Temperature-Dependent Release of Drug from Copolymers of N-Isopropylacrylamide Containing Liposome (리포솜이 함유된 N-이소프로필아크릴아마이드의 공중합체로부터 온도에 따른 약물의 방출)

  • Thermosensitive poly(N-isopropylacrylamide) gels containing temperature-sensitive liposomes showing temperature-dependent sol-gel transition were prepared. The surface of temperature-sensitive liposome was modified with copolymers of N-isopropylacrylamide and octadecylacrylate, which exhibited a lower critical solution temperature at around 30 $^{\circ}C$ After mixing the modified temperature-sensitive liposomes with poly(N-isopropylacrylamide) solution, the temperature-sensitive 1iposomes formed physically cross-linked gels through heating the solution above their lower critical solution temperatures. The release of drug from temperature-sensitive liposomes was determined by measuring fluorescence intensity. The drug release from temperature-sensitive liposomes in poly(N-isopropylacrylamide) gel gradually showed sustained-release with increasing temperature.

  • Thermosensitive block copolymers of ethylene oxide and N-isopropylacrylamide (NIPAM) were synthesized. A five armed star shape oligo(ethylene oxide) initiator with a cyclotriphosphazene core was prepared and used for the atom transfer radical polymerization (ATRP) of NIPAM. The lower critical solution temperatures (LCSTs) of the copolymers were 36 to 46 ${^{\circ}C}$, higher than that of PNIPAM (32 ${^{\circ}C}$), depending on their molecular weights. The copolymers were soluble in water below the LCSTs but formed micelles above the LCSTs. The thermosensitive micellization behaviors of the polymers were investigated by fluorescence spectroscopy. With increasing the temperature of an aqueous solution of P2 and pyrene above the LCST, the peak of 333 nm red-shifted to appear around 339 nm and its intensity increased significantly, indicating the micelle formation. The transfer of pyrene into the micelles was also confirmed by a confocal laser scanning microscope. The fluorescence image obtained from P2 in an aqueous pyrene solution exhibited a green emission resulting from the pyrene transferred into the micelles. Salt effects on the solubility of the copolymers in an aqueous solution were investigated. The LCST of P2 decreased sharply as the concentration of sodium chloride increased, while decreased slowly with potassium chloride.

Saccharide Effect on the Lower Critical Solution Temperature of Poly(organophosphazenes) with Methoxy-poly(ethylene glycol) and Amino Acid Esters as Side Groups

  • The lower critical solution temperature (LCST) of thermosensitive poly(organophosphazenes) with methoxypoly(ethylene glycol) (MPEG) and amino acid esters as side groups was studied as a function of saccharide concentration in aqueous solutions of mono-, di-, and polysaccharides. Most of the saccharides decreased the LCST of the polymers, and the LCST decrease was more prominently observed by saccharides containing a galactose ring, such as D-galactose, D-galactosamine and D-lactose, and also the polysacccharide, 1-6-linked D-dextran effectively decreased the LCST of the polymers. Such an effect was discussed in terms of intramolecular hydrogen bonding of saccharides in polymer aqueous solution. The saccharide effect was found to be almost independent on the kinds of the amino acid esters and MPEG length of the polymers. Such a result implies that the polymer-saccharide interaction in aqueous solution is clearly influenced by the structure of sacchardes rather than by that of the polymers. The acid saccharides such as D-glucuronic and D-lactobionic acid increased the LCST, which seems to be due to their pH effect.

  • In some binary solution, closed miscibility loop of temperature-composition phase diagram occurs where both an upper critical solution temperature and a lower critical solution temperature exist. It is known that this phenomena occurs if specific interaction between molecules exists. There are several ways describing the specific interaction. In this work it is assumed that the total number of specific interactions is distributed according to binomial distribution. In this case, exact mathematical conditions for closed miscibility loop phase behavior are derived when the specific interaction is applied to regular solution theory, quasichemical theory and Flory-Huggins lattice theory. And we investigated the effect of parameters on the phase diagram. The phase diagram of water-nicotine is calculated and compared with experimental data.

Well-Defined Thermoresponsive Copolymers with Tunable LCST and UCST in Water

  • A thermoresponsive polymer, poly(2-(dimethylamino)ethyl methacrylate) (PDMAEMA), was successfully synthesized by atom transfer radical polymerization (ATRP). Different amounts of 1,3-propanesultone were used as quaternization agent to transit the PDMAEMA into partially modified poly(zwitterions), resulting in p[DMAEMA-co-3-dimethyl(methacryloyloxyethyl)ammonium propanesulfonate] (PDMAEMA-co-PDMAPS). Molecular weight, molecular weight distribution, and degree of quarternization were determined by gel permeation chromatography (GPC) and 1H NMR spectroscopy. The transmission spectra of the 1.0 wt % aqueous solutions of the resulting polymers at 650 nm were measured as a function of temperature. Results showed that the lower critical solution temperature (LCST) and the upper critical solution temperature (UCST) could be easily controlled by the different composition of dimethylamino and zwitterion groups. The effect of partial quaternization on thermoresponsive properties was also studied by dynamic light scattering (DLS) with the same aqueous concentration (1.0 wt %) as employed for turbidimetry studies. The LCST and UCST values measured by DLS correlated well with those determined by turbidimetry.

