삼릉(三稜) 추출물의 인간 유방암 세포 성장 억제 효과 (Anti-proliferative effect of Sam-nueng(Sparganii Rhizoma) extract on MCF-7 cells)

  • Purpose : This investigation was undertaken to evaluate the antiproliferation, atoptosis of Sam-nueng(Sparganii Rhizoma) extract using MCF-7 human breast cancer cells. Methods : MCF-7 cells were cultured in Dulbecco's modified Eagle's medium/F12(DMEM/F12) supplemented with 10 % fetal bovine serum(FBS; Gibco) and antibiotics. At varying times after extract treatment, cells were harvested with scraper and processed for analysis of protein expression, proliferation, cytotoxicity and apoptosis. Results : Our results show that the extract of Sam-nueng(Sparganii Rhizoma) strongly inhibits the proliferation of MCF-7 cells in a dose and time-dependent manner. Sulforhodamine B showed that the addition of Sam-nueng(Sparganii Rhizoma) extract reduced the viability of MCF-7 cells in a dose-dependent manner. Poly[ADP(ribose)] polymerase(PARP) which serves as a marker of cells undergoing apoptosis, a major substrate for caspase-3 was extensively cleaved in the Sam-nueng(Sparganii Rhizoma)-treated cells. Conclusion : So, we can conclude that Sam-nueng(Sparganii Rhizoma) can have an inhibitive effect on MCF-7 human breast cancer cells by variable mechanisms.

블루베리가 정상유선세포와 유방암세포의 ROS 축적과 세포사멸에 미치는 영향 (The Effect of Blueberry on ROS Accumulation and Cell Death in Human Normal Breast Epithelial(MCF10A) and Breast Cancer(MCF7) Cells)

  • In an effort to elucidate the differential actions of blueberry(BB) in both normal and cancer cells, we utilized human breast cell lines to assess the accumulation of radical oxygen species(ROS) and ROS-associated apoptosis in both human normal breast epithelial(MCF10A) and breast cancer(MCF7) cells. BB extract was added to the cultures at a final concentration of $20{\mu}g/m{\ell}$ for 0(control), 6, 12, and 24 hr intervals. The MCF10A cells evidenced no marked ROS accumulation in the presence of BB, whereas the MCF7 cells evidenced clear ROS accumulation upon BB treatment from 12 hours forward. The number of dying or dead cells did not increase in the BB-treated MCF10A cell groups, whereas that number increased profoundly from 12 hr forward. Furthermore, the expression levels of certain stress-related, and pro- and antiapoptotic gene products evidenced differential responses to BB treatment between the MCF10A and MCF7 cell groups. These results indicate that the components of BB extract differentiate cancer cells by not preventing ROS accumulation within cells and by inducing ROS-associated cell death in cancer cells. However, no marked ROS accumulation or induction of cell death was noted in the normal breast epithelial cells. The fact that BB extract exerted a differential effect on cancer cells opens further directions of research regarding the specific components that exert the differential BB-mediated effects in the selective prevention of normal cells and therapy for cancer tissues in the physiological body.

항암제 내성 유방암 MCF7/adR 세포주에 대한 보정방암탕과 홍삼산성다당체의 세포고사 유도효과 (Apoptotic Effect of Ethanol Extracts of Bojungbangamtang and Acidic Polysaccharide of Korea Red Ginseng in a MCF7/adR Multidrug-resistance Breast Cancer Cells)

  • This study was undertaken to determine whether the 9 herbal complex induces apoptosis in human breast cancer MCF-7 cells and adriamycin-resistant MCF7/adR cells. Ethanol extracts of Bojungbangamtang (BBTE) and acidic polysaccharide of Red Ginseng (GIN) induced cell death in both MCF-7 and MCF7/adR cells. Ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng also induced $G_2/M$ cell cycle arrest and increased TUNEL positive cells in MCF7/adR cells. In addition, flow cytometric analysis revealed the decreased expression of P-glycoprotein (P-gp) in ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng treated MCF7/adR cells. Similarly, decreased protein levels of P-glycoprotein and multidrug resistance associated proteins-1 were also determined by immunocytometry in ethanol extracts of Bojungbangamtang treated MCF7/adR cells. Taken together these data indicate that ethanol extracts of Bojungbangamtang and acidic polysaccharide of Red Ginseng inhibit the function of ABC transporters such as multi drug resistance associated proteins (MRPs) and P-glycoprotein as well as induce apoptosis in MCF7/adR cells. Thus, these data suggest that ethanol extracts of Bojungbangamtang and polysaccharide of Red Ginseng can be candidates for the treatment of multidrug-resistant MCF7/adR cells.

