붉나무 순차 용매 추출물의 항산화 효과 비교 (Antioxidative Effect of us javanica Linne Extract by Various Solvents)

  • 75% ethanol 추출물에서 뚜렷한 항산화 효과가 확인된 붉나무 껍질로부터 hexane, ethyl acetate, methanol 그리고 99% ethanol 추출물을 얻고 이들을 팜유와 돈지에 ascorbic acid, citric acid 그리고 ${\delta}-tocopherol$과 함께 첨가하여 Rancimat으로 항산화 효과를 비교하였다. 용매별 추출 수율은 methanol이 14.53%(w/w)로 가장 높았다. hexane, ethyl actate, methanol 그리고 ethanol(99%) 추출물 600ppm을 ascorbic acid 200ppm과 함께 팜유에 첨가하였을 때 antioxidant index(AI : 항산화제첨가 유지의 유도 기간/대상 유지의 유도 기간)는 각각 1.83, 2.25, 2.81 그리고 2.85로 다른 처리구에 비하여 높았고 돈지의 경우는 추출물 600ppm에 ${\delta}-tocopherol$ 200ppm 첨가구가 가장 효과가 높아서 AI는 각각 3.64, 7.83, 7.34 그리고 9.30이었다. ethanol 및 ethyl actate 추출물의 용매별 분획물은 뚜렷한 항산화 증진 효과가 없었고 methanol 추출물을 첨가한 팜유 및 도니지의 $60^{\circ}C$ 저장 시험에서 POV 및 TBA가를 비교한 결과 산화 억제 효과가 뚜렷하였다.

방사선이 조사된 생쥐의 혈청성분 변화 (Change of the Serum Composition in Irradiated Mice)

  • 본 연구는 방사선 조사에 의해 생쥐의 혈청성분이 어떠한 변화를 일으키는가를 구명하는데 있고, 이를 위해 10MeV급 Linac 방사선치료장치를 사용하여 3Gy 방사선을 1회 전신 조사한 생쥐의 혈청(serum)을 채취하여 14가지 생화학적 분석을 실시한 후 paired t-test를 실시하였다. 그 결과 다음의 3가지 혈청성분에서 유의한 변화가 있다는 사실을 확인하였다. 첫째, glucose 측정에서 정상대조군(normal control group)의 $185.43{\pm}14.93$보다 방사선조사군(irradiation group)이 $220.00{\pm}17.58$로 높게 나타나 평균값에 매우 유의한 차이를 보였다(p<0.001). 둘째, BUN(blood urea nitrogen) 측정에서 정상대조군의 $19.61{\pm}1.65$보다 방사선조사군이 $15.70{\pm}1.48$로 낮게 나타나 평균값에 유의한 차이를 보였다(p<0.01). 셋째, albumin 측정에서 정상대조군의 $3.19{\pm}0.34$보다 방사선조사군이 $2.89{\pm}0.25$로 낮게 나타나 평균값에 유의한 차이를 보였다(p<0.05). 결론적으로, 3Gy 방사선이 조사된 생쥐의 혈청은 glucose, BUN(blood urea nitrogen), albumin 등 3가지 성분에서 유의할만한 변화를 일으켰다.

대전·충남지역 개에서 지알디아증 유병률 (Prevalence of Canine Giardiasis in the Daejeon and Chungnam Area)

  • 본 연구는 한국의 대전과 충남 지역에 있는 개 100마리에서 실내견 (25마리)과 실외견 (75마리)을 구분하여 지알디아증의 유병률을 조사하였다. 모든 샘플은 Giardia SNAP 키트($SNAP^{(R)}$ test, IDEXX Laboratories, Inc., Westbrook, ME)를 이용하여 감염여부를 조사하였다. 지알디아증은 실외견(11/75, 14.66%)이 실내견(1/25, 4.00%) 보다 유의성 있는 높은 유병률은 나타내었다. 또한 자견(< 2yr: 10/51, 19.60%)이 성견(> 2yr: 2/49, 4.08%) 보다 유의성 있는 높은 유병률은 나타내었다. 임상증상을 나타내는 개는 증상이 없는 개보다 높은 유병률을 나타내었으나 (증상: 4/22, 18.18%, 무증상: 9/78, 11.54%) 통계학적으로 유의하지는 않았다. 실외견 중 분리되어 단독으로 기르는 개(2/50, 4.00%)보다 합사하여 여러 마리가 같이 생활하는 개(9/25, 36.00%)에서 높은 유병률을 나타내었다. 세계 여러 나라의 다양한 지역에서 지알디아증 유병률에 대한 조사는 많이 진행되었으나, 사육 패턴에 따른 유병률을 평가한 적은 없기에 본 연구의 의의가 있다.

