Substituted Phenyl 2-Thiophenecarboxylates and Benzoates:Synthesis, NMR Spectra, and Aromaticity Index

  • A series of m-, and p-substituted phenyl 2-thiophenecarboxylates and benzoates was prepared by the reaction of the corresponding acyl chlorides and phenols. Their $^1H$ and $^{13}C$ NMR chemical shifts were analyzed using single substituent parameter (S SP) and dual substituent parameter (DSP) methods. The relative aromaticity index of thiophene was estimated to be 0.92 from the plot of the chemical shift of the carbonyl carbons of the thienoyl esters against chemical shift of the carbonyl carbons of the benzoyl esters.

JS-III-49, a hydroquinone derivative, exerts anti-inflammatory activity by targeting Akt and p38

  • Since previous studies have reported that hydroquinone (HQ) exerted immunosuppressive and anti-inflammatory activity, various HQ derivatives have been synthesized and their biological activities investigated. In this study, we explored the anti-inflammatory activity of JS-III-49, a novel HQ derivative, in macrophage-mediated inflammatory responses. JS-III-49 suppressed the production of the inflammatory mediators nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) and down-regulated the mRNA expression of the inflammatory enzymes cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) as well as the expression of the pro-inflammatory cytokines interleukin-6 (IL-6) and IL-$1{\beta}$ without cytotoxicity in LPS-stimulated RAW264.7 cells. JS-III-49 inhibited nuclear translocation of the $NF-{\kappa}B$ transcription factors p65 and p50 by directly targeting Akt, an upstream kinase of the $NF-{\kappa}B$ pathway, in LPS-stimulated RAW264.7 cells. However, JS-III-49 did not directly inhibit the kinase activities of Src and Syk, which are upstream kinases of Akt, in LPS-stimulated RAW264.7 cells. Moreover, JS-III-49 suppressed the nuclear translocation of c-Fos, one of the components of AP-1, by specifically targeting p38, an upstream mitogen-activated protein kinase (MAPK) in the AP-1 pathway in LPS-stimulated RAW264.7 cells. These results suggest that JS-III-49 plays an anti-inflammatory role in LPS-stimulated macrophages by targeting Akt and p38 in the $NF-{\kappa}B$ and AP-1 pathways, respectively.

SPECTROSCOPIC AND PHOTOMETRIC STUDY OF STARBURST GALAXIES: OPTICAL AND NEAR INFRARED PROPERTIES OF A BLUE COMPACT DWARF GALAXY MRK 49 IN THE VIRGO CLUSTER

  • We present optical and near-infrared imaging and long-slit spectroscopy for the blue compact dwarf galaxy (BCD) Mrk 49 in the Virgo Cluster. The surface brightness distribution analysis shows that Mrk 49 consists of an off-centered blue bright compact core of r = 10" and a red faint outer exponential envelope. The $H_{\alpha}$ image and color difference suggest that these two components have different stellar populations: a high surface brightness population of massive young stars and an underlying low surface brightness population of older stars. The redder near-infrared colors of the inner most region suggest that the near-infrared flux of Mrk 49 originates from evolved massive stars associated with the current star-forming activity. The total apparent magnitude is $B_T\;=\;14.32$ mag and the mean effective surface brightness is ${\mu}_{eff}(B)\;=\;21.56$ mag $arcsec^{-2}$. Long-slit spectroscopy shows that Mrk 49 rotates apparently as a solid body within r = 10" in a plane at position angle 55 degrees with an amplitude of about $20\;km\;sec^{-1}$. The measured radial velocity of Mrk 49 was derived as $1,535\;km\;sec^{-1}$; and the total mass of stars and gases is in the range of 3 to $6\;{\times}\;10^9\;M_{\odot}$. The mass-to-light ratios for the central region of Mrk 49 in I and B band are estimated 1.0 and 0.5, respectively. The upper limit of the dark matter to visible matter ratio seems to be < 5. The oxygen abundance is $12\;+\;\log(O/H)\;=\;8.21\;{\pm}\; 0.1$ which is about one quarter of the solar value while the relative helium abundance appears to be similar to that of the sun.

