Effect of Paclobutrazol on Growth, and High Temperature and Drought Stress in Perennial Ryegrass (Paclobutrazol 처리가 Perennial Ryegrass의 생육 및 고온과 건조 Stress에 미치는 영향)

  • This study was conducted to investigate the effect of paclobutrazol [(2 RS , 3 RS )1-(4- chlor-ophenyl )-4, 4- dimethyl -2- (1, 2, 4- triazol -1- yl )- pentan -3-01] on the tolerance of hi-gh temperature and drought stress as related to growth retardation , iranspiration rate , soil water content , nitrogen level and photosynthetic rate in perennial ryegrass ( Loliurn perenne L . ' Omega H , ). Plants were given a 30 ml soil drench of paclohutrazol at the concentrations of 0, 0.01, 0.1, 1.0, 10.. 0, mg / 6 .5cm- diameter pot . The rcsults were as follows : 1. Increasing concentrations of paclohutrazul reduced plant height , leaf area , fresh weight and dry weight , hut increased chlorophyll content per unit area . The number of tillers and leaf width were not affected hy the paclobutrazol concentrations . 2. The proper concentration of paclohatrazol on growth retardation in perennial ryegrass was about I mq /pot , hut leaf deformity and severe growth retardation were shown at high concentration of 10 mq / pot . 3. Perennial ryegrasses grown at 30˚C were shown significantly short plant height and low leaf nitrogen level compared with those grown at 20˚C. Increasing concentrations of paclohutrazol at 20˚C increased nitrogen level hut it could not increase nitrogen level at 30˚C . 4. During the drought stress , increasing temperatures significantly promoted transpiration rate and wilting time . It took about 5 days at 20˚C and 3 days at 30˚C to reach wilting time of leaves from water stress treatment . Soil water contents at wilting time of non-treated controls were averaged 6. 871% at 20˚C and 6. 17% at 30˚C 5. Paclohutrazol reduced transpiration rate at high temperature and drought stress . Wilting appeared at the lower water content of soil according to increasing concentrations of paclobutrazol at 30˚C hut there were no differences among concentrations of at 20˚C. 6.Paclohutrazol treatment at 1 rag /pot reduced injury rate of leaves from 67.1 % and 100 % in control plants to 15.7% and 80% at 20˚C and 3010, respectively. 7. Photosynthetic rate per unit area was significantly reduced at high temperature . Paclohutrazol stimulated photosynthetic rate with increase of concentrations at 20˚C but there was no increasing effect at 30˚C.

Combined Effects of Irradiance-Salinity and Temperature-Salinity on the Growth of Enteromorpha compressa (Chlorophyta) in Laboratory Culture (실내배양에서 해산 녹조 납작파래 (Enteromorpha compressa)의 생장에 미치는 광도와 염분 및 온도와 염분의 복합효과)

  • The effects of environmental factors on the growth of Enteromorpha compressa germlings from Daeyulri (34$^{\circ}$36'N; 127$^{\circ}$47'E), the southern cost of Korea were examined in laboratory culture through combinations of irradiance and salinity and temperature and salinity. They showed a maximum growth rate at 125 $\mu$E.m-2.s-1 and 32$\textperthousand$ of irradiance and salinity combination, and at 15$^{\circ}C$ and 32% of temperature and salinity combination. Optimal parameters for the growth of germlings were 15$^{\circ}C$, 125 $\mu$E.m-2s-1 and 32$\textperthousand$. The germlings of E. compressa were survived in a wide range of irradiance, temperature and salinity levels, even though they had relatively low irradiance optimum. In the field E. compressa occurred commonly during autumn and spring seasons and disappeared in summer, except for particular habitats. This may be caused by the salinity and water temperature of this area rarely drop below 26$\textperthousand$ and 8$^{\circ}C$ during winter. A broad tolerance to environmental and rapid growth of germlings made them a wide geographical distribution over the world and a survival in both the upper and lower intertidal zones.

  • SiC fiber-reinforced SiC matrix composite is a promising accident-tolerant fuel cladding material to improve the safety of light water nuclear reactors. Compared to the current zirconium alloy fuel cladding as well as metallic accident-tolerant fuel cladding, SiC composite fuel cladding has exceptional accident-tolerance such as excellent structural integrity and extremely low corrosion rate during severe accident of light water nuclear reactors, which reduces reactor core temperature and delays core degradation processes. In this paper, we introduce the concept, technical issues, and properties of SiC composite accident-tolerant fuel cladding during operation and accident scenarios of light water nuclear reactors.

