Hydrogel Dressing with a Nano-Formula against Methicillin-Resistant Staphylococcus aureus and Pseudomonas aeruginosa Diabetic Foot Bacteria

  • This study proposes an alternative approach for the use of chitosan silver-based dressing for the control of foot infection with multidrug-resistant bacteria. Sixty-five bacterial isolates were isolated from 40 diabetic patients. Staphylococcus aureus (37%) and Pseudomonas aeruginosa (18.5%) were the predominant isolates in the ulcer samples. Ten antibiotics were in vitro tested against diabetic foot clinical bacterial isolates. The most resistant S. aureus and P. aeruginosa isolates were then selected for further study. Three chitosan sources were tested individually for chelating silver nanoparticles. Squilla chitosan silver nanoparticles (Sq. Cs-Ag0) showed the maximum activity against the resistant bacteria when mixed with amikacin that showed the maximum synergetic index. This, in turn, resulted in the reduction of the amikacin MIC value by 95%. For evaluation of the effectiveness of the prepared dressing using Artemia salina as the toxicity biomarker, the LC50 was found to be 549.5, 18,000, and 10,000 μg/ml for amikacin, Sq. Cs-Ag0, and dressing matrix, respectively. Loading the formula onto chitosan hydrogel dressing showed promising antibacterial activities, with responsive healing properties for the wounds in normal rats of those diabetic rats (polymicrobial infection). It is quite interesting to note that no emergence of any side effect on either kidney or liver biomedical functions was noticed.

  • Whole oil and ketonic fraction (KF) of Leptospermum scoparium have been tested for their antimicrobial activity and combination effect with several antibiotics against various bacterial strains and fungi by using microbiological assay methods. Antibacterial activities of KF against a number of test strains were 2-3 fold stronger than those of whole oil. MICs of the KF were $65~125{\;}{\mu\textrm{g}}/ml$ against seven gram positive bacterial strains, $65~250{\;}{\mu\textrm{g}}/ml$ against 19 methicillin resistance Staphylococcus aureus strains, and $65~50{\;}{\mu\textrm{g}}/ml$ against 14 quinolone resistance strains. However, KF showed little or no activity against gram negative bacteria. MICs of the KF were $16~250{\;}{\mu\textrm{g}}/ml$ against more than 50% of the anaerobic bacterial strains tested. KF showed the higher antibacterial activity than bacitracin against 10 strains of Bacteroids thetaiotaomicron, or three strains of Bacteroides ovatus, and the more active than ciprofloxacin against one strain of Bacteroides thetaiotaomicron and three strains of Bacteroids ovatus. The MICs of KF was 63 and $250{\;}{\mu\textrm{g}}/ml$ against Aspergillus niger and Candida albicans, respectively. Antibacterial activities of KF in combination with 19 antibiotics against 14 strains and with four antifungal agents against one fungal strain were determined by paper strip diffusion method. While most of combination showed additivity, KF showed synergism with bacitracin, exfadroxil, cephradin, and meropenem for 29~57% of the strains tested. However, ofloxacin, enoxacin, sparfloxacin showed antagonism with KF for 43~71% of the strains. KF alone and in combination with bacitracin, gentamycin, neomycin, itraconazole, fluconazole, terfinafine and ketoconazole against five bacterial strains or one fungus strain synergistic effect was demonstrated against 33% of strains examined with FIC index value below 0.5 by checkerboard study. Synergistic effect of KF with gentamicin against Staphylococcus epidermidis 329 (QRS) was found by time-kill study.

Antimicrobial and Antiradical Activity of Nepalese Medicinal Plants

  • In this study fourteen ethanol extracts from Nepalese medicinal plants were screened for their in vitro antimicrobial and antiradical activity and their total phenolic content was evaluated. The antiradicalactivity was evaluated by free radical scavenging assay, using 2,2-diphenyl-1-picryl hydrazyl radical (DPPH). Plant extracts showed a wide range of radical scavenging activity, with $IC_50$ value ranging in between $5.38\; {\mu}g/\;mL$ - $429.61\;{\mu}g/mL$. Strong radical scavenging activity was shown by flower extract of Woodfordia fruticosa ($5.38\;{\mu}g/\;mL$) and stem bark extract of Azadirachta indica ($5.58 {\mu}g/\;mL$)that also contained high phenolic content. Most of the sample showed activity below the concentration of $100\; {\mu}g/mL$. For antimicrobial activity three test microorganisms namely Staphylococcus aureus, Streptococcus epidermidis, and Candida albicans were used. The minimum inhibitory concentration (MIC) of the plant extracts was determined. Most of the plant extracts were effective against bacterial strains only at higher concentration ($800\;-\;1,600\;{\mu}g/mL$) but none of these were effective against Candida albicans below $1,600\;{\mu}g/mL$.