Investigation of Antimicrobial Activity and Stability of Orixa japonica Thunb. Leaf Extract (상산나무 잎 추출물의 항균활성 및 안정성 조사)

  • The antimicrobial activity of Orixa japonica Thunb. leaf extract towards 13 microorganism strains was evaluated. Both methanol (MEex) and 70% ethanol extracts showed antimicrobial activity towards Streptococcus mutans, Bacillus cereus, Staphylococcus aureus, and Pseudomonas aeruginosa. MEex showed a higher antimicrobial activity than the 70% ethanol extract. In addition, the dichloromethane fraction (DCMfr) of the MEex also had an antimicrobial effect against the microorganisms examined. The minimum inhibitory concentrations (MICs) towards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 49.22, 24.61, 49.22, and 49.22 mg/mL, respectively. In contrast, the MICs of the DCMfr tpwards S. mutans, B. cereus, S. aureus, and P. aeruginosa were 3.31, 0.21, 1.7, and 1.7 mg/mL, respectively. The MEex antimicrobial activity was not affected by a 3 h exposures to pH in the range of 3-11 or by temperatures were maintained between $80^{\circ}C-100^{\circ}C$ for 6 h. However, the MEex antimicrobial activity decreased at a heat treatment of $121^{\circ}C$ 1 h.

Inactivation of Foodborne Pathogenic Bacteria in Corn Silk Tea Using a Microwave Plasma Sterilization System (Microwave Plasma Sterilization System 처리가 옥수수수염 차의 저장 중 Escherichia coli 및 Listeria monocytogens의 생육 저해에 미치는 영향)

  • Inactivation of foodborne pathogenic bacteria in corn silk tea was evaluated using a microwave plasma sterilization system (MPSS). Corn silk tea was inoculated with Escherichia coli and Listeria monocytogenes, treated with an MPSS treatment, and stored at 25$^{\circ}C$ for 12 days. The one, two, and three cycles of treatment with MPSS reduced the population of E. coli by 1.14, 2.49, and 5.72 log CFU/mL, respectively, compared to that of the control. In the case of L. monocytogenes, one, two, and three cycles of MPSS treatment reduced the population by 1.93, 4.49, and 6.62 log CFU/mL, respectively. Both E. coli and L. monocytogenes were eliminated within four cycles of treatment with MPSS, and even after 12 days of storage, the bacteria were not detected. Total polyphenol content in the corn silk tea did not change much among treatments, and turbidity of the corn silk tea improved following four cycles of MPSS treatment. These results suggest that MPSS treatment can be useful for improving the microbial safety and quality of corn silk tea during storage.

Effect of Insulin-like Growth Factor-I and Cumulus Cells on In Vitro Maturation in Porcine Oocytes (돼지난자의 체외성숙시 Insulin-like Growth Factor-과 난구세포의 영향)

  • The effects of insulin-like growth factor-I (IGF-I) and cumulus cells during in vitro maturation in porcine oocytes were examined. When follicular oocytes were cultured in medium with different concentrations of IGF-I, maturation rates were 60, 61 and 62 and 72% for 0, 15 and l0ng/$m\ell$ IGF-I. In medium with 10ng/$m\ell$ IGF-I, maturation rates were not significantly difference between oocytes with (68%) and without (52%) cumulus cells during the culture. In medium with-out IGF-I, however, the maturation rates in oocytes with cumulus cells (63%) was significantly (P<0.05) higher than oocytes without cumulus cells (32%). On the other hand, when IGF-I was added for first 24 h period or later 24 h period of culture, maturation rates were higher in oocytes with (61 and 49%) that than without (49 and 45%) cumulus cells. In experiment used medium without fetal calf serum (FCS) and porcine follicular fluid (PFF), the maturation rates in oocytes with cumulus cells for 48 h (48 and 67%) or first 24 h (46 and 63%) period after culture were significantly (P<0.01) higher than in oocytes without cumulus cells (16 and 18%) in the presence or absence of IGF-I. These results indicated that cumulus cells is essential on maturation in vitro in porcine oocytes, but IGF-I can promote oocytes maturation of oocytes without cumulus cells in medium with FCS and PFF.

Estimation of Glomerular Filtration Rate from Plasma Creatinine and Height in Children (소아에서 신장과 혈장 Creatinine 농도를 이용한 사구체여관율 측정)