A Semi-Interpenetrating Network for Temperature-Sensitive Polymer System

  • A semi-interpenetrating network (IPN) was prepared for a temperature-sensitive polymer system composed of sodium alginate and poly (N, N-dimethylaminoethyl methacrylate (DMAEMA)-co-ethyl acrylamide (EAAm)). The role of sodium alginate is to provide crosslinked network and that of poly(DMAEMA-co-EAAm) is to provide temperature responsiveness to the polymer system. Semi-IPN gel shows temperature-induced swelling transition at the same temperature of the lower critical solution temperature of poly(DMAEMA-co-EAAm) and its swelling kinetics is manipulated by the control of crosslinking densitv.

Thermo- and Acid/base-induced Spectral Switching of a Poly(N-isopropylacrylamide) Copolymer Containing Benzopyran-based D-π-A type Dye Units

  • A thermoresponsive poly(NIPAM-co-dye) labeled with benzopyran-based D-$\pi$-A type dye was prepared by typical radical copolymerization. It can be also constructed a acid/base-induced molecular switch by modulation of intramolecular charge transfer with protonation/deprotonation. The lower critical solution temperature behavior was investigated by means of UV-vis spectroscopy which allows the measurement of the phase transition from $25^{\circ}C$ to $45^{\circ}C$ in aqueous DMSO solution. The morphology of the internal microstructure of the poly(NIPAM-co-dye) hydrogel was observed by scanning electron microscopy. The reversible switch could be obtained by thermal and acid/base stimuli.

Deformation Property of TiC-Mo Solid Solution Single Crystal at High Temperature by Compression Test (TiC-Mo 고용체 단결정의 고온 압축변형 특성)

  • To investigate the deformation properties of TiC-(5-20) mol% Mo solid solution single crystals at high temperature by compression testing, single crystals of various compositions were grown by the radio frequency floating zone technique and were deformed by compression at temperature from 1250K to 2270K at strain rates from $5.1{\times}10^{-5}$ to $5.9{\times}10^{-3}/s$. The plastic flow property of solid solution single crystals was found to be clearly different among a three-temperature range (low, intermediate and high temperature ranges) whose boundaries were dependent on the strain rate. From the observed property, we conclude that the deformation in the low temperature range is controlled by the Peierls mechanism, in the intermediate temperature range by the dynamic strain aging and in the high temperature range by the solute atmosphere dragging mechanism. The work softening tends to become less evident with an increasing experimental temperature and with a decreasing strain rate. The temperature and strain rate dependence of the critical resolved shear stress is the strongest in the high temperature range. The curves are divided into three parts with different slopes by a transition temperature. The critical resolved shear stress (${\tau}_{0.2}$) at the high temperature range showed that Mo content dependence of ${\tau}_{0.2}$ with temperature and the dependence is very marked at lower temperature. In the higher temperature range, ${\tau}_{0.2}$ increases monotonously with an increasing Mo content.

  • Thermoresponsive polymers were successfully synthesized by a combination of atom transfer radical polymerization (ATRP) and Cu(I)-catalyzed 1,3-dipolar cycloaddition of azide and alkynes (click chemistry). Poly(2-hydroxyethyl methacrylate) (PHEMA) was synthesized by ATRP, followed by introduction of alkyne groups using pentynoic acid, leading to HEMA-alkyne. Homopolymers having secondary amine groups, tertiary amines with hydroxyethyl and hydroxypropyl groups were synthesized by adding 2-azido-N-ethyl-ethanamine, 2-[(2-azidoethyl)amino]ethanol, and 2-[(2-azidoethyl)amino]propanol, respectively, to the PHEMA-alkyne backbone using click chemistry. Molecular weight (MW), molecular weight distribution (MWD), and click reaction efficiency were determined by gel permeation chromatography (GPC) and $^1H$ NMR spectroscopy. The transmission spectra of the 1.0 wt% aqueous solutions of the resulting polymers at 650 nm were measured as a function of temperature. Results showed that the lower critical solution temperature (LCST) could be easily controlled by the length of the hydroxyalkyl groups.

Synthesis and Characterization of Poly(N-isopropylacrylamide) Containing Polydimethylsiloxane (Polydimethylsiloxane을 함유한 poly(N-isopropylacrylamide)? 합성 및 성질)

  • Poly(N-isopropylacrylamide) (PNIPAAm) containing polydimethylsiloxane (PDMS) was synthesized using PDMS as crosslinking agent, and characterized by IR and DSC. It seems that the copolymer has separated phases, PNIPAAm and PDMS. The $T_g$ of PNIPAAm was decreased in accordance with the increase of PDMS-contents. The swelling behavior of polymer in water was examined with the function of temperature and PDMS-contents as well. The equilibrium swelling ratio of polymer in water was decreased with increasing PDMS-contents, but lower critical solution temperature (LCST) was not significantly affected by the incorporated PDMS-contents.
