Deep Learning-based system for plant disease detection and classification (딥러닝 기반 작물 질병 탐지 및 분류 시스템)

  • Plant diseases and pests affect the growth of various plants, so it is very important to identify pests at an early stage. Although many machine learning (ML) models have already been used for the inspection and classification of plant pests, advances in deep learning (DL), a subset of machine learning, have led to many advances in this field of research. In this study, disease and pest inspection of abnormal crops and maturity classification were performed for normal crops using YOLOX detector and MobileNet classifier. Through this method, various plant pest features can be effectively extracted. For the experiment, image datasets of various resolutions related to strawberries, peppers, and tomatoes were prepared and used for plant pest classification. According to the experimental results, it was confirmed that the average test accuracy was 84% and the maturity classification accuracy was 83.91% in images with complex background conditions. This model was able to effectively detect 6 diseases of 3 plants and classify the maturity of each plant in natural conditions.

  • With the development of networks and the increase in the number of network devices, the number of cyber attacks targeting them is also increasing. Since these cyber-attacks aim to steal important information and destroy systems, it is necessary to minimize social and economic damage through early detection and rapid response. Many studies using machine learning (ML) and artificial intelligence (AI) have been conducted, among which payload learning is one of the most intuitive and effective methods to detect malicious behavior. In this study, we propose a preprocessing method to maximize the performance of the model when learning the payload in term units. The proposed method constructs a high-quality learning data set by eliminating unnecessary noise (stopwords) and preserving important features in consideration of the machine language and natural language characteristics of the packet payload. Our method consists of three steps: Preserving significant special characters, Generating a stopword list, and Class label refinement. By processing packets of various and complex structures based on these three processes, it is possible to make high-quality training data that can be helpful to build high-performance ML/AI models for security monitoring. We prove the effectiveness of the proposed method by comparing the performance of the AI model to which the proposed method is applied and not. Forthermore, by evaluating the performance of the AI model applied proposed method in the real-world Security Operating Center (SOC) environment with live network traffic, we demonstrate the applicability of the our method to the real environment.

Detection of a Thermal Stable-Soluble Protein (TSSP) as a Marker of Peanut Adulteration Using a Highly Sensitive Indirect Enzyme-Linked Immunosorbent Assay based on Monoclonal Antibodies

  • Food allergy represents a severe problem for many societies, including sensitive populations, academies, health authorities, and the food industry. Peanut allergy occupies a special place in the food allergy spectrum. To prevent consumption by consumers suffering from a peanut allergy, a rapid and sensitive detection method is essential to identify unintended peanut adulteration in processed foods. In this study, we produced four monoclonal antibodies (MAbs; RO 3A1-12, PB 4C12-10, PB 5F9-23, and PB 6G4-30) specific to thermo-stable and soluble proteins (TSSPs) of peanut and developed an enzyme-linked immunosorbent assay (ELISA) based on the MAbs. Among them, PB 5F9-23 MAb was firmly bound to Ara h 1, and other MAbs strongly reacted to Ara h 3 in the Western blot analysis. An antibody cocktail solution of the MAbs was used to enhance the sensitivity of an indirect ELISA, and the limit of detection of the indirect ELISA based on the antibody cocktail solution was 1 ng/ml and improved compared to the indirect ELISA based on the single MAb (11 ng/ml). The cross-reaction analysis revealed the high specificity of developed MAbs to peanut TSSPs without cross-reaction to other food allergens, including nuts. Subsequently, analyzing processed foods by indirect ELISA, all foods labeled as containing peanuts in the product description were confirmed to be positive. The results indicate that the developed antibodies exhibit high specificity and sensitivity to peanuts and can be used as bio-receptors in immunoassays or biosensors to detect intentional or unintentional adulteration of peanuts in processed foods, particularly heat-processed foods.

