Regulation of Cyclin D3 by Calpain Protease in Human Breast Carcinoma MDA-MB-231 Cells (인체 유방암세포에서 calpain protease에 의한 cyclin D3의 발현 조절)

  • The $Ca^{2+}-activated$ neutral protease calpain induced proteolysis has been suggested to play a role in certain cell growth regulatory proteins. Cyclin proteolysis is essential for cell cycle progression. D-type cyclins, which form an assembly with cyclin-dependent kinases (cdk4 and cdk6), are synthesized earlier in G1 of the cell cycle and seem to be induced in response to external signals that promote entry into the cell cycle. Here we show that cyclin D3 protein levels are regulated at the posttranscriptional level by calpain protease. Treatment of human breast carcinoma MDA-MB-231 cells with lovastatin and actinomycin D resulted in a loss of cyclin D3 protein that was completely reversible by the peptide aldehyde calpain inhibitor, LLnL. The specific inhibitor of the 26S proteasome, lactacystin, the lysosome inhibitors, ammonium chloride and chloroquine, and the serine protease inhibitor, phenylmethylsulfonylfluoride (PMSF), did not block the degradation of cyclin D3 by lovastatin and actinomycin D. Results of in vitro degradation of cyclin D3 by purified calpain showed that cyclin D3 protein is degraded in a $Ca^{2+}-dependent$ manner, and the half-life of cyclin D3 protein was dramatically increased in LLnL treated cells. These data suggested that cyclin D3 protein is regulated by the $Ca^{2+}-activated$ protease calpain.

Peptide H Reduces IL-6 Expression in Human Breast Cancer MDA-MB-231 Cells (인간 유방암 MDA-MB-231 세포에서 Peptide H에 의한 IL-6 발현 억제효과)

  • Chronic inflammation is involved in cancers, rheumatoid arthritis, and Crohn's disease. Inerleukin-6 (IL-6) plays major roles in inflammation. Chungkookjang, fermented soybean contains diverse peptides produced by cleavage of soybean proteins. The peptides can be bioactive compounds. Peptide (Gly-Val-Tyr-Tyr-Met-Tyr was purified from Chungkookjang, and modified to be 6mer H, Glu-Val-Tyr-Tyr-Met-Tyr (EVYYMY). Peptide H's activity to suppress IL-6 expression in a human breast cancer cell, MDA-MB-231 was determined. IL-6 Expression was reduced in the cell treated with peptide H 25 times less than controls which were not treated with peptide H. Proliferation of MDA-MB-231 cells was inhibited by peptide H, which is concentration-dependent. Blocking of IL-6 signals is known to be effective in reducing inflammation in rheumatoid arthritis, Crohn's disease, and cancers. Since peptide H can reduce inflammatory IL-6 expression, application of this study will contribute to drug development for diseases which are caused by excessive IL-6.

  • Chungkookjang, fermented soybeans, contains diverse peptides produced during fermentation. Fermented soybean extracts containing the peptides can affect cellular signal transduction. Proliferation of human breast cancer MDA-MB-231 cells were repressed dependent on concentrations of fermented soybean extracts. Since fermented soybean extracts inhibited breast cancer cell's growth, and inflammation is related to cancer, it is determined whether it can suppress inflammatory $TNF{\alpha}$ expression. $TNF{\alpha}$ expression in MDA-MB 231 cells treated with fermented soybean extracts was repressed by that extracts. $TNF{\alpha}$ inhibitors were developed as drugs for autoimmune diseases. Since fermented soybean extracts suppressed $TNF{\alpha}$ expression, it can be developed as those drugs.

Gender-Specific Changes of Plasma MDA, SOD, and Lymphocyte DNA Damage during High Intensity Exercise (고강도 운동 시 성별에 따른 혈장 MDA, SOD 및 임파구 DNA 손상 변화)

  • The purpose of this study was to investigate gender-specific changes of plasma MDA, SOD, and lymphocyte DNA damage during high intensity exercise. In this study, 17 healthy male and 18 healthy female college students ran on a treadmill at 85%$VO_{2max}$ until the point of all-out. Blood-collecting was carried out five times (Rest, Ex-Exha, R0.5h, R4h and R24h), and with the collected blood, plasma malondialdehyde (MDA), superoxide dismutase (SOD), and lymphocyte DNA damage were analyzed. Plasma MDA and SOD concentration increased significantly at the Ex-Exha (p<0.05), and there were no significant differences in gender. For the degree of lymphocyte DNA damage, all %DNA in the tail, tail length and tail moment increased significantly at the Ex-Exha (p<0.05), and %DNA in the tail and tail length were significantly higher in the male group than in the female group (p<0.05). These results suggest that acute high intensity exercise not only causes oxidative stress but also brings about lymphocyte DNA damage. In addition, it was found that males showed higher DNA damage than females in terms of oxidative stress subject to high intensity exercise. Nevertheless, further subsequent studies are required in order to better understand the mechanism behind DNA damage varying with gender, in a way that takes into consideration physical fitness, hormonal level, exercise intensity and duration - additional factors which might affect DNA damage.

