$aroP^{-}$변이가 E.coli에서 트립토판 방출에 미치는 영향 (Effects of $aroP^{-}$ mutation on the tryptophan excretion in escherichia coli)

  • 증폭된 재조합 trp operon의 발현을 위한 숙주박테리아 개발의 일환으로 숙수 E. coli에 $aroP^{-}$ 변이를 도입하였다. $aroP^{-}$ 변이의 유도에는 trans po son Tn10을 사용하였으며 P1Kc파아지를 이용하여 숙주박테리아에 형질도입하였다. General aromatic amino acid transport system이 결여된 $aroP^{-}$ 변이주는 $\beta$-thienylalanine ($(2{\times}10^{-4}M)$). p-fluor-phenylalanine ($(2{\times}10^{-4}M)$) 그리고 5-methyltryptophan에 저항성을 가졌다. $aroP^{-}$ 변이주는 $aroP^{-}$ 야생주에 비해서 〔$[^3H]$-tryptophan uptake가 상당히 감소하였다. 또한 NaN, ($(2{\times}10^{-4}M)$)를 처리하였을 때의 ($[^3H]$)-tryptophan uptake 비율은 aroP 변이주가 $aroP^{-}$야생주보다 덜 감소하였다. E. coli $trpR^{-ts}/ColE_1 -trp^+$ 균주에 aroP 형질을 도입하였을 때 트립토판 방출이 $aroP^{-}$ 야생주에 비해서 4 배나 증가하였다.

Microbial Conversion of Ginsenoside from the Extract of Korean Red Ginseng (Panax ginseng) by Lactobacillus sp.

  • Thirty-four strains of Lactobacillus species were isolated from soil and eight of these isolates (M1-4 and P1-4) were capable of growing on red ginseng agar. The M1 and P2 strains were determined to be L. plantarum and other strains (M2, M3, M4, P1, P3 and P4) were determined to be L. brevis. Fermentation of red ginseng extract (RGE) with strains M1, M2, P2 and P4 resulted in a low level of total carbohydrate content (174.3, 170.0, 158.8 and 164.8 mg/mL, respectively). RGE fermented by M3 showed a higher level of uronic acid than the control. The polyphenol levels in RGE fermented by M1, P1 and P2 (964.9, 941.7 and $965.3\;{\mu}g/mL$, respectively) were higher than the control ($936.8\;{\mu}g/mL$). Total saponin contents in fermented RGE (except M1) were higher than the control. RGE fermented by M2 and M3 had the highest levels of total ginsenosides (31.7 and 32.7 mg/mL, respectively). The levels of the ginsenoside Rg3 increased from 2.6 mg/mL (control) to 3.0 mg/mL (M2) or 3.1 mg/mL (M3). RGE fermented by M2 and M3 also had the highest levels of Rg5+Rk1 (7.7 and 8.3 mg/mL, respectively). Metabolite contents of ginsenoside (sum of CK, Rh1, Rg5, Rk1, Rg3 and Rg2) of M2 (13.0 mg/mL) and M3 (13.9 mg/mL) were also at a high level among the fermented RGE. Protopanaxadiol and protopanaxatriol content of ginsenoside of M2 (10.9 and 5.4 mg/mL, respectively) and M3 (11.0 and 5.7 mg/mL, respectively) were at higher levels than other fermented RGE.