  • In clinical practice, creatinine clearance(Ccr) remains the most commonly used laboratory assessment of glomerular function despite methodological and technical problems of urine collection. Schwartz et al. in 1976, reported that an accurate estimate of glomerular filtration rate(GFR) could be obtained from the simple determinations of plasma creatinine(Pcr) and body length(L) : GFR($m{\ell}/min/1.73m^2$=k L(cm)/Pcr(mg/$100m{\ell}$), (k=constant). The subject of this study were ill children admitted to our pediatric department from July, 1985 to June, 1987 and they were divided into three groups; group I, from 1 to 5 years old, group II, from 6 to 10 years old, group III. from 11 to 15 years old. The results were as following ; 1) Measured creatinine clearance($Ccr_M$, $m{\ell}/min/1.73m^2$) were $109.73{\pm}9.97$ in group I, $108.26{\pm}9.02$ in group II, $96.20{\pm}4.72$ in group III and $105.48{\pm}5.23$ in all age group. 2) Measured k($k_M$) obtained from $Ccr_M=k$ Ht/Pcr were $0.49{\pm}0.03$ in group I, $0.48{\pm}0.02$ in group II, $0.43{\pm}0.02$ in group III, and $0.47{\pm}0.02$ in all age group.(Ht ; height) 3) Linear equations and correlation coefficients between Ht/Pcr(x) and Ccr(y) were y=0.822x-65.63(r=0.99) in group I, y=0.61x-23.46(r=0.72) in group II, y=0.18x+54.44(r=0.54) in group III and y=0.58x-22.13(r=0.81) in all age group. 4) $Ccr_E$ was again estiamted from linear equations between Ht/Pcr and $Ccr_M$ and $k_E$ was calculated with Ht/Pcr and $Ccr_E$ were $0.48{\pm}0.01$ in group I, $0.49{\pm}0.01$in group II, $0.43{\pm}0.01$ in group III and $0.47{\pm}0.00$ in all age group. 5) Consistant values of $k_E$ and $k_M$ were highly significant as 95~97.5% in group I and II, 90~95% in group III and 97.5~99% in all age group. In summary, we could estimate GFR with height, plasma creatinine and measured k($k_M$) according to the age in easy and rapid way.

An Evaluation of Building Effect in 2-Dimensional Inundation Analysis Using GIS (GIS를 활용한 2차원 침수해석에서의 건물영향 분석)

  • In this study, 2-dimensional inundation analysis for Taehwa watershed in Ulsan metropolitan city was conducted to analyze flow behaviors, inundation depth and inundation stage, considering the building effect. Lidar having the interval of 1 m was employed to generate topographic data with 10m interval, and building data extracted from digital map was combined with the constructed topographic data for 2-dimensional inundation analysis. A few scenarios were constructed for the analysis to provide an effective and accurate inundation analysis method through analyzing the results. The disagreement based on the areas of inundation showed over 10% between the cases with and without consideration of building effect. The maximum inundation depth without considering the effects of buildings was 0.29m higher than that with considering the building effects. On the contrary, the maximum inundation stage with consideration of building effects was 0.49m higher than that without consideration of building effects.

Localization of Quantitative Trait Loci for Bone Mineral Density on Chromosome 13 in the Mongolian Population

  • Although the genetic basis for bone mineral density (BMD) has been studied by many groups so far, genes responsible for this complex trait has not been completely revealed. In order to localize quantitative trait loci (QTLs) for BMD variation in Asian population, the study was designed using a group of Mongolian population, a genetically closed population with a homogeneous lifestyle. BMD was measured at the left and right wrists and ankles using DEXA in 1,082 participants from 142 families. Genotyping of 13 polymorphic microsatellite markers on chromosome 13 (average spacing 8-9 cM) and two-point and multipoint linkage analysis were performed. In two-point linkage analysis, we identified two markers, D13S175 (6.03 cM) and D13S265 (68.73 cM) that had LOD scores greater than 1 for left ankle (LOD=2.09, LOD=1.49, respectively). We also found a marker D13S175 (6.03 cM) with a high LOD for left wrist (LOD=1.49) and the markers D13S265 (68.73 cM) and D13S217 (17.21 cM) for the right wrist (LOD= 1.82, LOD= 1.62, respectively). Among these significant marker regions, only two regions at 17 cM (13p11) and 65 cM (13q21) for the right wrist overlapped with major QTLs reported in following multipoint linkage analysis (LOD= 1.7549, LOD=1.4462, respectively). This study provides the possible evidence of the presence of QTLs affecting right wrist BMD in Mongolian populations on 13p11 and 13q21. Modest evidence was also found for genes affecting left ankle and left wrist BMD on 13p13.

In Vitro Antineoplastic Effects of Chitosan Hydrolysates on Various Tumor Cell Lines (키토산 가수분해물의 In Vitro 항종양성)

  • In this study, the antineoplastic effects of chitosan hydrolysates were assessed. The chitosan hydrolysates showed no cytotoxicity in in vitro trials using the normal cell line, Vero E6(Africa green monkey kidney cells). The $IC_{50}$ value of the chitosan hydrolysates on Vero E6 was 1,107.95 ${\mu}g/m{\ell}$. The hydrolysates exhibited in vitro antineoplastic activity in five human tumor (lung carcinoma, bladder carcinoma, colon carcinoma, stomach carcinoma, breast carcinoma) cell lines. The $IC_{50}$ values of the hydrolysates on A549, J82, SNU-C4, SNU-1, and ZR75-1 cells were 421.06, 417.99, 445.54, 380.65 and 460.49 ${\mu}g/m{\ell}$, respectively.
