Identification and Characterization of Hemolytic Bacillus cereus Isolated from Commercial Ssam-jang (시판 쌈장에서 분리한 용혈성 Bacillus cereus의 동정 및 특성 조사)

  • This study was undertaken to identify and characterize hemolytic Bacillus cereus isolated from commercial ssam-jang. The physiological and biochemical properties of isolate were first examined. Using the BIOLOG system, the isolate was identified and assigned to B. cereus MH-2. Phylogenetic tree of MH-2 was plotted based on 16S rRNA sequence comparisons. Hemolytic activity was observed around wells of sheep blood agar plates seeded with MH-2 cultures; the zone of hemolysis gradually increased with increasing incubation time of the cultures. Zymographic analysis estimated the molecular weight of the presumed hemolysis-causing molecule to be about 30 kDa. Survival rates of MH-2 cells decreased with increasing NaCl concentrations in the media. The stress shock proteins (e.g., DnaK and GroEL) induced by NaCl were reduced in proportion to the NaCl concentration and exposure period to B. cereus MH-2. Analysis of SDS-PAGE and Western blot revealed that the stress shock proteins, 70-kDa DnaK and 60-kDa GroEL were decreased proportionate to the NaCl concentrations as well as exposure period in exponentially growing cultures. Scanning electron microscopy demonstrated the presence of perforations and irregular rod forms with wrinkled surfaces in cells treated with NaCl.

  • Using a flue- cured tobacco variety, KF 109, effect of growth regulators(fatty alcohol and C- MH) on the change of protein, DNA, and RNA were investigated. Generally, inhibition of DNA synthesis was observed soon after become notably reduced when checked on 14 days after the treatment. Fatty alcohol treatment appeared to alleviate the inhibition of DNA synthesis caused by the C - MH treatment. It was also observed that in the tips DNA content increased slightly at the early stage after the C - MH treatment but evident reduction of it was resulted from 7th day after the treatment. RNA content in cutters and tips was increased initially but variable transcription inhibitory activities - not so obvious as was observed in DNA synthesis - according to leaf positions were shown thereafter. Ripening of leaves probably due to senescence was advanced by the treatment of the growth regulators. DNA content in root was relatively higher in plants treated with the growth regulators while it was clearly decreased in stalk, However, RNA contents in tissue of stalk and root was not different with that of foliage. Increase of protein content in foliage as well as in stalk was evident 14 days after dual treatment of fatty alcohol and C - MH.

Screening, Characterization, and Cloning of a Solvent-Tolerant Protease from Serratia marcescens MH6

  • solvent-tolerant bacterium strain, MH6, was isolated by hydrophilic organic solvent DMSO enrichment in the medium and identified as Serratia marcescens. The extracellular protease with novel organic-solvent-stable properties from strain MH6 was purified and characterized. The molecular mass of the purified protease was estimated to be 52 kDa on SDS-PAGE. The open reading frame (ORF) of the MH6 protease encoded 504 amino acids with 471 amino acid residues in the mature protease. Based on the inhibitory effects of EDTA and 1,10-phenathroline, the MH6 protease was characterized as a metalloproteinase. The enzyme activity was increased in the presence of $Ni^{2+}$, $Mg^{2+}$, and $Ca^{2+}$. The protease could also be activated by the nonionic surfactants Tween 80 (1.0%) and Triton X-100 (1.0%). The protease showed remarkable solvent stability in the presence of 50% (v/v) solutions of long-chain alkanes and long-chain alcohols. It was also fairly stable in the presence of 25% solutions of hydrophilic organic solvents. Owing to its high stability in solvents and surfactants, the MH6 protease is an ideal candidate for applications in organic catalysis and other related fields.

A Study on Optimized Size of a Mobile Harbor for South Korea Coastal Service

  • The aim of paper is to calculate the optimized size of Mobile Harbor(MH) which would be operated in South Korea coast area. MH is the combined entity which has the function of both ship and container port. In estimating the optimized size, the total cost concept is applied to the different size of MH. Trade-off factors for calculating total cost are MH cost and the over-capacity lost cost. The factors for MH cost estimation are the cargo demand, distance from origin to destination, voyage route and MH's fixed and variable cost in both sailing and port. The other cost is the over-capacity lost cost which is occurred from dead space in case of oversize compared with a voyage demand. The alternatives for the least cost are 250TEU, 500TEU, 750TEU and 1,000TEU sized vessel. The result of research is that 250TEU sized vessel is optimized in a South Korea costal service. If the coastal area be separated in terms of voyage distance or the specific area in considering trade, the optimized size is changed depending upon distance.

  • MetalHalid lamps have good efficiency, good color rendition and good focusing capability. But the shortcoming of metalhalid lamps is blown as acoustic resonance phenomena in the arc tube. Such acoustic resonance produce annoying fluctuations in the intensity and distribution of the emitted light, they can raise the voltage to the point where the arc is extinguished and they can move the arc close enough to the wall to cause local overheating and tube cracking. The objective of this research is to reduce acoustic resonance in the arc tube of the Pulse Start MetaIHaide lamp(MH200[W]). To reduce the acoustic resonance phenomena the electronic ballast was designed for high frequency operation with the constant frequency sinusoidal wave of 89[kHz] in the 84.6[kHz]∼94.2[kHz] range. Experimental results show that the acoustic resonance phenomena are not in the arc tube of Pulse Start MetalHalide lamp (MH200[W]) .

