Humic and fulvic acids present in soil of different depth were extracted and their characteristics were analyzed as a basic study to evaluate the effect of humic substances on the behaviour of radioactive elements deposited on soil. Molecular size distribution of the humic and fulvic acids was measured by stirred cell ultrafiltration technique and the structural informations were obtained from their UV-Vis., IR and synchronous fluorescence (SyF) spectral analysis. Main molecular size ranges of the soil 1) humic and fulvic acids were 30~100 kDa (46~56%) and 10~30 kDa (33~43%) respectively, and their overall molecular sizes were found to became smaller with increasing the soil depth. Absorptivities measured at 280 nm in the UV-Visible spectra of humic acids were 1.4~1.5 times higher than those of fulvic acids, and increased with increasing the soil depth. SyF spectral data showed two distinct peak components having maximum peak positions of 428 nm (type I) and 498 nm (type II) for the soil humic and fulvic acids. From the analysis of the peak components, it was found that humic molecules are mainly made up of aromatic compounds corresponding to longer wavelength (type II), and the molecular components increased with increasing the soil depth. Analysis of IR spectral data indicated that the humic molecules contain a higher relative concentration of carboxylic groups than those of fulvic molecules, and the carboxylic group contents are seen to increase as the soil depth increase.
The contamination pattern of indoor air was simulated when groundwater dissolving benzene was used for household activities. Indoor exposure scenario consisted of inhalation, ingestion, and dermal absorption. Physiologically based pharmacokinetic (PBPK) model was used to analyze how benzene exposed to human body was distributed in internal organs. Main exposure pathways contributing total internal dose were inhalation and ingestion while the contribution of dermal absorption was very small. Man showed higher exposure rate than woman due to his higher breath rate. For a short-term exposure, benzene concentration in venous blood of SPT, RPT and liver changed rapidly while slowly did in venous blood of adipose tissue at a low concentration. For a long-term exposure, woman accumulated about 2.1 times higher than man. Most of benzene exposed to human body was removed by exhalation and metabolism at lung and liver, respectively. For inhalation and ingestion, the benzene removals by exhalation were 69.8 and 48.4%, respectively. Relative importance of removal mechanism was different according to the inflow displacement of benzene. The results obtained from this study would help understand exposure, distribution, and removal phenomena and make plans for the reduction of the health risk associated with the contaminated groundwater by various organic compounds.

The Mume Fructus (MF) has been used for relieves cough, arrests arrest chronic diarrhea, treat fluid depletion, and treat ascariasis in Korea. In this study, a high-performance liquid chromatography (HPLC) method was established for simultaneous determination of six main components of MF. Additionally, we were investigated the anti-inflammatory and anti-allergic effects of MF extract on lipopolysaccharide (LPS)-treated RAW264.7 cells and tumor necrosis factor (TNF)-${\alpha}$/interferon (IFN)-${\gamma}$-treated HaCaT cells. The analytical column for separation was used a Gemini $C_{18}$ column maintained at $40^{\circ}C$. The mobile phase consisted of 1.0% (v/v) acetic acid in water (A) and 1.0% (v/v) acetic acid in acetonitrile (B). The flow rate was 1.0 mL/min and the detector was a photodiode array (PDA) set at 280 nm and 320 nm. We evaluated the inhibitory effect of MF extract on the production of inflammatory markers, nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) in LPS-stimulated RAW264.7 cells and thymus- and activation-regulated chemokine (TARC/CCL17) in TNF-${\alpha}$/IFN-${\gamma}$-treated HaCaT cells, respectively. We confirmed the genes expression related with TARC, macrophage-derived chemokine (MDC/CCL22) and regulated on activation, normal T cell expressed and secreted (RANTES/CCL5) in HaCaT keratinocyte cells by MF extract. The contents of the five compounds in MF were 0.22-1.01 mg/g. Also, the MF extract show inhibition of about 78% and 75% on NO and $PGE_2$ production at the concentration 1000 mg/mL in RAW264.7 cells. MF extract suppressed the hTARC level and genes expression such as TARC, MDC, and RANTES on TNF-${\alpha}$/IFN-${\gamma}$-treated HaCaT cells.

