육기의 기상조건에 따라 많은 편차가 있으나 다우조건과 관련이 깊은 역병 등 저온성 병해의 발생이 증가한 반면에 탄저병, 무름병 등의 발생은 뚜렷이 감소하는 경향을 보였다.

  • 2001년 기상의 특징은 3월부터 5월까지 계속된 극심한 가뭄과 생육기간 내내 계속된 고온현상 그리고 별다른 태풍의 내습 없이 9, 10월 수확기 맑은날의 지속으로 요약할 수 있다. 벼는 순조로운 기상 때문에 잎, 이삭도열병, 세균성벼알마름병, 흰잎마름병, 잎집무늬마름병의 발생이 예년에 비해 경미하였으나, 감비추진 정책에 의해 생육후기 깨씨무늬병이 발생하여 전국적으로 문제시되었으며 그동안 발생이 적었던 줄무늬잎마름병이 경기, 충청, 호남 서해안지역을 중심으로 많이 발생하였다. 고추는 생육기 비가 많이 왔던 전남북 지역에서 역병 발생이 심하였으며 토마토와 오이는 대부분 예년에 비해 병발생이 적었다. 수박의 경우는 꽃자리 부분이 썩는 푸른곰팡이병이 전국적으로 발생하였으며 CGMMV는 전년에 비해 상당폭 발생면적이 감소하였고 노지재배의 경우 탄저병 발생이 예년에 비해 심하였다. 참외와 딸기도 전년에 비하여 병발생이 경미하였으며 예년에 발생이 많았던 참외의 급성위조증상도 발생이 대폭 감소하였다. 마늘·양파의 흑색썩음균핵병은 봄철의 가뭄과 고온현상으로 병발생의 전년의 절반수준으로 감소하였다. 감자의 풋마름병은 발병포장이 증가하는 경향이었으며 고구마의 덩굴쪼김병도 발생이 감소하였다. 사과의 경우 전반적으로 병 발생이 예년에 비해 경미하였으며 배는 흰가루병이 많이 발생하였다. 맥류의 붉은곰팡이병은 예년처럼 거기 발생이 없었다.

Development and evaluation of a model for management of plant pests in organic cucumber cultivation

  • Crop protection strategies in organic horticulture aim to prevent insect pest and plant disease problems through utilization of non-chemical based control means. In order to develop a model for management of plant diseases and insects in organic cucumber cultivation, we compared efficacies between chemical pesticide spraying system and biological control means in semi-forcing and retarding cucumber cultivation during 2005 and 2006. Conventional chemical spray program using various chemical pesticides was applied 5 - 10 days intervals, while two different non-chemical pesticide application programs using two formulated biopesticides Topseed$^{TM}$ and Q-fect$^{TM}$, Suncho$^{TM}$, and Sangsungje$^{TM}$ (biocontrol agents 1) and using egg-yolk and cooking oil(EYCO), Bordeaux mixture, Suncho$^{TM}$, and Sangsungje$^{TM}$ (biocontrol agents 2) were applied 5 - 7 days intervals during entire cucumber cultivation period. Efficacy of both biocontrol agents programs was effective to comparable to conventional chemical pesitice spray program to control plant diseases such as powdery mildew and downy mildew as well as insect pests such as aphids and thrips which are known as major threats in cucumber organic cultivation. In this study, we established and evaluated an effective and economic crop protection strategy using various biological resources can be used to control plant diseases and pests simultaneously in organic cucumber cultivation field.

