Acute Toxicity Assessment of New Algicides of Thiazolidinediones Derivatives, TD53 and TD49, Using Ulva pertusa Kjellman

  • Objectives : This study was aimed to examine the acute toxicity assessment of two new algicides, thiazolidinediones derivatives (TD53 and TD49), which were synthesized to selectively control red tide, to the marine ecosystem. Methods : The assessment employed by a new method using Ulva pertusa Kjellman which has been recently accepted as a standard method of ISO. The toxicity was assessed by calculating the $EC_{50}$ (Effective Concentration of 50%), NOEC (No Observed Effect Concentration) and PNEC (Predicted No Effect Concentration) using acute toxicity data obtained from exposure experiments. $EC_{50}$ value of TD49 and TD53 was examined by 96-hrs exposure together with Solutol as a TD49 dispersing agent and DMSO as a TD53 solvent. Results : $EC_{50}$ value of TD53 was $1.65\;{\mu}M$. From the results, values of NOEC and PNEC were calculated to be $0.63\;{\mu}M$ and 1.65 nM, respectively. DMSO under the range of $0{\sim}10\;{\mu}M$, which is same solvent concentration used in examining TD53, showed no toxic effect. $EC_{50}$ value of TD49 was $0.18\;{\mu}M$ and that of Solutol was $1.70\;{\mu}M$. NOEC and PNEC of TD49 were $0.08\;{\mu}M$ and 0.18 nM, respectively and those for Solutol were $1.25\;{\mu}M$ and 1.25 nM, respectively. Conclusions : From the values of NOEC, PNEC of TD53 and TD49, TD49 showed 9 times stronger toxicity than TD53. On the other hand, DMSO showed no toxicity on the Ulva pertusa Kjellman, but Solutol was found to be a considerable toxicity by itself.

  • Tetrahydroisoquinoline (THI) compounds have various pharmacological actions in the cardiovascular system. Recently, we have synthesized 1-${\alpha}$-naphthylmethyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline, YS 49. In the present study, we evaluated the effect of YS-49 on positive inotropic and chronotropic action using isolated rat heart and on blood pressure and heart rate using anesthesized rabbit. Vasodilating action was also assessed in isolated rat thoracic aorta. YS 49, concentration-dependently relaxed rat aorta precontracted with phenylephrine (PE, 0.3 ${\mu}$M) and high potassium (high K$\^$+/, 65.4 mM). The 50% inhibitory concentration (IC$\sub$50/) of YS 49 in PE-induced and high K$\^$+/-induded contraction was 5.36 ${\mu}$M and 2.52 ${\mu}$M, respectively. In isolated rat atria, YS 49 increased both heart rate and force, and in anesthesized rabbit it decreased blood pressure but increased heart rate. In addition, to know the mechanism of action of the compound, propranolol, nonselective ${\beta}$-antagonist, and phentolamine, ${\alpha}$-blocker, were used. Furthermore, a comparison with the effect of higenamine, trimetoquinol on the vasodilating action in rat thoracic aorta was also made. The action of YS 49 was inhibited by the presence of propranolo, not pentolamine. These results indicate that cardiotonic and vasodilatory action of YS 49 is attributable, at least in part, for ${\beta}$-receptor stimulation.

Optimization of Hybrid Process of(Chemical Coagulation, Fenton Oxidation and Ceramic Membrane Filtration) for the Treatment of Reactive Dye Solutions (반응성 염료폐수 처리를 위한 화학응집, 펜톤산화, 세라믹 분리막 복합공정의 최적화)

  • This study investigated the effects of hybrid process(chemical coagulation, Fenton oxidation and ceramic UF(ultrafiltration)) on COD and color removals of commercial reactive dyestuffs. In the case of chemical coagulation, the optimal concentrations of $Fe^{3+}$ coagulant for COD and color removals of RB49(reactive blue 49) and RY84(reactive yellow 84) were determined according to the different coagulant dose at the optimal pH. They were 2.78 mM(pH 7) in RB49 and 1.85 mM(pH 6) in RY84, respectively. In the case of Fenton oxidation, the optimal concentrations of $Fe^{3+}\;and\;H_2O_2$ were obtained. Optimal $[Fe^{2+}]:[H_2O_2]$ molar ratio of COD and color removals of RB49 and RY84 were 4.41:5.73 mM and 1.15:0.81 mM, respectively. In the case of ceramic UF, the flux and rejection of supernatant after Fenton oxidation were investigated. After ceramic UF for 9 hr, the average flux of RB49 and RY84 solutions were $53.4L/m^2hr\;and\;67.4L/m^2hr$ at 1 bar, respectively. In addition, the permeate flux increased and the average flux recovery were 98.5-99.9%(RB49) and 91.0-97.3%(RY84) according to adopting off-line cleaning(5% $H_2SO_4$). Finally, COD and color removals were 91.6-95.7% and 99.8% by hybrid process, respectively.

