A Comparison between Extract Products of Magnolia officinalis on Memory Impairment and Amyloidogenesis in a Transgenic Mouse Model of Alzheimer's Disease

  • The components of Magnolia officinalis have well known to act anti-inflammatory, anti-oxidative and neuroprotective activities. These efficacies have been sold many products as nutritional supplement extracted from bark of Magnolia officinalis. Thus, to assess and compare neuroprotective effect in the nutritional supplement (Magnolia $Extract^{TM}$, Health Freedom Nutrition LLC, USA) and our ethanol extract of Magnolia officinalis (BioLand LTD, Korea), we investigated memorial improving and anti-Alzheimer's disease effects of extract products of Magnolia officinalis in a transgenic AD mice model. Oral pretreatment of two extract products of Magnolia officinalis (10 mg/kg/day in 0.05% ethanol) into drinking water for 3 months ameliorated memorial dysfunction and prevented $A{\beta}$ accumulation in the brain of Tg2576 mice. In addition, extract products of Magnolia officinalis also decreased expression of ${\beta}$-site APP cleaving enzyme 1 (BACE1), amyloid precursor protein (APP) and its product, C99. Although both two extract products of Magnolia officinalis could show preventive effect of memorial dysfunction and $A{\beta}$ accumulation, our ethanol extract of Magnolia officinalis (BioLand LTD, Korea) could be more effective than Magnolia $Extract^{TM}$ (Health Freedom Nutrition LLC, USA). Therefore, our results showed that extract products of Magnolia officinalis were effective for prevention and treatment of AD through memorial improving and anti-amyloidogenic effects via down-regulating ${\beta}$-secretase activity, and neuroprotective efficacy of Magnolia extracts could be differed by cultivating area and manufacturing methods.

후박(厚朴)의 약효(藥效)에 관한 연구(硏究) (Comparison of Pharmacological Effects of Magnoliae officinalis, Magnolia and Machilus Barks)

  • It has been known that Magnolia officinalis, Magnolia and Machilus Barks are reputed to be effective in 'Hagi', 'Sanman' and to be analgesic but there was differences of efficacy of these crude drugs against the kinds and habitates. For this reason, this study was conducted to compare the acute toxicities, analgesic effects and motilities of isolated intestine. This results obtained in this work were summarized as follow: 1. In the test of acute toxicies, Machilus bark was found to be more toxic than the others. 2. Both these was found to have analgesic effect. 3. In the effects against the motility of isolated intestine, Magnolia officinalis and Magnolia barks were found to be relaxant, Machilus bark was found to be contractive. From these results, Magnolia officinalis and Magnolia barks might have analgesic effect and relaxant effect of intestine.

후박 에탄올 추출물의 불안 및 인지기능에 대한 효과 (Effects of Ethanol Extract from Magnolia Officinalis on Anxiety and Cognitive Function)

  • The bark of Magnolia officinalis has been used in traditional oriental medicine to treat a variety of mental disorders including anxiety and depression. The purpose of this study was to examine the effect of M. officinalis ethanol extract on stress-induced alterations in learning and cognitive function using a passive avoidance test (PAT) and also on anxiety-related behavior using the elevated plus-maze test (EPM) in female rats . The degree of Tyrosine hydroxylase (TH) in the region of the ventral tegmental area (VTA) and the locus coeruleus (LC) was measured using an immunohistochemical method. Corticosterone concentrations in serum were also measured. The ethanol extract from Magnolia officinalis was orally administered to female rats 30 minutes before evaluating their immobilization stress and anxiety-related behavior using an elevated plus-maze test and a passive avoidance test. Time spent in the open arms of the EPM increased in the M. officinalis-treated group compared with that of the saline-treated control group. In the passive avoidance test, the memory and cognitive function improved in the M. officinalis extract-treated group. M. officinalis extracts reduced elevated corticosterone concentrations in serum. Also, stress-induced TH increases were suppressed in the M. officinalis extract-treated group in the LC and the VTA region. These results suggest that M. officinalis might prove to be an effective anxiolytic anti-stress agent.