유럽 유기농업 현황과 유기경종의 이론 및 핵심기술 - 독일을 중심으로 - (Situation of European Organic Agriculture and its Principle and Skills with special regard to Germany)

  • Within the paper, an overview of organic farming in Europe countries is given and the Principle and skills of organic agriculture is shortly reported with special regard to Germany. The overview information on European organic farming is covered such as (1)development of organic farming, (2) organic farming organizations, (3)standards and certification, (4)implementation of EU council regulation, (5)state support, (6)implementation of Agenda 2000, (7)training and education, (8)advisory service and research situation. In the paper the principle and skills for organic farming which are practiced actually in the German organic farms is also reported. How to maintain and increase the fertility and microbiological activity of the soil by (1)cultivation of legumes, green manures or deep-rooting crops in multi-annual rotation system, (2)incorporation in the soil organic material, by-products from livestock farming is one of the major principle to organic crop production. Pest and diseases and weeds are controlled by any one, or a combination of the following measure; (1) choice of appropriate species and varieties, (2)appropriate rotation programs, (3)mechanical cultivation, (4)protection of natural enemies of pests through provision of favourable habitat and ecological buffer zone, (5)diversified ecosystems, (6)flame weeding, (7)natural enemies, (8)bio-dynamic preparations, (9)mulching and mowing, (10)grazing of animals, (11)mechanical controls, (12)steam sterilization.

Application of Pac-Bio Sequencing, Trinity, and rnaSPAdes Assembly for Transcriptome Analysis in Medicinal Crop Astragalus membranaceus

  • Astragalus membranaceus (A. membranaceus) has traditionally been used as a medicinal plant in East Asia for the treatment ofvarious diseases. A. membranaceus belongs to the legume family and is known to be rich in substances such as flavonoids and saponins. Recent pharmacological studies of A. membranaceus have shown that the plant has immunomodulatory, anti-oxidant, anti-cancer, and anti-inflammatory effects. However, knowledge of major biosynthetic pathways in A. membranaceu is still lacking. Recently developed sequencing techniques enable high-quality transcriptome analysis in plants, which is recognized as an important part in elucidating the regulatory mechanisms of many plant secondary metabolic pathways. However, it is difficult to predict the number of transcripts because plant transcripts contain a large number of isoforms due to alternative splicing events, which can vary depending on the assembly platform used. In this study, we constructed three unigene sets using Pac-Bio isoform sequencing, Trinity and rnaSPAdes assembly for detailed transcriptome analysis mA. membranaceus. Furthermore, all genes involved in the flavonoid biosynthetic pathway were searched from three unigene sets, and structural comparisons and expression profiles between these genes were analyzed. The isoflavone synthesis was active in most tissues. Flavonol synthesis was mainly active in leaves and flowers, and anthocyanin synthesis was specific in flowers. Gene structural analysis revealed structural differences in the flavonoid-related genes derived from the three unigene sets. This study suggests the need for the application of multiple unigene sets for the analysis of key biosynthetic pathways in plants.

국내 지황에 발생하는 바이러스병 발생 현황 (The Incidence of Virus Diseases in Rehmannia glutinosa in Korea)

  • 최근 국내 지황에서는 Rehmannia mosaic virus (ReMV), Youcai mosaic virus (YoMV), Broad bean wilt virus 2 (BBWV2), Plantago asiatica mosaic virus (PlAMV), Rehamnnia virus 1 (ReV1) 등 5종의 바이러스 감염이 확인되었다. 국내 주요 지황 재배지역의 포장에서 지황 바이러스의 발병률을 조사하기 위해 2017년부터 2018년까지 여섯 지역의 포장에서 바이러스병 조사를 실시하였다. 지황에 나타나는 주요 바이러스 증상은 잎에 모자이크와 노란 반점 및 변색이었으며 생육이 저조해지는 증상이 관찰되었다. 총 145개의 시료를 채집하여 5종 바이러스에 대한 유전자 진단을 실시한 결과, 전 시료에서 2종 이상의 바이러스가 복합 감염되어 있음을 확인하였다. 지황은 뿌리로 영양번식하는 재배 특성 상 품종별 종근에서도 바이러스 감염여부를 조사하였으며, 종근에서도 5종의 바이러스가 검정됨에 따라 지황에서의 바이러스병 관리를 위해서는 무병 종근의 생산 및 보급이 매우 중요할 것으로 판단된다.