  • We study the tensor product algebra Pk(x1) ⊗ Pk(x2) ⊗ ⋯ ⊗ Pk(xm), where Pk(x) is the partition algebra defined by Jones and Martin. We discuss the centralizer of this algebra and corresponding Schur-Weyl dualities and also index the inequivalent irreducible representations of the algebra Pk(x1) ⊗ Pk(x2) ⊗ ⋯ ⊗ Pk(xm) and compute their dimensions in the semisimple case. In addition, we describe the Bratteli diagrams and branching rules. Along with that, we have also constructed the RS correspondence for the tensor product of m-partition algebras which gives the bijection between the set of tensor product of m-partition diagram of Pk(n1) ⊗ Pk(n2) ⊗ ⋯ ⊗ Pk(nm) and the pairs of m-vacillating tableaux of shape [λ] ∈ Γkm, Γkm = {[λ] = (λ1, λ2, …, λm)|λi ∈ Γk, i ∈ {1, 2, …, m}} where Γk = {λi ⊢ t|0 ≤ t ≤ k}. 비타민 $B_2$ 부족이 운동중 연료의 이용과 회복에 미치는 영향 (The effect of Vitamin $B_2$ Deficiency on the Utilization and Recuperation of Fuel in Exercising Rats)

  • The purpose of this study was to investigate the effect of vitamin B2 deficiency on the utilization and recuperation of fuel in exercising rats. Thirthy six rats were fed either a vitamin B2 deficient diet (-B2) or a control diet (+B2) for 3 weeks and then subdivided into 3 groups : non-exercise group(NE), exercise group (ES), exercise and recuperation group (ER). ES group were exercised on treadmill (10$^{\circ}$, 0.5-0.8km/h) for 2 hours and ER group were recuperated three days with the respective diet after exercise. Glucose (GLU), glycogen(GLY), protein(PRO), triglyceride(TG) and free fatty acid(FFA) were compared in plasma(P), liver(L) and skeletal muscle(M) of rats. Compared to + B2 rats, in NE group, the level of P-GLU and L-GLU of -B2 rats was lower, L-PRO and M-GLY was higher and there were no differences in P-PRO, P-FFA, L-TG and M-TG. In ES group, the level of P-GLU was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLU, L-TG, M-GLY, M-TG and M-PRO. In ER groups, the level of P-GLU and L-TG was lower, P-FFA was higher and there were no differences in P-PRO, P-TG, L-GLY, L-PRO, M-GLY, M-TG and M-PRO. These results suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise suggest that a vitamin B2 deficiency may impair the utilization of stored fuel during exercise and may lead a sluggish recuperation related to fuel stores after exercise.

아세트산염 배지에서의 검정곰팡이(Aspergillus niger)의 포자형성에 관한 연구 (Studies on the Spore Formation of Aspergillus niger in Potassium Acetate Medium)

  • 1. 검정곰팡이 (Aspergillus niger)를 실험균주로 하여 동조적으로 액침배양한 결과 포자의 발아로 부터 균사의 생장, 생식기관의 성숙 및 경자까지의 분화를 재현시킬 수 있었으며 그 다음의 아세트산염 배지에서 농도와 pH가 적절할 때 포자의 형성을 볼 수 있었다. 2. pH 5.5, 6.0, 6.5 및 7.0로 조절하고 각각 농도를 대조구, 20mM, 40mM, 80mM 및 160mM로 조절한 아세트산염 배지에서 검정곰팡이를 $28^{\circ}C$의 온도로 4일간 진탕배양하면서 균체량과 pH변화, 포자개시 시간을 측정하였다. 아세트산염 배지에서 적응이 되면 균체량은 증가하였으며 성장이 진행되면서 PH가 증가하다가 포자가 형성되면 pH는 감소하였다. pH 7.0의 아세트산염 배지에서는 균이 적응할 수 없고 pH 증가가 너무 지나치게 되어 균체량은 점차 감소하였다. 또 포자형성개시 시간은 pH 5.5의 아세트산염 배지에서는 그 농도가 20mM일 때 1일, 40mM일 때 2일, 80mM일 때 3일, 160mM일 때 4일이 소요되었으며 pH 6.0의 아세트산염 배지에서는 그 농도가 40mM일 때 2일만에, 80mM일 때 1일만에, 160mM일 때 4일만에 포자가 형성되었다. 또 pH 6.5의 아세트산염 배지에서는 80mM의 농도일 때 3일만에, 160mM일 때 4일만에 포자가 형성되었고 pH 7.0의 아세트산염 배지에서는 포자가 형성되지 않았다. 모든 pH의 아세트산염 배지에서 대조구에서는 포자가 형성되지 않았다.