Evaluating and predicting net energy value of wheat and wheat bran for broiler chickens

  • Objective: It is crucial to accurately determine the net energy (NE) values of feed ingredients because the NE system is expected to be applied to the formulation of broilers feed. The NE values of 5 wheat and 5 wheat brans were determined in 12-to 14-day old Arbor Acres (AA) broilers with substitution method and indirect calorimetry method. Methods: A total of 12 diets, including 2 reference diets (REF) and 10 test diets (5 wheat diets and 5 wheat bran diets) containing 30% of test ingredients, were randomly fed to 864 male AA birds with 6 replicates of 12 birds per treatment. These birds were used to determine metabolizable energy (ME) (8 birds per replicate) in the chicken house and NE (4 birds per replicate) in the chamber respectively at the same time. After a 4-d dietary and environment adaptation period, growth performance, energy values, energy balance and energy utilization were measured during the following 3 d. Multiple linear regression analyses were further performed to generate prediction equations for NE values based on the chemical components and ME values. The NE prediction equation were also validated on another wheat diet and another wheat bran diet with high correlation (r = 0.98, r = 0.75). Results: The NE values of 5 wheat and 5 wheat bran samples are 9.34, 10.02, 10.27, 11.33, and 10.49 MJ/kg, and 5.37, 5.17, 4.87, 5.06, and 4.88 MJ/kg DM, respectively. The equation with the best fit were NE = 1.968AME-0.411×ADF-14.227 (for wheat) and NE = -0.382×CF-0.362×CP-0.244×ADF+20.870 (for wheat bran). Conclusion: The mean NE values of wheat and wheat bran are 10.29 and 5.07 MJ/kg DM in AA broilers. The NE values of ingredients could be predicted by their chemical composition and energy value with good fitness.

Accuracy of posteroanterior cephalogram landmarks and measurements identification using a cascaded convolutional neural network algorithm: A multicenter study

  • Objective: To quantify the effects of midline-related landmark identification on midline deviation measurements in posteroanterior (PA) cephalograms using a cascaded convolutional neural network (CNN). Methods: A total of 2,903 PA cephalogram images obtained from 9 university hospitals were divided into training, internal validation, and test sets (n = 2,150, 376, and 377). As the gold standard, 2 orthodontic professors marked the bilateral landmarks, including the frontozygomatic suture point and latero-orbitale (LO), and the midline landmarks, including the crista galli, anterior nasal spine (ANS), upper dental midpoint (UDM), lower dental midpoint (LDM), and menton (Me). For the test, Examiner-1 and Examiner-2 (3-year and 1-year orthodontic residents) and the Cascaded-CNN models marked the landmarks. After point-to-point errors of landmark identification, the successful detection rate (SDR) and distance and direction of the midline landmark deviation from the midsagittal line (ANS-mid, UDM-mid, LDM-mid, and Me-mid) were measured, and statistical analysis was performed. Results: The cascaded-CNN algorithm showed a clinically acceptable level of point-to-point error (1.26 mm vs. 1.57 mm in Examiner-1 and 1.75 mm in Examiner-2). The average SDR within the 2 mm range was 83.2%, with high accuracy at the LO (right, 96.9%; left, 97.1%), and UDM (96.9%). The absolute measurement errors were less than 1 mm for ANS-mid, UDM-mid, and LDM-mid compared with the gold standard. Conclusions: The cascaded-CNN model may be considered an effective tool for the auto-identification of midline landmarks and quantification of midline deviation in PA cephalograms of adult patients, regardless of variations in the image acquisition method.

산청목 메탄올 추출물의 진통 효과 (Anti-nociceptive Activity of Acer tegmentosum MeOH Extract)

  • Acer tegmentosum which is belongs to Aceraceae has been widely used as a traditional medicine for the treatment of lots of diseases including pain management. In this study, we evaluated the anti-nocicepitve effects of methanolic extract of A. tegmentosum (MAT) in mice using various pain models. MAT presented strong and dose-dependent anti-nociceptive activities on thermal nociception models such as tail-immersion test and hot plate test. Moreover, acetic acid-induced chemical nociception was signigicantly reduced by MAT treatment. We could confirm MAT's central and peripheral analgesic properties by formalin test. We also found that the pre-treatment of opioid receptor antagonist did not alter the MAT's anti-nociception, suggesting opioid receptor is not involved in analgesic activity of MAT. Based on our results, we could conclude that MAT may be possibly used as an anti-nociceptive agent for the treatment of various nociceptive pains.