  • Background : Early diagnosis and proper antibiotic treatment are very important in the management of ventilator-associated pneumonia (VAP) because of its high mortality. Bronchoscopy with a protected specimen brush (PSB) has been considered the standard method to isolate the causative organisms of VAP. However, this method burdens consumer economically to purchase a PSB. Another useful method for the diagnosis of VAP is quantitative cultures of aspirated specimens through bronchoscopic bronchoalveolar lavage (BAL), for which the infusion of more than 120 m1 of saline has been recommended for adequate sampling of a pulmonary segment. However, occasionally it leads to deterioration of the patient's condition. We studied the diagnostic efficacy of minibronchoalveolar lavage (miniBAL), which retrieves only 25 ml of BAL fluid, in the isolation of causative organisms of VAP. Methods: We included 38 consecutive patients (41 cases) suspected of having VAP on the basis of clinical evidence, who had received antibiotics before the bronchoscopy. The two diagnostic techniques of PSB and miniBAL, which were performed one after another at the same pulmonary segment, 'were compared prospectively. The cut-off values for quantitative cultures to define causative bacteria of VAP were more than $10^3$ colony-forming units (cfu)/ml for PSB and more than $10^4$ cfu/ml for BAL. Results: The amount of instilled normal saline required to retrieve 25 ml of BAL fluid was $93{\pm}32 ml$ (mean${\pm}$SD). The detection rate of causative agents was 46.3% (19/41) with PSB and 43.9% (18/41) with miniBAL. The concordance rate of PSB and miniBAL in the bacterial culture was 85.4% (35/41). Although arterial blood oxygen saturation dropped significantly (p<0.05) during ($92{\pm}10%$) and 10 min after ($95{\pm}3%$) miniBAL compared with the baseline ($97{\pm}3%$), all except 3 cases were within normal ranges. The significantly elevated heart rate during ($l25{\pm}24$/min, p<0.05) miniBAL compared with the baseline ($1l1{\pm}22$/min) recovered again in 10 min after ($111{\pm}26$/min) miniBAL. Transient hypotension was developed during the procedure in two cases. The procedure was stopped in one case due to atrial flutter. Conclusion: MiniBAL is a safe and effective technique to detect the causative organisms of VAP.

Analysis of Good Agricultural Practices (GAP) in Panax ginseng C.A. Mayer (인삼의 GAP (우수농산물인증) 관련요소 분석)

  • For the analysis of hazard factors and the development of GAP (Good Agricultural Practices) Ginseng, 10 ginseng cultural farms wished certified GAP were selected at Geumsan-gun area, a representative site of ginseng cultivation in Korea. In order to verify the safety of GAP ginseng, possible contamination of pesticide and heavy metal residues, and microbial hazard were analyzed. Soil and water around ginseng cultivation field, and ginseng were investigated. Eighty-one pesticides including carbendazim were used as typical pesticide against plant pathogens and insect pests of ginseng plant and general crops. There was no excess the maximum residue limit (MRL) in residue figure of the soil. Including the residue figure of the arsenic (0.81 ml/kg) and 7 other heavy metals was also suitable to cultivate the ginseng plant. The irrigation water and dilution water for pesticide application were also safety level for GAP. Fresh ginsengs from the farms were sampled and investigated pesticide residues and contaminations of bacteria. Among 23 pesticides tested, we didn't detect any kinds of pesticide residues, but tolclofos-methyl was frequently found in the other ginseng field. On the investigation of microorganism hazards, 2 gram negative bacteria and 1 gram positive bacterium were found in the fresh ginseng. Number of total bacteria was $1.5{\times}10^3$ cfu/ml, which was less than the other agriculture products. At these results, 10 selected ginseng farms were good cultural places for GAP ginseng production and the ginseng cultured from Geumsan-gun area were a good safe far human.

