A Study on the 2-D distribution of Dynamic Poisson's Ratio using 3-C Geophones (3성분 지오폰을 이용한 동포아송비의 2차원 분포 연구)

  • In order to acquire 3 components data which has the good signal to noise ratio with only one shot, 3-C geophones were used, As a result, the vertical component showed the distinct first arrival of P-wave, and the horizontal component was improved the signal to noise ratio of S-wave, while was attenuated P-wave. The 2-D Poisson's ratio section was computed from P- and S-wave cell velocities included velocity tomograms of the P- and S-waves. The Poisson's ratio values were computed in the range of $0.2{\~}0.3$. With one shot, we can obtain 2-D distribution of dynamic Poisson's ratio as well as velocity tomograms of P- and S-waves.

Effects of Feeding Monensin in Combination with Zeranol Implants on Performance, Carcass Traits and Nutrient Digestibility of Growing Lambs

  • Thirty-six Naeimi ram lambs were equally and randomly allotted to four treatment groups with three replications per treatment to determine the simple and additive effects of monensin and zeranol on growth performance, carcass characteristics and nutrient digestibility. The treatment groups were: basal diet-fed lambs (C), monensin-fed lambs (M) where the basal diet was supplemented with 33 mg monensin per kilogram DM, lambs implanted with 12 mg zeranol (Z), and monensin-fed lambs implanted with zeranol (MZ). Lambs fed monensin-containing diet consumed 10.5% less (p<0.05) DM/100 kg weight and were 8.3% more (p<0.05) efficient in converting feed than lambs fed control diet. Zeranol implanted lambs tended to grow 35.2% (p<0.05) faster, consumed 5.1% more (p<0.05) feed and were (p<0.05) 21.9% more efficient in their feed conversion than control lambs. Responses of lambs to monensin and zeranol implants were not additive. Except for Z treatment, there were no marked differences in all carcass characteristics among the various treatment groups. Z-lambs produced 12.7% heavier (p<0.05) carcasses compared with those from C treatment. Also fat parameters, namely, kidney and pelvic fat (KP), body wall thickness and fat thickness, indicated trends for higher finish in Z treatment lambs (p<0.05) than for those lambs from other treatments. Except for CF and ADF, no significant differences in nutrients digestibility were noticed between various treatments; feeding monensin resulted in 24.5% and 8.5% depressions (p<0.05) in CF and ADF digestibility, respectively in comparison to C treatment. Nitrogen retention as percentage of total N-intakes was averaging 7.5 and 20.2% higher (p<0.05) in lambs implanted with zeranol than those fed the M and C diets, respectively.

Effects of Galgeungyulpitang on Cellular Production of Melanin and Elastase

  • Background: This study was designed to investigate the potential effects of Galgeungyulpitang for whitening and elasticity treatment by examining its effect on melanoma cells. Methods: The effects of Galgeungyulpitang on B16/F10 melanoma cell viability, production of melanin, tyrosinase and elastase, were investigated. Cell viability was measured by colorimetric assay that assesses cell metabolic activity (MTT assay). Melanin was measured by Hosei's method, tyrosinase was measured by Yogi's method and elastase was measured by James's method. Results: At concentrations higher than $500{\mu}g/mL$ Galgeungyulpitang, cell viability was significantly reduced ($p{\leq}0.05$). At concentrations of $500{\mu}g/mL$ and lower, morphological changes were not observed. The rate of melanin synthesis was significantly reduced to $73.49%{\pm}2.92%$ at a concentration of $500{\mu}g/mL$ Galgeungyulpitang compared with untreated cells (p < 0.05). Extracellular tyrosinase production was not significantly decreased in vitro, however, intracellular tyrosinase production was significantly reduced to $76.06%{\pm}2.17%$ when treated with Galgeungyulpitang at a concentration of $500{\mu}g/mL$ compared with the control (p < 0.05). Elastase Type 1 production was significantly reduced to $74.98%{\pm}3.24%$ and $69.62%{\pm}4.66%$ at concentrations of 250 and $500{\mu}g/mL$ Galgeungyulpitang, respectively (p < 0.05). Elastase Type 4 production was significantly reduced to $72.77%{\pm}3.52%$ at concentrations of 250 and $500{\mu}g/mL$ (p < 0.05). Conclusion: The results in this study showed that Galgeungyulpitang may inhibit melanin and tyrosinase synthesis, and inhibit elastase production, suggesting that Galgeungyulpitang may be potentially beneficial for skin whitening and loss of skin elasticity treatments.

