Concentrations of metavolite in the leachate from pilot scale burial (실험용 매몰지 침출수의 대사체물질 농도변화)

  • At the end of November 2010, livestock infected with foot and mouth disease (FMD) were buried, increasing concern about secondary pollution of drinking water and groundwater with odor. Hence, measures to reduce pollution of drinking water and groundwater are needed. In this study, based on livestock type and soil texture, five pilot-scale burial sites were monitored for the past three years from Feb 2012 to 2015. Leachate from the site was analyzed for 90 compounds. A detailed investigation has been completed, looking into organic matter decomposition characteristics of the buried bodies, relevant trace elements thereof, and in particular metabolite concentration changes such as the degree of decomposition of a carcass. From the results, no detectable heavy metals, including cadmium and arsenic, were measured. Among animal types, the rate of cattle decomposition is faster than swine decomposition, and the decomposition of the bodies were performed in sequence from sand, clay loam, sandy loam.

  • Outbreak of animal infectious diseases such as foot-and-mouth disease, avian influenza are becoming prevalent worldwide. For prevent the further infection, tremendous numbers of the infected or culled stocks are buried around farm. This burial method can generate a wide range of detrimental components such as leachate, nutrient, salt, and pathogenic bacteria, consequently. In this study, for the stabilization of livestock carcasses leachate, advanced oxidation processes utilizing the Fenton reaction was investigated in lab-scale experiments for the treatment for $COD_{Cr}$ of livestock carcass leachate. $COD_{Cr}$ reduction by the Fenton oxidation was investigated response surface methodology using the Box-Begnken methods were applied to the experimental results. A central composite design was used to investigate the effects of the independent variables of pH ($x_1$), dosage of $FeCl_2{\cdot}4H_2O$ ($x_2$) and dosage of $H_2O_2$ ($x_3$) on the dependent variables $COD_{Cr}$ concentration ($y_1$). A 1 M NaOH and $H_2SO_4$ was using for pH control, $FeCl_2{\cdot}4H_2O$ was used as iron catalyst and NaOH was used for Fenton reaction. The optimal conditions for Fenton oxidation process were determined: pH, dosage of $FeCl_2{\cdot}4H_2O$ and dosage of $H_2O_2$ were 3, 0.6 g (0.0151 M) and 7 mL(0.259 M), respectively. Statistical results showed the order of significance of the independent variables to be pH > initial concentration of ferrous ion > initial concentration of hydrogen peroxide.

Characteristics of the Antibacterial Substance Produced by Paenibacillus polymyxa JB115 (Paenibacillus polymyxa JB115로부터 생산된 항균물질의 특성)

  • The culture broth of a ${\beta}$-glucan-producing bacterium, Paenibacillus polymyxa JB115, was confirmed to show the antibiosis against pathogenic bacteria of livestock disease. The antibacterial substance produced by P. polymyxa JB115 exhibited strong bactericidal or bacteriostatic effect on the growth of livestock pathogenic bacteria including Staphylococcus aureus, Salmonella choleraesuis, Escherichia coli and Pseudomonas aeruginosa. This antibacterial substance also showed high stabilities in broad pH range (pH 3-11) and in broad temperature range $(40-121^{\circ}C)$, which is good enough to apply spray-dry method for the formulation of culture broth. It was also found that the antibacterial substance was very stable in artificial gastric fluid and bile acid, which implies the anticipated antibacterial activity against gastrointestinal bacteria harmful for livestocks. In conclusion, the culture broth of P. polymyxa JB115 can be developed as a multifuctional feed additive containing immune-enhancing ${\beta}$-glucan as well as antibacterial agent against livestock pathogenic bacteria.

  • Efficient gene transfer into hematopoietic stem cells is a great tool for gene therapy of hematopoietic disease. Retrovirus have been extensively used for gene delivery and gene therapy. However, current in vitro gene transfer has some obstacles suck as induction of differentiation loss of self-renewal capacity, and down-regulation of homing efficiency for in vitro hematopoietic stem cells transplantation. To overcome these problems, we developed efficient in vitro retroviral transfer technique by direct intra-bone marrow injection (IBM). We identified effective retrovirus gene transfer in bone marrow hematopoietic cells in vitro. Two weeks after retrovirus transfer via IBM injection, we observed stable EGFP gene expression in bone marrow, lymph node, spleen, and liver cells. In addition, $6.4{\pm}2.7%$ of hematopoietic stem/progenitor cells were expressed EGFP transgene from flow cytometry analysis. Our results demonstrate that in vitro retrovirus gene transfer via IBM injection can provide a viable alternative to current or moo gene transfer approach.