The Neurological Safety of Epidural Pamidronate in Rats

  • Background: Pamidronate is a potent inhibitor of osteoclast-mediated bone resorption. Recently, the drug has been known to relieve bone pain. We hypothesized that direct epidural administration of pamidronate could have various advantages over oral administration with respect to dosage, side effects, and efficacy. Therefore, we evaluated the neuronal safety of epidurally-administered pamidronate. Methods: Twenty-seven rats weighing 250-350 g were equally divided into 3 groups. Each group received an epidural administration with either 0.3 ml (3.75 mg) of pamidronate (group P), 0.3 ml of 40% alcohol (group A), or 0.3 ml of normal saline (group N). A Pinch-toe test, motor function evaluation, and histopathologic examination of the spinal cord to detect conditions such as chromatolysis, meningeal inflammation, and neuritis, were performed on the 2nd, 7th, and 21st day following administration of each drug. Results: All rats in group A showed an abnormal response to the pinch-toe test and decreased motor function during the entire evaluation period. Abnormal histopathologic findings, including neuritis and meningeal inflammation were observed only in group A rats. Rats in group P, with the exception of 1, and group N showed no significant sensory/motor dysfunction over a 3-week observation period. No histopathologic changes were observed in groups P and N. Conclusions: Direct epidural injection of pamidronate (about 12.5 mg/kg) showed no neurotoxic evidence in terms of sensory/motor function evaluation and histopathologic examination.

Development of Simultaneous Analysis for the Multi-residual Pesticides in the Ginseng Extract using Gas Chromatography (인삼농축액에서 GC를 이용한 잔류농약 동시다성분 분석법의 개발)

  • The simultaneous analysis of multi-residual pesticides was developed using a gas chromatography (GC) method. In this study, a simple and reliable methodology was improved to detect 154 kinds of pesticides in sinseng extract sample by using a liquid-liquid extraction procedure, open column chromagraphy and chromatographic analysis by CC electron capture detector (ECD) and GC nitrogen-phosphorus detector (NPD). The 154 kinds of pesticides were classified in 4 groups according to the chemical structure. The extraction of pesticides was experimented with $70\%$ acetone and dichloromethane/petroleum ether in order, and cleaned up via open column chromatography $(3\times30cm)$ packed with florisil $(30g,\;130^{\circ}C,\;12hrs)$. The final extract was concentrated in a rotator evaporator at $40^{\circ}C$ until dryness. Then the residue was redissolved to 2ml with acetone, and analyzed by GC-ECD and GC-NPD. The applied concentration of pesticides was over $1\~10{\mu}g/ml$. The recovery tests were ranged from $70.7\%$ to $115.2\%$ with standard deviations between 0.3 and $5.7\%$ of the standard spiked to the ginseng extract sample (Group $I\~IV$). The limit of detection (LOD) ranged from 0.001 to $0.099{\mu}g/ml$ (Group $I\~IV$). The 9 kinds of pesticides were not detected. The developed method was applied satisfactory to the determination of the 154 kinds of pesticides in the ginseng extract with good reproducibility and accuracy.

Isolation of Bovine Spermatozoal Components by Physical or Chemical Treatments (물리.화학적 처리에 의한 소 정자세포구성분의 분리)

  • An understanding of the structure and function of mammalian spermatozoa requires the iso-lation of these components. In this study, frozen-thawed bovine spermatozoa were treated by physical treatments (vortexing, 26 gauge needle, strained 26 gauge needles and freezing-thawing) or chemical treatments (trypsin, dithiothreitol, sodium dodecylsulfate and $\beta$-mercaptoethanoJ) to yield free heads and tails. The most effective treatment was repeated pumping of sperm suspension through a strained 26 gauge needle conneted to a syringe. Spermatozoa by this treatment were mainly broken at the junction of the head and the tail, resulting in 90-100% yields. Also, sperm head surface did not modify during strained 26 gauge needle treatment when either spermatozoa or sperm heads were incubated in 250${\mu}\textrm{g}$/ml of FITC-UEA 1 for 1 h at room temperature to detect the modification of sperm surface components. Other physical treatments were less efficient for the breakdown of spermatozoa. The effects of chemical treatments on bovine spermatozoa are not noticeable. Dissected sperm heads and tails should be fractional leading to nearly pure components by sucrose gradient centrifugation at 1,000 rpm for 15 min. The result suggest that the established method may be useful for the biochemical study of spermatozoal components, and the understanding of oocyte activation mechanism either by spermatozoal components during fertilization or microinjection of isolated components.