Production of IL-6 and IL-8 in Human Fibroblasts Stimulated with Mycoplasma Lysates and Bacterial Toxins (세균독소와 Mycoplasma 항원으로 자극시킨 사람 섬유아세포의 Interleukin-6와 Interleukin-8 생성의 변화)

  • Cytokines are hormone-like proteins which mediate and regulate inflammatory and immune responses. The purpose of this study was to investigate the effect of lipopolysaccharide (LPS), Staphylococcus enterotoxin B (SEB), and Mycoplasma lysates on regulation of IL-6 and IL-8 production by human nasal fibroblasts. Primary cultured cells were incubated with LPS ($1.0\;{\mu}g/ml$) from E.coli, SEB ($1.0\;{\mu}g/ml$) from S.aureus, or Mycoplasma lysates (M.pneumoniae, Mp; M. fermentans, Mf; M. hominis, Mh, each $1.0\;{\mu}g/ml$). The culture supernatants were collected at 2, 6, and 24 hr and assessed for IL-6 and IL-8 production by enzyme-linked immunosorbent assay. The production of IL-6 in the culture supernatant was downregulated by LPS, SEB, or Mycoplasma lysates. But IL-6 was upregulated by mixed exposure with Mp+LPS (2 hr), Mp+LPS+SEB (24 hr), Mf+LPS (24 hr), Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+SEB (24 hr), or Mh+LPS+SEB (24 hr). The production of IL-8 in the culture supernatant was similar to that of IL-6 by same stimulants. But IL-8 was upregulated by mixed exposure with Mf+LPS+SEB (2 hr), Mh+LPS (24 hr), Mh+ SEB (24 hr), or Mh+LPS+SEB (24 hr). These studies show that costimulation of LPS or SEB with Mycoplasma whole cell lysates upregulates the production of IL-6 and IL-8.

  • There are two approaches, HA(Home Agent)-based and FA(Froegn Agent)-based mulicasting in mobile computing environments based on mobile IP. If we consider efficiency of routing and bandwidth the FA-based model is better than the HA-based model. But in the case of a MH(Mobile Host) moves frequently it shows bad connection durability because of join and graph delays. To overcome this and to support effective mobile multicasting we propose an extended FA-based multicast routing. In our model previous FA for wards multicast packets to MH. while new FA processes a jion request from MH. That is in general case our model supports FA-based multicasting for efficiency but it provides unicast-based mulicasting for connection durability when MH moves to new FN(Roreign Network). To evaluate our model, we analyze connecction durability, bandwidth, overheads and routing sosts in many situations and compare with previous works.

  • Using a flue-cured tobacco variety, BU 109, effect of growth regulators(Fatty Alcohol and C-MH) on the change of invertase activity and sugar metabolites were investigated. Invertase activity in untreated leaf tissue was decreased along with maturation of leaf, However, a momentary increase of the activity was observed in leaves by the dual treatment of fatty alcohol and C-MH regardless of leaf position while sole C-MH treatment resulted in activity increase by 14 days after the treatment. Similar tendency was observed in stalk. Sugar content in leaf was increased immediately after the treatment but no significant increase at large resulted until 14 days after treatment. After harvest, reducing sugar was increased by the growth regulators. Nevertheless, in case of dual treatment, the total sugar content was not different with that of untreated control. R/T ratio was gradually increased after topping stage and reached maximum at 14 days after treatment of growth regulators. It decreased a little after harvest but the RR ratio was relatively higher due to increase of reducing sugar resulted by the treatment of the chemicals. Upon treatment of growth regulators, reducing sugar content was increased in lugs and leaves compared to untreated control and the content of sugar metabolites was increased by the use of the chemicals either at lower or higher than recommended dose.   • Characteristics of a commercial metal hydride (MH) hydrogen supply system have been investigated and an operating strategy was developed based on the experimental data. As a prior step, charging/discharging capacity, thermal properties such as heat capacity, heat of reaction of MH system were experimentally measured. And then P-C-T data for various operating conditions were collected and a correlation between P, C and T predicting the behavior of MH was derived. Based on the basic experimental data, an operating strategy of MH system was developed, in which the hot water temperature supplied into the water jacket of MH was controlled depending on the pressure of MH, thereby the pressure of MH could be maintained at a suitable range. By adjusting the temperature of hot water from $40^{\circ}C$ to $60^{\circ}C$, the maximum discharging capacity of hydrogen could be increased by 4.7%, and consequently more stable hydrogen supply and longer operation time of fuel cell system could be achieved.

  • Recently, researches on resource allocation algorithms operating in a distributed way are widely conducted because of the increasing number of network nodes and the rapidly changing the network environment. In this paper, we propose Multi-Hop DESYNC(MH DESYNC), that is bio-inspired TDMA-based resource allocation scheme operating in a distributed manner in multi-hop networks. In this paper, we define a frame structure for the proposed MH DESYNC algorithm and firing message structure which is a reference for resource allocation and propose the related operating procedures. We show that MH DSYNC can resolve the hidden-node problem effectively and verify that each node shares resources fairly among its neighboring nodes. Through simulation evaluations, it is shown that MH DESYNC algorithm works well in a multi-hop networks. Furthermore, results show that MH DESYNC algorithm achieves better performance than CSMA/CA algorithm in terms of throughput.