A Study on Estimation of Factors Affecting Duration of Milk Flow and Milk Flow Rate and Their Relationships with Milk Yields of Dairy Cattle (유우의 착유 지속시간과 유속에 미치는 요인 및 산유량과의 상관관계 추정에 관한 연구)

  • The objective of this study was to investigate the environmental effects on duration time of milk flow, peak milk flow, and average milk flow in teats, and to estimate their relationships with milk yields in morning and evening milking, and to provide fimdamental information for the further study on their relationships with clinical mastitis and somatic cell in milk. A total of 6,768 milking records were studied in 72 Holstein cows. The influences of season, parity, lactation month, and milking interval on characteristics of milk flow considering in linear model were significant(p<.05). Duration of milk flow was longest at milking in fall, past first parity and second month of lactation, and with milking interval over than 13.5hrs. Average milk flow rate and peak milk flow rate were highest at milking in summer, past first parity and 8${\sim}$10 months of lactation, and with milking interval over than 13.5hrs. Milk flow rate was positively correlated to milk yield, and negatively correlated to the duration of milk flow. However duration milk flow was positively correlated to the milk yield with high level of correlation coefficient(+.60). For the establishment of optimum selection criteria on these traits, other aspects such as the udder health, disease and respective economic weights of milk flow characteristics in this study must be considered to develop the indices.

Seroprevalence of Brucellosis among Risk Population in Gyeongsangbuk-do, 2006 (경상북도 거주 고위험군에서 브루셀라 감염에 관한 혈청 역학적 연구)

  • Objectives : Cases of human brucellosis in Korea have recently increased due to the increasing incidence of bovine brucellosis. The authors conducted this study to elucidate the status of brucellosis through seroepidemiologic study. Methods : We selected our study population from a high risk group. We conducted a questionnaire survey and obtained blood samples to determine the seroprevalence of brucellosis antibodies for 10 days in February, 2005. The titers of brucellosis were measured by the combination of standard tube agglutination test (STA) and enzyme-linked immunosorbent assay (ELISA) test. Results : Our study subjects comprised 1,075 cases: 971 livestock workers, 51 veterinarians, and 53 artificial inseminators. In the STA test, 27 cases (2.5%) had titers of greater than or equal to 1:20. Of 1,068 cases (7 cases were excluded due to previous brucellosis), 7 cases of brucellosis were diagnosed with titers of 1:160, giving a seroprevalence of brucellosis of 0.66%. The seroprevalence in the male group was 0.95%, and that of livestock workers, veterinarians, and artificial inseminators was 0.52%, 4.17%, and 0.00%, respectively. The Spearman's correlation coefficient between the positive rate of bovine brucellosis per capita and household and human brucellosis was 0.806 and 0.744, respectively. The concordance rate between the Korea National Institute of Health and the Gyeongsangbuk-do Institute of Health and Environment by the STA and ELISA tests was 94.7% and 100.0%, respectively. Conclusions : The study results indicated in higher seroprevalence rate among veterinarians than among livestock workers and artificial inseminators. Because veterinarians may be exposed to this high risk, effective working guidelines for veterinarians to guard against brucellosis must be developed. Moreover, more extensive epidemiologic research for laboratory workers and meat handlers is needed.

Locational Characteristics of Highly Pathogenic Avian Influenza(HPAI) Outbreak Farm (고병원성 조류인플루엔자(HPAI) 발생농가 입지특성)

  • This study was conducted to identify the location characteristics of infected farms in the areas where livestock diseases were clustered(southern Gyeonggi-do and Chungcheong-do), analyze the probability of disease occurrence in poultry farms, find out the areas corresponding to the conditions, and use them as the basis for prevention of livestock diseases, selection of discriminatory prevention zones, and establishment of prevention strategies and as the basic data for complementary measures. The increase of one poultry farm within a radius of 3-kilometers increases the risk of HPAI infection by 10.9% compared to the previous situation. The increase of 1m in distance from major roads with two lanes or more reduces the probability of HPAI infection by 0.001% compared to the previous situation. If the distance of the poultry farm located with 15 kilometers from a major migratory bird habitat increases by 15 to 30 kilometers, the chance of infection with HPAI is reduced by 46.0%. And if the distance of the same poultry farm increase by more than 30 kilometers, the chances of HPAI infection are reduced by 88.5%. Based on the results of logistic regression, the predicted probability was generated and the actual area of the location condition with 'more than 15 poultry farms within 3km a radius of, within 1km distance from major roads, and within 30km distance from major migratory birds habitat was determined and the infection rate was measured. It is expected that the results of this study will be used as basic data for preparing the data and supplementary measures when the quarantine authorities establish discriminatory quarantine areas and prevention strategies, such as preventive measures for the target areas and farms, or control of vehicles, by identifying the areas where livestock diseases are likely to occur in the region.