Effect of Antibiotic Combination Therapy on Metallo-${\beta}$-Lactamase Producing Imipenem Resistant Pseudomonas aeruginosa (Metallo-${\beta}$-lactamase를 생성하여 Imipenem에 내성인 Pseudomonas aeruginosa에 대한 항균제 병합요법의 효과)

  • This study was to detect MBL (metallo-${\beta}$-lactamase) among glucose non-fermenting Gram-negative bacilli isolated from clinical specimen and to search antimicrobial combination therapy against MBL producing Pseudomonas aeruginosa. Among fifty one isolates of Gram-negative bacilli with reduced imipenem susceptibility ($MIC{\ge}8{\mu}g/ml$), nine isolates have shown positive results in MBL detection test. They were seven Achromobacter xylosoxidans subsp. xylosoxidans and two P. aeruginosa. The results from EDTA-DDST coin-cided with those of PCR and nucleotide sequence analysis which showed the presence of $bla_{VIM-2}$. The combination of aztreonam (AZT) and piperacillin-tazobactarn (TZP) or AZT and amikacin (AN) screened by one disk synergy test showed no synergistic effect. Triple antibiotic combination therapy with AZT, TZP and AN, however, was shown to be effective and the most synergistic after 8 hrs of exposure. This result strongly suggest that the triple combination therapy of AZT, TZP, and AN could be useful for the treatment of infection caused by MBL producing Gram-negative bacilli.

Spatio-Temporal Variations of Harmful Algal Blooms in the South Sea of Korea

  • Harmful algal blooms (HAB) caused by the dominant species Cochlodinium polykrikoides (C. polykrikoides) appear in the South Sea of Korea and are particularly present in summer and fall seasons. Environmental factors such as water temperature, weather conditions (air temperature, cloud cover, sunshine, precipitation and wind) influence on the initiation and subsequent development of HAB. The purpose of this research was to study spatial and temporal variations of HAB in the Yeosu area using environmental (oceanic and meteorological) and satellite data. Chlorophyll-a concentrations were calculated using Sea-viewing Wide Field-of-view Sensor (SeaWiFS) images by an Ocean Chlorophyll 4 (OC4) algorithm, and HAB were estimated using the Red tide index Chlorophyll Algorithm (RCA). We also used the surface velocity of sequential satellite images applying the Maximum Cross Correlation method to detect chlorophyll-a movement. The results showed that the water temperature during HAB occurrences in August 2002-2008 was $19.4-30.2^{\circ}C$. In terms of the frequency of the mean of cell density of C. polykrikoides, the cell density of the HAB found at low (<300 cells/ml), medium (300-1000 cells/ml), and high (>1000 cells/ml) levels were 27.01%, 37.44%, and 35.55%, respectively. Meteorological data for 2002-2008 showed that the mean air temperature, precipitation, wind speed and direction, and sunshine duration were $22.39^{\circ}C$, 6.54 mm/day, 3.98 m/s (southwesterly), and 1-11.7 h, respectively. Our results suggest that HAB events in the Yeosu area can be triggered and extended by heavy precipitation and massive movement of HAB from the East China Sea. Satellite images data from July to October 2002-2006 showed that the OC4 algorithm generally estimated high chlorophyll-a concentration ($2-20\;mg/m^3$) throughout the coastal area, whereas the RCA estimated concentrations at $2-10\;mg/m^3$. The surface velocity of chlorophyll-a movement from sequential satellite images revealed the same patterns in the direction of the Tsushima Warm Current.