  • Let (M,F) and (M',F') be two foliated Riemannian manifolds with M compact. If the transversal Ricci curvature of F is nonnegative and the transversal sectional curvature of F' is nonpositive, then any transversally harmonic map ${\phi}:(M,F){\rightarrow}(M^{\prime},F^{\prime})$ is transversally totally geodesic. In addition, if the transversal Ricci curvature is positive at some point, then ${\phi}$ is transversally constant.

A Study on Prediction of Moment Developed in Bottom of Foundations between Pile and Heterogeneous Soils (말뚝기초와 이질지반 경계부 기초저판에서의 발생모멘트 예측에 관한 연구)

  • To reinforce bearing capacity-changed section or different foundation in the same building, empirical or simple tools have been used. To solve this problem, an analytical solution which can evaluate and reinforce the stability of foundation is introduced. To suggest a clue for the problems, current foundation reinforcing method is studied through recent literature studies and the structural analyses of foundation slab are performed on the pile foundation of 49$m^2$, 59$m^2$ and 84$m^2$ I type apartments in 15 story building. The analyses are conducted with SAP 2000, a computer program for ordinary structural analysis. To predict the moments of slab by ground non-uniformity, the structural analysis results for the foundation slab of 3 types 15 story apartment buildings in 49$m^2$, 59$m^2$ and 84$m^2$ I type on non-uniformity ground are shown in the diagrams.

Some Factors Affecting Bone Mineral Density of Korean Rural Women (농촌 성인 여성들의 골밀도에 영향을 미치는 요인 분석 연구)

  • The factors affecting bone mineral density (BMD) of 103 rural women aged 30 to 76 years were investigated. Data for food and nutrient intake was obtained by 24-hour recall method. BMD of lumbar spine(L1-L4), femoral neck, ward's triangle and trochanter was measured by XR-series x-ray bone densitometer. Serum was collected and analyzed for total Ca, P and ionized Ca (Ca++)content. Relationship between the factors and BMDs was analyzed by Pearson's correlation coefficient(r) and multiple regression analysis. The results are summarized as follows. 50% of the subjects under 50 years of age($\leq$49 yr group) and 86.4% of the subjects from 50 up($\geq$50yr group) were classified as osteopenia or osteoporosis. Mean body weight, height and BMI were 153.1cm, 59.1kg and 25.0kg/$m^2$ in $\leq$49 yr group and 151.9cm, 55.9kg, and 24.2kg/$m^2$ in $\geq$50 yr group. BMDs of lumbar spines and femurs ranged from 0.84 to 1.05g/$m^2$ and from 0.67 to 1.16g/$m^2$ in $\leq$49 yr group, and ranged from 0.67 to 0.85kg/$m^2$ and from 0.68 to 0.44g/$m^2$ in $\geq$50 yr group, respectively. On the whole, the BMDs were reduced to 83.8 to 94.2% of peck bone mass in $\leq$49 yr group and 55.2 to 78.9% of those in $\geq$50 yr group. Mean daily intake of Ca was much less than the Korean RDA, \67.2% in $\leq$49 yr group and 62.3% in $\geq$50 yr group. The average concentration of total Ca, P and Ca++ in serum were within normal range in both age group. Both age and height were significantly related with BMD in both age group but the relationship tended to be stronger in $\geq$50 yr group than in $\leq$49 yr group. Body weight was also a potent determinant of BMD only in 50 yr group. In $\leq$49 yr group, total food intake was positively related with BMDs of ward's triangle, L1 and L2 and intake of cereals and grain products, sugars and sweets, milk and dairy products was positively related with BMDs measured in this study. On the contrary, intake of eggs, oil and fats were positively related with a few BMDs in 50 yr group. The BMDs were positively affected by intake of energy, protein, carbohydrate, Ca, P and Fe in $\leq$49 yr group and those of protein, fat Ca, P, vitamin B1, vitamin B2 and vitamin C in $\geq$50 yr group. It was noteworthy that serum Ca++ concentration was positively related with BMDs of lumbar spine in boty age groups. According to multiple regression analysis, the four factors, age, body weight, height and BMI additionally accounted for 21% of the variance in BMD of trochanter in $\leq$49 yr group and only two factors, age and C a intake accounted for 38% of that of femoral neck in $\geq$50 yr group. Further investigation is necessary to make sure of the relations between BMD and serum Ca++ level.

