In this study, the toxic effects of PFOA and PFOS potassium salt on Mesocentrotus nudus using 10 min-fertilization rate and 48 h-normal embryogenesis were confirmed through the calculation of toxicity values such as Non-observed effective concentration, Low-observed effective concentration, and 50% of effective concentration. The case of 10 min-fertilization rate and 48 h-normal embryogenesis showed the concentration-dependent reduction pattern when exposed to PFOA and PFOS potassium salt, in tested concentration, respectively. The EC50 values of 10 min-fertilization rates for PFOA and PFOS potassium salt were 1346.43 mg/l and 536.18 mg/l, respectively, and the EC50 values of 48 h-normal embryogenesis were 42.67 mg/l and 17.81 mg/l, respectively. Both toxicity test methods showed high toxicity sensitivity to PFOS potassium salt. Recent studies have shown that the concentration of PFOA and PFOS in the marine environment has continuously decreased, and it is not enough to show acute toxicity to sea urchin. However, PFOA and PFOS have a very long half-life and can accumulate throughout the life of marine life, so it is still observed at a high concentration in shellfish. Therefore, a study on chronic toxicity through the whole-life cycle of marine organisms in coastal environments should be needed.
This study was carried out to investigate the optimal nitrogen concentration level suitable for forage rice growth by hydroponic cultivation in the salinity concentration of 0.1~0.3% which is similar to that of Muan reclaimed paddy field, and based on this results, to estimate optimal nitrogen fertilization level by field experiment in Muan reclaimed paddy for maximum forage production by cultivation of Yeongwoo rice. As a result of the growth response to the salt and nitrogen concentrations in the hydroponic cultivation experiment, the growth amount increased as the nitrogen concentration increased in the range of 0~24 me/L in the absence of salt stress. However, at a salt concentration of 0.1~0.3%, the growth amount was the highest at a nitrogen concentration of 12 me/L, and at higher nitrogen concentrations of that, the rice growth decreased as the nitrogen concentration increased. Therefore, nitrogen concentration of 12 me/L was judged to be an appropriate concentration for forage rice growth at salt concentration of 0.1~0.3%, and a nitrogen fertilization amount level corresponding to a nitrogen concentration of 12 me/L was actually applied to the Muan reclaimed paddy field for forage rice cultivation during two years. The amount of nitrogen fertilizer was tested with three treatments, which are 18 kg/10a considered appropriate, and 1.5 times and 2 times of the appropriate amount, and the planting density was tested with 2 treatments of 15 hills/m2 and 26 hills/m2. As a result of the reclaimed paddy field experiment, the yield was the highest when nitrogen fertilizer was applied at 18 kg/10a in the planting density of both treatments. Looking at the yield according to planting density, the high planting density plot yielded higher than the low planting density plot. In other words, when the planting density was 26 hills/m2 and the nitrogen fertilization amount was 18 kg/10, the highest dry matter yield of 1,763 kg/10a was obtained. From the results of hydroponics and reclaimed field experiments, we could conclude that the productivity of forage rice decreased more as the nitrogen concentration increased when the nitrogen concentration was higher than the optimal level under salt stress.

To restore reclaimed land, it needs to be supplemented with organic matter; this is especially true for Korea, where organic matter constitutes only one-tenth of conventional agricultural soils. The giant Miscanthus, a perennial grass known for its extensive biomass, shows signs of being an excellent source of organic matter for restoring reclaimed land. Therefore, the objectives of this study were to (i) evaluate the feasibility of using the giant miscanthus as an organic resource within the context of re-using reclaimed land for agricultural purposes (i.e., potato cultivation), and (ii) determine the optimum fertilization rate for the potatoes while the giant miscanthus is being used as an organic resource. Our results show that after 180 days, giant miscanthus lost 23-47% of its original dry weight, with the extent of the loss dependent on soil salinity. Nutrient concentrations (Mg2+, Na+) continued to increase until the end of the study period. In contrast, potassium (K+) and the ratio of carbon to nitrogen (C/N) decreased until the end of the study period. Specifically, after 180 days, low salinity topsoil treatments had the lowest C/N ratio. In the first year, 150 % of standard N rates were required for the potatoes to achieve maximum productivity; however in the 2nd year, standard rates were sufficient to achieve maximum productivity. Overall, this implies that even though the application of giant miscanthus did eventually improve soil quality, increasing crop yields, N fertilization is still necessary for the best outcomes.