Synthesis of Two Nitro Analogs of Tranylcypromine: Relations of Aromatic Substitution of Nitro Groups to MAO-Inhibitory Activity

  • Two new nitro analogs of tranylcypromine, (E)-2-(p-nitrophenyl)cyclopropylamine ((E)-p-NTCP) and (E)-2-(m-nitrophenyl)cyclopropylamine ((E)-m-NTCP) were synthesized in order to examine the effect of aromatic nitro substitution on the MAO-inhibitory activity of 2-phenylcyclopropylamines. The compounds were obtained by treating t-butyl (E)-2-(p-nitrophenyl) cyclopropanecarbamate and t-butyl (E)-2-(m-nitrophenyl)cyclopropanecarbamate with p-toluenesulfonic acid in $CH_3$CN. Inhibitions of rat brain mitochondrial MAO-A and B by the compounds were examined using serotonin and benzylamine as the substrate at both in vitro and ex vivo levels. It was found from in vitro measurements that (E)-p-NTCP at $6.0{\times}10^{-5}M$ elicited merely 22.5% inhibition against MAO-B without any effect on MAO-A. In contrast, (E)-m-NTCP showed fair degrees of inhibitions of MAO-A and B with $IC_{50}$ values, $2.5{\times}10^{-7}M\;and\;1.4{\times}10^{-6}M$, respectively. It was also noted from (E)-m-NTCP that m-nitro substitution caused a shift of selectivity of the inhibition toward MAO-A. According to ex vivo measurements at 1.5, 3, 6, and 12 hr following the administration of a dose of 0.015 mmol/kg, i.p. to the rats, the inhibition percents of MAO-A by (E)-m-NTCP were 58.6, 63.7 63.6, and 46.6%, slightly lower than those observed by tranylcypromine. Whereas, (E)-m-NTCP at the same dose level did not show significant inhibitions against both MAO-A and MAO-B. Possible reasons for the difference in potencies between (E)-m-NTCP and (E)-p-NTCP were sought in relation to differing electron withdrawing effects of m- and p-substituents which will influence electron density of the side chain amino functions and the partitions.

Furyl furamide 가 여름철 김치세균에 미치는 영향을 보기 위하여 두가지 여름김치에서 분리(分離)한 세균 Bacillus group 과 Lactic acid bacteria중 Bac. megaterium, Bac. subtilis 그리고 Bac. licheniformis등이 발효 초기(初期)의 젓갈김치에 우세 하였고 국물김치에서는 Lact. plantarum 과 Lact. buhneri등이 시종(始終) 매우 우세하였다. 2. 김치에서 분리(分離)된 Bacillus group 은 10 p.p.m. 이하(以下)의 AF-2 저농도(低濃度)에서 대단히 민감(敏感)하게 억제되었으나 Lactic acid bacteria의 group은 50 p.p.m.의 고농도(高濃度)에서도 저항(抵抗)하고 있었다. 3. 김치의 AF-2 첨가허용농도(添加許容濃度)는 최고(最高) 10 p.p.m.을 넘지 않아야 할 것이다. 4. 김치에 첨가(添加)된 고농도(高濃度)(50 p.p.m.)의 AF-2는 김치 발효를 heterofermentation 에서 Homofermentation으로 변화(變化)시키는 경향이 있었다. 5. AF-2 10 p.p.m. 농도(濃度)의 김치에서 bacterial flora는 약간 영향(影響)을 받고 있었으나, 50 p.p.m.의 고농도(高濃度)에서도 하절(夏節)김치의 산패(酸敗)는 억제할수가 없었다. 6. $23^{\circ}{\sim}25^{\circ}C$ 상태의 김치발효액중(醱酵液中)의 AF-2 소장(消長)은 산성(酸性)에서 비교적(比較的) 안전(安全)하다고 하나 4 일후(日後)에 약(約) 25%가 감소(減少)되고 pH 변화곡선(變化曲線)과 거의 평행(平行)한 변화(變化)를 보였다.

