Effects of Elicitors on Seedling Growth, Total Polyphenol and Chlorophyll Content and Antioxidant Activity of Barley (Hordeum vulgare L.)

  • This study is focused on the evaluation of growth parameters, total polyphenol content (TPC), chlorophyll content as well as the DPPH (1,1-diphenyl-2-picryhydrazyl) free radical scavenging activity of young barley seedling (YBS) affected by elicitation. Salicylic acid (SA), methyl jasmonate (MJ), amino acid liquid fertilizer (ALF) and microbial metabolism activator (MMA) were used. Elicitation was conducted for two times and various concentrations were used in this study. The result revealed that, MJ 1 ml/L treated-YBS gave the longest seedling length of 1.33 cm, followed by the ones treated with SA 1.38 mg/L and ALF 2 ml/L, respectively. ALF 3 ml/L treatment gave the highest fresh weight of 10 seedlings, followed by MJ 5 ml/L and SA 13.8 mg/L treatment with 1.56 g, 1.55 g and 1.53 g respectively. SA 138.12 mg/L elicitor treated-YBS gave the highest Chl a, Chl b content of $8.57{\mu}g/mg$ and $3.83{\mu}g/mg$, respectively while the highest carotenoid content was found in MJ ml/L treatment with $1.62{\mu}g/mg$. Among elicitor treated-YBS, SA showed better TPC. The highest TPC was found in SA 1.38 mg/L treatment with 18.82 mg/g TAE. Likewise, SA 1.38 mg/L showed the highest DPPH free radical scavenging activity among all the treatments. However, the lowest TPC was found in ALF 1ml/L treated-YBS with 9.46 mg/g TAE, which was even lower than the control (14.31 mg/g TAE).

  • Lactococcus lactis SA72 from Jeot-gal (Korean traditional fermented fish foods) produces lacticin SA72. The influence of several parameters on the fermentative production of lacticin SA72 by Lactococcus lactis SA72 was studied. MRS medium among several media was selected for enhanced bacteriocin production. The mean growth rate and bacteriocin productivity of L. lactis SA72 increased as the initial pH of the media increases. The highest lacticin SA72 activity was detected 3,200 AU/ml at pH 6.0, $32^{\circ}C$, and 1% (inoculum size, v/v) in the jar fermenter. Enhanced production of lacticin SA72 was investigated by a fed-batch cultivation with the intermittent feeding of the concentrated glucose solution. Under the optimized conditions, lacticin SA72 activity finally reached to 6,400 AU/ml.

A Study on the Preparation of High Purity Silicic Acid by Solvent Extraction from Sodium Silicate (규산 나트륨으로부터 유기 용매 추출에 의한 고순도 규산 제조에 관한 연구)

  • Silicic acid(SA) was extracted with organic solvents from aqueous sodium silicate solutions acidified with dilute sulfuric acid. Tetrahydrofuran(THF), isopropyl alcohol and acetone were used as organic solvents. The extraction degree of Si and the removal efficiency of Na is determined according to kinds of solvent and the mixing ratios of solvent, sodium chloride and $H_2O$. Optimum conditions for the preparation of high purity SA were THF as an organic solvent, volume ratio of organic solvent/SA : 1 (organic solvent 10ml/10ml SA) and the amount of NaCl addition 2.5g/10ml SA. The extraction degree of Si and the removal efficiency of Na at that point were 86.2% and 99.95% respectively and the purity of SA was 99.96%.

The Effect of Yeast(Saccharomyces exiguus SJPAF1) on Odor Emission and Contaminants Reduction in Piggery Slurry (효모(Saccharomyces exiguus SJPAF1) 첨가에 따른 돈분뇨의 악취제거 및 오염물질 감소 효과)

  • The aim of this study was to evaluate the effect of yeast(Saccharomyces exiguus SJPAF1, referred to as SA) addition on odor emission and contaminants reduction in piggery sluny. Four different rates of yeast addition were compared: no addition(SA0), 0.7L(SA0.7), 1.0L(SA1.0), and 1.5L(SA1.5) to one tone of piggery slurry. Odor emission tended to decrease with increasing the yeast application with concurrent effects of changes in temperature on outside of reactors. Particularly, reduction in ammonia emission was proportional to the yeast application rate; it reduced from 161.1 ppm in SA0 to 47.1 ppm in SA1.5 after 6 days of treatment Decomposition of piggery shiny by yeast increased to 13.8% more in SA1.5, and total amounts of piggery slurry decreased to 12.5% in SA1.5. Total coliforms were detected below 30MPN $ml^{-1}$ in SA1.5, while $8.3{\times}10^3$ MPN $ml^{-1}$ of Total coliforms were found in SA0. However, the effect of yeast addition in piggery slurry seemed to have no influence on the removal efficiency of contaminants such as BOD, COD, $NO_3^{-}-N$, $NH_4^{+}-N$, $PO_4^{-}P$. Consequently, the yeast(Saccharomyces exiguus SJPAF1) addition of 1.5% in the piggery sluny seems to have potential applicability for improving agent of pig-farm environment.

