Free Radical Scavenging Effect and Extraction Condition of Ethanol Extracts of Epimedium Koreanum Nakai Containing Different Icariin Quantity (Icariin 함량에 따른 삼지구엽초 추출물의 추출조건 및 항산화 효과)

  • This study was conducted to determine the optimum condition of extraction and antioxidant activity of ethanol extracts of Epimedium koreanum Nakai based on the icariin quantity. Also, further organic solvent fractions of hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of Epimedium koreanum Nakai at different temperatures. Total ethanol extraction yield of wild grape seed ranged from 11.8% to 39.3% depending on the concentration of icariin as well as different ethanol concentration, extraction temperature and time condition. Among different extraction temperatures and time, the highest extraction yield of 39.3% was obtained at 70% ethanol for 3 hour at $50^{\circ}C$ in the sample containing the 0.596% icariin (KE9412). In the meantime, the strongest free radical scavenging effect $(RC_{50})$ with $18.9{\mu}g/mL$ in the KE9412 sample was observed in 70% ethanol extract of Epimedium koreanum Nakai extracted for 7 hour at $70^{\circ}C$, while $RC_{50}$ with $35.2{\mu}g/mL$ was observed in the KE9405 (0.20% icariin content) sample at same condition. Also, antioxidant activity of ethanol extracts of Epimedium koreanum Nakai increased as icariin concentration increased. Among each fraction obtained from organic solvents, butanol fraction was found to have the strongest $RC_{50}\;(39.2{\mu}g/mL)$ and followed by ethylacetate $(49.0{\mu}g/mL)$, water $(118.8{\mu}g/mL)$, ethylacetate $(119.8{\mu}g/mL)$, and chloroform fraction $(138.5{\mu}g/mL)$ respectively.

Effects of Temperature and lncoming Concentrations on the Removal of Volatile Organic Compounds in a Biofilter Packed with Peat (Peat가 충진된 고정층 생물반응기에서 온도 투입농도가 휘발성 유기화합물의 분해에 미치는 영향)

  • Biofiltration of volatile organic compounds (VOCs) was performed for 80 days in a biofilter packed with peat. The empty bed residence time was 3.2 min. for a gas mixture of isoprene, dimethyl sulfide, chloroform. benzene, trichlorethylene, toluene, m0xylene, o-xylene and styrene. After 34 days of acclimatization the removal efficiency for a 83 g/㎥ gas input was 93% at $25^{circ}C$ and 73% at $45^{circ}C$, respectively. The maximum cell density at $25^{circ}C$ was 1.12$\times$10(sup)8 cells/g. Removal efficiencies of m-xylene and toluene (91%) were better than that of benzene (86%). The first quarter of the packed column removed 60% of the incoming VOCs.

Antioxidative Activity and Componential Analysis of Chamaecyparis obtusa Leaf Extract (편백나무 잎 추출물의 항산화 활성과 성분분석에 관한 연구)

  • In this study, the evaluation of antioxidative activity and componential analysis of C. obtusa leaf extracts was carried out. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of C. obtusa leaf extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($OSC_{50}$; 0.22 ${\mu}g/mL$) and aglycone fraction of C. obtusa leaf extracts (0.20 ${\mu}g/mL$) showed about 7 times more prominent ROS scavenging activity than L-ascorbic acid (1.50 ${\mu}g/mL$). The cellular protective effects of fractions obtained from C. obtusa leaf extracts on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of C. obtusa leaf extracts showed the cellular protective effects in a concentration dependent manner (5~25 ${\mu}g/mL$). The inhibitory effect ($IC_{50}$) of ethyl acetate fraction and aglycone fraction on tyrosinase exhibited 74.43 and 53.80 ${\mu}g/mL$, repectively. The aglycone fraction showed four times higher tyrosinase inhibitory effect than arbutin (226.88 ${\mu}g/mL$), known as a whitening agent. The aglycone fraction of C. obtusa leaf extracts showed three bands in TLC chromatogram and three peaks in HPLC chromatogram (360 nm). Three compounds were identified as taxifolin, quercetin and kaempferol. These results indicate that the fractions of C. obtusa leaf extracts can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against reactive oxygen species. The fractions of C. obtusa leaf extracts can be applicable to new functional cosmetics for antioxidan and whitening effects.

