Immunomodulatory Activity of Crude Polysaccharides from Makgeolli (막걸리에서 분리한 다당의 면역자극 활성에 미치는 효과)

  • In this study, the immunomodulatory activities of crude polysaccharides from makgeolli were investigated. Crude polysaccahrides from makgeolli (RWW) were isolated by hot water extraction ($100^{\circ}C$, 30 min), ethanol precipitation (four volumes of 95% ethanol), dialysis (MWCO: 6,000~8,000), and lyophilization. The major constituents in RWW were neutral sugar (87.3%), uronic acid (2.5%), and protein (10.2%). RWW showed potent anti-complementary activity as well as increased cell proliferation of RAW 264.7 macrophages. The immunomodulatory effects of RWW were also analyzed based on cytokine production of macrophages. Macrophages stimulated with RWW produced cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factor-${\alpha}$ in a dose-dependent manner. These data indicate that RWW may have immunomodulatory effects through activation of the complement system and macrophages, which are a part of natural immunity.

Changes in Plasma Levels of Inhibin and Follicle Stimulating Hormone in Buffaloes Superovulated with eCG

  • The present study was undertaken to investigate the effect of stimulation of follicular development with eCG on the peripheral levels of inhibin and FSH in Murrah buffaloes. Estrus was synchronized in five normally cycling females by insertion of Crestar (Intervet, Boxmeer, Holland) implants for nine days. Estradiol valerate was administered i.m. on the day of implant insertion. On the 10th day of the induced estrous cycle a single dose of 3000 IU eCG (Folligon, Intervet, Boxmeer, Holland) was given, followed by treatment with 25 mg of $PGF_2$ alpha (Lutalyse, Upjohn, Belgium) 48 h later. Blood samples were obtained during the induced estrus, on cycle day 10 (luteal phase), at the superovulatory estrus (43 h after PGF) and during the periovulatory period (64 h after PGF). Ultrasonography was done daily to monitor follicular development. Plasma concentrations of inhibin and FSH were determined by specific radioimmunoassays. Differences between $mean{\pm}SEM$ values of different phases of the cycle were compared by ANOVA. The mean number of small (2-5 mm), medium (6-9 mm) and large (>10 mm) follicles observed two days after eCG treatment and on the day of superovulatory estrus was $2.8{\pm}0.31$, $5.2{\pm}0.30$ and $1.4{\pm}0.09$ and $1.9{\pm}0.21$, $2.8{\pm}0.40$ and $5.0{\pm}0.83$, respectively. The mean number of ovulations was $3.6{\pm}0.37$ and the mean number of unovulated follicles was $6.1{\pm}0.47$. Most of the follicles >10 mm in diameter had ovulated (72%). The mean ${\pm}SEM $ of plasma inhibin concentration $(2584.15{\pm}17.92pg/ml)$ during the superovulatory estrus was significantly higher $(p{\leq}0.05)$ than during the induced estrus $(749.87{\pm}17.29pg/ml)$, the luteal phase $(1099.54{\pm}24.98pg/ml)$ and periovulatory period $(1682.71{\pm}29.88pg/ml)$, respectively. $Mean{\pm}SEM$ plasma FSH concentration during the induced estrus $(10.35{\pm}0.41ng/ml)$ was not different from that during the superovulatory estrus $(8.52{\pm}0.39ng/ml)$, but was significantly higher $(p{\leq}0.05)$ than during the luteal phase $(2.81{\pm}0.42ng/ml)$ and periovulatory period $(5.7{\pm}0.28ng/ml)$. These data indicate that treatment with eCG in buffaloes for inducing superovulation results in a significant elevation in plasma inhibin levels and a decrease in plasma FSH levels during the superovulatory estrus. Thus, we suggest that the elevated plasma inhibin coming from fully developed follicles continued for a long time which results in inhibition of FSH leading to poor ovulation in the remaining follicles, which may be the cause of suboptimal superovulatory response.