Mechanisms of Chilling Tolerance in Relation to Antioxidative Enzymes in Rice

  • In order to examine the mechanistic basis for differential sensitivities to chilling and subsequent recovery between two rice (Oryza sativa L.) cutivars, a chilling-tolerant japonica type (Ilpumbyeo) and a chilling-susceptible indica type (Taebaekbyeo), changes of physiological responses and antioxidant enzymes were investigated. Both cultivars at 3 leaf stage were exposed at a low temperature of $5^{\circ}C$ for 3 days and subsequently recovered in a growth chamber at a $25^{\circ}C$ for 5 days with 250 mmol $m^{-2}$ $s^{-1}$. Physiological parameters such as leaf fresh weight, relative water content, cellular leakage, lipid peroxidation, and chlorophyll a fluorescence showed that the chilling tolerant cultivar had a high tolerance during chilling. However, the chilling-susceptible cultivar revealed severe chilling damages. The chilling-tolerant cultivar was also faster in recovery than the chilling-susceptible cultivar in all parameters examined. We analyzed the activity and isozyme profiles of four antioxidant enzymes which are: superoxide dismutase (SOD), caltalase (CAT), ascorbate peroxidase (APX), and glutation reductase (GR). We observed that chilling-tolerance was due to a result of the induced or higher antioxidant enzyme system, CAT and APX in leaves and SOD, CAT, APX, and GR in roots. Especially, we observed the most significant differences between the chilling-tolerant cultivar and -susceptible cultivar in CAT and APX activity. Also in isozyme profiles, CAT and APX band intensity in the chilling-tolerant cultivar was distinctively higher than in the chilling-susceptible cultivars during chilling and recovery. Thus, the cold stability of CAT and APX are expected to contribute to a tolerance mechanism of chilling in rice plants. In addition, the antioxidative enzymes activity in roots may be more important than in that of leaves to protect chilling damage on rice plants.

Evaluation of Cold Tolerance-Related Traits of Recombinant Inbred Lines in Rice (벼 재조합 자식계통의 내냉성 관련 형질 분석)

  • This study was carried out to construct cold-tolerance characteristics. The RILs were developed from progenies of a cross between cold-susceptible Tongil-type rice variety, Milyang 23 and cold-tolerant Japonica rice variety, Stejaree 45 by single seed descent methods. The 175 RILs $(F_8)$ were evaluated for cold tolerance traits by field screening under cold-water irrigation. Frequency distribution of RILs in leaf discoloration, heading delay, culm length reduction and number of spikelets reduction displayed nearly normal distributions with transgressive segregations to either side of parents, while the spikelet fertility reduction and panicle exsertion at low-temperature showed the more or less skewed continuous distribution toward the susceptible parent. Higher heritabilities over $60\%$ were observed in leaf discoloration, spikelet fertility reduction, panicle exsertion, while relatively lower heritabilities less than $40\%$ were observed in culm length reduction, number of spikelets reduction and grain yield reduction. Some cold-tolerance RILs were selected effectively by cold water irrigation, which are expected to be good materials in breeding program for cold tolerance.

Comparative Transcriptome Analysis Reveals Differential Response of Phytohormone Biosynthesis Genes in Glumous Flowers of Cold-Tolerant and Cold-Sensitive Rice Varieties Upon Cold Stress at Booting Stage

  • Low temperature stress is one of the major negative factors affecting vegetative and reproductive growth of rice. To better understand responses of rice plants to low temperature we analyzed transcriptome expression patterns in glumous flower of cold-tolerant japonica rice variety, Stejaree45, and cold-susceptible variety, HR19621-AC6 at booting stage under cold water irrigation. A total of 2,411 probes were differentially expressed by low temperature in glumous flowers of the two varieties. Some important genes involved in hormone biosynthesis showed variety-specific regulation. Expression of GA20ox3 and GA2ox, among the genes involved in GA biosynthesis, was regulated differentially in the two varieties. Among the genes involved in IAA biosynthesis, YUCCA1 and TAA1:1 showed variety-specific regulation. Among the genes involved in cytokinin biosynthsis and signaling, expression of LOG, HK1 and HK3 was significantly down-regulated only in the cold-susceptible variety. Among the genes involved in ABA biosynthesis, NSY and AAO3 were down-regulated only in the cold-tolerant variety. In general, genes involved in GA, IAA and cytokinin biosynthesis responded to cold temperature in such a way that capacity of those bioactive hormones is maintained at relatively higher levels under cold temperature in the cold-tolerant variety, which can help minimize cold stress imposed to developing reproductive organs in the cold-tolerant variety.