Simultaneous determination of amphetamine-like drugs in human urine by SPE and GC/MS (고체상추출과 GC/MS를 이용한 소변 중 암페타민계 마약성분 동시분석법)

  • Although liquid-liquid extraction (LLE) method has been used routinely for the analysis of amphetamine-like drugs (amphetamine; AP, methamphetamine; MA, 3,4-methylenedioxyamphetamine; MDA, 3,4-methylenedioxymethamphetamine; MDMA, 3,4-methylenedioxyethylamphetamine; MDEA), a solid-phase extraction (SPE) method, which can be automated, was applied for the simultaneous determination by GC/MS in human urine. Urine samples (3 mL) and 0.1 M phosphate buffer (1 mL, pH 7.0) were extracted by an automated SPE system. The eluent was evaporated, derivatized with trifluoroacetic anhydride (TFAA), and analyzed by GC/MS. The calibration curves was linear with correlation coefficient ($r^2$) above 0.994 in the ranges of 34.0 (AP), 28.0 (MDA)~1000.0 ng/mL for AP, MDA, and 50.0~2000.0 ng/mL for MA, MDMA, and MDEA. The limits of detection ranged from 4.0 to 10.0 ng/mL, and the limits of quantitation ranged from 12.0 to 34.0 ng/mL. The relative recoveries were 93.5~107.7 %. The precisions and accuracies were 1.9~14.8 % and -8.7~14.8 %, respectively. The present method was successfully applied to identify the MA or Ecstasy (MDMA) abusers in exact as well as rapid.

A Convergence Study on the Effects of Improving Buckwheat Dietary Fiber in Mice with Hyperlipidemia and Oxidative Stress (고지혈증과 산화적 스트레스가 유도된 생쥐에서 메밀 식이섬유의 개선 효과에 대한 융합 연구)

  • The effect of buckwheat dietary fiber (BDF) as hypolipidemic and antioxidant agent were determined in C57BL/6 mice fed a high fat diet (HFD) with different doses of 500 (low, BDF-L) or 1,000 (high, BDF-H) mg/kg of body weight, compared with the HFD-diet control group (HFD). The negative control group (ND) was fed the basal diet. Body weights in the BDF-L and BDF-H groups were significantly decreased as compared to those in the HFD group (p<0.05). BDF also improved the lipid profile in a dose-dependent manner; serum lipid profiles and levels of insulin, glucose, and free fatty acid were significantly decreased in the BDF-L and BDF-H groups, whereas HDL-C and adiponectin significantly increased as compared to the HFD group (p<0.05). Meanwhile, BDF lowered serum malondialdehyde (MDA) in comparison with the HFD group (p<0.05). The results demonstrate that the intake of BDF might prevent obesity and its related metabolic disorders by inducing dyslipidemia and oxidative stress.

Umbilical Cord Arterial Concentrations of Isoprostane(8-iso-PGF2α) in Newborn Infants (신생아에서 제대 동맥혈 Isoprostane(8-iso-PGF2α) 농도에 관한 연구)

  • Purpose : We measured the umbilical cord arterial concentrations of isoprostane($8-iso-PGF_{2{\alpha}}$) and intended to decide whether the umbilical cord arterial concentrations of isoprostane could be used as a useful parameter for lipid peroxidation in newborn infants. Methods : The isoprostane and malondialdehyde(MDA) concentrations of the umbilical cord were measured by enzyme immunoassay and TBARS(thiobarbituric acid reactive substance) assay in 33 preterm and 28 term infants, respectively. The concentrations of isoprostane and MDA were compared between preterm infants and term infants, and were analysed for association with perinatal risk factors and neonatal complications. Results : Umbilical cord arterial concentrations of isoprostane were $704.7{\pm}635.6pg/mL$ and $421.9{\pm}306.5pg/mL$ in preterm and term infants, respectively. Umbilical cord arterial concentrations of MDA were $44.0{\pm}22.9{\mu}M/L$ and $26.2{\pm}10.7{\mu}M/L$ in preterm and term infants, respectively. Umbilical cord arterial concentrations of isoprostane and MDA in preterm infants were significantly higher than those in term infants(P<0.05). The umbilical cord arterial concentrations of isoprostane were significantly associated with perinatal risk factors such as fetal distress, oligohydramnios, and breech delivery in preterm infants and pregnancy-induced hypertension in term infants(P<0.05). Conclusion : Umbilical cord arterial concentrations of isoprostane in preterm infants were higher than those in term infants, and those are significantly associated with some perinatal risk factors.