Phytochemical Screening and Biological Studies of Boerhavia Diffusa Linn

  • Hexane, ethyl acetate and methanol extracts of whole plant of Boerhavia diffusa were screened for phytochemical and biological activities. Qualitative phytochemical screening via colorimetric method and the quantitative estimation of phenolic and flavonoid content were performed. Antioxidant assay using DPPH scavenging method was studied. Antimicrobial screening of plant extracts was done by cup diffusion technique. Cytotoxic activity of B. diffusa was studied by brine shrimp bioassay and anthelminthic activity was evaluated in vitro in Pheretima posthuma. This study revealed B. diffusa as a source of various phyto-constituents such as alkaloids, glycosides, saponins, tannins, carbohydrates, cardiac glycosides, flavonoids and terpenoids. Quantitative estimation of total phenol was found to be maximum in BEE i.e. $29.73{\pm}0.88$, BME $19.8{\pm}2.02$ and in BHE $9.15{\pm}0.304mgGAE/g$. Similarly, the total flavonoid content was found to be $17.44{\pm}0.75$ in BEE, $14.43{\pm}0.23$ in BHE and 3.678 mg QE/g in BME. Ethyl acetate extract showed its antibacterial activity against all tested pathogens except Escherichia coli whereas Staphylococcus aureus and Salmonella Typhi were resistant to methanol and hexane extract. The zone of inhibition (ZOI) of ethyl acetate extract against S. Typhi and B. cereus was found to be 18 mm and 14 mm respectively. The MIC value of BEE in S. Typhi was $3.125{\mu}g/ml$ and in B. cereus was $12.5{\mu}g/ml$. The preliminary screening of anticancer property of B. diffusa i.e. BSLT in methanol was found to be $165.19{\mu}g/ml$. B. diffusa was also found to contain anthelmintic property. The study helped in further exploration of medicinal properties of B. diffusa by phytochemical screening and biological activities paving the path for study and investigation in this plant.

Antibacterial effects of two cecropin type peptides isolated from the silkworm against Salmonella species

  • In insect defense system, antimicrobial peptides (AMPs) are one of important biological molecules to survive in a variety of environments. Insect can synthesize AMPs to protect against invading pathogens in humoral immune response. Taking more advantage of biological antimicrobial molecules, we report antibacterial activity of two cecropin type peptides, cecropin and moricin, isolated from the silkworm against four salmonella species. In this work, we purified antimicrobial candidate peptides (AMCP) from the extracts of immune challenged silkworm larval hemolymph by two-step chromatographic purification procedure, cation exchange and gel permeation chromatography. The molecular weights of purified peptides were estimated to be about 4 ~ 5 kDa by Tricin SDS-PAGE analysis, and identified as silkworm cecropin and moricin by NCBI BLAST homology search with their N-terminal amino acid sequences. As antibacterial activity assay, the purified peptides showed stronger antibacterial activity against Salmonella pathogens with an MIC value of $1{\sim}4{\mu}g/mL$. Therefore two cecropin type peptides purified from the silkworm will be valuable potential materials for development of new natural antibiotics.

HPLC, NMR Based Characterization, Antioxidant and Anticancer Activities of Chemical Constituents from Therapeutically Active Fungal Endophytes

  • Fungi generate different metabolites some of which are intrinsically bioactive and could therefore serve as templates for drug development. In the current study, six endophytic fungi namely Aspergillus flavus, Aspergillus tubigenesis, Aspergillus oryzae, Penicillium oxalicum, Aspergillus niger, and Aspergillus brasiliensis were isolated and identified from the medicinal plant, Silybum marianum. These endophytic fungi were identified through intra transcribed sequence (ITS) gene sequencing. The bioactive potentials of fungal extracts were investigated using several bioassays such as antibacterial activity by well-diffusion, MIC, MBC, anti-biofilm, antioxidant, and haemolysis. The Pseudomonas aeruginosa PAO1 was used to determine the antibiofilm activity. The ethyl acetate extract of Aspergillus flavus showed strong to moderate efficacy against Staphylococcus aureus, Escherichia coli, P. aeruginosa, and Bacillus spizizenii. Aspergillus flavus and Aspergillus brasiliensis exhibited significant antibiofilm activity with IC50 at 4.02 and 3.63 mg/ml, while A. flavus exhibited maximum antioxidant activity of 50.8%. Based on HPLC, LC-MS, and NMR experiments kojic acid (1) and carbamic acid (methylene-4, 1-phenylene) bis-dimethyl ester (2) were identified from A. flavus. Kojic acid exhibited DPPH free radical scavenging activity with an IC50 value of 99.3 ㎍/ml and moderate activity against ovarian teratocarcinoma (CH1), colon carcinoma (SW480), and non-small cell lung cancer (A549) cell lines. These findings suggest that endophytic fungi are able to produce promising bioactive compounds which deserve further investigation.

