Changes in the RNA and Protein Synthesis at the Pre- and Post fertilization Stages of a Sea Urchin, Hemicentrotus pulcherrimus (말똥성게 (Hemicentrotus polcherrimus)의 수정전과 초기 발생동안 RNA 및 단백질합성의 변화)

  • Syntheses of RNA and protein were studied to examine changes in activating stored mRNAs during the early development of a sea urchin, Hemicentrotus pulcherimus. The rates of RNA and protein syntheses are very low in the unferilized eggs but the protein synthesis is activated upon the fertilization, while RNA synthesis remains still inactive at the same stage. These rates increase drastically at the blastula and gastrula stages, although the increases are not exactly in parallel. The protein synthesis was found to be also changing in quality during the early development from the studies by the two-dimensional gel electrophoresis.

  • Pigs are considered an ideal source of human disease model due to their physiological similarities to humans. However, the low efficiency of in vitro embryo production (IVP) is still a major barrier in the production of pig offspring with gene manipulation. Despite ongoing advances in the associated technologies, the developmental capacity of IVP pig embryos is still lower than that of their in vivo counterparts, as well as IVP embryos of other species (e.g., cattle and mice). The efficiency of IVP can be influenced by many factors that affect various critical steps in the process. The previous relevant reviews have focused on the in vitro maturation system, in vitro culture conditions, in vitro fertilization medium, issues with polyspermy, the utilized technologies, etc. In this review, we concentrate on factors that have not been fully detailed in prior reviews, such as the oocyte morphology, oocyte recovery methods, denuding procedures, first polar body morphology and embryo quality.

Polscope-Assisted Enucleation for Nuclear Transfer in Mice

  • Efficiency of somatic cell nuclear transfer was investigated in mice. First, oocyte activation was induced by SrCl₂, and the rate of development was compared with embryos from normal fertilization. Although more than one half of SrCl₂-treated oocytes developed to blastocysts (146/262, 55.7%), the rate of blastocyst formation was significantly lower than normal fertilization controls (59/79, 74.6%). Second, enucleation of oocytes was performed using Polscope that enables non-invasive visualization of metaphase spindles. Such approach could not only avoid damage of oocytes during an exposure to UV light often employed in conventional enucleation procedures, but could also assure the removal of nuclei from all oocytes operated because of monitoring the location of spindles during an entire process of enucleation. Morphologically normal blastocysts were obtained from the transfer of cumulus cell nuclei into enucleated oocytes. However, the rate of development into the blastocyst stage was still low (4/93, 4.3%). This reflects that the nuclear transfer procedure used in this study was not sufficiently optimized, and other factors may also impact greatly the efficiency of nuclear transfer. Including an induction of oocyte activation and method of enucleation tested in this study, a lot more elements are remained to be optimized to improve the efficiency of somatic cell nuclear transfer in mice.

Industrialization possibilities of purified pig sperm hyaluronidase

  • The goals of the present study were to develop a simple method for obtain highly purified pig sperm hyaluronidase (pHyase) and to assess its activity, function, and safety. In mammals, sperm-specific glycophosphatidylinositol (GPI)-anchored Hyase assists sperm penetration through the cumulus mass surrounding the egg and aids in the dispersal of the cumulus-oocyte complex. Recently, Purified bovine sperm hyaluronidase (bHyase) has been shown to enhance therapeutic drug transport by breaking down the hyaluronan barrier to the lymphatic and capillary vessels, thereby facilitating tissue absorption. Commercially available Hyase is typically isolated from bovine or ovine; which have several disadvantages, including the risk of bovine spongiform encephalopathy, low homology with human Hyase, and the requirement for relatively complex isolation procedures. This study successfully isolated highly purified pHyase in only two steps, using ammonium sulfate precipitation and fast protein liquid chromatography. The isolated Hyase had activity equal to that of commercial bHyase, facilitated in vitro fertilization, and effectively dissolved high molecule hyaluronic acid. This simple, effective isolation method could improve the availability of pHyase for research and clinical applications.