  • Stable MCF-7-ERE cells, in which pERE-Luc reporter gene has been stably integrated into the genome of the MCF-7 cells, were used to detect the estrogenic activity of various dietary flavonoids.in either pure chemical or mixtures.   • In the course of screening for microbial metabolites employing human cancer cell line, we identified a mycelial extract of Streptomyces sp. 1365, which are effective on growth inhibition and morphological change of MCF-7, human breasr cancer cell line. By repeased column chromatography and recrystallization process, yellow needle crystals were obtained as an active compound and identified as resistomycin by spectral analysis.

환경호르몬에 의해 유도된 인체 유방암세포의 증식에 대한 당귀로부터 분리한 Decursin 억제효과 (Anti-Proliferation Effects of Decursin from Angelica gigas Nakai in the MCF-7 Cells Treated with Environmental Hormones)

  • 당귀로부터 분리한 decursin을 환경호르몬에 의해 증식을 유도한 인체 유방암세포(MCF-7)에 처리한 후 그 억제효과를 조사하였다. 인체 유방암세포주인 MCF-7은 20, 40, 60, 80 및 100 ${\mu}g/mL$ 농도로 decursin의 처리 시 20 ${\mu}g/mL$ 이상에서 농도 의존적으로 그 증식이 억제되었다. 호르몬이 제거된 배지로 배양한 세포에 0, 0.01, 0.1 및 1 ${\mu}M$의 농도로 환경 호르몬 $17{\beta}$-estradiol과 bisphenol을 처리한 결과 호르몬이 제거된 배지로 배양한 세포에 비해 세포의 증식을 유도하였으며, Yamada(21)와 본 실험 결과가 유사한 세포성장을 보여 암세포의 성장억제 효과를 측정하기 위한 환경호르몬의 농도를 0.1 ${\mu}M$로 하였다. 환경호르몬에 의해 증식이 유도된 MCF-7 세포에 당귀 메탄올추출물 및 decursin을 1, 3, 10 및 30 ${\mu}g/mL$ 농도로 처리한 결과 농도에 비례하여 세포의 증식을 억제하였으며, 10 ${\mu}g/mL$의 농도 이상에서는 대조구의 증식보다도 낮은 생존율을 나타내어 강한 세포독성을 나타내었다. 또한 hoechst 염색을 통하여 세포 핵의 변화를 알아본 결과 decursin 처리군에서 핵의 응축과 apoptic body가 관찰되어 decursin은 apoptosis를 유도함으로써 환경호르몬이 처리된 MCF-7 세포의 증식을 억제하는 것으로 판단된다. 이들 결과는 decursin이 환경호르몬에 의해 증식이 유도되는 MCF-7 세포의 성장을 apoptosis에 의해 억제한다는 것을 나타낸다.

Bioassays of Polycyclic Aromatic Hydrocarbons using CYP1A1-luciferase Reporter Gene Expression System in Human Breast Cancer MCF-7 Cells

  • Biological activities of PAHs are not known although PAHs are considered as carcinogens. Recent industrial society has human widely exposed to PAHs (polynuclear aromatic hydrocarbons) that are comming from the incomplete combustion of organic material as wider spread environmental contaminants. Our laboratory have been studied the effect of PAHs in the human breast cancer MCF-7 cells. In this study, we examined the human breast cancer MCF-7 cells as a new system to evaluate bioactivity of PAHs. We have selected 13 PAHs to examine bioassay using CYP1A1-luciferase reporter gene expression system where CYP1A1 1.6 Kb 5flanking region DNA was cloned in front of luciferase reporter gene and this plasmid was transfected into MCF-7 cells transiently. This cells then used for the study to observe the effect of PAHs. We demonstrated that PAHs induced the CYP1A1 promoter, CYP1A1 mRNA and 7-ethoxyresolufin O-deethylase (EROD) activities in a concentration-dependant manner. None of PAHs that we have tested showed stronger stimulatory effect on CYP1 gene expression than TCDD. Benz(a)anthracene and benzo(b)fluoranthene were weak responders to CYP1A1 promoter activity stimulation, CYP1A1 mRNA and EROD induction in MCF-7 cells and these chemicals seemed to respond less either CYP1A1 mRNA or EROD than CYP1A1 promoter activity. Benzo(k)fluoranthene, chrysene, and dibenzo(a, h)anthracene showed strong response to CYP1A1 promoter activity stimulation, CYP1A1 mRNA increase and also EROD induction in MCF-7 cells. Results of dose response study suggested that two strong responding PAHs, such as benzo(k)fluoranthene and dibenzo(a, h)anthracene might be mediated through Aryl hydrocarbon receptors system in MCF-7 cells.

