Screening of Anti-angiogenic Activity from Plant Extracts (식물 추출물의 혈관신생 억제 효능 검색)

  • We examined anti-angiogenic effects of water extracts from 32 plant materials (20 Korean medicinal plants and 12 western herbs) using cell-based anti-angiogenic assay, HUVEC tube formation assay, and then we found that 7 plant extracts inhibited HUVEC tube formation strongly. The plant materials which showed anti-angiogenic effects are Cinnamomi Ramulus, Atractylodis Rhizoma alba, Polygalae Radix, Myristicae Semen, Artemisiae Iwayomogii Herba, leaves of Rosmarinus officinalis, and leaves of Melissa officinalis. We also investigated inhibitory effects of these anti-angiogenic herbal extracts on MMP (matrix metalloproteinase) activity which has important roles in angiogenesis. Among extracts tested in this study, water extract of Melissa officinalis showed the most potent anti-angiogenic and MMP inhibitory activity.

  • 1. Objectives: The purpose of this study is to understand the origin, changes and the constructive principles of Hyangsayangyitang (XiangShaYangWeiTang). 2. Methods: Hyangsayangyi-tang and other related prescriptions were analyzed in terms of pathology, based on "Donguibogam(東醫寶鑑)", "Donguisusebowon Chobongwon(東醫壽世保元 草本卷)", "Donguisusebowon Gabobon(東醫壽世保元 甲午本)", "Donguisusebowon Sinchukbon(東醫壽世保元 辛丑本)". 3. Results and Conclusions: (1) The origin of Hyangsayangyi-tang is Hyangsayukgunja-tang from "Gongshin Uigamseo(龔信 醫鑑書"). Hyangsayangyi-tang inherited some of the principles from"Euihangangmok(醫學綱目)" Jeonssiyigong-san's (全氏異功散) application. (2) "Gabobon(甲午本)"'s prescription emphasizes the effect of lossen up by using Pogostemonis Herba(藿香). "Sinchukbon(辛丑本)"'s prescription emphasizes the effect of warm up stomach by using Zingiberis Rhizoma(乾薑). (3) Ginseng Radix(人蔘), Atractlodis Rhizoma White(白朮), Glycyrrhizae Radix(甘草), Paeoniae Radix Alba(白芍藥) support Soeumin's spleen root (脾元), and Zingiberis Rhizoma(乾薑), Myristicae Semen(肉豆蔲) warm up stomach, and Pinelliae Rhizoma(半夏), Citri Pericarpium(陳皮) circulate Qi, Crataegi Fructus(山楂), Amomi Fructus(砂仁), Cyperi Rhizoma(香附子) encourage digestion to maintain descending Yin.

The Crystal Structure of a $\beta$-Allyl Type Phenylpropanoid 2-(4-allyl-2, 6-dimethoxyphenoxy)-1-(4-hydroxy-3-methoxyphenyl) propan-1-ol, from the Seeds of Myristica fragrans

  • The structure of a $\beta$-allyl type phenylpropanoid was determined by single crystal X-ray diffraction analysis. The compound was recrystallized from a mixture of n-hexane and benzene in monoclinic crystal system with a = 24.782 (2), b = 10.537 (1), c = 7.871 (1) ${\AA}, \beta=95.74$ (1)$^\circ, $D_x$=1.216, $D_m$=1.22g/$cm^3$, space group $P2_1/a$, and Z=4. The structure was solved by direct method and refined by least-squares procedure to the final R value of 0.054 for 2824 observed reflections {$F{\geq}3\sigma(F)$}. The molecular geometry shows a most stable trans-form with respect to the bulky phenyls, and this conformation is settled by an intramolecular hydrogen bond. In the crystal, the molecules are arranged along with the screw axis, and stabilized by the $O{\cdot}H{\cdots}O$ type intermolecular hydrogen bonds. The other intermolecular contacts appear to be the normal van der Waals' interactions. The compound is a dimeric phenylpropanoid, and belongs to the neolignan analogues.

Myristicae Semen Extract Protects Excitotoxicity in Cultured Neuronal Cells

  • Myristica fragrans seed from Myristica fragrans Houtt (Myristicaceae) has various pharmacological activities peripherally and centrally. The present study aims to investigate the effect of the methanol extract of Myristica fragrans seed (MF) on kainic acid (KA)-induced neurotoxicity in primary cultured rat cerebellar granule neuron. MF, over a concentration range of 0.05 to $5\;{\mu}g/ml$ inhibited KA $(500\;{\mu}M)-induced$ neuronal cell death, which was measured by trypan blue exclusion test and 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. MF $(0.5\;{mu}g/ml)$ inhibited glutamate release into medium induced by KA $(500\;{\mu}M)$, which was measured by HPLC. Pretreatment of MF $(0.5\;{mu}g/ml)$ inhibited KA $(500\;{\mu}M)-induced$ elevation of cytosolic calcium concentration $([Ca^{2+}]_c)$, which was measured by a fluorescent dye, Fura 2-AM, and generation of reactive oxygen species (ROS). These results suggest that MF prevents KA-induced neuronal cell damage in vitro.