Effects of Plum Fruits Extracts at Different Growth Stages on Quinone Reductase Induction and Growth Inhibition on Cancer Cells (생육시기별 피자두 추출물의 암세포 증식 억제 효과 및 Quinone Reductase 유도 활성에 미치는 영향)

  • The plum (Prunus salicina L., cv. ‘Soldam’) fruits were harvested at different growth stages, and then extracted using 80% methanol, respectively. The methanol extracts of plum were investigated for their growth inhibition on 6 kinds of human cancer cells using MTT assay and for their activity to induce quinone reductase (QR) in murine Hepa1c1c7 cells. Among various methanol extracts of plum, the plum 1~4 (immature fruit), which thin out 10~25 days before final harvest, showed higher anticarcinogenic activity against 5 kinds of cancer cells than plum 5~9 (intermediate-mature and mature fruit). Especially, plum 1 and 2 were exhibited the strongest growth inhibiting activities to AGS, HepG2 and MDA cancer cells. Also the plum extracts induced the activity of QR, an anticarcinogenic marker enzyme, in Hepa1c1c7 cells while the induction of QR activities by adding plum extracts were shown to be a little difference depending on growth stages. These results suggested that methanol extracts of immature plum can be considered as an effective natural cancer chemoprevention materials.

Difference in Growth, SOD Activity and MDA Content Between Ozone Tolerant and Sensitive Families of Open-Pollinated Pinus densiflora (소나무 풍매차대묘의 오존 내성 및 민감성 가계간 생장, SOD 활성 및 MDA 함량 차이)

  • This study was conducted to understand response of damage and tolerance to ozone between ozone tolerant and sensitive families of P. densiflora which had been selected by the based on visual injury and growth. Five individuals were selected in each group, and were exposed to 100ppb ozone for 90 days. Every 30 days after ozone fumigation, diamter at root collar (ORC), superoxide dismutase (SOD) activity, and malondialdehyde (MDA) content were measured. In early state of ozone fumigation tolerant families showed three times higher relative DRC growth rate than sensitive families. And during whole fumigation period growth rate of tolerant families was superior to growth rate of sensitive families. SOD activity in sensitive families was increased 30 days after fumigation, and in accordance with fumigation period extended it was decreased continuously. MDA content in ozone treatment showed higher than in control, and MDA content of tolerant families was higher than that sensitive families. In our results tolerant families has higher antioxidative activity than sensitive families. Therefore tolerant families can restrain lipid peroxidation and damage on physiological activity.

Differences in Urine Cadmium Associations with Renal Damage Markers According to the Adjustment with Specific Gravity or Urinary Creatinine (요비중 또는 크레아티닌 보정에 따른 요중 카드뮴과 신장손상지표와의 관련성 비교)

  • In general, specific gravity (SG) and urinary creatinine (CR) have been used to adjust urinary cadmium (Cd) concentrations. However, the validity of correction methods has been controversial. We compared the two adjustments to evaluate associations between urinary Cd and various renal damage markers and to evaluate the relationship between urinary Cd concentration and renal disease markers, such as estimated glomerular filtration rate (eGFR), in a relatively large general population sample. Among the 1,086 volunteers who were enrolled in this study, 862 healthy volunteers who did not have kidney disease were included in the final analysis. Urinary Cd, malondialdehyde (MDA), and N-acetyl-${\beta}$-D-glucosaminidase (NAG) concentrations were measured, the creatinine-based eGFR was calculated, and the relationships between these markers were subsequently analyzed. This study showed the use of urinary Cd concentration adjusted with SG rather than with urinary creatinine may be appropriate in studies evaluating renal function based on Cd exposure. Urinary Cd concentration adjusted with SG had a positive correlation with urinary MDA levels and a negative correlation with eGFR. This relationship was relatively stronger in women than in men. This study showed that urinary Cd level was associated with decreased eGFR in the general population, and oxidative stress was likely to act as an intermediator in this process. These results suggest that eGFR can be a very good indicator of kidney damage caused by Cd exposure in the general population.
