Immunomodulatory activity of Salicornia herbacea L. Components

  • Salicornia herbacea is an annual herb growing in salt marshes and on muddy seashores. Salicornia herbacea has been used as a fork medicine as well as a seasoned vegetable. In fork medicine, Salicornia herbacea has been used to treat a variety of diseases such as constipation, obesity, diabetes, asthma, arthritis and cancer. However, the biological mechanisms for these activities have not been characterized, nor the active components. The immunomodulatory activity of Salicornia herbacea components were studied in the present study. The components of Salicornia herbacea were prepared from the whole plant by passage through a fine screen, and then dialyzed against PBS overnight. Immunomodulatory activities of the Salicornia herbacea components were examined on a mouse macrophage cell line, RAW 264.7 cells. The Salicornia herbacea components were shown to stimulate cytokine production, nitric oxide release, and expression of surface molecules in a dose dependent manner. The Salicornia herbacea components also induced further differentiation of slightly adherent RAW 264.7 cell into strongly adherent macrophages. These results indicate that Salicornia herbacea contains immunomodulator(s) that induces activation of macrophages.

Effect of the addition temperament drugs of Yeongyopaedock-san(連翹敗靑散加味方)on acne in the state of inflammation (連翹敗毒散加味方이 炎症狀態의 面胞에 미치는 影響)

  • In the age of puberty or 20-30th young people who are sensitive to outward appearance, acne is serious problem at beauty and has social and psychological influence on that people. So this experiment is carried out for test whether the addition temperament drugs of Yungyopaedock-san(YP) have an anti-inflammatory effect and have suppression effect on immunocyte in the state of inflammation which induced by acne. The results was as follows. 1. YP has suppress inflammatory reaction induced by carageenan. 2. YP has suppress increasing activation of abdominal cavity macrophage in the carageen and zymosan induced inflammation. 3. YP has suppress increasing activation of spleen cell in the carageenan and zymosan induced inflammation. Based on the above result, YP was improved its suppression effect to the inflammatory reaction through the suppression of spleen cell and macrophage activation. So we concluded that YP is prospected as a anti-inflammatory agent to cure inflammation induced by ance.

Effect of Polysaccharide Extracted from Panax ginseng on Murine Hematopoiesis (인삼 다당체가 생쥐의 조혈과정에 미치는 영향)

  • We previously reported that acidic polysaccharide from Panax ginseng induced the proliferation lymphocytes and the generation of activated killer cells. Here we found that polysaccharide (PG-75) precipitated with 75% EtOH from water extract of Panax ginseng also has both in vitron and in vivo hematopoietic activities. In vitro studied with bone marrow cells from BALB/c mouse revealed that PG-75 had direct effect on hematopoietic colony-forming cell(CFC) growth, increased granulocyte macrophage-colony forming cell numbers by 1.59 fold over than non-treated. the ability of PG-75 to modulate hematopoiesis in vivo was evaluated the bone marrow and spleen celluarity, granulocyte-macrophage progenitor cells. BALB/c female mice were administered G-75 intraperitoneally, PG-75 was found to significantly increase the number of BM cells, spleen cells, GM-CFU on 3 hours after injection. PG-75 was also able to induce significant augmentation of GM-CSF and IFN-${\gamma}$, production in sera. These studies illustrate than PG-75 has hematopoietic activities and that this agent may be useful in the prevention and/or treatment of radio- or chemotherapy-associated myelosuppression.

Isolation and Identification of Inhibitory Compounds on TNF-$\alpha$ Production from Magnolia fargesii

  • Three TNF-$\alpha$inhibitory lignans were isolated from the flower buds of Magnolia fargesii through bioassay-guided isolation. They were identified as eudesmin, magnolin and lirioresinol-B dimethylether on the basis of their spectroscopic data. All three lignans showed inhibitory effects on TNF-$\alpha$ production in LPS-stimulated murine macrophage cell line, RAW264.7 and eudesmin showed the strongest activity ($IC_{50}=51{\mu}M)$.