Transmission of Multiple Drag-Resistance in Hospital Pseudomonas aeruginosa by Intraspecies Conjugation System (병원녹농균(病院綠膿菌)의 동종균접합(同種菌接合)에 의한 다제내성(多劑耐性)의 전달(傳達))

  • Two hundred and ninety-five strains of Peudomonas aeruginosa isolated from clinical sources were tested for drug resistance and demonstration of R plasmids by intraspecies conjugation system. Sixty strains were found highly resistant to two or more of drugs. The rate of resistant strains were 38.9% to kanamycin(km), 33.2% to streptomydn(sm), 22.7% to sulfisomidine(Sa), 14.2% to chloramphenicol(Cp), 13.8% to tetracycline(Tc), 3.0% to carbenicillin(Cb), and to gentamicin(Gm), respectively. But no strains was resistant to nalidixic acid and colistine. They were resistant to per milliliter to more than $400{\mu}g$ per ml. of Tc, $800{\mu}g$ per ml of Cp and of Sm, $6,400{\mu}g$ per ml. of Sa, $200{\mu}g$ per ml. of Cb, $100{\mu}g$ per ml. of Gm, and $25{\mu}g$ per ml. of colistine. Forty-three strains of Pseudomonas aeruginosa could be transferred their resistance to Pseudomonas aeruginosa 2-70, 1005 rifampin resistant FP-auxotrophic mutant. Of sixty multiple resistant strains, forty-three(71.6%) demonstrated R plasmids; nineteen carried resistance to(Tc Cp Sm Sa), six to(Tc Cp Sm), three to(Tc Cp Sa), and Cp, five to(Tc Sm Sa), two to(Tc Sa), (Cp Sm) and Tc, and one to(Cp Sm Sa). Degree of resistance of recipients recieving R plasmids from donors were almost the same level of resistance as the donor in regardless of mating temperature at $25^{\circ}C$ and $37^{\circ}C$. Resistance to Tc, Sm, and Sa were transferred to a very few of recipient cells at five minutes after mating with donor and recipient cells but resistance to Cp were transferred to the majority of recipient cells. The transfer frequency of Tc, Cp, Sm, and Sa resistance from donors to recipients were from $1.0^{-1.4}\;to\;1.0^{-3.5}$ at $25^{\circ}C$ for 18 hours of incubation and were from $1.0^{-1.5}\;to\;1.0^{-3.5}$ at $37^{\circ}C$ for 18 hours of incubation.

Dose Related Effects of Poloxamer/Sodium Alginate Mixture in Prevention of Postoperative Adhesion Formation in Dogs (개에서 Poloxamer/Sodium Alginate 혼합물의 용량에 따른 복강 유착방지 효과)

  • This study was performed to determine the minimum dose of Poloxamer/Sodium alginate (PX/SA) mixture on preventing intraperitoneal adhesions to evaluate organ toxicity. Twenty five healthy adult mongrel dogs (weighing 4.68${\pm}$1.67 kg) were divided into five experimental groups composed of five dogs respectively; negative control group (NC, non-treated), positive control group (PC, 2% carboxymethyl chitosan solution treated), and experiment 1 group (E1, 0.25 ml PX/SA mixture of abraded area), experiment 2 group (E2, 0.5 ml PX/SA mixture of abraded area), experiment 3 group (E3, 1.0 ml PX/SA mixture of abraded area). Venous blood specimens were collected from all experimental animals for hematologic and biochemical analysis: WBC, fibrinogen, AST, ALT, ALP, BUN and creatinine. The anti-adhesion effect was evaluated using a serosa abrasion model. The denuded ileum was coated with PX/SA mixture, carboxymethyl chitosan solution or neither. The tensile strength of the adhesion site was evaluated with a tensiometer. For histopathological examination, tissue samples of the liver and kidney were collected from all dogs. According to the results, the frequency and tensile strength values for adhesion separation in PX/SA group were significantly lower than those in negative control group (p < 0.05). In E2 group, the tensile strength was significantly decreased in consideration of PX/SA dose. The values of AST, ALT, ALP, BUN and creatinine of the control and the experimental groups showed no statistical differences. No obvious microscopic differences were noted among tissue sections obtained from all groups. The results suggest that PX/SA mixture may be effective on reducing peritoneal adhesion formation in dog and that 0.5 ml PX/SA mixture of abraded area is most effective dose. Moreover, PX/SA mixture was considered not to have toxicity for the liver and the kidney.