Acute Toxicity Assessment of New Algicides, Thiazolidinedione Derivative (TD49) to Marine Ecosystem (신규 살조물질인 Thiazolidinedione 유도체 (TD49)의 해양생태계에 대한 급성독성평가)

  • A thiazolidinedione derivative, TD49 with the highly selective algicide to red tide was newly synthesized and its acute toxicity was examined in order to evaluate the effect on aquatic ecosystems of coast. Major three species having important role in the food chain of marine ecosystem, such as Skeletonema costatum of microalgae, Daphnia magna of crustacea, Paralichthys olivaceus of flatfish fingerling were employed for the acute toxicity assessment. $EC_50$ or $LC_50$ as the assessment criterion was investigated to each specie, and NOEC (No Observed Effect Concentration) and PNEC (Predicted No Effect Concentration) from most sensitive specie to toxicity of TD49 were further calculated. $EC_50$ of S. costatum in 96-hour, $EC_50$ of D. magna in 48-hour, and $LC_50$ of P. olivaceus in 72-hour for TD49 were $0.34\;{\mu}M$, $0.68\;{\mu}M$, and $0.58\;{\mu}M$, respectively. NOEC from the results of S. costatum was estimated to be $0.20\;{\mu}M$ and PNEC was estimated as 3.40 nM by applying factor value of 100 to $EC_50$ $0.34\;{\mu}M$ of S. costatum. In addition, it was revealed that Solutol used as the dispersing agent of TD49 had very little toxic influence under the concentration range of $0{\sim}0.4\;{\mu}M$ used in TD49 toxicity experiment. Although the estimated concentration of TD49 that will be sprayed onto the coastal field for the algicide is higher than NOEC value, it is considered that the spraying concentration would not be a considerable problem due to a dilution effect by tide at the opened coast.

Strain Improvement by Interspecific Fusion of Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 producing $\alpha$-Amylase Inhibitor ($\alpha$-Amylase 저해제 생성균 Streptomyces actuosus DMCJ-49와 Streptomyces minoensis DMCJ-144의 종간 융합에 의한 균주 개량)

  • Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 produce the .alpha.-amylase inhibitor. Inerspecific protoplast fusion technique was used to increase the productivity of .alpha.-amylase inhibitor. Four auxotrophic mutants were obtained respectively from two strains by N-methyl-N'-nitor-N-nitrosoguanidine(3mg/ml) treatment. The optimum conditions for the protoplast formation of Streptomyces actuosus DMCJ-49 ade was as follows; 1.2% w/v of glycine, 3mg/ml of lysozyme, and 30 min of lysozyme treatment followed by 36 hr. incubation in the protop-last formation medium. In case of DMCJ-144-his those were 1.2%w/v, 3 mg/ml, 30 minutes and 60 hours, respectively. Regeneration was accomplished with hypertonic soft agar medium that contained 0.4M sucrose, 20mM CaCl$_2$, 50 mM MgCl$_2$ and low levels of phosphate. Fusion of protoplasts carrying different auxotrophic markers was achieved by treatment with polyethylene glycol. The optimum concentration of polyethylene glycol 1450 for the production of recombinants was 40%w/v. When the protoplasts was treated with 40% polyethylene glycol for 30 minutes, the frequency of recombinants was 6.5$\times$$10^{-3}$ and the $\alpha$-amylase inhibition activity of $ade^-his^-$ No. 4, which is the fusant with the most improved activity increased from 33 to 125 I.U./ml.

Rapamycin-resistant and torin-sensitive mTOR signaling promotes the survival and proliferation of leukemic cells

  • The serine/threonine kinase mTOR is essential for the phosphoinositide 3-kinases (PI3K) signaling pathway, and regulates the development and function of immune cells. Aberrant activation of mTOR signaling pathway is associated with many cancers including leukemia. Here, we report the contributions of mTOR signaling to growth of human leukemic cell lines and mouse T-cell acute leukemia (T-ALL) cells. Torin, an ATP-competitive mTOR inhibitor, was found to have both cytotoxic and cytostatic effects on U-937, THP-1, and RPMI-8226 cells, but not on Jurkat or K-562 cells. All cells were relatively resistant to rapamycin even with suppressed activity of mTOR complex 1. Growth of T-ALL cells induced by Notch1 was profoundly affected by torin partially due to increased expression of Bcl2l11 and Bbc3. Of note, activation of Akt or knockdown of FoxO1 mitigated the effect of mTOR inhibition on T-ALL cells. Our data provide insight on the effect of mTOR inhibitors on the survival and proliferation of leukemic cells, thus further improving our understanding on cell-context-dependent impacts of mTOR signaling.