Antibacterial Activity of Caryophylli Flos on the Growth of Dental Caries Bacteria, Streptococcus mutans KDJ-50 (구강균 Streptococcus mutans KDJ-50에 대한 정향의 향균효과)

  • This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from various medicinal plants of which safety was indetified. Medicinal plants used in this study was dried and grinded after purchased at Daegu yakryung sijang and Kyungsan jungang sijang. Medicinal plants extracted with 80% ethanol at boiling point for 3 hrs was used as antibacterial agent after freeze dried. This study was conducted to find antibacterial agent against growth of dental caries bacteria. St. mutans KDJ-50 from 32 medicinal plants of which safety was indetified. The result of using paper disc method. Caryophylli Flos. Coptidis Rhizoma. Phellodendri Cortex. Schizamdrae Fructs. Myristicae Semen, Crataegi Fructus and Acori Graminei Rhizoma was selected as antibacterial agent. The result of viable cell counting method. the antibacterial activity of Caryophylli Flos was highest among tested 7 medicinal plants followed by Phellodendri Cortex and Coptidis Rhizoma. The extinction effect of the St. mutans by Caryophylli Flos was shown with the addition of 0.5 (w/v) in the medium. The high antibacterial activity was acquired at high extraction temperature and long extraction temperature. The antibacterial activity of Caryophylli Flos was not effected by the concentration of ethanol.

Attenuation of airway hyperreactivity and inflammation by Cheongsangbiyeum administration in a mouse model of asthma (마우스 천식모델에서 청상비음(淸上秘飮)의 기도 과민반응 및 염증의 억제 효과)

  • Objective : Although management of asthma has become increasingly effective, its cure remains elusive, necessitating a new modality to prevent or eliminate causes triggering clinical progress. Based in the clinical experiences, a novel decoration Cheongsangbiyeum (CSB), has been developed to treat asthma, which consists of Polyporus, Semen Myristicae, Pericarpium citri Reticulatae, Rhizoma Cimicifugae, Cortex Albizziae, Fructus Rubi, Rhizoma Zedoariae, and Rhizoma Rhei. In the current study, its anti-asthmatic efficacy was evaluated using a mouse model of asthma. Methods : Experimental allergic asthma was induced by repeated intraperitioneal sensitization and intranasal challenge of ovalbumin (OVA). Water extract of CSB (1 mg/mouse/day) was administrated orally whereas control mice on given with identical volume of phosphate-buffered saline (PBS) for 5 days during the course of antigen challenge. When airway hyperreactivity(AHR) measured by ${\bata}-methacoline-induced$ airflow obstruction was compared, AHR of CSB-treated mice was significantly lower than those of control mice, indicating that CM extract can attenuate an asthmatic symptom. Airway recruitment of leukocytes and eosinophils was also markedly reduced by CSB treatment suggesting that oral treatment of CSB can alleviate the airway inflammation. For a better understanding of possible mechanisms underlying anti-asthmatic effet of CSB, cytokine (IL-4, IL-5, IL-13 and $IFN{\gamma}$ levels in bronchoalveola lavage fluid (BALF) and lung tissues were determined. Results : The results showed that cytokine levels were significantly lowered by CSB treatment. Additionally, number of draining lymph node cells was significantly lower than those of control mice. These data indicate that CSB suppress in vivo allergen-specific response. However, notably, levels of type 2 cytokines such as IL-5 and IL-13 were more profoundly influenced. Moreover, in vitro OVA-specific proliferative response and type 2 cytokine (IL-4, IL-5 and IL-13) production lymph node cells was markedly decreased in CSB-treated mice, whereas their $IFN{\gamma}$ production was not significantly altered Thrse data clearly showed a preferential inhibition of type 2 T cell (Th2) response by CSB treatment. This finding was also supported by serum antibody data showing that levels of OVA-specific type 2 antibodies, IgE and IgG1, in CSB-treated mice were significantly lower than in control mice, while type 1 antibody, IgG2a level m rather higher than controls, although the difference was in significant. Conclusions : In conclusion, oral administration of CSB attenuates asthmatic manifestations including AHR ad airway recruitment of eosinophils in a mouse model which possibly results from selective inhibition of Th2 cell response to allergen. Our data suggest a potential clinical application of CSB for control of allergic asthma.

Inhibitory Effects of the Seed Extract of Myristicae Semen on the Proliferation of Human Tumor Cell Lines (II) (육두구 추출물의 암세포증식 저해 효과 (제 2보))

  • The methanol extract from seed of Myristica fragrans (Myristicaceae) demonstrated a potent inhibition on the pro-liferation of cultured human tumor cells such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT-15 (colon) in vitro. By the continuous effort to purify the active components responsible far the anti-proliferative effect on tumor cell lines, we have isolated eleven kinds of lignan components, i. e., safrole (1), machilin A (2), licarin B (3), macelignan (4), mere-dihydroguaiaretic acid (5), mγnstargenol A (6), methoxyeugenol (7), machilin F (8), licarin A (9), nectandrin B (10), and 2-(4-allyl-2,6-dimethoxyphenoxy)-1-(4-hydroxy-3-methoxyphenyl)propan-1-ol (11) together with a novel furan fatty acid, (E)-3-(3-methyl-5-pentylfuran-2-yl) acrylic acid (12) from seed extract of M. fragrans. Chemical structures of the isolated components (1-12) were established bγ the aid of NMR spectroscopic analyses, i. e., COSY HMQC and HMBC. Each of the Isolates demonstrated a potent inhibition on the proliferation of cultured human tumor cells such as A549 (non small cell lung), SK-OY-3 (ovary), SK-MEL-2 (melanoma) and HCT-15 (colon) in vitro.