농약 노출과 건강 (Pesticide Exposure and Health)

  • Although pesticides have increased crop production and controlled disease, they have produced a number of adverse health effects. Pesticides have potential human toxicity and a variety of groups, such as farmers or industrial workers, as well as the general population, are exposed to pesticides. The purpose of this article, therefore, is to provide an overview of pesticide exposure and health through a literature review, focusing on exposure assessment, acute poisoning, chronic health effects, and future research needs. The exposure types and levels of pesticides vary by study subjects and need to be assessed by integrating several methods focused on the epidemiological purpose. Acute pesticide poisoning is a major public health problem in the world. Paraquat is the main causative pesticide for acute poisoning in Korea and should be banned in order to save several thousands of lives every year. Occupational pesticide exposure also causes numerous chronic diseases among farmers and industrial workers, including cancers, respiratory diseases, depression, retinal degeneration, diabetes, and abnormal menstrual cycle. However, controversy exists regarding the long-term effects of low-dose environmental pesticide exposure. The area of pesticide study is relatively new with broad study populations and it has received little academic and policy attention, particularly in Korea. More detailed studies investigating the risk of pesticide-related health effects and intervention efforts to reduce the problems are needed in Korea.

Evaluation of Cell Based Anti-oxidation Assay of Functional Components Derived from Domestic Major Potato Varieties

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. Potatoes have a short growing season, high production per unit area, relatively strong environmental adaptability, and are cultivated in more than 130 countries around the world. It is the world's fourth-largest crop, following rice, wheat, bean and maize. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by cell based anti-oxidation assay. The smallest amount of ROS(Reactive oxygen species) was generated when the compound was derived from 'Haryung' and 'hongyoung' and strong SOD(Superoxide dismutase) activity was observed in 'Sumi' and 'Jayoung'. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers

잔디 토양전염성병원진균에 대한 길항미생물의 분리 및 길항효과 (Isolation of Antibiotic-producing Microorganisms Antagonistic to Soilborne Pathogenic Fungi of Bentgrass and Their Antifungal Activity)

  • Recently, the importance of management and cultivation of grasses has been increased in Korea. Among these cultural practices, the appropriate control of diseases is considered more important than other cultivation techniques such as fertilization and irrigation. The damages of brown patch and large patch caused by Rhizoctonia spp. and Pythium blight caused by Pythium spp. are serious in the major cultivation area of turfgrass in Korea. Since these diseases are difficult to control by agrochemicals, the damages are very serious if these are occured. The periodic spray of agrochemicals, to protect and control these diseases could make some problems of toxicity and environmental pollution as well as rising of non-target diseases. Therefore, the biological methods to control diseases have been required to decrease problems resulted from overuse of agrochemicals, to conserve natural ecosystem, and to control effectively diseases of grasses in the long period. The number of studies about biological control using antagonistic microorganisms have been increased for last half century. However, the application of biological control method has been very limited. In this study, thirteen isolates of R. cerealis, 8 isolates of R. solani and 3 isolates of Phthyn spp. have been isolated from diseased turfgrass in golf course and grass-culture area that have patch and wilting symptoms of zoysia grass and creeping bentgrass. Isolation frequency of R. cerealis and R. solani was high in especially zoysiagrass, while Pythym spp. was isolated from bent grass at low frequency but showed high pathogenicity. Totally, 205 isolates of soil microorganisms were isolated in this study as primary antagonistic microorganism by Herr's triple agar layer plate and dual culture method using rhizosphere of grasses, soil of crop field as the source of antagonistic microorganisms. Among the 205 isolates, 23 isolates were actinomycetes and 182 isolates were bacteria. All of the actinomycetes were isolated by Herr's method. Antagonistic effect of primary isolated microorganisms was tested for in vitro mycelial growth inhibition against pathogenic fungi isolated from grasses and for inhibition of disease occurrence in 24 well tissue culture plate and pot experiment. Then, four isolated of bacteria which are BG23, BG74, BG136 and BG171 were selected as antagonistic microorganisms against soil-born pathogenic fungi of bentgrass.