형태특징 및 RAPD분석에 의한 후박(厚朴) 약재의 진위감별 (Discrimination of Genuines and Misuses of Hu-Bak medicines by Morphological Characters and Random Amplified Polymorphic DNA Analysis)

  • For identification of genuine and misuse samples of Hu-Bak medicines, an morphological characters, and RAPD (random amplified polymorphic DNA) were performed. In this study, three samples were collected, of which two were genuine and one was misuse sample. Genuine samples of 'Hu-Bak' were Magnolia obovata Thunberg and Magnolia officinalis Rehder et Wilson. Misuse samples was Machilus thunbergii S. et Z. In the morphological characters, M. obovata Thunberg and M. officinalis Rehder et Wilson were similar, but M. thunbergii S. et Z. was different with cortex, cambium, and fibrous layer. The result of RAPD analysis, seven primers can distinguish between genuines and counterfeit samples of Hu-Bak medicines.

후박의 항우울 효과에 대한 실험적 연구 (Experimental Study on the Antidepressant Effects of Magnolia Officinalis Extracts)

  • Objectives : The purpose of this study was to investigate the protective effects of Magnolia Officinalis extracts on the animal model of depression induced by immobilization stress. Methods : The subjects were divided into 4 groups : normal, saline solution-administered during immobilization stress, 200 mg/kg of magnolia extracts-administered (magnolia extract 200), and 400 mg/kg of magnolia extracts- administered (magnolia extract 400). During 2 days of immobilization stress treatment, they underwent forced swimming test (FST) and tail suspension test (TST). The number of serotonin (5-HT) immunostained nuclei in the dorsal raphe nucleus regions was measured by immunohistochemistry. Superoxide dismutase (SOD) and glutathione peroxidase (GPX) in blood were measured. Results : In FST, magnolia-administered groups showed significantly decreased immobilization. In TST, the magnolia extract 400 group showed decreased immobilization. The stress group showed significantly decreased number of 5-HT immunostained nuclei in the dorsal raphe nucleus regions, while magnolia extract 400 group showed increased number of 5-HT immunostained nuclei. Stress group showed decrease in serum level of SOD and GPX, while the magnolia extract 200 group showed increase in serum level of SOD and GPX. Conclusions : These results suggest potent effectiveness of magnolia extracts in the treatment of depression.

Inhibitory Effect of Methanol Extract of Magnolia officinalis on Matrix Metalloproteinase-2

  • Matrix metalloproteinase-2 (MMP-2) is a key enzyme involved in tumor invasiveness. The plant of Magnolia officinalis Rehd. et Wils. is often included as an ingredient in various herbal remedies recommended for cancer theraphies in Korea. Various extracts prepared from stems of M. officinalis were tested for cytotoxic activity on human hepatocellular carcinoma cell line, SK-Hep cells using the XTT assay method. Then, the inhibitory effect was examined on MMP-2 activity using gelatin zymography. Methanol (MeOH) extract of M. officinalis caused the strongest inhibition of the MMP-2 activity, as measured by gelatin zymography method for enzyme activity. $IC_{50}$ values of fractions on MMP-2 activity were in a range of $4.9{\sim}11.3\;{\mu}g/mL$. Among each fraction, butanol and ethylacetate (EtOAc) fractions showed the strong inhibitory activities ($IC_{50}=10.7\;and\;4.9\;{\mu}g/mL$, respectively). When the M. officinalis's constituents such as magnolol, honokiol, (-)-epigallocatechin gallate (EGCG) and ovovatol were examined for inhibitory effects on MMP-2 activity, EGCG showed strong inhibitory activity. However, MeOH extract of M. officinalis was dose-dependently inhibited to MMP-2 activity. The MeOH extract, hexane and EtOAc fractions $(IC_{50}\;of\;>200\;{\mu}g/mL)$ exhibited weak cytotoxicity activity, while butanol $(IC_{50}=80\;{\mu}g/mL)$ and chloroform fractions $(IC_{50}=90\;{\mu}g/mL)$ exhibited relatively strong cytotoxic activity. From these results, M. officinalis could be suitable for cancer treatment and chemopreventive drugs.

