Various microorganisms play important roles in food fermentation, food spoilage, and agriculture. In this study, the biotype of 54 yeast and bacterial strains having high potential for utilization in food and agriculture, including Candida spp., Lactobacillus spp., and Acetobacter spp., were characterized by matrix-assisted laser desorption/ionization time-of flight mass spectrometry (MALDI-TOF MS). This characterization using a fast and robust method provides much-needed information on the selected microorganisms and will facilitate effective usage of these strains in various applications. Importantly, the unique protein profile of each microbial species obtained from this study was used to create a database of fingerprints from these species. The database was validated using microbial strains of the same species by comparing the mass spectra with the created database through pattern matching. The created reference database provides crucial information and is useful for further utilization of a large number of valuable microorganisms relevant to food and agriculture.
In this study, we present the preparation and characterization of oxidized fullerene and fullerene dimer [$C_{120}$]. From the XRD data, other weak peaks with pristine fullerene [$C_{60}$] peaks were observed in the X-ray diffraction patterns for fullerene dimer [$C_{120}$]. SEM micrographs for the fullerene dimer [$C_{120}$] indicated that practically all the surface state was shown the drastic morphology changes and its outer surface is clearly visible and resulted in clogging and frost-like formation. From the MALDI-TOF mass spectra, the differences in the spectra recorded on two kinds of fullerene are due to the oxidation including chemical bonding and bridging between the $C_{60}$ molecules. We also obtained additional information from FT-IR spectra on functional component on the chemically modified surface of oxidized fullerene and fullerene dimer [$C_{120}$].
The unique Br signature was utilized for C-terminal amino acid sequencing of model peptides. C-terminal carboxyl group was selectively derivatized in peptides, containing side chain carboxyl group, using 1-ethyl-3-[3-dimethylaminopropyl]carbodiimide hydrochloride (EDC) and Br was introduced using 4-bromophenylhydrazine hydrochloride (BPH) in a one pot reaction. Matrix-assisted laser desorption/ionization (MALDI) time-of-flight (TOF) tandem mass spectra were obtained carrying the Br signature in the y-series ions. The Br signature facilitated C-terminal sequencing and discrimination of C-terminal carboxyl groups in the free acid and amide forms.
Characterization of the various chemical aspects of composite polymers is important for quality control of manufactured polymers. In this study, we compared three suitable matrices (α cyano-4-hydroxycinnamic acid [CHCA], 2,5 dihydroxy benzoic acid [2,5-DHB], and dithranol), to characterize various synthetic polymers by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. Although the spectra obtained with the CHCA and 2,5-DHB matrices were generally good, in certain samples ghost peaks disappeared only when dithranol was used as the matrix. Furthermore, we examined the use of sodium trifluoroacetate (NaTFA) as an additive to reduce interference by metals and copolymers in the spectra. In conclusion, appropriate selection of a matrix, according to the characteristics of the polymer, and the use of additives to improve sensitivity are important considerations for polymer analysis and development.
An extracellular protease was identified from Bacillus subtilis KCCM 10257 by N-terminal sequencing and mass spectral analysis. The molecular mass of the extracellular protease was estimated to be 28 kDa by SDS-PAGE. Sequencing of the N-terminal of the protease revealed the sequence of A(G,S,R)QXVPYG(A)V(P,L)SQ. The N-terminal sequence exhibited close similarity to the sequence of other proteases from Bacillus sp. A mass list of the monoisotopic peaks in the MALDI-TOF spectrum was searched after peptide fragmentation of the protease. Six peptide sequences exhibiting monoisotopic masses of 1,276.61, 1,513.67, 1,652.81, 1,661.83, 1,252.61, and 1,033.46 were observed from the fragmented protease. These monisotopic masses corresponded to the lytic enzyme L27 from Bacillus subtilis 168, and the Mowse score was found to be 75. A doubly charged Top product (MS) at a m/z of 517.3 exhibiting a molecular mass of 1034.6 was further analyzed by de novo sequencing using a PE Sciex QSTAR Hybrid Quadropole-TOF (MS/MS) mass spectrometer. MS/MS spectra of the Top product (MS) at a m/z of 517.3 obtained from the fragmented peptide mixture of protease with Q-star contained the b-ion series of 114.2, 171.2, 286.2, 357.2, 504.2, 667.4, 830.1, and 887.1 and y-ion series of 147.5, 204.2, 367.2, 530.3, 677.4, 748.4, 863.4, and 920.5. The sequence of analyzed peptide ion was identified as LGDAFYYG from the b- and y-ion series by de novo sequencing and corresponded to the results from the MALDI-TOF spectrum. From these results the extracellular protease from Bacillus subtilis KCCM 10257 was successfully identified with the lytic enzyme L27 from Bacillus subtilis 168.
