Early Morphological Development of the Brown Croaker, Miichthys miiuy (Basilewsky): Fin Differentiation, Head Dimensions, and Squamation (민어, Miichthys miiuy의 초기 형태 발달: 지느러미 분화, 두부 계측 및 비늘 도포)

  • We describe early morphological development in laboratory-reared specimens of the brown croaker, Miichthys miiuy, in relation to fin differentiation, head dimensions, and squamation. From the yolk sac stage to the flexion larval stage (a period of 12 days following hatching, at which time the larvae were <4.2 mm in total length; TL) we observed the presence of a fin-fold around the body, while the caudal fin appeared rounded and lacked scales. Rays developed in the dorsal, anal, and pectoral fins in a process that was almost complete in larvae 12 days, while ray segmentation occurred between 26 and 29 days of age. Elongation of the middle rays of the caudal fin was initiated at 32 days, and the rays were remarkably elongated by 37 days. By 68 days the caudal fin was lanceolated (50.7 mm TL). Scales began to develop from the midlateral lines of the caudal peduncle at 9.1mm TL (28 days), eventually encompassing the entire operculum (22.1 mm TL; 44 days). The head dimensions were largely stabilized at >12 mm TL (30 day).

Tolerance Capacity to Salinity Changes in Adult and Larva of Oryzias dancena, a Euryhaline Medaka (광염성 송사리 Oryzias dancena 성체 및 자어의 염분도 변화에 대한 내성)

  • Osmoregulatory capabilities of a euryhaline medaka, Oryzias dancena (Beloniformes; Teleostei), was examined with a particular emphasis on adult and larval viability during direct salinity changes. O. dancena adults were highly capable for hyper-osmoregulation as well as hypo-osmoregulation, as evidenced by no adverse effect on their viability during the direct transfer either from complete freshwater ($0^{\circ}/_{\circ\circ}$) to $40^{\circ}/_{\circ\circ}$ salinity, or from $70^{\circ}/_{\circ\circ}$ to $0^{\circ}/_{\circ\circ}$. Furthermore, the phased increase of external salinity with acclimation periods allowed them to survive at a salinity as high as $75^{\circ}/_{\circ\circ}$. However, tolerant capability to acute salinity increase in early larval stage was much less than in adult stage, based on the finding that the tolerance range of salinity increase was only $15^{\circ}/_{\circ\circ}$ from freshwater, indicating that the hyper-osmoregulation system might not be fully developed in the early larval stage. On the contrary, the hypoosmoregulation system could be more solidified in O. dancena larvae, as evidenced by their good survival even after direct transfer from $45^{\circ}/_{\circ\circ}$ to $0^{\circ}/_{\circ\circ}$. Knowledge achieved in this study could form the basis for a wide scope of researches including ecotoxicogenomics and geneexpression assay using this model species.

Spawning Performance, Embryonic Development and Early Viability under Different Salinity Conditions in a Euryhaline Medaka Species, Oryzias dancena (서로 다른 염분도 조건하에서 광염성 송사리 Oryzias dancena의 산란, 발생 및 초기 생존)

  • Effects of different salinity levels on spawning performance, embryonic development and early viability of a euryhaline medaka species, Oryzias dancena, were examined. O. dancena were able to spawn eggs in a wide range of salinity from 0 to 70$^{\circ}/_{\circ\circ}$, however, the spawning frequency was lowered in complete freshwater (0$^{\circ}/_{\circ\circ}$) and in highly salted water (70$^{\circ}/_{\circ\circ}$). Fertilization success was negatively affected when the environmental salinity was higher than the salt concentration found in normal seawater. Embryonic viability and hatching success were also inversely related with the salinity levels. Typical abnormality was observed in developing embryos incubated at high salinities (30, 45 and 60$^{\circ}/_{\circ\circ}$). In addition, the time to hatch was significantly delayed with increasing salinities: peak hatching occurred at 12~14 days post fertilization (dpf) in freshwater and at least at 17 to 18 dpf in 60$^{\circ}/_{\circ\circ}$. Mean survival rates of the hatched larvae up to 7 days post hatching (dph) were at least 97% in salinity levels ranging from 0 to 30$^{\circ}/_{\circ\circ}$. However, larvae reared in 45 and 60$^{\circ}/_{\circ\circ}$ experienced significant mortality, especially in the early phase, resulting in only 75% and 64% survival rates up to 7 dph, respectively.

Morphometric Characteristics and Fin Dimorphism between Male and Female on the Marine Medaka, Oryzias dancena

  • Sexual dimorphism is the most conspicuous difference between the sexes. This study examines possible sexual dimorphism and the relative growth patterns of morphometric characteristics in the marine medaka, Oryzias dancena for their potential to help differentiate between males and females of this species. The von Bertalanffy growth parameters estimated by a non-linear regression method were $L_{\infty}=30.2mm$, K=3.22/year, and ${\tau}_0=-0.05$. All 18 characteristics measured showed a difference between males and females from 70 days after hatching. Each of these characteristics were significantly different between sexes (ANCOVA, P<0.05), and the ratio of standard length between sexes showed that males were larger than females for all five morphometric measurements. Fin length measurements were taken for 21 distances of anal fin and 7 distances of dorsal fin between landmarks. There were all differences for all dorsal fin rays between the males and the females and there is significant difference in 70 days after their hatch when the sexual dimorphism is presented. The significant difference (P<0.05) in fin ray for male and female was more greatly seen as they grow. Male marine medaka showed more rapid growth than females, with longer length, dorsal fins and anal fins. Differences in these characteristics will be useful during experiments when it is necessary to differentiate between sexes of marine medaka.