Cultural Conditions for Endotoxin Production by Bacillus thuringiensis serovar. thuringiensis (Bacillus thuringiensis serovar. thuringiensis의 내독소 생산조건)

  • Bacillus thuringiensis var. thuringiensis H1 (BTT) strain was cultured in the 4 different fermentation media and then measured their growths and the productions of endotoxin crystals from the culture media. Out of the 4 media, the productions of the endotoxin crystals and spores were maximal in the pH9-M-3 medium. The wet weight of BTT cells grown in the 150ml culture was approximately 3.218g and the number of the viable spores was $3.3{\times}10^{10}/ml$, and the ratio of the endotoxin weight over total cell weight was 20.05%. The generation time of the BTT bacteria in the M-1 was about 47.6 minutes in the M-2, 132.9 minutes in the M-3 and 110.2 minutes in the M-4. The proper pH for the production of the endotoxin by BTT appeared to be pH 6.5.

PCBs에 오염된 연안해양 Microcosm에서의 PCBs분해 유전자조작 Pseudomonas putida AC30(pMFB2)의 동태해석

  • PCBs를 분해하는 bphABC유전자를 plasmid vactor pMFB2에 유전자조작한 Pseudomonas putida AC30(pMFB2)를 PCBs에 오염된 연안해역의 해수와 저니로 만든 microcosm에 도입한 결과, 각각 도입 4일과 7일만에 사멸하였다. 그러나, 도입한 P. putida AC30(pMFB2)는 사멸하였지만, 연안해수와 저니 microcosm에서 plasmid pMFB2가 전이한 토착미생물이 검출되었다. 도입한 P. putida AC30(pMFB2)의 생잔실패의 원인을 분석한 결과 공경 0.2$\mu\textrm{m}$의 filter를 통과하는 물질과 생물이 가장 크게 영향을 미치는 것으로 나타났다. 유전자조작 P. putida AC30(pMFB2)의 도입과 bphABC유전자의 토착미생물로의 전이에 따른 토착미생물군집에 미치는 영향을 개체수 변동으로 조사한 결과, 토착미생물 군집에 미치는 영향은 보이지 않았다. P. putida AC30(pMFB2)의 도입에 의한 PCBs의 생분해성을 분석하였다. 그러나, 도입한 유전자조작 균주가 생잔에 실패함으로써 잔류하고 있는 PCBs의 농도변화는 보이지 않았다.

The Effects of Self-esteem and Social Capital on Self-support Program Participants' Will of Self-reliance

  • This study was conducted in order to identify factors that influence the will of self-reliance among workers participating in self-support programs. For this purpose, data were collected from self-support program participants in 2012 (1,301 participants in self-supporting labor programs and 128 in self-support community programs). Input variables analyzed as factors influencing the will of self-reliance were socio-demographic variables, self-esteem, and social capital. According to the results, first, self-supporting labor program participants' self-esteem was 2.328 out of 4 (S.M= .402) and it was somewhat lower than 2.406 (S.M=.404) in self-support community participants, but the difference was not significant. Self-supporting labor program participants' social capital was 2.980 out of 5 (S.M=.844), significantly lower than 3.356 (S.M=.815) in self-support community program participants. Self-supporting labor program participants' will of self-support was 4.150 out of 5 (S.M=.602), lower than 4.314 (S.M=.521) in self-support community program participants. Second, according to the results of regression analysis on self-support program participants' will of self-reliance, age (B=-.198, p<.001), self-esteem (B=.236, p<.001), and social capital (B=.166, p<.001) were found to have a significant effect. That is, the will of self-support was higher when age was young, when self-esteem was high, and when social capital was high. Based on these findings, this study suggested self-support policies, education systems, differentiated programs, etc. for enhancing self-support program participants' will of self-reliance.

