연두금파리 종령유충의 복신세포의 미세구조 (Ultrastructure of the Ventral Nephrocytes in the Larva of Lucilia illustris Meigen)

  • Ventral nephrocytes in the larva of the Lucilia illustris comprise ellipsoid cells situated onto the salivary glands. The cells are $60{\sim}100{\mu}m$ in diameter. Junctional complex beneath the basement membrane hold the plasma membrane in a even contour. Intracellular channels from the juntion complex are well developed at the cortex part of the cell. Coated vesicles pinched off from the channels seems to be connected with the ${\alpha}$-vacuoles via the tubular elements, which is regared as selective absorption system from the hemolymph. Two nuclei are sometimes observed in the medulla part of the cell. Ventral nephrocytes contain well-developed rough endoplasmic reticulum and Golgi complex, and numerous mitochondria. These cellular organelles synthesize lysosome. The lysosome not only digest some cell organells but also seems to be related with the ${\beta}$-vacuoles.

An aural myiasis case in a 54-year-old male farmer in Korea

  • A 54-year-old male farmer residing in Chunchon, Korea. complaining of blood tinged discharge and tinnitus in the left ear for two days. was examined in August 16, 1996. Otoscopic examination revealed live maggots from the ear canal. The patient did not complain of any symptoms after removal of maggots. Five maggots recovered were identified as the third stage larvae of Lucilia sericata (Diptera: Calliphoridae). This is the first record of aural myiasis in Korea.

  • On July 2009, 5 fly larvae were discovered inside the nose of a 76-year-old female. She was living in Cheonansi, and in a state of coma due to rupture of an aortic aneurysm. Surgery was performed on the day of admission, and the larvae were found 4 days later. By observing their posterior spiracle, the larvae were identified as Lucilia sericata. Considering the rapid development of this species, the infection was likely acquired during hospitalization. Further investigation on the hospital environment should be needed to know the origin of the infection.

  • 연두금파리의 유충-번데기-성충의 발생에 따른 cyclic AMP(CAMP)와 ecdysteroids의 농도 및 cyclic AMP-dependent protein kinase(PKA)의 활성도를 High Performance Liquid Chromatography(HPLC)와 Liquid Scintillation Counter(LSC)를 이용하여 측정하고 상관 기능 변화를 조사하였다. CAMP의 농도와 PKA의 활성도는 종령유충 및 성충에서 낮은 갓(0.29 UM/g, 5.52~5.59 unit/mg)을 나타내었고 번데기 0일(0.49 UM/g. 92.22 unit/mg)과 7일(0.50UM/g, 24.45 unit/mg)에서 최고치를 보였으며 번데기 4일에서 최저수준(0.13 $\mu$M/g3.23 unit/mg)을 나타내었다. Ecdysone 농도는 번데기 2일에 최고치(37 84 USlg)를 4일에 최저치(18.46 Ugyg)를 보인 후, 5일에 24.37 1519로 상승하였으며 성충에서는 낮은 값을 나타내었다 그러나 20-hydroxvecdysone 농도는 번데기 4일(23.66 UgyS)과 번데기 6일(14.90 Ugyg)에 최고치를 이루었고, 7일에서 최저치(1.21 USi9)를 나타내었다.

Forensically Important Blow Flies Chrysomya pinguis, C. villeneuvi, and Lucilia porphyrina (Diptera: Calliphoridae) in a Case of Human Remains in Thailand

  • This is the first study to report Chrysomya pinguis (Walker) and Lucilia porphyrina (Walker) (Diptera: Calliphoridae) as forensically important blow fly species from human cadavers in Thailand, in addition to Chrysomya villeneuvi (Patton) already known in Thailand. In 2016, a fully decomposed body of an unknown adult male was discovered in a high mountainous forest during winter in Chiang Mai province. The remains were infested heavily with thousands of blow fly larvae feeding simultaneously on them. Morphological identification of adults reared from the larvae, and molecular analysis based on sequencing of 1,247 bp partial mitochondrial cytochrome c oxidase subunit 1 gene (CO1) of the larvae and puparia, confirmed the above mentioned 3 species. The approving forensic fly evidence by molecular approach was described for the first time in Thailand. Moreover, neighbor-joining phylogenetic analysis of the CO1 was performed to compare the relatedness of the species, thereby affirming the accuracy of identification. As species of entomofauna varies among cases in different geographic and climatic circumstances, C. pinguis and L. porphyrina were added to the list of Thai forensic entomology caseworks, including colonizers of human remains in open, high mountainous areas during winter. Further research should focus on these 3 species, for which no developmental data are currently available.

무균 구리금파리 유충 생산용 배지의 선발과 알 및 유충의 저장을 위한 온도 및 기간 (Selection of Rearing Media, Proper Temperature and Period for Storage of Sterile Lucilia sericata (Diptera: Calliphoridae) Eggs and Larvae)

  • 구더기 치료에 널리 이용되고 있는 무균 구리금파리 [Phaenicia (=Lucilia) sericata (Meigen)]를 보다 손쉽게 사육하기 위한 배지를 선발하고, 알과 유충의 저장조건을 실험하였다. 시판되고 있는 8가지의 미생물 배양용 선택배지에서 실험한 결과, 알의 부화율은 배지 간에 차이가 없었고, 유충의 생존율은 1, 2차 실험결과 blood agar(BA), sabouraud dextrose agar 및 brucella blood agar 배지에서 가장 높았다. BA 및 chocolate agar 배지에서 양의 혈액 함량을 20${\sim}$40%로 높일수록 유충의 생육이 좋았다. 알은 부화율의 저하 없이 $8^{\circ}C$에서는 12일까지 저장할 수 있었고, 3령 초기 유충은 생존율의 저하 없이 $6^{\circ}C$에서 15일까지 저장할 수 있었다.

  • 연두금파리의 난세포성숙에 따른 단백질의 변화와 난특이성단백질의 특성을 확인하기 위하여 gel filtration, 전기영동 및 분자량측정, 아미노산과 지방산함량을 측정하여 얻은 결과는 다음과같다. 연두금파리 암컷성충의 난소단백질은 단백질원을 섭식시킨 후 72시간 이후 빠르게 증가하였고, 완전한 성숙이 일어나는 96시간에 최고의 함량을 나타냈다. DEAE-cellulose와 Sephacryl 5-200으로 gel filtration하고 7.5% native polyacrylamide gel에서 전기영동한 결과 난소에서 분리된 특이단백질은 ${R}_{f}$ 0.4에서 혈림프 및 난소와 다른 밴드가 확인되었으며, 분자량은 110,000 dalton이였다. 분리된 난특이성단백질내 아미노산 조성은 asparagine 외 모두 13종이 검출되었으며, asparagine, glutamic acid와 함께 tyrosine이 특이하게 높게 나타났다. 지방산은 난소와 함께 난특이성단백질에서 palmitic acid의 4종이 분리되었다. 따라서, 연두금파리의 난에는 지방체에서 합성, 분비된 난황단백질이외에 난소에만 존재하는 특이단백질이 있음을 알 수 있다.