Monoterpenoids were recently found as main biologically active compounds which is responsible for various physiological effect in goat's beard (Aruncus dioicus). Ethyl acetate extract of A. dioicus (ADE) was treated to B16F10 melanoma cells for the examination of whitening activity. MTT assay was performed to evaluate cell toxicity and the result showed that slight cell toxicity (> 10%) by over $500{\mu}g{\cdot}mL^{-1}$. Thus, 0, 5, 10, or $50{\mu}g{\cdot}mL^{-1}$ ADE was used for further experiments. We found that tyrosinase activity was decreased according to ADE concentration, and the total melanin content was also dramatically reduced. Especially with $50{\mu}g{\cdot}mL^{-1}$ ADE treatment tyrosinase activity was reduced to 35.6%, and 58.8% of melanin content was lowered. In addition, whitening related proteins including tyrosinase, tyrosinase related protein 1 (TRP1), TRP2, microphthalmia associated transcription factor (MITF) and cAMP and protein kinase A (PKA) were reduced by ADE treatment. It caused decreased phosphorylation of cAMP response binding protein (CREB) but increased phosphorylation of extracellular signal related kinase (ERK). Therefore, in this paper we would like to suggest the potent usage of A. dioicus natively grown in Ulleungdo, Korea as materials of functional cosmetics by confirming whitening activity related with melanin content.
Volatile flavor components in three varieties (shingo(niitaka), mansamgil (okusankichi) and chuwhang pears) of Pear (Pyrus pyrifolia N.) were extracted for 24 hours with pentane-diethylether (1 : 1, v/v) using the LLEP (liquid-liquid extraction & perforation). Neutral fraction was separated from the extract and then analyzed by GC-FID and GC/MS equipped with a fused silica capillary column (Carbowax 20M, HP). Individual components were identified by mass spectrometry and their retention indices. The totals of 52, 47 and 22 volatiles were identified in shingo, mansamgil and chuwhang pears, respectively. Ethyl acetate, propyl acetate, hexanal, 1-hexanol, ethyl butanoate, ethyl-3-hydroxy butanoate, ethyl-2-hydroxy propanoate were the main components in each samples, though there were several differeces in composition of volatile compounds. Total contents of volatile components isolated in shingo, mansamgil and chuwhang pears were 6.972, 2.776 and 2.653 mg/kg of pears.

Modem drug discovery requires rapid pharmacokinetic evaluation of chemically diverse compounds for early candidate selection. This demands the development of analytical methods that offer high-throughput of samples. Naturally, liquid chromatography / tandem mass spectrometry (LC-MS/MS) is choice of the analytical method because of its superior sensitivity and selectivity. As a result of the short analysis time(typically 3-5min) by LC-MS/MS, sample preparation has become the rate- determining step in the whole analytical cycle. Consequently tremendous efforts are being made to speed up and automate this step. In a typical automated 96-well SPE(solid-phase extraction) procedure, plasma samples are transferred to the 96-well SPE plate, internal standard and aqueous buffer solutions are added and then vacuum is applied using the robotic liquid handling system. It takes only 20-90 min to process 96 samples by automated SPE and the analyst is physically occupied for only approximately 10 min. Recently, the ultra-high flow rate liquid chromatography (turbulent-flow chromatography)has sparked a huge interest for rapid and direct quantitation of drugs in plasma. There is no sample preparation except for sample aliquotting, internal standard addition and centrifugation. This type of analysis is achieved by using a small diameter column with a large particle size(30-5O ${\mu}$m) and a high flow rate, typically between 3-5 ml/min. Silica-based monolithic HPLC columns contain a novel chromatographic support in which the traditional particulate packing has been replaced with a single, continuous network (monolith) of pcrous silica. The main advantage of such a network is decreased backpressure due to macropores (2 ${\mu}$m) throughout the network. This allows high flow rates, and hence fast analyses that are unattainable with traditional particulate columns. The reduction of particle diameter in HPLC results in increased column efficiency. use of small particles (<2 urn), however, requires p.essu.es beyond the traditional 6,000 psi of conventional pumping devices. Instrumental development in recent years has resulted in pumping devices capable of handling the requirements of columns packed with small particles. The staggered parallel HPLC system consists of four fully independent binary HPLC pumps, a modified auto sampler, and a series of switching and selector valves all controlled by a single computer program. The system improves sample throughput without sacrificing chromatographic separation or data quality. Sample throughput can be increased nearly four-fold without requiring significant changes in current analytical procedures. The process of Bioanalytical Method Validation is required by the FDA to assess and verify the performance of a chronlatographic method prior to its application in sample analysis. The validation should address the selectivity, linearity, accuracy, precision and stability of the method. This presentation will provide all overview of the work required to accomplish a full validation and show how a chromatographic method is suitable for toxirokinetic sample analysis. A liquid chromatography/tandem mass spectrometry (LC-MS/MS) method developed to quantitate drug levels in dog plasma will be used as an example of tile process.