SOME CLASSES OF REPEATED-ROOT CONSTACYCLIC CODES OVER 𝔽pm+u𝔽pm+u2𝔽pm

  • Constacyclic codes of length $p^s$ over $R=\mathbb{F}_{p^m}+u\mathbb{F}_{p^m}+u^2\mathbb{F}_{p^m}$ are precisely the ideals of the ring $\frac{R[x]}{<x^{p^s}-1>}$. In this paper, we investigate constacyclic codes of length $p^s$ over R. The units of the ring R are of the forms ${\gamma}$, ${\alpha}+u{\beta}$, ${\alpha}+u{\beta}+u^2{\gamma}$ and ${\alpha}+u^2{\gamma}$, where ${\alpha}$, ${\beta}$ and ${\gamma}$ are nonzero elements of $\mathbb{F}_{p^m}$. We obtain the structures and Hamming distances of all (${\alpha}+u{\beta}$)-constacyclic codes and (${\alpha}+u{\beta}+u^2{\gamma}$)-constacyclic codes of length $p^s$ over R. Furthermore, we classify all cyclic codes of length $p^s$ over R, and by using the ring isomorphism we characterize ${\gamma}$-constacyclic codes of length $p^s$ over R.

  • We define a projective representation $M_1{^{\rightarrow}_{\rightarrow}}M_2{\rightarrow}M_3$ of a quiver $Q={\bullet}{^{\rightarrow}_{\rightarrow}}{\bullet}{\rightarrow}{\bullet}$ and consider their properties. Then we show that any projective representation $M_1{^{\rightarrow}_{\rightarrow}}M_2{\rightarrow}M_3$ of a quiver $Q={\bullet}{^{\rightarrow}_{\rightarrow}}{\bullet}{\rightarrow}{\bullet}$ is isomorphic to the quotient of a direct sum of projective representations $0{^{\rightarrow}_{\rightarrow}}0{\rightarrow}P,\;0{^{\rightarrow}_{\rightarrow}}P{\rightarrow\limits^{id}}P$ and $P{^{\rightarrow}_{\rightarrow}}^{e1}_{e2}P{\oplus}P{\rightarrow\limits^{id_{P{\oplus}P}}}P{\oplus}P$, where $e_1(a)=(a,0)$ and $e_2(a)=(0,a)$.

고추내 함유 Pectinesterase, Polygalacturonase, Lipoxygenase와 Peroxidase의 특성에 관한 연구 (A Study of Pectinesterase, Polygalacturonase, Lipoxygenase and Peroxidase in Hot Pepper)

  • This study was carried out 19 investigate the characteristics of pectinesterase (PE), polygalacturonase (PG), lipoxygenase(LOX) and peroxidase (POD) in hot pepper to know the effect of hot pepper on food quality during food processing and storage. The results were as follows : 1. The optimum pH of PE was pH 7.5 and the activity of PE below pH 5.5 was revealed scarcely, The concentration of NaCl and $CaCl_2$ that showed the highest activity of PE were 0.2M and 0.05M, respectively. 2. The optimum pH of PG was pH 6.0 and the activity of PG in acidity was higher than that in alkalinity. The activity of PG was maximum at 0.3M NaCl and 0.2mM $CaCl_2$. Above the concentration of 0.5M NaCl and 0.5M $CaCl_2$, the activity of PG was lower than that of PG not adding these salts 3. The optimum pH of LOX was pH 7.0 and pH 8.5. 4. The optimum pH of POD was pH 6.0 and the activity of POD was higher in weak acidity and neutrality than in alkalinity. POD activity was slightly decreased by the increase of NaCl and $CaCl_2$ concentration.