The Effect of a Natural Zeolite (Clinoptilolite) on the Performance of Broiler Chickens and the Quality of Their Litter

  • The objective of this study was to assess the effect of clinoptilolite (a natural zeolite) on growth and performance of broilers as well as on the quality of their litter. A total of 5,200 one-day old broiler chickens (Cobb 500) were used in two consecutive rounds lasting 42 days each. The broilers were given free and continuous access to a nutritionally non-limiting diet (in meal form) that was either a basal diet (B) or a 'zeolite diet' (Z; the basal diet supplemented with clinoptilolite at a level of 2%). A $2{\times}2$ factorial design consisted of two feeding treatments (B and Z) and two bedding types, sawdust (S) and sawdust with zeolite (Sz, which was the result of adding $2kg\;zeolite/m^2$), was used. In each round the broilers were randomly assigned to one of four (n = 650), treatment groups: two fed on the basal diet (B) and had bedding of either sawdust (group BS) or sawdust and zeolite (group BSz) and two fed on the Z diet and had as bedding either sawdust (ZS) or sawdust and zeolite (ZSz). Average growth rates were significantly (p<0.05) different between broilers of different groups; broilers that were fed on the 'zeolite diet' (Z) and were placed either in a compartment with sawdust bedding or sawdust bedding and zeolite (ZS and ZSz) as well those that were fed on the basal diet in a compartment with sawdust bedding and zeolite (BSz) grew at a faster rate (p<0.05) compared with those of the control group (BS). The incorporation of NZ in broilers diets and in their bedding material decreased the organic content in litter samples throughout the experimental period. The lowest organic content was recorded in group ZSz where NZ was added in both feed and litter. Mean ammonia concentration (ppm) was significantly higher in group ZS in comparison to groups BSz and ZSz (27.00 vs. 20.55 and 21.71 respectively). The results of this study showed that the incorporation of the clinoptilolite both in feed and into the litter had a positive effect on broiler growth and also on the quality of their litter.

An Retrospective Analysis of 1549 patients under the Cooperative Medical Care consulted to the Department of Acupuncture and Moxibustion in a Korean medical hospital (한방병원 침구과로 협진의뢰된 한양방 협진 환자 1,549명에 대한 후향적 분석)

  • Objectives : The aim of this study is to give a statistical basis for the characteristics of patients who received cooperative medical care after being referred from Western medical departments to the Department of Acupuncture and Moxibustion in a Korean medical hospital. Methods : The request records were searched for cooperative medical treatment at the Department of Acupuncture and Moxibustion in a Korean medical hospital between September 1st, 2011 and August 31, 2016, based on the Order Communication system(OCS). The records were reviewed and retrospectively analyzed according to the patient's general characteristics, the chief diagnosis code based on the 7th Korean standard classification of diseases(KCD-7) before and after the cooperative medical care in the Department of Acupuncture and Moxibustion, and the actual treatment measures which patients received as a result of the cooperative medical care. Results : Among the 2702 records, 1549 patients were included in the analysis. The male to female ratio was 1 : 1.17. Sexagenarians formed the highest age group and the number of patients whose age was over 50 was 1176(75.92 %). The surgery department requested the cooperative care from the department of Acupuncture and Moxibustion for the highest number of patients, 1194(77.08 %). When analysing specific medical departments, Neurosurgery, General Surgery, Neurology, Otorhinolaryngology, and Physical medicine and rehabilitation followed. The frequency of chief diagnosis code used before the cooperative medical care was in the order of I, C, G, S, and M code, from highest to lowest. The order of the codes after the cooperative medical care was the same. Acupuncture treatment was the most frequently-used measure after the cooperative medical care. Following were infrared, moxibustion, transfer, herbal acupuncture, cupping, electroacupuncture, and so on. Conclusion : This study provides a statistical basis for the demands placed on the Department of Acupunture and Moxibustion for cooperative medical care with the Western medical department. Based on the present condition of cooperative medical care, more preparation and effort is required for its activation.