Fomitopsis pinicola 균사체배양액의 Helicobacter pylori에 대한 저해물질의 분리 및 특성 (Isolation and Characterization of Inhibition Helicobacter pylori from Culture Media of Fomitopsis pinicola)

  • Helicobacter pylori에 대하여 항균효과가 가장 높은 Fomitopsis pinicola를 동정하고 저해물질의 성질에 관한 연구를 수행한 결과 다음과 같은 결론이 얻어졌다. H. pylori에 대한 항균력이 가장크게 나타난 네팔에서 채취한 F. pinicola의 동정결과는 F. pinicola AY854083로 밝혀졌다. F. pinicola의 균사체는 액상배양을 시작한 후 5일이 경과하면서 H. pylori에 대한 항균력이 나타났으며 8일 이후부터 가장 높았다. 그러나 균사체 추출물에서는 항균력이 나타나지 않았다. F. pinicola 균사체 8일 배양액의 H. pylori에 대한 최소억제 농도는 $0.25\;mg/m{\ell}$ 이었고, 배양액의 낮은 pH (pH 2.0)가 H. pylori의 생육억제에 영향을 주지만 중화를 하여도 H. pylori에 대한 항균력은 유지되었다. F. pinicola 균사체 8일 배양액을 DEAE-sephadex A-25 LC로 분리하여 Fp-P1, Fp-P2 및 Fp-P3를 얻었으며 H. pylori에 대한 항균력은 Fp-P1에서만 나타났다. Fp-P1을 thin layer chromatography로 분리하였으며 H. pylori에 대한 항균력은 $R_{f}$ 값이 0.67인 Fp-T3에서만 나타났다. F. pinicola 균사체 8일 배양액, Fp-P1, Fp-T3를 HPLC의 Waters symmetry $300\;C_{18}\;5\;{\mu}m$ column ($3.9{\times}150\;mm$)으로 분리한 결과 Fp-T3에서 단일 peak만 남았으며 이것이 H. pylori에 대한 항균력을 확인하였다. H. pylori에 대한 항균력을 나타내는 F. pinicola 균사체 배양액에서 분리 정제한 물질은 BioLC 분석과 TLC 발색결과로써 아미노기를 포함한 당으로 사료된다.

다중검출기유도결합플라즈마질량분석기를 이용한 Pu 동위원소비 정밀 분석법 (The Improved Method for Precise Determination of Pu Isotope Ratio using MC-ICP-MS)

  • Pu 동위원소는 기원에 따라 각기 다른 $^{240}Pu/^{239}Pu$ 동위원소비를 나타내는 특성이 있다. 질량분석법은 극미량 장반감기핵종 분석에 매우 유용한 방법으로서 검출기 특성상 낮은 검출한계를 갖고 있으며, 소량의 시료를 이용하여 짧은 시간에 측정을 할 수 있다는 장점을 갖고 있으므로 Pu을 비롯한 장반감기 핵종 분석에 유용하다. 다중검출기유도결합플라즈마질량분석기(Multicollector ICP-MS)는 다중검출시스템을 이용하여 분석하고자 하는 여러 핵종의 동시분석이 가능하며, 특별히 본 연구에서는 극미량 핵종의 검출이 가능한 multiple ion counter (MIC)를 사용하여 Pu 동위원소 분석에 적용하였다. 본 연구방법의 검출한계(detection limit)는 최적화된 조건에서 $^{239}Pu$$^{240}Pu$ 각각 $0.10\;fg\;ml^{-1}$ ($0.24\;{\mu}Bq\;ml^{-1}$), $0.12\;fg\;ml^{-1}$ ($0.97\;{\mu}Bq\;ml^{-1}$)이었으며, 240Pu 기준으로 약 200 cps 정도의 매우 낮은 농도수준에서 1%이하의 $^{240}Pu/^{239}Pu$ 동위원소비 계측편차를 나타내었다. 본 연구방법은 표준용액 및 다양한 매트릭스의 표준물질 분석에 적용함으로써 검증 확인하였다.