MCF-7 유방암 세포에서 AMPK 활성에 의한 conjugated linoleic acid의 apoptosis 유도에 관한 연구 (Conjugated Linoleic Acid Induces Apoptosis by Activating AMPK in MCF-7 Breast Cancer Cells)

  • 본 연구는 쇠고기와 유제품에 들어 있는 CLA의 항암효과를 조사하기 위하여 수행되었다. 이 실험을 위하여 MCF-7 인체 유방암 세포주를 사용하였으며, CLA를 처리했을 때 MCF-7 세포의 증식은 CLA의 농도가 증가할수록, 또한 일정한 농도에서는 시간이 경과함에 따라 의존적으로 억제되었다. 이와 같이 암세포의 증식이 억제되는 이유는 Hoechst 33342 염색을 이용한 chromatin staining 및 ROS의 활성 측정실험 결과, apoptosis와 연관이 있는 것으로 확인되었다. CLA 처리에 의한 apoptosis가 AMPK 및 COX-2 단백질의 활성 발현과는 어떤 연관성이 있는지를 조사하기 위하여 Western blot 실험을 실시한 결과, CLA 처리에 따라 AMPK의 활성이 증가되었고, COX-2의 발현은 감소됨으로써, MCF-7 세포에서 apoptosis가 유도되었다는 것을 알 수 있었다. 본 연구를 통하여 조사한 CLA의 항암효과로부터, 향후 다른 식품에 포함된 성분들에서도 암세포의 증식 억제와 apoptosis의 유도를 연구할 수 있는 기초 자료를 제시한 것이라고 할 수 있다.

고삼의 인체 유방암세포에 미치는 효과 (Effects of Sophorae Radix on Human Breast Adenocarcinoma Cells)

  • Objective : The purpose of this study was to investigate the anti-cancer effects of Sophorae Radix and the effects of Doxorubicin (DOX) in human breast adenocarcinoma cells (MCF-7). Method : We used human breast adenocarcinoma cell line, MCF-7 cells. We examined cell death by MTT assay and caspase 3 assay with Sophorae Radix. To examine the inhibitory effects of Sophorae Radix, cell cycle analysis was done the MCF-7 cells after three days with Sophorae Radix. The reversibility of Sophorae Radix was examined on one day to five days treatment with 100 ${\mu}g/ml$ Sophorae Radix. Result : Sophorae Radix inhibited the growth of MCF-7 cells in a dose-dependent fashion. Also we showed that Sophorae Radix induced apoptosis in MCF-7 cells by MTT assay, caspase 3 assay and sub-G1 analysis. Sophorae Radix combined with DOX markedly inhibited the growth of MCF-7 cells compared to Sophorae Radix or DOX alone. After 3 days treatment of MCF-7 cells with Sophorae Radix, the fraction of cells in sub-G1 phase was much higher than that of the control group. Conclusion : Our findings provide insight into unraveling the effects of Sophorae Radix in human breast adenocarcinoma cells and developing therapeutic agents against breast cancer.

Cytokinetic Study of MCF-7 Cells Treated with Commercial and Recombinant Bromelain

  • Background: Breast cancer is a leading cause of death in women. The available chemotherapy drugs have been associated with many side effects. Bromelain has novel medicinal qualities including anti-inflammatory, anti-thrombotic, fibrinolytic and anti-cancer functions. Commercially available bromelain is obtained through tedious methods; therefore, recombinant bromelain may provide a cheaper and simpler choice with similar quality. Materials and Methods: This study aimed to assess the effects of commercial and recombinant bromelain on the cytokinetic behavior of MCF-7 breast cancer cells and their potential as therapeutic alternatives in cancer treatment. Cytotoxic activities of commercial and recombinant bromelain were determined using (sulforhodamine) SRB assay. Next, cell viability assays were conducted to determine effects of commercial and recombinant bromelain on MCF-7 cell cytokinetic behavior. Finally, the established growth kinetic data were used to modify a model that predicts the effects of commercial and recombinant bromelain on MCF-7 cells. Results: Commercial and recombinant bromelain exerted strong effects towards decreasing the cell viability of MCF-7 cells with $IC_{50}$ values of 5.13 ${\mu}g/mL$ and 6.25 ${\mu}g/mL$, respectively, compared to taxol with an $IC_{50}$ value of 0.063 ${\mu}g/mL$. The present results indicate that commercial and recombinant bromelain both have anti-proliferative activity, reduced the number of cell generations from 3.92 to 2.81 for commercial bromelain and to 2.86 for recombinant bromelain, while with taxol reduction was to 3.12. Microscopic observation of bromelain-treated MCF-7 cells demonstrated detachment. Inhibition activity was verified with growth rates decreased dynamically from 0.009 $h^{-1}$ to 0.0059 $h^{-1}$ for commercial bromelain and to 0.0063 $h^{-1}$ for recombinant bromelain. Conclusions: Commercial and recombinant bromelain both affect cytokinetics of MCF-7 cells by decreasing cell viability, demonstrating similar strength to taxol.