Induction of Extracellular Polyphenol Oxidase from Two White-rot Fungi (목재(木材) 부후균(腐朽菌)의 목질소(木質素) 분해효소(分解酵素) 유도(誘導)에 관하여)

  • Among the representative phenolic compounds in relation to lignin derivatives and protein synthesis inhibitors, the most effective inducer for the extracellular polyphenol oxidase (PO) of Lentinus edodes JA01 was gallic acid and ferulic acid for Pleurotus ostreatus. Optimum concentration of these inducers was 2.0 mM and 1.0 mM, respectively. Addition of gallic acid after two days culture had the best effect on production of PO enzyme of L. edodes JA01 and for P. ostreatus, and addition of ferulic acid after three days culture had the best effect. Also, in case of L. edodes JA01, polyphenol oxidase activity was parallel to growth curve, whereas the maximum enzyme activity of P. ostreatus was shown at exponential growth phase and declined thereafter.

  • An intracellular inulase ( fJ-fructofuranoside fructohydrolase, EC 3.2.1.26) from Bacillus subtilis has been partially purified and its mode of action and general properties were studied. The enzyme had an apparent molecular weight of 49,000 as estimated by gel filtration and its pI point was 5.2. Substrate concentration studies showed an apparent Km of 10 mM for sucrose and of 18 mM for raffinose. The enzyme was an acid-labile protein with a pH optimum of 6.6. The optimum temperature was 50$^{\circ}$C. The enzyme acts on straight chain oligo- and poly-fructosides of the inulin series via a exo-wise cleavage mechanism, as well as on sucrose.

Enzymatic Study on Acetanilide p-Hydroxylase in Streptomyces fradiae

  • S. fradiae exhibited the highest acetanilide p-hydroxylation activity among the Streptomyces spp. screened. Studies with inhibitors (metyrapone, 2. 6-dichloroindophenol, $\alpha,\alpha'$-dipyridyl, o-phenanthroline) and an absorption peak after CO treatment suggested that S. fradiae hydroxylase activity was due to cytochrome p-450. This hydroxylase activity was increased to ten times in the cell extract containing 0.5 mM sodium azide. Furthermore, the sedimentary activity in $105,000\times{g}$ centrifugal forces and solubilization of the activity with Triton-X 100 implied that this enzyme was membrane bound monooxygenase. pH Optimum of the enzyme was 6.5 in membrane bound state.

Antibacterial Activity of Essential Oils from Zanthoxylum piperitum A.P. DC. and Zanthoxylum schinifolium

  • This study was carried out to investigate the potential use of Zanthoxylum schinifolium and Zanthoxylum piperitum A.P. DC. as a source of antimicrobial agents against food borne pathogens. Essential oils of Z. schinifolium and Z. piperitum A.P. DC. were collected by steam distillation and analyzed by GC-MS. The antimicrobial activity of the essential oils was examined using the agar diffusion and micro-dilution assays. The effectiveness of Z. schinifolium essential oil was greater against Bacillus cereus, Staphylococcus aureus, and Vibrio parahaemolyticus than other pathogens, and the minimal inhibitory concentration (MIC) values were 1.25, 2.5, and 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Z. piperitum A.P. DC. essential oil was the most effective against all pathogens tested except for Escherichia coli O157:H7, and the MIC values against B. cereus, Salmonella choleraesuis, and V. parahaemolyticus were 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Limonene, the major component of Z. piperitum A.P. DC. essential oils, had the highest inhibitory activity toward V. parahaemolyticus with a MIC value of $0.15\;{\mu}g/mL$. Meanwhile, citronellal and geranyl acetate, major components of both essential oils, displayed antibacterial activity against only B. cereus with MIC values of 1.25 and $5\;{\mu}g/mL$, respectively. Therefore, these essential oils could be useful as antimicrobial agents against foodborne pathogens.