Advanced Wastewater Treatment Process Using Aerobic Granular Sludge (AGS-SBR) (호기성 그래뉼 슬러지를 이용한 하수고도처리기술(AGS-SBR))

  • Aerobic granular sludge (AGS) can be classified as a type of self-immobilized microbial aggregates measuring more than 0.2 mm. It offers the option to simultaneously remove COD, N, and P that occur in different zones inside a granule. Also, AGS is characterized by high precipitability, treatability with high organic loading, and high tolerance to low temperature. In this study, a sequencing batch reactor inoculated with AGS (AGS-SBR) is a new advanced wastewater treatment process that was proven to grow AGS with integrated nutrient removal and low C/N ratio. A pilot plant, AGS-SBR with a capacity of 225 ㎥/d was installed at an S sewage treatment plant in Gyeonggi-do. The results of the operation showed that the water quality of the effluent indicated that the value of BOD5 was 1.5 mg/L, CODMn was 11.4 mg/L, SS was 6.2 mg/L, T-N was 13.2 mg/L, and T-P was 0.197 mg/L, and all of these values reliably satisfied an effluent standard (I Area). In winter, the T-N treatment efficiency at a lower temperature of less than 11℃ also showed reliability to meet the effluent standard of the I Area (20 mg/L or less). Analysis of microbial community in AGS showed a higher preponderance of beneficial microorganisms involved in denitrification and phosphorus accumulation compared with activated sludge. The power consumption and sludge disposal cost were reduced by 34.7% and 54.9%, respectively, compared to the domestic SBR type sewage treatment plant with a processing capacity of 1,000 ㎥/d or less.

Physiological and Ecological Studies on the Low Temperature Damages of Rice (Oryza sativia L.) (수도의 저온장해에 관한 생리 생태학적 연구)

  • Experiments were conducted to investigate rice varietal response to low water and air temperatures at different growth stages from 1975 to 1980 in a phytotron in Suweon and in a cold water nursery in Chooncheon. Germination ability, seedling growth, sterility of laspikelets, panicle exertion, discoloration of leaves, and delay of heading of recently developed indica/japonica cross(I/J), japonica, and indica varieties at low air temperature or cold water were compared to those at normal temperature or natural conditions. The results are summarized as follows: 1. Practically acceptable germination rate of 70% was obtained in 10 days after initiation of germination test at 15\circ_C for japonica varieties, but 15 days for IxJ varieties. Varietal differences in germination ability at suboptimal temperature was greatest at 16\circ_C for 6 days. 2. Cold injury of rice seedlings was most severe at the 3.0-and 3.5-leaf stage and it was reduced as growth stage advanced. A significant positive correlation was observed between cold injury at 3-leaf stage and 6-leaf stage. 3. At day/night temperatures of 15/10\circ_C seedlings of both japonica and I/J varieties were dead in 42 days. At 20/15\circ_C japonica varieties produced tillers actively, but tillering of I/J varieties was retarded a little. At 25/15\circ_C, both japonica and I/J varieties produced tillers most actively. Increase in plant height was proportional to the increase in all varieties. 4. In I/J varieties the number of differentiated panicle rachis branches and spikelets was reduced at a day-night temperature of 20-15\circ_C compared to 25-20 or 30-25\circ_C, but not in japonica varieties although panicle exertion was retarded at 20-15\circ_C. The number of spikelets was not correlated with the number of primary rachis branches, but positively correlated with that of secondary rachis branches. 5. Heading of rice varieties treated with 15\circ_C air temperature at meiotic stage was delayed compared to that at tillering stage by 1-3 days and heading was delayed as duration of low temperature treatment increased. 6. At cold water treatment of 17\circ_C from tillering to heading stage, heading of japonica, I/J, and cold tolerant indica varieties was delayed 2-6, 3-9, and 4-5 days, respectively, Growth stage sensitive to delay of heading delay at water treatment were tillering stage, meiotic stage, and booting tage in that order, delay of heading was greater in indica corssed japonica(Suweon 264), japonica(Suweon 235), and cold tolerant indica(Lengkwang) varieties in that order. Delay of heading due to cold water treatment was positively correlated with culm length reduction and spikelet sterility. 7. Elongation of culms and exertion of panicles of rice varieties treated with low air temperature 17\circ_C. Culm length reduction rate of tall varieties was lower than that of short statured varieties at low temperature. Panicle exertion was most severaly retarded with low temperature treatment at heading stage. Generally, retardation of panicle exertion of 1/1 varieties was more severe than that of japonica varieties at low temperature. There was a positive correlation between panicle exertion and culm length at low temperature. 8. The number of panicles was increased with cold water treatment at tillering stage, but reduced at meiotic stage. As time of cold water treatment was conducted at earlier growth stage, culm length was shorter and panicle exertion poorer. 9. Sterility of all rice varieties was negligible at 17\circ_C for three days but 30.3-85.2% of strility was observed for nine-day treatment at 17\circ_C. Among the tested varieties, sterility of Suweon 264 and Milyang 42 was highest and that of Suweon 290 and Suweon 287 was lowest. The most sensitive growth stage to low temperature induced sterility was from 15 to 5 days before heading. There was positive correlation between sterility of rice plants treated with low temperature at meiotic and heading stage. 10. Percentage of spikelet sterility was greatest at cold water treatment at meiotic stage (auricle distance -15~-10cm) and it was higher in 1/1 (Suweon 264, Joseng tongil), japonica (Nongbaek, Towada), and cold tolerance indica(Lengkwang) varieties in the order. Level of cold water and position of young-ear affected on the sterility of varieties at meiotic stage; percentage of spikelet sterility of variety, Lengkwang, of which young-ear was located above the cold water level was high, but that of short statured variety, Suweon 264, of which young-ear was located in the cold water was lower. 11. Percentage of ripened grains was not reducted at 15\circ_C air temperature for three days at full heading stage in all varieties. However, at six-day low temperature treatment Suweon 287, Suweon 264 showed percentage of ripended grains lower than 60%, but at nine-day low temperature treatment all varieties showed percentage of ripened grains lower than 60%. Low temperature treatment of 17\circ_C from 10 days after heading for 20 days did not affect on the ripening of all varieties. 12. Uptake of nitrogen, phosphorous, potassium, calcium, and magnesium in whole plants was higher at average air temperature of 25\circ_C, but concentration of the elements was lower compared to those at 19\circ_C. However, both total uptake and concentration of manganese were higher at 19\circ_C compared to 25\circ_C. 13. Higher application of nitrogen, phosphorus, silicate, and compost increased yield of rice due to increased number of panicles and spike let fertility in cold water irrigated paddy.