후박 추출물의 지방세포 분화 억제 효능에 관한 연구 (Inhibition of Adipogenesis in 3T3-L1 Adipocytes with Magnolia officinalis Extracts)

  • 후박추출물은 중국 원산인 당후박에서 제조되며, 항산화 및 항염증 효과로 알려져 있다. 후박의 성분으로는 폴레페놀인 honokiol 및 magnolol 등이 알려져 있다. 이 연구에서, 우리는 후박추출물이 triglyceride (TG)의 축척을 감소시킴으로써 지방세포 분화를 억제한다는 표면적인 결과를 얻었다. 또한, 후박 추출물이 hormone sensitive lipase (HSL) 단백질 수준을 증가시키고 adipocyte에 specific한 transcription factors로 peroxisome proliferator activated receptor (PPAR)-${\gamma}$의 단백질과 mRNA 수준을 억제하였다. 결과적으로, 후박추출물이 항비만제로서 adipogenic transcription factor와 그들의 특이적인 유전자의 발현을 억제함으로써 지방세포 분화를 억제한다고 사료된다.

후박(厚朴)의 생약학적 연구 (Pharmacognostical Studies on the ‘Hoo Bak’)

  • 'Hoo Bak' is one of the Chinese crude drugs used mainly to cure a headache, apoplexy and dyspepsia. With regard to the botanical origin of 'Hoo Bak', it has been considered to be Machilus thunbergii of Lauraceae in Korea. But there has no pharmacognostical confirmation on it. To clarify the botanical origin of 'Hoo Bak', the anatomical characteristics of the bark of Machilus thunbergii, Magnolia officinalis and Magnolia obovata were studied. As a result, it was clarified that 'Hoo Bak' from Korea was the bark of Machilus thunbergii of Lauraceae.

네트워크 약리학을 이용한 소양증을 동반한 피부 염증에 대한 창출(蒼朮) 및 후박(厚朴)의 잠재적 치료기전 탐색 (Analysis of Potential Active Ingredients and Treatment Mechanism of Atractylodes Lancea(Thunb.) D.C and Magnolia Officinalis Rehder et Wilson for Dermatitis Accompanied by Pruritus Using Network Pharmacology)

  • Objectives : To investigate the active compounds and therapeutic mechanisms of Atractylodes Lancea(Thunb.) D.C. and Magnolia Officinalis Rehder et Wilson in the treatment of dermatitis accompanied by pruritus, as well as their potential to complement or replace standard drugs. Methods : We conducted the network pharmacological analysis. We selected effective ingredients among the active compounds of research target herbs. Then we explore pathway/terms of the common target proteins among research target herbs, fexofenadine and disease. Results : We selected 9 active compounds are selected from Atractylodes lancea and identified 231 target proteins. Among them, 74 proteins are associated with inflammatory skin diseases that cause pruritus. These proteins are involved in various pathways including, 'Nitric-oxide synthase regulator activity', 'Hydroperoxy icosatetraenoate dehydratase activity, Aromatase activity', 'RNA-directed DNA polymerase activity', 'Arachidonic acid metabolism', 'Peptide hormone processing', 'Chemokine binding' and 'Sterol biosynthetic process'. Additionally, coregenes are involved in 'IL-17 signaling pathway'. Similarly, we selected 2 active compounds from Magnolia officinalis and identified 133 target proteins. Among them, 33 proteins are related to inflammatory skin diseases that cause pruritus. These proteins are primarily involved in 'Vascular associated smooth muscle cell proliferation' and 'Arachidonic acid metabolism'. There is no significant difference between the pathways in which coregenes are involved. Conclusions : It is expected that Atractylodes Lancea will be able to show direct or indirect anti-pruritus and anti-inflammatory effects on skin inflammation accompanied pruritus through suppressing inflammation and protecting skin barrier. Meanwhile, it is expected that Magnolia Officinalis will only be able to show indirect anti-inflammation effects. Therefore, Atractylodes Lancea and fexofenadine are believed to complement each other, whereas Magnolia Officialinalis is expected to provide supplementary support on skin disease.

A Cytotoxic Monoterpene-Neolignan from the Stem Bark of Magnolia officinalis

  • A monoterpene-neolignan, piperitylmagnolol (1), was isolated from the stem bark of Magnolia officinalis, together with syringaresinol (2), caffeic acid (3), and sinapaldehyde (4). The isolated compounds were established on the basis of spectroscopic and physicochemical analyses including 1D- and 2D-NMR techniques, as well as on comparing the spectral data with those in the literature and of authentic samples. Compounds 1 - 4 were tested for their cytotoxic activity against the HeLa, K562, A549, and HCT116 cancer cell lines in vitro. Of the isolates, piperitylmagnolol (1) exhibited cytotoxic activity against the tested cancer cell lines with $IC_{50}$ values of 7.7 - 9.5 ${\mu}g/ml$.
