  • We investigated 52 livestock farming complexes in Gyeong-Gi and Incheon provinces based on low, medium, and high livestock density and groundwater quality. The objective of this study was to evaluate a relationship between nitrate N concentration in groundwater and animal factors, such as livestock density and animal species. 2,200 groundwater samples for 3 years from 2012 to 2014 at Gyeong-Gi and Incheon provinces were collected and analyzed for pH, EC, DO, ORP, temperature, major anions and cations, such as $NO_3-N$, ${HCO_3}^-$, ${PO_4}^-$, ${SO_4}^{2-}$, $Cl^-$, $NH_4-N$, $K^+$, $Na^+$, $Ca^{2+}$, $Mg^{2+}$, T-N, and TOC. Average concentration of total N for generated load density was $23,973g\;day^{-1}\;km^{-2}$ for cattle, $51,551g\;day^{-1}\;km^{-2}$ for pig, and $52,100g\;day^{-1}\;km^{-2}$ for poultry. For animal feeding species, average ratio for generated load over discharge load was 16.1% for cattle, 7.8% for pig, and 7.1% for poultry. Therefore, cattle feeding region is highly vulnerable for water pollution compared to pig and poultry feeding areas. The concentrations of chloride, nitrate, and total N in the groundwater samples were higher at high animal farming regions than other regions. The average concentration of nitrate, and chloride in groundwater samples was $5.0mg\;L^{-1}$, $16.6mg\;L^{-1}$ for low livestock density, $6.9mg\;L^{-1}$, $17.7mg\;L^{-1}$ for medium livestock density and $7.6mg\;L^{-1}$, $22.7mg\;L^{-1}$ for high livestock density and total nitrogen (T-N) was $7.7mg\;L^{-1}$ for low livestock density, $9.4mg\;L^{-1}$ for medium livestock density, $10.7mg\;L^{-1}$ for high livestock density. In conclusion, based on this research, for managing groundwater quality near livestock farming regions, $Ca-(Cl+NO_3)$ group from the Piper diagram is more efficient than using 19 factors for water quality standard.

  • Objective: An experiment was conducted to determine the effect of reduced energy density of close-up diets on metabolites, lipolysis and gluconeogenesis in cows during the transition period. Methods: Thirty-nine Holstein dry cows were blocked and assigned randomly to three groups, fed a high energy density diet (HD, 1.62 Mcal of net energy for lactation $[NE_L]/kg$ dry matter [DM]), a medium energy density diet (MD, $1.47Mcal\;NE_L/kg\;DM$), or a low energy density diet (LD, $1.30Mcal\;NE_L/kg\;DM$) prepartum; they were fed the same lactation diet to 28 days in milk (DIM). All the cows were housed in a free-stall barn and fed ad libitum. Results: The reduced energy density diets decreased the blood insulin concentration and increased nonesterified fatty acids (NEFA) concentration in the prepartum period (p<0.05). They also increased the concentrations of glucose, insulin and glucagon, and decreased the concentrations of NEFA and ${\beta}-hydroxybutyrate$ during the first 2 weeks of lactation (p<0.05). The plasma urea nitrogen concentration of both prepartum and postpartum was not affected by dietary energy density (p>0.05). The dietary energy density had no effect on mRNA abundance of insulin receptors, leptin and peroxisome proliferator-activated $receptor-{\gamma}$ in adipose tissue, and phosphoenolpyruvate carboxykinase, carnitine palmitoyltransferase-1 and peroxisome proliferator-activated $receptor-{\alpha}$ in liver during the transition period (p>0.05). The HD cows had higher mRNA abundance of hormone-sensitive lipase at 3 DIM compared with the MD cows and LD cows (p = 0.001). The mRNA abundance of hepatic pyruvate carboxy-kinase at 3 DIM tended to be increased by the reduced energy density of the close-up diets (p = 0.08). Conclusion: The reduced energy density diet prepartum was effective in controlling adipose tissue mobilization and improving the capacity of hepatic gluconeogenesis postpartum.

  • A low-cost device using the light-extinction method was developed to measure the optical smoke density in various fire experiments in the present study. The relative measurement accuracy of low-cost device was evaluated through the comparison of optical density measured by a high-cost standard device consisting of He-Ne laser, photo detector and various optical components. The low-cost device was composed of laser module, photocell and acrylic board. From the experiments using a smoke generator can be easily adjusted the smoke concentration, it was found that the low-cost device could measure the smoke density within the range of ${\pm}10%$, compared to the standard device. In addition, the reliability of low-cost device was also confirmed in the experiment using a polyethylene flame. Finally, it is expected that the low-cost device developed with real-time measurement and simple installation for measuring the smoke density will be used instead of the high-cost standard device.