Background: Asarum sieboldii Miq., a species of forest understory vegetation, is an herbaceous perennial belonging to the family Aristolochiaceae. The metapopulation of A. sieboldii is distributed sparsely and has a short seed dispersal distance by ants as their seed distributor. It is known that many flowers of A. sieboldii depend on self-fertilization. Because these characteristics can affect negatively in genetic structure, investigating habitat structure and assessment of genetic structure is needed. A total of 27 individuals in a valley were sampled for measuring genetic diversity, genetic distance, and genetic differentiation by RAPDPCR. Results: The habitat areas of A. sieboldii metapopulation were relatively small (3.78~33.60 m2) and population density was very low (five to seven individuals in 20×20 m quadrat). The habitat of A. sieboldii was a very shady (relative light intensity = 0.9%) and mature forest with a high evenness value (J = 0.81~0.99) and a low dominance value (D = 0.19~0.28). The total genetic diversity of A. sieboldii was quite high (h = 0.338, I = 0.506). A total of 33 band loci were observed in five selected primers, and 31 band loci (94%) were polymorphic. However, genetic differentiation along the valley was highly progressed (Gst = 0.548, Nm = 0.412). The average genetic distance between subpopulations was 0.387. The results of AMOVA showed 52.77% of variance occurs among populations, which is evidence of population structuring. Conclusions: It is expected that a small-scale founder effect had occurred, an individual spread far from the original subpopulation formed a new subpopulation. However, geographical distance between individuals would have been far and genetic flow occurred only within each subpopulation because of the low density of population. This made significant genetic distance between the original and new population by distance. Although genetic diversity of A. sieboldii metapopulation is not as low as concerned, the subpopulation of A. sieboldii can disappear by stochastic events due to small subpopulation size and low density of population. To prevent genetic isolation and to enhance the stable population size, conservative efforts such as increasing the size of each subpopulation or the connection between subpopulations are needed.

In recent years, an increasing number of studies on pig in vitro maturation(IVM) and in vitro fertilization(IVF) have been separated. the wide range of new technologies, including that in applied molecular genetics, has increased this interest. the production of viable porcine embryos in vitro is a prerequisites for the successful production of transgenic pigs to date. The efficiency of IVM/IVF techniques in the porcine is lower than that obtained in other species such as cattle and mouse. The several problems are generally thought to be the cause of poor results: the low rate of MPN formation derived from inadequate IVM of oocytes, the high incidence of polyspermy after IVF and cell blocking at 4 cell during embryos culture. For there reasons overcoming, many studies have been conducted to improve in vitro embryo-genic competence of oocytes. In the last several years, many maturation culture media have been evaluated and various exogenous factors such as hormones and grows factors have been tested to improve the efficiency of porcine in vitro system. In the study several antioxidants have been examined to improve in vitro fertilization and development of porcine oocytes. In this study, several antioxidants were examined to determine the effects on the development of oocytes to the cleavage, morula and blastocyst stage when added at the maturation(IVM) or in vitro fertilization(IVF) or in vitro culture(IVC) of porcine embryos. Porcine oocytes were matured, fertilized and embryos were cultured in defind conditioned medium in vitro with or without supplementation with the antioxidents of cysteine, catalase and glutathione. 1. Significant improvement of blastocyst rate (27.2% versus 15.4%, p<0.05) were achieved when catalase(500U/$m\ell$) were added to TCM-199 medium and morula rate(72.0% versus 53.9%, p<0.05) were significantly higher when glutathione(1.0mM/$m\ell$) were added to TCM-199 medium than those of control. 2. In mTBM medium for oocytes fertilization, the addition of cysteine, catalase and glutathione had no positive effect on embryonic development. glutathione had no positive effect on embryonic development. In conclusion, this study shows that addition of catalase, gluththione during IVM improved the rate of porcine embryo development.

This pot experiment was conducted to find out the effects of boron application($B_{0}$ ; control, $B_1$; 0.2, $B_2$; 2.0, B$_3$; 6.0, $B_4$; 15.0 boron me/pot) on the forage performance of pure and mixed cultures of orchardgrass and white clover. The third part was concerned with the changes in the contents and yields of nitrogen compounds(crude/pure protein and soluble N-compound) in forages. The results obtained are summarized as follows: 1. With no additional fertilization, especially nitrogen, in a pure culture, the $B_{0}$ and $B_4$ treatments on white clover decreased the amount of crude/pure protein, and showed nitrogen deficiency symptoms. However, the optimum boron application($B_2$) positively resulted in the increase of crude and pure protein, especially pure protein, and the content ratio of pure protein/soluble N-compounds. With additional fertilization, especially nitrogen, differences were not found among the boron treatments($B_{0}$, $B_2$, and $B_4$). 2. Owing to the decline of white clover as affected by the additional fertilization, especially nitrogen, in the grass-clover mixed cultures, the effects of B-application on these contents of white clover were different and relatively low, compared with the pure cultures. But the positive effect of $B_2$ treatment tended to be similar to the pure cultures. Also, it was recognized that the $B_2$ treatment resulted in the increase of their contents in orchardgrass, however, the effect was relatively minor compared with that of white clover. 3. The optimum B application(B$_2$) on white clover influenced relatively better on the yields of crude and pure protein than on the dry matter yields, especially with no additional fertilization. The effects of boron application on the contents and yields of crude and pure protein were different according to the forage species, whether it was a pure or mixed culture, and additional fertilization.