비수리 추출물의 항균 및 항산화 활성 (Antibacterial and Antioxidative Activity of Lespedeza cuneata G. Don Extracts)

  • 본 연구에서는 비수리 추출물의 피부 상재균에 대한 항균 작용과 항산화 활성, tyrosinase 저해 효과 등에 관한 조사를 수행하였다. 피부 상재균에 대한 항균활성 측정결과, P. ovale에 대한 ethyl acetate 분획의 MIC는 0.125%로 나타났으며, 이는 methyl paraben보다 큰 항균활성을 나타내었다. 추출물의 free radical(1,1-diphenyl-2-picrylhydrazyl, DPPH) 소거활성($FSC_{50}$)은 aglycone 분획(14.63 ${\mu}g$/mL)에서 가장 큰 활성을 나타내었고, 또한 Luminol-의존성 화학발광법을 이용한 $Fe^{3+}$-EDTA/$H_2O_2$ 계에서 생성된 활성산소종(reactive oxygen species, ROS)에 대한 비수리 추출물의 총항산화능은 aglycone 분획(0.07 ${\mu}g$/mL)에서 가장 큰 활성을 나타내었다. 비수리 추출물에 대하여 rose-bengal로 증감된 사람 적혈구의 광용혈에 대한 억제 효과를 측정하였고, 농도 의존적(1 ~ 50 ${\mu}g$/mL)으로 세포보호 효과를 나타내었다. Tyrosinase의 활성 저해 효과($IC_{50}$)는 비수리 추출물의 ethyl acetate 분획과 aglycone 분획이 각각 104.83 ${\mu}g$/mL, 27.55 ${\mu}g$/mL으로 나타났다. 이상의 결과들은 비수리 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거하고, ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 시사한다. 또한 tyrosinase 저해활성과 피부 상재균에 대한 항균작용으로부터 항산화, 항노화 및 항균성 화장품 소재로서의 응용 가능성을 확인하였다.

식품부패 및 병원성미생물에 대한 천연약용식물 추출물의 항균효과 (Antimicrobial Activities of Natural Medicinal Herbs on the Food Spoilage or Foodborne Disease Microorganisms)

  • 민간요법 또는 식품가공 공정에서 그 안정성이 확보되어 생약재료 또는 식품가공재료로서 사용되고 있는 약용식물에 대하여 항균력을 검색하였고 그중 가장 항균력을 나타낸 황련을 사용하여 활성성분을 분획 추출물한 다음 이 추출물로 부터 여러 종류의 부패 미생물 및 병원성균에 대한 항균성을 검색하였다. 산사, 황련, 측백, 창출 및 석창포 에탄올 추출물은 그람 양성 또는 음성세균 모두에 대하여 강한 항균성을 나타내었으며, 토사자 에탄올추출물은 그람음성 세균에서만 항균성을 나타내었고 사군자, 숙지황 및 백제 에탄올추출물은 거의 또는 모든 시험균에 대하여 항균력을 나타내지 않았다. 황련의 에탄올추출물은 기존에 보고된 많은 한약제의 에탄올추출물과 비교하여 볼 때 적은 농도에서도 매우 다양하고 강한 항균력을 나타내었으며 특히 그람양성 또는 음성세균 모두 강한 항균력을 보였다. 한편, 황련 에탄올 추출물은 $100^{\circ}C$에서 각각 30분 또는 1시간 동안, $121^{\circ}C$에서 15분간 열처리한 후 Listeria monocytogenes균에 대한 생육저해환을 측정한 결과 상기의 모든 열처리에 의해서도 무열처리군과 비교하여 볼 때 항균력의 차이가 없어 열에 매우 안정 하였다. 또한 생육배지의 pH 변화가 Listeria monocytogenes균의 생육억제에 대한 황련 에탄올 추출물의 항균활성에 대하여 미치는 영향을 조사한 결과 배양배지의 pH 가 중성에서 알칼리성으로 갈수록 황련 에탄올 추출물의 항균활성은 더욱 증가되었고 약산성으로 갈수록 감소하는 경향을 나타내었다. 따라서 황련 에탄올추출물은 알칼리성 조건하에서 더욱 강한 항균력을 나타내었다. 황련 에탄올추출물로부터 부분정제한 물질의 항균력은 검색된 미생물중 그람양성세균, 그람음성세균, 효모 및 곰팡이균 모두에 항균력을 나타내었으나 Candida utilis, Rhizopus javanicus에는 억제효과를 나타내지 않았다. 식중독세균인 Listeria monocytogenes과 Staphylococcus aureus에 대한 부분정제물질의 생육저해 효과를 조사한 결과 두균주 모두 $100{\;}{\mu}g/mL$의 농도에서는 무첨가 대조균과 비교하여 차이가 없었으며 $500{\;}{\mu}g/mL$ 첨가구에서는 Listeria monocytogenes균은 36시간까지는 생육을 억제하였으나 그 이후부터는 급속한 생육을 하였고 Staphylococcus aureus균은 완전히 생육이 억제되었고 $1000{\;}{\mu}g/mL$ 첨가구에서는 두 균주 모두 완전히 생육이 억제되었다.