Fish pathogens cause not only economic damages to fish farming but also infectious pathogens known as a zoonotic agent. Since the continued use of antibiotics to control fish pathogens entails side effects, materials of natural origin need to be developed. The purpose of this study is to discover coniferous essential oils with excellent antibacterial effects in order to develop antibiotic alternatives. We have extracted essential oils using hydro-distillation from the leaves of Abies holophylla, Pinus thunbergii, Pinus parviflora, Tsuga sieboldii, and Pinus rigitaeda, which are all Pinaceae family. And, we have evaluated antibacterial activity with the extracted essential oils against Edwardsiella tarda, Photobacterium damselae, Streptococcus parauberis, and Lactococcus garvieae, which are fish pathogens. As a result, the essential oils from A. holophylla and P. thunbergii showed the selectively strong antibacterial activity against E. tarda and P. damselae, which are gram-negative bacteria. From GC-MS analysis, it was identified that main component of A. holophylla essential oils are (-)-bornyl acetate (29.45%), D-limonene (20.47%), and camphene (11.73%), and that of P. thunbergii essential oils is α-pinene (59.81%). In addition, we found three compounds: neryl acetate, (-)-borneol, and (-)-carveol, which are oxygenated monoterpenes. These exist in a very small amount but exhibit the same efficacy as essential oil. Therefore, we expect that A. holophylla and P. thunbergii essential oils having excellent growth inhibitory effect against gram-negative fish pathogens can be used as biological products such as feed additives and fishery products.
The essential oils from the aerial parts of Tagetes minuta L. from two different locations in Korea were obtained by hydrodistillation in 0.52%(Goheung, Chonnam) and 0.48% yields(Puan, Chonbuk) as a dry weight base, respectively. A total of 69 components, accounting in 94.7% and 92.1% of the oil, respectively were identified by gas chromatography-mass spectrometric analysis and comparison of retention indices with those of authentic compounds. The main components in both samples(Goheung, Chonnam and Puan, Chonbuk) were limonene(5.83% and 6.41%, respectively), $cis-{\beta}-ocimene$ (4.87% and 47.73%), dihydrotagetone(14.78% and 52.83%), trans-tagetone(0.64% and 2.99%), cis-tagetone(1.13% and 2.50%), trans-tagetenone(1.15% and 11.45%) and cis-tagetenone(0.69% and 1.41 %). Although these components were present in both samples, the results showed large differences in the percentage composition of these components.

To evaluate the efficiency of the decontamination plant for the removal of uranium compounds deposited on the internal surface of $UF_6$ cylinder for its reuse, two demonstration tests of the plant with different ratio of ${Na_2}{CO_3}$ and ${H_2}{O_2}$ were carried out, and each test had 5 steps. The main chemical form removed by the tests was to be identified as ${Na_4}{UO_2}(CO_3)_3$. More than 50% of uranium was removed by water of the first step, and at the following steps the removal amounts were exponentially decreased. On the other hand, the result shows that the injected amount of ${Na_2}{CO_3}$, compared with that of the removed uranium, was stoichiometrically excessed. This suggests that the injected amounts of ${Na_2}{CO_3}$, the generation rate of decontaminated waste, and the decontamination steps could be reduced by a process optimization of the plant.
The optimum regeneration conditions for the regeneration of three way spent catalysts (TWCs), which were taken from automobiles with different driving conditions, were investigated to evaluate the suitability as alternative catalysts for removing VOCs. The spent catalysts were washed with five different acids ($HNO_3$, $H_2SO_4$, $C_2H_2O_4$, $C_6H_8O_7$, and $H_3PO_4$) to remove contaminants and examine the optimum conditions for recovering the catalytic activity. The physicochemical properties of spent and its regenerated TWCs were evaluated by using nitrogen adsorption-desorption isotherms, XRD, and ICP. The relative atomic ratios of contaminants and platinum group metals (PGMs) of the spent TWCs were greatly dependent on the placed positions. The main contaminants formed were lubricant oil additives and metallic components. Also, the regeneration treatment increased the PGMs ratio, BET surface area, and average pore diameter of TWCs. The catalytic activity results indicated that the spent TWCs have the possibility for removing VOCs. Moreover, the employed acid treatments greatly enhanced the catalytic activity of the spent TWCs. Especially, nitric and oxalic acids provided the most improvement in the catalytic behavior. The catalytic activities of the regenerated TWCs were significantly influenced by the containing platinum ratios rather than the removal ratios of contaminants and the changes in the structural properties offered by the acid treatments.