Synthesis and Electrochemical Properties of $LiNi_{1-y-z}(M_1)_y(M_2)_zO_2$ for the Development of Cathode Materials with Large Capacity (고용량 양극재료 개발을 위한 $LiNi_{1-y-z}(M_1)_y(M_2)_zO_2$의 합성과 전기화학적 특성)

  • [ $LiNi_{0.995}Al_{0.005}O_2$ ], $LiNi_{0.990}Ti_{0.010}O_2$ and $LiNi_{0.0990}Al_{0.005}Ti_{0.005}O_2$ were synthesized with a combustion method by calcining in an $O_2$ stream at $750^{\circ}C$ for 36 h. The X-ray diffraction patterns of these synthesized samples showed $-NaFeO_2$ structure of rhombohedral system(space group; $R{\bar{3}}\;m$) with no evidence of impurities. Among these samples, $LiNi_{0.995}Al_{0.005}O_2$ exhibited comparatively high first discharge capacity and discharge capacity, and the best cycling performance. $LiNi_{0.995}Al_{0.005}O_2$ had the first discharge capacity of 165.2 mA h/g and a discharge capacity of 116.7 mA h/g at the 50th cycle at 0.1C rate. It showed the first discharge capacity of 141.0 mA h/g and a discharge capacity of 93.5 mA h/g at the 50th cycle at 0.5C rate.

he Study of Nucleic Acid Extraction Method from Archival Paraffin Blocks (보존된 파라핀 블록에서 핵산 추출기법에 관한 연구)

  • It designed a study to examine the efficiency of DNA and RNA extraction from archival formalin-fixed, paraffin-embedded tissues using an non-heating and heating method. Archival paraffin blocks of liver, kidney, colon were randomly selected. Each paraffin block was prepared in 20 microtubes. For each paraffin blocks were tested non-heating DNA extraction to 10 microtubes and heating protocol under pH 7.0 and $100^{\circ}C$ to 10 microtubes. Evaluation of the results of DNA extraction was carried out by measuring concentration by UV spectrophotometry and then PCR amplification. DNA extraction content that non-heating method was liver $5{\pm}0.7{\mu}g/mL$, kidney $2{\pm}0.3{\mu}g/mL$, colon $6{\pm}0.4{\mu}g/mL$ and heating method was liver $12{\pm}0.6{\mu}g/mL$, kidney $7{\pm}0.5{\mu}g/mL$, colon $10.{\pm}0.3{\mu}g/mL$. Successful RNA extraction was observed, by ${\beta}$-actin amplification, in 46.7% sections for samples treated by the heating method versus 30.0% using non-heating DNA extraction. The extracted nucleic acid showed better values for samples heated at $100^{\circ}C$. Therefore heating extraction of nucleic acid is reliable, quick and efficiency.

In vitro anticancer and antioxidant effects of acetone extract of Eucommia ulmoides oliver leaves (두충잎 아세톤 추출물의 in vitro 항암 및 항산화 효과)

  • In vitro anticancer and antioxidant effects of acetone extract from leaves of Eucommia ulmoides Oliver were investigated. The extraction yield and total phenolic content of the acetone extract were $1.13{\pm}0.033%$ (w/w) and $36.7{\pm}1.96mg$ gallic acid equivalents/g-extract, respectively. $GI_{50}$ values of the acetone extract for human non-small cell lung cancer cells (A549), human colon cancer cells (SNU-C4), human cervical cancer cells (HeLa), and human embryonic lung epithelial cell (L132) were 53.4, 53.8, 88.3, and $153.9{\mu}g/mL$, respectively. The acetone extract effectively inhibited the proliferation of human non-small cell lung cancer (A549) and colon cancer (SNU-C4) cells in a concentration-dependent manner, but was less cytotoxic with human normal cells (L132). $EC_{50}$ values of the acetone extract for free radical scavenging, reducing power, and lipid peroxidation inhibition were about 2,000, 275.8, and $257.9{\mu}g/mL$, respectively. The acetone extract showed a potent reducing power and lipid peroxidation inhibitory activity in a concentration-dependent manner.