  • The flavonoids are one of the most numberous and widespread groups of natural consituents. The low molecular weight of benzo-${\gamma}$-pyrone derivative are ubiquitous in plants and are vegetables nuts, seeds, leaves, flowers, and bark. The flavonoids constitute of a large class of compounds ubiquitous in plants containing a number of phenolic hydroxyl groups attached to ring structures conferring the antioxidant activity. Epidemiologic studies suggest that the dietary intake of antioxidants constitutes a risk factor for vasclar disease indicating that oxidation may be important in the pathogenesis of human athero-sclerosis. Elevated plasma low density lipoprotein (LDL) cholesterol concentration are associated with accelerated atherosclerosis, LDL is oxidized by smooth muscle cells resulting in several chemicals and physical changes of LDL. Oxidized LDL is responsible for cholesterol loading of macrophages foam cells formation and atherogenesis. There have been insulficient tests of the protective effects of flavonoids against LDL oxidation to make definitive statements about their structure activity relationships. How-ever hydroxylation of the flavone uncleas can appears to be advantageous because polyhydroxylated aglycone flavonoids are potent inhibitor of LDL modification. This identification may lead to new and more effective antioxidant strategies for abrogating the atherosclerotic process the leading cause of death and disability in industrialized societies.

  • The concentration changes of discharged wastewater effluents due to ambient current flows and density stratifications in an outfall mixing zone have been investigated by using the outfall mixing zone analysis of Huang et al.[1996]. This analysis was applied to Masan sea outfall case and the concentration distributions of wastewater effluent discharges were simulated using three month period of current-meter data measured in the outfall site. Hourly concentration distributions of wastewater effluents were averaged for the period of 15 days which covers the flow conditions of the neap and the spring tidal currents in Masan Bay. The results show that the wastewater concentrations in the Masan outfall mixing zone were very low due to the higher dilution rates during the period of strong ambient currents and less density stratifications. The higher concentrations in the mixing zone were found in August because of strong density stratifications with low ambient currents. The mixing zone was extended to the west coast beach area because of major tidal current directions. This result can be used to explain the dynamical reasons for the depositional distribution of the contaminated sediments in Masan sea outfall area.

  • This study was designed to develope the antioxidative activity on oxidation of human low density lipoprotein(LDL) from marine microbials. Bacillus KS-96 producign antioxidant have been isolated and identified from seawater, Bacillus sp. KS-96. The optimal medium pH was 7.0 and incubation temperature was 30$^{\circ}C$. The antiosidant of potential substance produced extracellularly in the culture broth by Bacillus sp. KS-96 was obtained by elution of silica gel culumn chromatography with hexane, ethylacetate and water. The ethylacetate faction are shown at highest level of antioxidant activity using thiocyanate method among them. By IR, NMR, and GC/MS, antioxidant purified from ehtylacetate fraction was identified and named as 2,6-dimethoxyphenol. 2,6-dimethoxyphenol inhibited the metal mediated oxidation of human LDL at concentration of 50∼100 ${\mu}$g/mL in the presence of 5uM CuSO4 with macrophage or J774 cells.

  • The methanol extract of Taraxacum coreanum Nakai (Compositae) was examined on the in vitro oxidation of human low density lipoprotein (LDL). It is well known that LDL oxidation induced artherosclerosis, if we can protect LDL oxidation process, excess plasma lipoprotein accumulation into the arterial lesion prone areas can be blocked. The methanol extract was treated with oxidized LDL which was incubated with $16\;{\mu}M$ $Cu^{2+}$ for metal catalyzed oxidation and TBA value, mobility on agarose gel and formation of conjugated diene and change of vitamin E were determined for the evaluation. The extract showed antioxidative effect at concentration $200\;{\mu}g/ml$ on LDL oxidation.

  • Immunosensor for the determination of LDL(Low-Density Lipoprotein), a good indicator for the diagnosis of atherosclerosis and hypercholesterolemia, was developed by using quartz crystal microbalance(QCM). The immunosensor consists of flow-through cell, oscillating circuit, oscilloscope, and frequency counter. FIA(Flow Injection Analysis) was applied to the QCM system for the measurement of LDL in liquid phase. Antibody showing binding affinity against LDL was immobilized on the gold electrode of a quartz crystal by covalent coupling via polyethylenimine / glutaredehyde. LDL was injected and bound to the antibody immobilized on the QCM immunosensor. The response of the immunosensor (F0 - F1) was found to be proportional to the LDL concentration from 200 $\mu\textrm{g}$/ml to 800 $\mu\textrm{g}$/ml. Operational conditions for the operation of immunosensor were also investigated in terms of sensitivity and non-specific binding.

  • This study was undertaken to evaluate an antioxidative activity of silymarin and silybin obtained from Silybum marianum against oxidation of human low density lipoprotein (LDL). The electrophoretic mobility observed apparently was higher phase for LDL oxidized by macrophages compared to native LDL. Silymarin and silybin inhibited the copper-catalysed oxidation of human LDL in a dose-dependent manner. Silymarin and silybin at the concentration of 50 $\mu$M/ml also inhibited the copper catalysed oxidation of LDL induced by the cell J774 and macrophages. LDL reisolated from the cell incubation in the presence of silymarin or silybin was degraded at rates similar with native LDL. Silymarin or silybin found to be potential inhibitors against oxidation of $^{125}$I-LDL by macrophages and endothelial cells.