Cytotoxic Effect of Taxol on Malignant Bone Tumor Cell Lines (악성 골종양 세포주들에 대한 Taxol의 세포독성)

  • Taxol, the extract from the Taxus brevifolia which is a Pacific yew tree has aroused the interest of the tumor investigators since the 1960s. As well, it is shown to have broad antitumor activity in preclinical experimental models. Its action mechanism is an anti-microtubule effect by duplication of tubulin. The most impressive antitumor activity of taxol has been observed in advanced ovarian cancer and metastatic breast cancer. The purpose of this study was to determine how taxol acts on malignant bone tumor cell lines, to compare its cytotoxic effect with those of other chemotherapeutic agents, and to ascertain the its combination effect with adriamycin. Cell lines used in this study were G-292(osteosarcoma, human), SaOS-2(osteosarcoma, primary, human), and HT-1080(fibrosarcoma, human). Methotrexate, adriamycin, cisplatinum, ifosfamide and taxol were used as testing chemotherapeutic agents and their maximum test concentration were $500{\mu}g/ml$, $200{\mu}g/ml$, $500{\mu}g/ml$, $1000{\mu}g/ml$, and $600{\mu}g/ml$, respectively. The media for cell culture was RPMI-1640 with 10% fetal bovine serum and gentamycin. The results were as follows. The $IC_{50}$ of methotrexate, ifosfamide, cisplatinum, adriamycin and Taxol in G-292 were $2.3{\times}10^{-1}{\mu}g/ml$, $8.0{\times}10^0{\mu}g/ml$, $3.5{\times}10^0{\mu}g/ml$, $9.8{\times}10^{-1}{\mu}g/ml$, $2.7{\times}10^{-2}{\mu}g/ml$ respectively, in SaOS-2 $3.5{\times}10^{-1}{\mu}g/ml$, $1.5{\times}10^1{\mu}g/ml$, $2.8{\times}10^0{\mu}g/ml$, $9.9{\times}10^{-2}{\mu}g/ml$, $1.0{\times}10^{-2}{\mu}g/ml$, respectively, in HT-1080 $4.2{\times}10^{-2}{\mu}g/ml$, $5.4{\times}10^1{\mu}g/ml$, $3.8{\times}10^0{\mu}g/ml$, $5.5{\times}10^{-3}{\mu}g/ml$, $1.1{\times}10^{-3}{\mu}g/ml$, respectively. In conclusion, taxol had very potent cytotoxic effect on the malignant bone tumor cell lines with adriamycin, and was more potent than methotrexate, cisplatinum and ifosfamide. There were synergistic antitumor effects on G-292 and SaOS-2 cell lines in combination test of taxol and adriamycin. From the above results, it would be estimated that taxol could be a new antitumor drug for the malignant bone tumors, providing measures against the side effects and followed by the clinical tests.

Channel Function of TRPML1 Prompts Lipolysis in Mature Adipocytes

  • Increased intracellular levels of $Ca^{2+}$ are generally thought to negatively regulate lipolysis in mature adipocytes, whereas store-operated $Ca^{2+}$ entry was recently reported to facilitate lipolysis and attenuate lipotoxicity by inducing lipophagy. Transient receptor potential mucolipin1 (TRPML1), a $Ca^{2+}$-permeable non-selective cation channel, is mainly expressed on the lysosomal membrane and plays key roles in lysosomal homeostasis and membrane trafficking. However, the roles of TRPML1 in lipolysis remains unclear. In this study, we examined whether the channel function of TRPML1 induces lipolysis in mature adipocytes. We found that treatment of mature adipocytes with ML-SA1, a specific agonist of TRPML1, solely upregulated extracellular glycerol release, but not to the same extent as isoproterenol. In addition, knockdown of TRPML1 in mature adipocytes significantly reduced autophagic flux, regardless of ML-SA1 treatment. Our findings demonstrate that the channel function of TRPML1 partially contributes to lipid metabolism and autophagic membrane trafficking, suggesting that TRPML1, particularly the channel function of TRPML1, is as therapeutic target molecule for treating obesity.

Effect of Hexane Extract of Acori graminei Rhizoma on Chloramphenicol Acetyltransferase of Staphylococcus aureus SA2 (석창포 헥산 추출물이 Staphylococcus aureus SA2의 Chloramphenicol Acetyltransferase 에 미치는 영향)

  • One subfraction from the hexane fraction of Acri graminei Rhizoma, the E4 fraction which is mainly consisted of acorenone, showed a potential inhibitory activity against chloramphenicol acetyltransferase (CAT) of S. aureus SA2 that is a multidrug-resistant strain to 10 usual antibiotics. The combination therapy of this fraction with chloramphenicol resulted in reduction of the minimal inhibitory concentration from 128 $\mu\textrm{g}$/ml to 8 $\mu\textrm{g}$/ml. The E4 fraction also revealed to prevent the induction of CAT from this strain.

Effects of Essential oils of Several Aromatic Plants on the Growth of Antibiotic Resistant Staphylococcus aureus SA2 (몇몇 식물 정유성분이 항생제내성균주 Staphylococcus aureus SA2의 성장에 미치는 영향)

  • The essential oil fractions from six plant parts including leaf of Zanthoxylum piperitum and flower of Lindera obtusiloba have revealed to possess resistance inhibitory activity on antibiotic resistant Staphylococcus aureus SA2 when combined with ohloramphenicol (Cm). The combination of Cm and essential oil mixtures showed potent resistance inhibition in the level of 10∼20 $\mu\textrm{g}$/ml.
