  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

  • Discospondylitis is an infection of an intervertebral disk with concurrent osteomyelitis of contiguous vertebrae. Clinical signs are variable and include pain, fever, anorexia, weight loss, depression, stilted gait and paresis or paralysis. A seven-year-old, intact female Yurkshire terrier dog was refered to the Vfterinary Medicine Teaching Hospital of Seoul National University because of intermitent pain of unknown cause for 2 weeks. On physical examination, pain was evident when spines were manipulated, especially thoracolumbar junction part. No neurologic deficits were deteced in the general neurologic test. Spinal radiography demonstrated the bony lysis of the vertebral end plates and sclerosis of the $T_{12-13}$ vertebral body. Based on the results of examinations, the dog was diagnosed as discospondylitis and recoverd following antibiotic therapy.

  • A castrated male, 9-year-old Yorkshire terrier was presented with a depression and bilateral hind limbs lameness. On physical examinations, upper motor neuron signs and stiffness of the hind limbs, back pain and progressive paresis were identified. Marked periosteal new bone formations and lysis include the first lumbar vertebra to the sacrum, bilateral iliums acetabulums and bilateral femoral heads were observed in survey radiographs. After death with septicemia suspected, renal infarction and the 5th vertebral osteomyelitis include pelvic periostitis were diagnosed in histological examination.

  • Many useful biomaterials like enzymes are contained in yeast cells. However, the release of these intracellular biomateriais from the cells is required to recover them with hot water, solvent or various cell breakage methods of mechanical or non mechanical ones. The cell lysis or breakage of yeast is usually made by solvent like ethyl acetate and mechanical disintrgration with high pressure homogenizer or agitating beads mill. The separation of cell debris (i.e. solid liquid separation) is done by centrifuge or membrane depending on the recovery conditions. The features of both separation methods are shown in Tables 1 and 2. As it is often difficult to obtain a clear supernatant by centrifuge from the suspension containing cell debris, the membrane separation is also often used to gel a clear supernatant. In this report we introduce the several applications of membrane separation to separate the cell debris of yeast disintegrated chemically or mechanically and to recover the intracellular biomaterials.

  • A temperate phage was isolated from emetic Bacillus cereus NCTC 11143 by mitomycin C and characterized by transmission electron microscopy and DNA and protein analyses. Whole genome sequencing of Bacillus phage 11143 was performed by GS-FLX. The phage has a dsDNA genome of 39,077 bp and a 35% G+C content. Bioinformatic analysis of the phage genome revealed 49 putative ORFs involved in replication, morphogenesis, DNA packaging, lysogeny, and host lysis. Bacillus phage 11143 could be classified as a member of the Siphoviridae family by morphology and genome structure. Genomic comparisons at the DNA and protein levels revealed homologous genetic modules with patterns and morphogenesis proteins similar to those of other Bacillus phages. Thus, Bacillus phages might have a mosaic genetic relationship.

  • This paper is to test the modulating effect of organizational culture on the relationship between personality and incremental innovation, and study of the relationship between incremental innovation and service Quality in service industry. Based on 507 data gathered in service companies in Korea, analysis results and showed that procedural, interactional, and innovative culture have affirmative relation with incremental innovation. Therefore innovative culture appeared to have some modulating effect on the relationship between personality and incremental innovation. And ana-lysis results of relationship between incremental innovation and service quality are followed; service innovation and process innovations showed positive effects except some operation innovations.

  • Direct analysis of microbes such as either gram-positive or gram-negative bacteria without cell lysis was investigated using capillary electrophoresis. Bacteria cells were directly introduced into the microbore fusedsilica capillary, then separated under high electric field in less than 15 min. It was found that a proper dispersion of bacteria cells was important for reproducible results. Migration behavior of bacteria at different storage condition was investigated and many unexpected peaks were observed from bacteria stored at room temperature due to the distortion of cells. This phenomenon was attributed to the change of size and shape of the same bacterium and confirmed by the scanning electron microscopic images.

  • Temporomandibular disorder(TMD) is described as a cluster of disorders characterized by pain in the preauricular area and/or the muscles of mastication; limitations or deviations in mandibular range of motion; and noises in the TMJ during mandibular function. The most common symptom in TMD patients is pain that is aggravated by chewing or other jaw function. These symptoms are appeared when the stimuli loaded in TMJ are over the physiologic tolerance. The primary goal in treatment of TMD is to alleviate pain and lor mandibular dysfunction. TMD treatment can be divided into 2 categories: reversible and irreversible methods. Reversible methods include medication, thermal therapy, habit modification, physical therapy, appliance therpy and arthrocentesis and lavage and irreversible methods include arthroscopic lysis, surgery, occlusal adjustment et al. It is widely accepted that reversible methods are ther first choice of treatments. However if reversible ones are not effective, irreversible methods are considered.

  • Helicobacter pylori (Hp), an important etiologic agent in a variety of gastroduodenal diseases, produces a large amount of urease, which is believed to neutralize gastric acid by producing ammonia for the survival of the bacteria. Up to 30% of the enzyme is associated with the surface of intact cells by lysis of neighboring bacteria. However, the role of the external enzyme has been a subject of controversy, because the enzyme is irreversibly inactivated below pH 5.(omitted)

  • Tolaasin is a 1.9 kDa peptide produced by Pseudomonas tolaasii and causes a brown blotch disease on cultivated oyster mushrooms. These molecules form channels in the plasma membranes of various cells including red blood cells and destroy cellular structure, known as 'colloid osmotic lysis'. In order to understand the molecular mechanism of tolaasin-mediated channel formation, the effect of Zn$^{2+}$ was investigated on hemolysis and channel formation since Zn$^{2+}$ has been known to block the tolaasin activity.(omitted)ted)