Age-dependent immune response in pigs against foot-and-mouth disease virus in vitro

  • Foot-and-mouth disease, one of the most contagious diseases in cloven-hoofed animals, causes significant economic losses. The pathogenesis of foot-and-mouth disease virus (FMDV) infection is known to differ with age of the animals. In this study, we aimed to reveal the difference in immunological response in the initial stage of FMDV infection between piglets and adult pigs. Peripheral blood mononuclear cells (PBMCs) were isolated from 3 piglets (8 weeks old) and 3 pigs (35 weeks old) that were not vaccinated against FMDV. O-type FMDV (2 × 102 median tissue culture infectious dose) was inoculated into porcine PBMCs and the cells were incubated at 37.0℃ under 5% CO2 for various time periods (0, 1, 3, 6, 12, 24, and 48 h). The total RNA was obtained from the FMDV-inoculated PBMCs after each time point, and the virus titer was investigated in these RNA samples. Furthermore, dynamics of mRNA expression of the six tested cytokines (interferon [IFN]-α, IFN-γ, interleukin [IL]-6, IL-8, IL-10, and tumor necrosis factor [TNF]-α) in FMDV-inoculated porcine PBMCs were evaluated by time-series analysis to determine the differences, if any, based on the age of the pigs. The PBMCs of piglets contained the highest quantity of FMDV mRNA at 6 hours post-inoculation (hpi), and the PBMCs of pigs had the highest quantity of FMDV mRNA at 3 hpi. The mean cycle threshold-value in the PBMCs steadily decreased after the peak time point in the piglets and pigs (6 and 3 hpi, respectively). The dynamics of mRNA expression of all cytokines except TNF-α showed age-dependent differences in FMDV-inoculated PBMCs. The mRNA expression of most cytokines was more pronounced in the piglets than in the pigs, implying that the immune response against FMDV showed an age-dependent difference in pigs. In conclusion, within 48 hpi, the 8-week-old piglets responded more rapidly and were more sensitive to FMDV infection than the 35-week-old pigs, which could be associated with the difference in the pathogenesis of FMDV infection among the pigs. These results provide valuable insights into the mechanisms underlying the age-dependent differences in immune response in pigs against FMDV infection.

Survey of antibody on Orientia Tsutsugamushi among wild rodents in Gyeongnam area and detection by nested polymerase chain reaction (경남지역 야생들쥐에서 Orientia tsutsugamushi에 대한 항체 조사 및 PCR에 의한 검색)

  • As a part of epidemiologic investigation of tsutsugamushi disease, the wild rodents which were captured in Gyeongnam area were diagnosed with indirect immunofluorescent antibody assay (IFA) and Polymerase Chain Reaction (PCR) to find if they have an antibody against Orientia tsutsugamushi. The conclusion was drawn as followings. (1) The captured 58 wild rodents showed that the subspecies distribution of Apodemus agrarius was 86.2%, Microtus fortis was 8.6% and Crocidura lasiura was 5.2%. (2) The antibody positive rate of O. tsutsugamushi Gilliam, Karp, Karto and Boryong by IFA method was 32.0% in Apodemus agrarius among 50 wild rodents and 40.0% in Microtus fortis among 5 wild rodents, respectively. It was negative in the case of all the 3 Crocidura lasiura. (3) The antibody titers on Apodemus agrarius, Microtus fortis and Crocidura lasiura against Gilliam, Karp, Karto and Boryong were measured between 1:20 and 1:640. The antibody titer against each antigen was in the order Boryong>Gilliam>Karp. (4) O. tsutsugamushi was detected from the blood, spleen and kidney from the artificially infected mice by IFA and PCR. IFA showed the positive response between 3 and 18 days after inoculation. On the other hand, positive response was found from all the samples by PCR. (5) From PCR of the genomic DNA extracted from the blood, spleen and kidney samples of the captured wild rodents, Boryong-specific amplification product with size of 210 bp, which is particular in Boryong, was detected from spleen and kidney samples, but not detected in the blood. (6) Boryong-specific amplification product was detected from spleen and kidney samples which were obtained at 3, 6, 12, 18 and 24 days after the infection with Boryong. But, it wasn't detected from the uninfected samples. (7) From PCR of spleen and kidney samples of the captured wild rodents, it was found that positive rate of O. tsutsugamushi in Apodemus agrarius and Microtus fortis were 25.0% (4/16) and 20.0% (1/5), respectively. From the above results, it can be concluded that Apodemus agrarius resided in Gyeongnam area carried O. tsutsugamushi and PCR method might be a simple, precise, rapid and useful diagnostic tool than IFA for the diagnosis of O. tsutsugamushi.

Brucellosis in sheep and goat of Bogra and Mymensingh districts of Bangladesh

  • Brucellosis is the most important bacterial disease of livestock in Bangladesh. The present study was conducted to determine the seroprevalence of brucellosis in goat and sheep in Mymensingh and Bogra districts of Bangladesh using slow agglutination test and Rose Bengal test as screening test and indirect enzyme linked immunosorbent assay as confirmatory test. Questionnaire based data on age, gender, area, client's complaint, number of animals in herds, disease history, reproductive problems such as abnormal uterine discharge, abortion or previous abortion in sheep and goat and their reproductive diseases were recorded. A total of 200 sera samples were collected from 80 sheep and 120 goats. The prevalence of brucellosis in goat was 2.50% and 1.25% in sheep. Positive reactors were only detected in female of both goat and sheep. In this study, there existed a significant association among abortion and the prevalence of brucellosis (p < 0.01). The prevalence of brucellosis in sheep and goat in Bangladesh is not negligible, and it is therefore worth considering the adoption of preventive measures.