Low temperature preservation of bovine ovaries on in vitro development of oocytes (소 난소 저온 보존이 난자의 체외 발달에 미치는 영향)

  • During the ovary preservation in low temperature, the cumulus oocyte complexes(COCs) lose their developmental competences after in vitro fertilization. We used phosphate-buffered saline (PBS) as a basic solutions of at various temperatures (25, 15 or $5^{\circ}C$) and supplemented them with 1mM glucose and 0.5mM glutamine as a source of carbohydrate metabolites. After recovery of COCs and in vitro fertilization, a significantly higher number of oocytes developed into blastocysts. The developmental competence of embryos that were originated from ovaries preserved at $15^{\circ}C$ was increased compared to those of 25 or $5^{\circ}C$. The maturation rate of oocytes was not differed between 24 and 36 h at $15^{\circ}C$ but showed lower than control group (71% versus 78%). In vitro-fertilized oocytes from ovaries stored at $25^{\circ}C$ for 24 h or at $5^{\circ}C$ for 24 h had a significantly decreased developmental potentials, but at $15^{\circ}C$ did not (27% versus 29% of blastocysts to develop into day 8). With these results, bovine ovaries can be preserved at $15^{\circ}C$ for 36 h without decreasing developmental capacity of in vitro-fertilized oocyte at least to the blastocyst stage. This information provides valuable information of preserving ovaries for embryo transfer or in vitro embryo production.

Water and Nutrient Mass Balances in Paddy Field with Groundwater Irrigation in Low-Rainfall Year (저강우연도 지하수 관개 필지논에서 수도재배기간 동안의 물질수지)

  • Field experiment was performed to investigate water and nutrient mass balances in paddy field with groundwater irrigation from May to October, 2001. The total water inflow was about 1,183mm in which rainfall, overflow from upstream paddy, and groundwater irrigation accounted for 43, 30, and 27%, respectively. Notice that the precipitation of the study period was less than the average annual precipitation. The total drainage was almost balanced with the inflow and more than half of it was occurred by surface drainage. From the nutrient mass balance analysis, the T-P output (17.56kg/ha) was estimated slightly lower than the input (20.90kg/ha) and the T-N output (130.41kg/ha) was slightly greater than the input(129.24kg/ha). However, the difference was within the expectation and the nutrient mass was thought to be balanced considering uncertainties in field experiment and other activities not included in the study such as algae and soil microorganisms. The surface discharge of nutrient, which was about 10% of total nutrient output, was mainly affected by fertilization and rainfall runoff. Therefore, prudent surface drainage plan might be necessary particularly for the fertilization period to prevent degradation of receiving water quality. The study was performed under abnormally low rainfall compared to the average annual rainfall record, and further monitoring in diverse rainfalls and irrigation methods is recommended to estimate nutrient behavior in the paddy field more reasonably.

Studies on the Grassland Development in the Forest XII. Effect of nitrogen fertilization on nitrate nitrogen concentration and water soluble carbohydrate content of grasses by growing season and growth stage under shading condition (임간초지 개발에 관한 연구 XII. 차광조건하에서 질소시비수준이 계절별 , 생육시기별 목초의 질산태질소함량과 수용성탄수화물함량에 미치는 영향)

  • This experiment was carried out to determine the effects of nitrogen(N) fertilization levels(0. 120, 240 and 360 kg/ha/y) on seasonal nitrate nitrogen($NO_3$-N) concentration and water soluble carbohydrate (WSC) content of grasses grown under shading condition. The plants were sampled on 7. 14, 21, 28 and 35 days after harvesting in spring(May), summer(July) and autumn(September), 1988. respectively. Shading degree was controlled artificially ca. 45-50%, and each annual level of N was distributed 5 times equally. The $NO_3$-N concentration of grasses was very low(0.064 %) in spring, high in summer(O.l98 %) and autumn(0.234%), and NO,-N was slightly decreased with growing of grasses. Also the NO,-N concentration of grasses was increased significantly with increasing level of N. In spring, the $NO_3$-N of grasses was very safety(less than 0.1 %), regardless of plant height and N level. In summer and autumn, however, plant heights of below 40 cm(summer) and 44 cm(autumn), and N levels of over 210 kg(summer) and 140 kg(autumn) were over toxic level to animals, respectively. The WSC content of grasses was slightly decreased with grass growth. and significantly decreased with high level of N. From the above results, it is suggested that 200 kg/ha of N in this experiment is effective for forage quality. Also a little more amount of N(50-70kgltimes) in spring. and low level of N(20-30 kgltimes) in summer and autumn season may be desirable on woodland posture.