  • Tumor immune evasion is a complex process that involves various mechanisms, such as antigen recognition restriction, immune system suppression, and T cell exhaustion. The tumor microenvironment contains various immune cells involved in immune evasion. Recent studies have demonstrated that granulocyte colony-stimulating factor (G-CSF) and granulocyte-macrophage colony-stimulating factor (GM-CSF) induce immune evasion in lung cancer by modulating neutrophils and myeloid-derived suppressor cells. Here we describe the origin and function of G-CSF and GM-CSF, particularly their role in immune evasion in lung cancer. In addition, their effects on programmed death-ligand 1 expression and clinical implications are discussed.

  • Antigen presenting cells (APCs), dendritic cells (DCs) and macrophages, playa critical role not only in the initiation of immune responses, but also in the induction of immune tolerance. In an effort to regulate immune responses through the modulation of APC function, we searched for and characterized APC function modulators from natural products. Bifidobacterium pseudocatenulatum SPM1204 (SPM1204) isolated from feces of healthy Korean in the age of 20s was used in this experiment. DCs and macrophages were cultured in the presence of supernatants of SPM 1204 and then examined for their activities for the presentation exogenous antigen in association with major histocompatibility complexes (MHC) and macrophage activation. SPM1204 increased class I MHC-restricted presentation of exogenous antigen (cross-presentation) in a DC cell line, DC2.4 cells. The RAW 264.7 cell line was used to test the nonspecific effect of immune reinforcement of SPM1204 as a source of biological regulating modulator for the macrophage activation, include nitric oxide (NO) production and cytokine production. Results showed that the production of NO, tumor necrosis factor (TNF)-$\alpha$, interleukin 1 (IL-1)-$\beta$ and morphological changes in macrophages were largely affected by SPM1204 in a dose-dependent manner. Our results demonstrated that SPM1204 promote cross-presentation of dendritic cells as well as the induction of NO, TNF-$\alpha$ production, and activation of macrophage.

Effects of Angelicae Pubescentis Radix Water Extract on Immune Property in RAW 264.7 Macrophages (독활 물 추출물이 대식세포 면역 활성에 미치는 영향)

  • Objectives: The purpose of this study was to investigate the effects of Angelicae pubescentis Radix water extract (ACE) on immune properties in macrophage cells. Methods: The cells were divided into two groups: As a control, the first was not treated with ACE, and the other was treated with ACE. Together with the cell viability, productions of nitric oxide (NO) and cytokines such as interleukin (IL)-$1{\beta}$, IL-6, and tumor necrosis factor (TNF)-${\alpha}$ by treating of ACE were monitored. Results: 1. There was no decrease of the cell viability after 24 hr incubation, but a significant decrease after 48 hr incubation with all four concentrations (25, 100, 200, and $400\;{\mu}g/m{\ell}$) of ACE. 2. A significant increase in the production of NO was observed in the concentrations above $50\;{\mu}g/m{\ell}$ of ACE after 24 hr incubation. 3. Further, after 48 hr incubation, the critical concentration of ACE for the increase was reduced to $25\;{\mu}g/m{\ell}$. 4. The production of (IL)-$1{\beta}$ significantly increased with the ACE concentrations of 100 and $200\;{\mu}g/m{\ell}$ after 24 hr incubation. 5. The production of IL-6 significantly increased with the ACE concentration of $200\;{\mu}g/m{\ell}$ after 24 hr incubation. 6. A significant increase in the production of (TNF)-${\alpha}$ was detected with ACE concentrations of 50, 100, and $200\;{\mu}g/m{\ell}$ after 24 hr incubation. Conclusions: These show that ACE increases mouse macrophage NO production at concentrations above $50\;{\mu}g/m{\ell}$, and the cytokines ((IL)-$1{\beta}$, IL-6, and (TNF)-${\alpha}$) at concentrations above $200\;{\mu}g/m{\ell}$. These results suggest that ACE improves macrophage immune property.