Background and Objective: Protein expression in colon and rectal cancer (CRC) and paired normal tissues was examined by two-dimensional gel electrophoresis (2-DE) to identify differentially expressed proteins. Materials and Methods: Five fresh colorectal cancer and paired adjacent normal tissues were obtained and differentially expressed protein spots were determined using PDQuest software, with identification on the basis of MALDI-TOF mass spectra. Results: Compared with normal colorectal mucosa, protein abnormal expression of 65 spots varying more than 1.5 times were found in 2-DE gels from colorectal cancer samples (P<0.05); forty-two proteins were up-regulated and 23 were down-regulated; twelve protein spots were identified using mass spectrometry, of which 8 were up-regulated, includimng HSPB1and Annexin A4, while 4 were down-regulated, the results being consistent with Western blot findings. Conclusions: Two-dimensional electrophoresis reference maps for CRC tissues and adjacent normal mucosa (NMC) were established and 12 differentially expressed proteins were identified. Up-regulated HSPB1 and Annexin A4 may play many important roles in the pathogenesis of colorectal cancer.
피부 활성 물질로 넓이 알려져 있는 vitamin C (L-Ascorbic acid)는 콜라겐 생합성 촉진과 강한 항산화 작용으로 피부 항노화 및 주름 개선 효과 있을 뿐만 아니라 멜라닌 세포 활성 억제, 자외선 차단, 상처치유 등의 효능을 갖고 있다. 그러나 물성 면에서 피부 자극과 수분 및 공기, 빛에 불안정하여 쉽게 산화된다는 단점을 갖고 있다 이러한 문제점을 해결하기 위해 많은 연구가 진행되어 왔고, 그 결과 다양한 vitamin C 유도체가 개발되었다. 그러나 아직까지도 vitamin C의 불안정한 물성을 해결하면서 그 자체의 효능을 피부에 적용시키기에는 미흡한 점이 많다는 평을 받고 있다. 본 연구에서는 이러한 vitamin C의 불안정한 물성을 개선시키고 그 효능을 피부에 적용시킬 수 있는 vitamin C 유도체를 개발하기 위해, 피부 친화성이 우수한 스핑고지질류 중 하나인 phytosphingosine을 이용하여 산염기 반응에 의한 vitamin C의 OH기와 phytosphingosine의 -NH$_2$이온 결합시킨 새로운 vitamin C 유도체인 phytosphingosine ascorbate (SP-VC)를 합성하였다. 이렇게 합성된 phytosphingosine ascorbate (SP-VE)는 원소 분석(C58.3 : H9.3 : N2.8 : 029.5) 및 mass spectrosocopy (Maldi TOF-MS), UV/vis spectra (268.5nm), $^1$H NMR, FT-IR, 열분석 (m.p=154$^{\circ}C$), HPLC 등을 통하여 구조 및 물성을 확인하였다 또한 효능면에서는 우선적으로 phytosphingosine ascorbate(SP-VC)의 항균 및 항산화 효과를 확인하였다. 이러한 결과를 토대로 vitamin C와 phytosphingosine의 효능을 동시에 갖으며 불안정한 물성과 자극을 개선시킬 것이라 예상되는 새로운 소재를 합성하였다.

Photoresponsive arylether dendrimers Bis-azo-Gn(3,5) 1a-1c and Bis-azo-Gn(3,4,5) 2a-2c (n = 1-3) with an azobenzene unit at the core and several vinyl groups (3,5-bis(but-3-enyloxy)phenyl groups or 3,4,5-tris(but-3- enyloxy)phenyl groups) at the periphery have been prepared. Their structures and reversible trans-cis isomerization behaviors have been investigated by $^1H$-NMR, $^{13}C$-NMR, MALDI-TOF-Mass, and UV-vis spectra. All six azobenzene-cored dendrimers carried out very fast trans $\rightarrow$ cis photoisomerization on irradiation of 350 nm light and reached to the photostationary state within 180 s. During the dark incubation, slow thermal back reversion from cis to trans form is observed for all six dendrimers and is completed within 3 days for 1a-1c and 1 day for 2a-2c. Isomerization efficiency decreases with increasing generation. However, the initial reaction rates of both trans $\rightarrow$ cis photochemical isomerization and cis $\rightarrow$ trans thermal isomerization increases significantly with increasing generation for dendrimers for 1a-1c but only slightly for 2a-2c.