The Efficacy of Low-dose Aspirin Therapy for Controlled Ovarian Hyperstimulation in IVF-ET (체외수정시술 환자의 과배란유도시 저용량 아스피린 투여의 효용성에 관한 연구)

  • Objective : To evaluate the efficacy of low-dose aspirin on IVF outcome and endometrium in patients undergoing IVF-ET. Materials and Methods : From February, 2001 to Jun, 2001, 60 infertile patients were randomly divided into study group (28 cycles) and control group (32 cycles). The study group received a daily oral dose of 25 mg of aspirin for at least 2 weeks from first visiting day. Controlled ovarian hyperstimulation was initiated in all patients with the GnRH agonist starting in the midluteal phase of the previous cycle. Results: There were no significant differences in age of the patients, basal serum E2, LH, FSH level and endometrial thickness among two groups. There were no statistically significant differences between the study group and the control group respectively in dosage ($26.5{\pm}4.8$ vs $26.2{\pm}5.3$ amples) and duration ($10.4{\pm}4.2$ vs $9.8{\pm}5.3$ days) of gonadotropin administration, serum E2 level on the hCG administration day ($1823{\pm}342$ vs $1854{\pm}543$), LH ($14.5{\pm}2.7$ vs $14.8{\pm}3.1$), FSH ($16.7{\pm}3.4$ vs $18.3{\pm}4.7$), the number of follicles > 15 mm ($13.2{\pm}6.3$ vs $12.8{\pm}5.9$), the number of oocytes retrieved ($9.2{\pm}2.4$ vs $8.4{\pm}1.7$), the number of embryos transferred ($4.7{\pm}2.0$ vs $4.7{\pm}2.0$), fertilization rate (68.4% vs 64.5%), implantation rate (21.3% vs 17.6%), and clinical pregnancy rate (28.4% vs 26.2%). The endometrial thickness and the percentage of endometrial trilaminar pattern on hCG day were significantly higher in study group than control group ($12.9{\pm}3.7mm$ vs $10.4{\pm}2.8mm$, 78.3% vs 64.5%). Conclusion: Many reports suggest that low-dose aspirin improve ovarian response, implantation rate, fertilization rate, implantation rate, and pregnancy rate by increasing the blood flow, but we couldn't prove the significant effect of low-dose aspirin on the IVF outcome except on endometrium. This may be affected by dose of aspirin, duration, and number of patients studied. This trial is small, so our results highlight the need for a large randomized controlled trial to identify the effect of low-dose as pirin on IVF-ET outcome.

Influence of Controlled-release Fertilizer Levels on Rice Growth, Weed Control and Nitrogen Efficiency in Paper Mulching Transplanting (벼 종이멀칭이앙 시 완효성비료 수준이 벼 생육, 잡초방제 및 질소효율에 미치는 영향)

  • Recently we have interest on rice products developed by environment-friendly management. The technology of paper mulching was practised without herbicide in machine transplanting cultivation of paddy. A field experiment was conducted on Gangseo series (coarse loamy, mixed, nonacid, mesic family of Aquic Fluventic Eutrochrepts) at the National Institute of Crop Science (NICS), RDA, Suwon, Gyeonggi province, Republic of Korea in 2004. This experiment was carried out to evaluate rice growth, weed control and nitrogen efficiency by the different controlled-release fertilizer levels in paper mulching transplanting. Treatments consisted of conventional fertilization, controlled-release fertilizer (100%, 80%, 60%) compared with nitrogen amount ($110kg\;ha^{-1}$) of conventional fertilization and no nitrogen plot. Mulching paper consisted of recycled paper which was coated with biodegradable plastics. There were no differences between conventional rice transplanting and paper mulching on missing hills. Weed occurrence and control were diverse and low as fertilizer amount increased. Plant height and tiller number per hill increased as fertilizer amount decreased. There were no difference between controlled-release fertilizer 80% and conventional fertilization plot on rice growth traits. Leaf color and $NH_4{^+}-N$ in soil had similar trends. There was no difference in yield between controlled-release fertilizer 80% and conventional fertilization plot. Agronomic nitrogen-use efficiency was high as fertilizer amount decreased. Therefore, these results suggested controlled-release fertilizer 80% should be optimum amount under paper mulching transplanting of rice considering weed control, rice growth and nitrogen efficiency.

  • This experiment was carried out to increase productivity of low productive fields (now producing less than 180kg per 10a ) at the Eumseong Tobacco Experiment Station in 1976- 1977. We could produce 220kg or more of yields (Hicks ) per 10a by increasing plant density to 2778 plants/10a and applying 20% more fertilizer than the recommended amount.