Effect of Ga-doping on the properties of ZnO films grown on glass substrate at room temperature by radio frequency magnetron sputtering (RF 마그네트론 스퍼터링 방법으로 상온에서 유리기판 위에 성장시킨 ZnO의 성질에 미치는 Ga 도핑 효과)

  • We present the effect of Ga-doping on the electrical, structural and optical properties of ZnO layers with a thickness of ${\sim}500nm$ deposited on glass substrates. Polycrystalline ZnO and Ga-doped ZnO (GZO) layers were deposited by radio frequency (rf) magnetron sputtering at room temperature. Based on the X-ray diffraction (XRD) and transmission electron microscopy (TEM) data, the crystalline quality of Ga-doped ZnO film was improved and GZO film has a preferred orientation along with the (002) crystal direction. The transmittance of the GZO film was enhanced by 10% in the visible region from that of the ZnO film. From photoluminescence (PL) data, the ratio of intensity of near band edge (NBE) emission to deep level (DL) emission was as high as 2.65:1 and 1.27:1 in the GZO and ZnO films, respectively. The res istivities of GZO and ZnO films were measured to be 1.27 and 1.61 $\Omega{\cdot}cm$, respectively. The carrier concentrations of ZnO and GZO film were approximately 1018 and 1020 $cm^2$/Vs, respectively. Based on our experimental results, the Ga-doping improves the electrical, structural and optical properties of ZnO film with potential application.

Comparison of Incidence of PSE Pork by Fattening Period, Transport Time and Lairage Time and Lairage Time (돼지의 출하일령, 수송시간 및 계류시간에 따른 PSE 발생율 비교)

  • The incidence of PSE(Pale, Soft, Exudative) pork by slaughter weight and handling conditions at preslaughtering stage such as fattening period, transport and lairage time were compared. The incidence of PSE pork were 24.9% when pigs were fed under 180 days and 18.9% when pigs were fed over 180 days for fattening. In the transport time from farm to slaughter house, the incidence of PSE pork were 46.6% for shorter than 30min and 45.65% for 30 min${\sim}$60 min and 35.44% for longer than 60min. The incidence of PSE pork were 55.6% for pigs had no lairage before slaughter, 33.9% for pigs held in lairage overnight. Therefore, the incidence of PSE pork were decreased by 6% with the extension of the fattening period and 9% with the transport time of longer than 1hr and 22.1% with lairage treatment before slaughter. The incidence of PSE pork were different in different location of farms and there were no significantly different in live weight, carcass weight and carcass yield between groups of lairage treatment. In conclusion, handling condition at preslaughtering stage such as extension of fattening period and lairage time were important to control pork quality and lairage treatment was not decreased the carcass yield.

Creation of Consistent Burn Wounds: A Rat Model

  • Background Burn infliction techniques are poorly described in rat models. An accurate study can only be achieved with wounds that are uniform in size and depth. We describe a simple reproducible method for creating consistent burn wounds in rats. Methods Ten male Sprague-Dawley rats were anesthetized and dorsum shaved. A 100 g cylindrical stainless-steel rod (1 cm diameter) was heated to $100^{\circ}C$ in boiling water. Temperature was monitored using a thermocouple. We performed two consecutive toe-pinch tests on different limbs to assess the depth of sedation. Burn infliction was limited to the loin. The skin was pulled upwards, away from the underlying viscera, creating a flat surface. The rod rested on its own weight for 5, 10, and 20 seconds at three different sites on each rat. Wounds were evaluated for size, morphology and depth. Results Average wound size was $0.9957cm^2$ (standard deviation [SD] 0.1845) (n=30). Wounds created with duration of 5 seconds were pale, with an indistinct margin of erythema. Wounds of 10 and 20 seconds were well-defined, uniformly brown with a rim of erythema. Average depths of tissue damage were 1.30 mm (SD 0.424), 2.35 mm (SD 0.071), and 2.60 mm (SD 0.283) for duration of 5, 10, 20 seconds respectively. Burn duration of 5 seconds resulted in full-thickness damage. Burn duration of 10 seconds and 20 seconds resulted in full-thickness damage, involving subjacent skeletal muscle. Conclusions This is a simple reproducible method for creating burn wounds consistent in size and depth in a rat burn model.