Acute Oral Toxicity of dsRNA to Honey Bee, Apis mellifera (꿀벌에 대한 dsRNA의 급성섭식독성 평가)

  • BACKGROUND: RNA interference (RNAi) eliminates or decreases gene expression by disrupting the target mRNA or by interfering with translation. Recently, RNAi technique was applied to generate new crop traits which provide protection against pests. To establish the environmental risk assessment protocol of RNAi LMO in lab scale, we developed dsRNA expression system using E. coli and tested acute oral toxicity assay to honey. METHOD AND RESULTS: The dsRNA expression vector, L4440, was chosen and cloned 240 bp of Snf7 and GFP gene fragment. To develop the maximum dsRNA induction condition in E. coli, we tested induction time, temperature and IPTG concentration in media. To estimate the risk assessment of dsRNA to honey bee, it has been selected and cultured with dsRNA supplement for 48 hours according to OECD guideline. As a result, the optimum condition of dsRNA induction was $37^{\circ}C$, 4 hours and 0.4 mM IPTG concentration and the difference between Snf7 and GFP dsRNA molecules from E. coli was not significant in survival and behavior to honey bee. Furthermore, blast search results indicated that effective match of predicted dsRNA fragments were not existed in honey bee genome. CONCLUSION: In this study, we developed and tested the acute oral toxicity of dsRNA using E. coli expression system to honey bee.

A Study on Ensuring Biosafety of Biotechnology Product under Debate about Trade and the Environment (DDA 무역-환경 논의와 생명공학제품의 안전성 확보)

  • This paper analyze problems about scope of specific trade obligations(STOs), principle of dispute settlement procedure, and non-parties in context of the Cartagena Protocol on Biosafety(POB), which based on sub-paragraph 31(i) of DDA WTO Ministrial Declaration. The implications based on result of this study are as follows. First, to accept the wider scope of STOs under POB in Korea, importing country, won't be harmful to LMOs and Bioindustry. Instead, it will ensure a high level of biosafety concerning the import of LMOs. Exporters can take different kinds of trade measures to countervail adverse effect on the export of LMOs in this case. Therefore importer will endure the aftereffect. However, if korea were in exporter's place, to accept the wider scope STOs under POB will not have a good influence on the export of LMOs. Korea, therefore, should devise scheme for responding to debate about the STOs in MEAs, which have to be based on cost-benefit analysis and scenarios taking into account of speed and level in biotechology progress, status and trend of LMOs R&D and production, and condition of other industries. Second, it is not easy to agree with applying to what's rule between the POB and WTO for settlement dispute. Because there is the incompatibility between the POB characterized according to social rationality and WTO's rules for safety and environmental protection characterized according to scientific rationality. This issue have to be discussed for long period due to gap like that. Accordingly Korea, one of major LMOs importing countries, should suggest continuously that the effort is needed to ensure an adequate level of protection in transboundary movements of LMOs and scientific, environmental and socio-economic study. Third, in case of dispute between party and non-party of the POB, the duties under the WTO of non-party of the POB(if WTO member country) is valid. The country, therefore, will try to settle dispute based on WTO's rules. However, international society have to ensure for sound and safe use of LMOs in the field of transboundary movements. Accordingly Korea should devise scheme for preventing the possibility of dispute between party and non-party of the POB(if WTO member country), which is supported by policy options under the POB.

Classification, Analysis on Attributes and Sustainable Management Plan of Biotop Established in Pohang City (포항시 비오톱의 유형 구분, 속성 분석 및 복원 방안)

  • Biotope, which represents the characteristic habitats of living organisms, need to be identified as essential for the efficient creation and sustainable management of urban ecosystems. This study was carried out to provide the basic information for ecological urban planning by analyzing types and attributes of the biotop established throughout the whole area of the Pohang city, a representative industrial city in Korea. The biotop established in Pohang city is composed of 12 types including forests (coniferous, deciduous, and mixed forests), agricultural fields (rice paddy and upland field), green facilities, river, reservoir, bare ground, residential area, public facilities, commercial area, industrial area, roads, and schools. As a result of analyzing the properties according to biotop types, industrial, commercial and residential areas, which represent urban areas, was dominated by introduced vegetation. Moreover the introduced vegetation is usually composed of exotic plants or modified forms for landscape architecture and horticulture rather than native plants, which reflects ecological property of both region and site. As the distance from the urban center increases, the agricultural field showed a form of typical farmland, whereas the closer it is, the more form of greenhouse farming. Natural green spaces were divided into riparian vegetation established along the stream and forest vegetation. Forest vegetation is consisted of secondary forests (seven communities) and plantations (three communities). The urban landscape of Pohang city is dominated by the industrial area. Among them, the steel industry, which occurs large amounts of heat pollution and carbon dioxide, occupies a large proportion. On the other hand, green space is very insufficient in quantity and inferior in quality. This study proposed several restoration plans and further, a green network, which ties the existing green spaces and the green space to be restored as a strategy to improve the environmental quality in this area.