The Effects of Water Level and Temperature on Seed Germination and Early Seedling Development of Rice (Oryza sativa I.)

  • The application of direct seeding cultivation reduces time, labor, and cost. However, this application often has poor seedling establishment and leads to lower yield as compared to transplanting system. The tolerance to anaerobic and low temperature germination is important to improve seedling establishment and the wide-spread application of direct seeding method. This study was carried out to evaluate the responses of three japonica cultivars to different temperatures (15℃, 18℃, 21℃, 24℃, and 27℃) and different flooding levels (1 cm, 4 cm, 7 cm, 10 cm) during germination and seedling development. The mean survival percentage significantly increased (P<0.05) when the flooding level decreased and when temperature increased. There were significant effects of the interaction between temperature and water depth on survival percentage and seedling height. When temperature decreased from 27℃ to 15℃, the germination duration significantly increased from 6.4 days to 16.3 days while the germination speed, survival percentage, and seedling height decreased from 5.3 seeds day-1, 61.9% and 190.6 cm to 2.2 seeds day-1, 33.2%, and 47 cm, respectively. The increase in temperature under submergence condition was associated with the increased expression of Amy3D and ALDH2a but the decreased expression of ADH1 and PDC1. The results of this research would be used for further studies and breeding programs to improve rice seedling establishment and the application of direct seeding cultivation.

A Survey on the Aged Consumers' Needs for an Elderly Friendly and Sensitive Bathtub Alternative (노인 친화적 감성용 욕조 대안에 대한 노인 소비자의 요구 및 평가)

  • This study finds out the aged consumers' needs on the developing elderly friendly and sensitive bathtub. This bathtub focuses on enhancement of multi-senses and enhancement of health, safety & convenience during bath. The enhancing multi-senses would have four functions; therapy with color and aroma, the massage with micro water bubbles, the visual and auditory with TV & cinema, and the auditory with music and radio. Also, enhancing others of bathtub would have seven functions; emergency bell, blood-pressure check system for health condition, water temperature indicator, keeping water warm, reading a book in that, and controlling of house entrance door and a getting the phone during bath. The aged consumers' needs find throughout a questionnaire survey, and 497 data was analyzed. The main results are as follow. They consider a full bath behavior as a healthcare. They have an intention to purchase or use the elderly and friendly bathtub, and they prefer using it in common facilities to using it at home. They highly need health related function of bathtub, which are massage (64.4%), blood-pressure check system (55.1%) and aroma therapy (45.7%). The emergency bell (67.0%), keeping water warm (62.2%), and water temperature indicator (49.7%), related to safety & information function of bathtub, are needed. Also, they think tolerance for error and low physical effort in bathtub design alternative, opening door and inside sitting support, to be important.