Infectivity of the sparganum treated by praziquantel, gamma-irradiation and mechanical cutting (프라지콴텔, 감마선 조사 및 기계적 절단으로 처리한 스파르가눔의 마우스에 대한 감염력)

  • An experimental study was performed to observe the infectivlty of sparganum (plerocercold of Spirometra erinncel) treated by prazlquantel, gamma-irradiation and mechanical cutting. The spargana were obtained from the naturally Infected European grass snake, Rhabdophis tigrina, or from the expenmentally infected mice. A total of 83 mice (ICR straine were divided Into 3 experimental groups by the source of the damage, fed each UH 5 spargana, and sacnaced 1 month later for worm recovery. In the praziquantel group, the worms were incubated In the concentration of 10㎍/ml (control: Tyrode for 4 hours) for 0.5. 1.2 and 4 hours at 36℃. and fed to mice. The recovery rate from mice in prazlquantel group was not different from that (80%) of control poop and in the range of 76-100%. In the gamma-Irradiation group, the worms were irradiated by 10-1000 Gy with Csl37. The average recovery rates of 69-100% were not different from that of control up to 100 Gy. The rate was 56% under 150 Gy, and 5% by 1000 Gy. In the mechanical cutting group, the worms were cut at 0.5, 1, 2 and 3 m from the anterior end of the scolex. The average recovery rates In each group were 70-90% and that of control was 90%. The present folding suggests that the sparganum be highly resistant to prazlquantel, gamma- irradiation and mechanical cutting. The vitality center of the sparganum must be at the anterior end of Its scolex.

Myo-inositol increases the plating efficiency of protoplast derived from cotyledon of cabbage (Brassica oleracea var. capitata)

  • This study describes the effect of myo-inositol on sustained cell division and plant regeneration from cotyledon-derived protoplast of cabbage (Brassica oleracea var. capitata). Freshly isolated protoplasts were cultured in modified Murashige and Skoog (MS) medium removed ammonia ions and containing $0.4\;mg\;l^{-1}$ thiamine HCl, $100\;mg\;l^{-1}$ myo-inositol, $2\;mgl^{-1}$ 2,4-D, $0.5\;mgl^{-1}$ BA, $30\;gl^{-1}$ sucrose and several concentrations of myo-inositol (2, 4, 6, 8, 10% (w/v)) as an osmotic stabilizer. After 3 weeks of culture in the dark at $25^{\circ}C$, the plating efficiency of cabbage protoplasts reached to $22.5{\pm}2.9%$ when cultured in modified MS medium supplemented with $2\;mgl^{-1}$ 2,4-D, $0.5\;mgl^{-1}$ BA, $30\;gl^{-1}$ sucrose and 8% (w/v) of myo-inositol at a density of $2{\times}10^5$ protoplasts/ml. Rapidly growing cell colonies after 3 weeks of culture were transferred to the same culture medium removed osmoticum. To induce shoot regeneration from calluses, calluses with about 2 mm in diameter were transferred to the MS medium containing $2\;mgl^{-1}$ BA and $0.5\;mgl^{-1}$ NAA. After further three weeks of incubation onto the medium in the light, green shoots were formed on the surface of calluses at a frequency of 30%. Upon transfer to half-strength MS basal medium, roots were formed onto the bottom of regenerated shoots without auxin treatments. These regenerated plantlets were successfully acclimatized to soil transfer, grown to normal mature plants. The cabbage protoplast culture system established in this study could be applied for production of somatic hybrids or cybrids by asymmetric protoplast fusion and mass proliferation of elite somatic clones of cabbage.

Analytical Method Development and Monitoring of Residual Solvents in Dietary Supplements (건강기능식품 중 잔류용매 분석법 개발 및 모니터링)

  • Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.