벼종자의 brassinosteroid 활성물질 (Brassinosteroid substances in immature oryza sativa seeds)

  • 벼종자에 존재하는 brassinosteroid 활성물질을 탐색하기 위하여 동진벼 미숙종자를 MeOH로 추출하고, 이 추출물을 rice inclination test를 지표로, 용매분획, silica gel adsorption chromatography, Sephadex LH-20 chromatography, charcoal adsorption chromatography 등으로 brassinosteroid 활성물질을 정제한 다음, silica gel 흡착 chromatography로 활성성분을 분리하였다. HPLC에 의해 활성본체를 구명한 결과, castaserone, teasterone, 6-deoxocastasterone이 최초로 동정되었다. 벼종자에 함유된 brassinosteroid 활성물질의 함량은 brassinolide로 환산하여 생체중량 g당 $0.5{\sim}1.5\;ng$ 수준이었다.

  • 결명자에 존재하는 brassinosteroid 활성물질을 탐색하기 위하여 미숙종자를 MeOH로 추출하고, 이 추출물을 rice inclination test를 지표로, 용매분획, silica gel adsorption chromatography, Sephadex LH-20 chromatography, charcoal adsorption chromatography, Bondesil chromatography 등의 수법으로 brassinosteroid 활성물질을 정제한 다음, silica gel 흡착 chromatography로 활성성분을 분리하고, 각 활성획분에 대해 TLC 및 $C_{18}$ column과 ODS column을 사용한 HPLC에 의해 활성본체를 구명한 결과, 결명자의 brassinosteroid 활성물질로 최초로 teasterone, castasterone, brassinolide 가 동정되었다. 결명자에 함유된 brassinosteroid의 함량은 brassinolide로 환산하여 생체중량g당 $3.5{\sim}5.5\;ng$ 정도였으며, 동정된 3종의 brassinosteroid는teasterone, castasterone, brassinolide순으로 존재하였다.

One-Step Simultaneous Immunochromatographic Strip Test for Multianalysis of Ochratoxin A and Zearalenone

  • Individual immunochromatographic assays (ICG) for ochratoxin A (OTA) and zearalenone (ZEA) were optimized and used in the development of a one-step simultaneous immunochromatographic assay (OS-ICG) for the rapid multianalysis of two mycotoxins in corn samples. The nitrocellulose membrane of the OS-ICG was treated with OTA-bovine serum albumin (BSA), ZEA-ovalbumin (OVA), and anti-mouse IgG in the OTA test, ZEA test, and control zones, respectively. Monoclonal antibody-gold conjugates (OTA3 MAb-gold and ZEA2C5 MAb-gold) were sprayed onto the conjugate pad. The visual detection limits were 2.5 and 5 ng/ml for OTA and ZEA, respectively, and the results were obtained within 15 min after starting the analysis. An efficient, simple, and rapid extraction method using 30% MeOH/PBS was established and validated by analyzing the corn samples spiked with OTA/ZEA mixtures (0/0, 5/10, 10/20, and $20/30\;{\mu}g/kg$). The cut-off values of the OS-ICG for the spiked corn were 5 and $10\;{\mu}g/kg$ for OTA and ZEA, respectively. Natural corn samples were analyzed by OS-ICG, direct competitive enzyme-linked immunosorbent assay (DC-ELISA), and HPLC. Results of the OS-ICG were in good agreement with those obtained by DC-ELISA and HPLC. The developed OS-ICG offers a rapid, easy-to-use, and portable analytical system and can be used as a convenient qualitative tool for the on-site simultaneous determination of OTA and ZEA in cereals, food, and agricultural products in one analytical cycle.

황련해독탕 중 3종 생리활성 물질의 HPLC-DAD 동시 정량분석법 확립 (Simultaneous Analysis of three Marker Components in Hwangryunhaedok-tang by HPLC-DAD)

  • In this study, a high performance liquid chromatography-diode array detector method was established, for simultaneous determination of three compounds, berberine, palmatine and geniposide in Hwangryunhaedok-tang, To develop and validate method, $C_{18}$ column (5 ${\mu}M$, 4.6 mm${\times}$250 mm) was used with gradient mobile phase, water containing 0.1% trifluoroacetic acid (TFA) and MeOH at the column temperature of $30^{\circ}C$. UV wavelength was set at 230 and 280 nm. Validation of the chromatography method was evaluated by linearity, precision and accuracy test. Calibration curve of standard components showed good linearity ($R^2$ > 0.9999). The limits of detection (LOD) and limits of quantification (LOQ) varied from 0.05 to 0.17 ${\mu}g/ml$ and 0.15 to 0.53 ${\mu}g/ml$, respectively. The relative standard deviations (RSDs) data of intra-day and inter-day test were in less than 2.99% and 1.90%, respectively. The results of the accuracy test were in the range of 98.36 to 102.52% with RSDs values 0.32 to 1.98%. The results of validation indicated that this method was a very accurate and sensitive assay.