We evaluated the hatchability of fertilized eggs from six hybrid combinations of highly valued grouper species inhabiting temperate and warm waters, with the goal of establishing a novel hybrid with enhanced growth and viability during the culturing period in the temperate waters of Korea. Hybrid combinations with red-spotted grouper females exhibited high hatchability with high a fertilization and hatching rate of fertilized eggs and a low deformity rate of hatched larvae. Conversely, hybrid combinations with kelp grouper females had very low hatching rates and very high deformity rates; commercial production of seed from such crosses would be difficult without improving hatchability. The hatchabilities of convict grouper ♀×giant grouper ♂ and kelp grouper ♀×red-spotted grouper ♂ were lower than those of maternal purebreds, but these two hybrid combinations were expected to produce potentially large quantities of hatched larvae. In the above evaluation, promising hybrid combinations were identified for commercial production of seed. For these hybrids to contribute to the development of Korea's mariculture industry, mass production of fertilized eggs and seeds is necessary, along with the development of advanced rearing techniques, such as the identification of a suitable rearing temperature.

To develop policies regarding fertility and pregnancy that will be effective in preventing further declines in fertility rates in the context present-day Korea, current policies in Japan were analyzed and compared with those now being implemented in Korea. This study was structured to involve (a) comparison of maternal health projects in Korea and Japan, and (b) comparison of infertility support policies based in regional cities in Korea and Japan. Korea's Health Plan 2030 emphasizes strengthening healthcare for high-risk pregnant women, expanding investments to benefit vulnerable groups, and establishing a support system for infertile couples. In Japan, government programs involving treatment targeting infertility specifically were implemented nationwide in 2006. Wide dissemination of accurate knowledge related to pregnancy is emphasized. Also, counseling centers specializing in infertility were established by 67 local governments. We have confirmed that Korean policies include decentralization, while Japan is implementing the central government's infertility policy uniformly in all regions. Japan also adjusted its policy out of concern that problems related to infertility and childbirth will worsen due to the social disaster of COVID-19. The results indicate that providing additional support for psychological counseling may be preferable to increasing the number of in-vitro fertilization procedures. The physical burden on women may be minimized by benchmarking policies in Japan. Step-by-step application of these procedures should be systematically supported to achieve the best results.
Objective: The aim of this study was to compare the complication rates of oocyte pick-up (OPU) procedures via transvaginal ultrasonography in patients with different levels of ovarian reserve. Methods: In total, 789 patients who underwent OPU procedures for in vitro fertilization (IVF) were included in the study. Results: Individuals with normal ovarian reserve had a 2.947-fold higher risk of complications in OPU procedures than individuals with low ovarian reserve, and individuals with high ovarian reserve had a 7.448-fold higher risk of complications than individuals with low ovarian reserve. In addition, a higher number of IVF trials was associated with an increased risk of complications. Conclusion: The results of this study show that OPU has a higher risk of complications, particularly severe pain, in patients with high ovarian reserve. It is thought that complications can be reduced by preferring mild stimulation in patients with high ovarian reserve. Collecting fewer oocytes is also associated with a lower risk of complications from OPU. Even if a patient's reserve is very good, fewer and higher-quality oocytes should be targeted with the use of the lowest possible dose of drugs.
The development of efficient method for the production of transgenic mice has been investigated in our laboratory. This study was conducted to develop the artificial insemination in the mouse. Spermatozoa were collected from the cauda epididymis of ICR males(age:12~15 weeks, Body weight : 30g) and artificially inseminated into the intrauterine via cervix of hormone-primed ICR females(age: 6~8 weeks, body weight: 25g) using the capillary tube, 200~300 $\mu$m in inner diameter. The effect of concentration of sperm(80$\times$104, 40$\times$104, 20$\times$104, 10$\times$104, 5$\times$104, 3$\times$104, 1$\times$104/20${mu}ell$. The artificial insemination was succeeded but fertilization rate was very low(5~15%) compared to the natural mating and 59 normal youngs born from 60 females. Therefore, our findings suggest that it is possible to produce the great number of mice from the same orgin of male by artificial inseminatin. However, the lower pregnancy rate has to be solved to used broadly the artificial insemination in mouse.