The Effects of Flammulina velutipes Water Extract on the Activation of Spleen Cell and Macrophage in Mice (팽이버섯 열수추출물이 마우스 비장세포와 대식세포의 증식 및 활성에 미치는 효과)

  • Flammulina velutipes is an edible mushroom and contains a lot of fiber, vitamin $B_1$, $B_2$, niacin and folic acid. This study was conducted to explore the effects of the Flammulina velutipes mushroom on immune cells and immunity. Th1 cytokine productions as $IFN-{\gamma}$, $TNF-{\alpha}$, and IL-2 were measured in an activated macrophage by Flammulina velutipes water extract in seven concentrations (0, 5, 10, 50, 100, 250, 500, and $1,000{\mu}g/mL$). Also, the splenocyte proliferation index was measured at 48 hours after treatment of the Flammulina velutipes water extract in seven concentrations or mitogen, LPS and ConA. The $IFN-{\gamma}$ and $TNF-{\alpha}$ productions were increased by treatment of the Flammulina velutipes water extract. The $TNF-{\alpha}$ production was significantly higher in the $50{\sim}1,000{\mu}g/mL$ Flammulina velutipes water extract treated macrophages. The $IFN-{\gamma}$ production of macrophages treated with the Flammulina velutipes water extract increased significantly in all groups, and the highest $1000{\mu}g/mL$ concentration. The splenocyte proliferation index was enhanced when the $10{\sim}1,000{\mu}g/mL$ Flammulina velutipes water extracts were treated compared to the control. These primary results suggest that Flammulina velutipes may enhance the immune function by activation of the macrophage and spleen cell.

Effects of Prunellae Spica Water Extract on Immune Response in Macrophage Cells (하고초 열수추출물이 대식세포 면역만응에 미치는 영향)

  • Purpose: The purpose of this study was to investigate the effects of Prunellae Spica Water Extract(PSE) on immune response in macrophage cells. Methods: We had devided two group the one is normal group; not treated with PSE, and the other is experimental group; treated with PSE. We measured the cell viability of PSE on RAW 264.7 cells and investigated production of nitric oxide(NO) and cytokines such as interleukin(IL)-$1{\beta}$, IL-6 and tumor necrosis factor (TNF)-$\alpha$ with sample PSE. Results: 1. Cell viability of PSE on RAW 264.7 cells was significantly decreased in both 24 hr and 48 hr incubation. 2. NO production of PSE on RAW 264.7 cells was significantly increased in both 24 hr and 48 hr incubation. 3. IL-$1{\beta}$ production of PSE on RAW 264.7 cells was significantly increased under concentration over $50\;{\mu}g/m{\ell}$ in 24 hr incubation. 4. IL-6 production of PSE on RAW 264.7 cells was significantly increased under concentration over $50\;{\mu}g/m{\ell}$ in 24 hr incubation. 5. TNF-$\alpha$ production of PSE on RAW 264.7 cells was significantly increased under concentration over $50\;{\mu}g/m{\ell}$ in 24 hr incubation. Conclusion: NO, IL-$1{\beta}$, IL-6 and TNF-$\alpha$ production of PSE on RAW 264.7 cells was significantly increased. This study suggest that PSE stimulates the macrophage and enhances the immune response.

Increased Lymphocyte Infiltration in Rheumatoid Arthritis Is Correlated with an Increase in LTi-like Cells in Synovial Fluid

  • In this study, we compared the immune cell populations in rheumatoid arthritis (RA) synovial fluid, which shows lymphoid tissue-like structure, with those in tonsils, which are normal secondary lymphoid tissues. Firstly, we found that $CD4^-CD11b^+$ macrophages were the major population in RA synovial fluid and that B cells were the major population in tonsils. In addition, synovial fluid from patients with osteoarthritis, which is a degenerative joint disease, contained $CD4^+CD11b^+$monocytes as the major immune cell population. Secondly, we categorized three groups based on the proportion of macrophages found in RA synovial fluid: (1) the macrophage-high group, which contained more than 80% macrophages; (2) the macrophage-intermediate group, which contained between 40% and 80% macrophages; and (3) the macrophage-low group, which contained less than 40% macrophages. In the macrophage-low group, more lymphoid tissue inducer (LTi)-like cells were detected, and the expression of OX40L and TRANCE in these cells was higher than that in the other groups. In addition, in this group, the suppressive function of regulatory T cells was downregulated. Finally, CXCL13 expression was higher in RA synovial fluid than in tonsils, but CCL21 expression was comparable in synovial fluid from all groups and in tonsils. These data demonstrate that increased lymphocyte infiltration in RA synovial fluid is correlated with an increase in LTi-like cells and the elevation of the chemokine expression.