  • Let G be a group and $\mathbb{C}$ the field of complex numbers. Suppose ${\sigma}:G{\rightarrow}G$ is an endomorphism satisfying ${{\sigma}}({{\sigma}}(x))=x$ for all x in G. In this paper, we first determine the central solution, f : G or $G{\times}G{\rightarrow}\mathbb{C}$, of the functional equation $f(xy)+f({\sigma}(y)x)=2f(x)+2f(y)$ for all $x,y{\in}G$, which is a variant of the quadratic functional equation. Using the central solution of this functional equation, we determine the general solution of the functional equation f(pr, qs) + f(sp, rq) = 2f(p, q) + 2f(r, s) for all $p,q,r,s{\in}G$, which is a variant of the equation f(pr, qs) + f(ps, qr) = 2f(p, q) + 2f(r, s) studied by Chung, Kannappan, Ng and Sahoo in [3] (see also [16]). Finally, we determine the solutions of this equation on the free groups generated by one element, the cyclic groups of order m, the symmetric groups of order m, and the dihedral groups of order 2m for $m{\geq}2$.

  • Let M be a real hypersurface with almost contact metric structure $(\phi,g,\xi,\eta)$ in a complex space form $M_n(c)$, $c\neq0$. In this paper we prove that if $R_{\xi}L_{\xi}g=0$ holds on M, then M is a Hopf hypersurface in $M_n(c)$, where $R_{\xi}$ and $L_{\xi}$ denote the structure Jacobi operator and the operator of the Lie derivative with respect to the structure vector field $\xi$ respectively. We characterize such Hopf hypersurfaces of $M_n(c)$.

  • Shaking cultivation was carried out in 250 mL Erlenmeyel' flasks containing 60 mL of culture solution for 7 days. Artificial seawater was used as a basal culture medium. The cultivation was performed under the conditions that the temperature ranged from $4^{\circ}C$ to $32^{\circ}C$, the agitation speed 50 rpm to 200 rpm, and the sugar concentration 5 g/L to 35 g/L. The medium condition was cultured that sugar and nitrogen ratio 1.25 to 10. The biomass. the lipid content in biomass, and the DBA content of lipid were investigated according to the cultivation conditions. The lipid in biomass was distributed between 10 and 349'0, the DHA in lipid 34% to 43% of the lipid. and the biomass concentration 0.425 g/L to 4.50 g/L.

Optimization of γ-Aminobutyric Acid (GABA) Production Using Immobilized Lactobacillus plantarum K154 in Submerged Culture of Ceriporia lacerata (Ceriporia lacerata 배양액과 고정화 Lactobacillus plantarum K154를 이용한 감마아미노뷰티르산 생산 최적화)

  • The production of GABA was optimized by co-cultivation of immobilized Lactobacillus plantarum K154 (ILK) with Ceriporia lacerata cultures. The mycelial culture of C. lacerata was performed in a defined medium containing 3% glucose, 3% soybean flour, and 0.15% $MgSO_4$ in a submerged condition for 7 days at $25^{\circ}C$, resulting in the production of 29.7 g/L mycelia, 3.1 g/L exopolysaccharides, 2% (w/w) ${\beta}$-glucan, 68.96 unit/mL protease, and 10.37 unit/mL ${\alpha}$-amylase. ILK in C. lacerata culture showed viable cell counts of $3.13{\time}10^9CFU/mL$ for immobilized cells and $1.48{\time}10^8CFU/mL$ for free cells after 1 day. GABA production in the free and immobilized cells was 9.96 mg/mL and 6.30 mg/mL, respectively, after 7 days. A recycling test of ILK in the co-fermentation was consequently performed five times at $30^{\circ}C$ for 15 days, resulting in the highest production of GABA. GABA could also be efficiently overproduced by co-cultivation with the produced polysaccharides, ${\beta}$-glucan, peptides, and probiotics.
