Optimization of liquid-liquid extraction conditions for paclitaxel separation from plant cell cultures (식물세포배양으로부터 Paclitaxel 분리를 위한 액-액 추출 조건의 최적화)

  • In this study, the process parameters of liquid-liquid extraction were optimized to obtain a high purity and yield of paclitaxel in a pre-purification step. The optimal solvent ratio (methylene chloride/concentrated methanol extract ratio), extraction times, mixing time, and standing time for liquid-liquid extraction were 0.28 (v/v), 3(times), 30 min, and 40 min, respectively. The polar impurities from the biomass extraction were efficiently removed by liquid-liquid extraction. The complete concentration of liquid-liquid extract by rotary evaporator was reliable enough to obtain a high purity and yield of paclitaxel for subsequent purification steps.

Thermal Inactivation of Listeria monocytogenes in Liquid Cultures during Microwave Radiation (Microwave 조사에 의한 Listeria monocytogenes의 불활성에 관한 연구)

  • The purpose of this study was to determine the thermal inactivation of L. monocytogenes KCTC3443 in liquid culture heated in the controlled microwave system and in the conventional heating method. Furthermore, we have carried out a comparative study on the thermal and nonthermal microwave effects on microorganisms, pasteurized using a controlled microwave energy specially designed apparatuses and a water bath. For the automatic temperature control during microwave heating, the real time data acquisition and computation system is designed with BASIC routine. The automatic temperature control system used in the experiments perform relatively stable control at the experiment temperature of 55, 65, $75^{\circ}C$ and $85^{\circ}C$ for 30 minutes. The effects of microwave heating on liquid cultures was compared with that of conventional heating. The results show that microwave radiation, while being slightly quicker than conventional heating, still reduces effectively the number of pathogenic bacteria during heating for a limit time in liquid cultures. While no particular differences between microwave heating and conventional heating was not observed in the thermal inactivation of L. monocytogenes at 55, 65, $75^{\circ}C$ and $85^{\circ}C$ for 30 min., respectively. Microwave heating is, therefore, substantially not effective in inactivating L. monocytogenes in liquid culture than conventional heating method.

Variability of Azadirachtin in Azadirachta indica (neem) and Batch Kinetics Studies of Cell Suspension Culture

  • Seeds of neem were collected from different parts of India and analyzed for their azadirachtin content by High Performance Liquid Chromatography (HPLC). In order to assess the effects of genotypic and geographical variation on azadirachtin content in cell cultures, callus development was attempted in the seeds containing high and low concentration of azadirachtin. The concentration of azadirachtin in callus cultures was significantly affected by the explant source. Seed kernels with higher azadirachtin content produced higher azadirachtin content in callus cultures and lower azadirachtin content was seen in callus cultures produced from seed kernels with low azadirachtin content. The protocol for development of elite stock culture of Azadirachta indica was established with the objective of selecting a high azadirachtin-producing cell line. The highest azadirachtin-producing cell line was selected and the effects of different media and illumination conditions on growth and azadirachtin production were studied in shake flask suspension culture. Detailed batch growth kinetics was also established. These studies provided elite starter culture and associated protocols for cultivation of A. indica plant cell culture in the bioreactor.

Berberine Production by Cell Suspension Cultures of Cork Tree (Phellodendron amurense Rupr)

  • Various culture conditions for cell growth and berberine production in cork tree (Phellodendron amurense Rupr.) were investigated. Callus was induced from cambium tissue of cork tree, and cultured on LS liquid medium supplemented with 0.5 mg/1 2,4-D, 0.1mg/1 BA, and 3% sucrose. Several factors enhancing berberine production and cell growth in cork tree cell cultures were found. Some of them enhanced both cell growth and berberine production, but others resulted in a decoupling of cell growth and berberine production with significant in the specific levels. High level of nitrate (80mM), high level of phosphate (8.98mM), and sucrose (7%), 1.0mg/l IAA were effective in berberine production, whereas low level of nitrate (40mM), and phosphate (2.25mM), and high level of sucrose (7%) in the medium were effective in cell growth. Two stage culture(first stage for cell growth, and second stage for berberine production) increased berberine production almost twice (5.06mg/g dry weight) as much as single stage cultures in berberine production.

Tannin-Reducing Effect and Changes of Physicochemical Properties in Aronia Homogenate after Treatment with Liquid Cultured Mushroom Mycelia (버섯균사체 배양액 처리에 의한 아로니아 균질액의 탄닌 감소 및 이화학성 변화)

  • Although aronia (Aronia melanocarpa (Michx.) Elliott) contains higher levels of polyphenols and more antioxidant activity than other berries, it is a berry that is difficult to eat raw due to its strong astringent taste and lack of sweetness. Therefore, in this study, we investigated the effect of tannin reduction of aronia by bioconversion method using mushroom mycelia cultures. Aronia and liquid cultures of Lentinula edodes and Phellinus linteus mycelia were mixed and then treated for 48 hours at 60℃. Tannin content, total polyphenol, total flavonoid and antioxidant activities (DPPH, ABTS radical-scavenging activities and FRAP activities) were investigated. The tannin content decreased from 64.2 mg ECE/g to 57.9 mg ECE/g (9.8% reduction) when treated with liquid culture of L. edodes and from 77.3 mg ECE/g to 47.9 mg ECE/g (38.1% reduction) from treatment with a liquid culture of P. linteus. Therefore treatment with mushroom mycelia culture solution may improve the palatability of aronia reducing the astringent taste.

  • Airlift bioreactors are extensively used in the fields of aerobic fermentation, animal and plant cell cultures. This review article evaluates the present research activities in the field of airlift bioreactors. The published research works on the design parameters such as types, location and properties of gas sparser, hydrodynamic properties such as phase holdups mixing, liquid circulation rate, mass and heat transfer rates are summarized. Also, recommendations are presented for designing airlift bioreactors.

  • In this study, the effects of auxin and casein hydrolysate (CH) on the growth of ginseng hairy root was elucidated. Ginseng hairy root was cultured under light and dark conditions in MS solid and liquid medium with various concentrations of auxin and CH for fifty days. After harvesting the cultures, the fresh and dry weight of cultures were examined, respectively. In the MS solid culture, 1 mg/L of IBA was most effective on the growth of ginseng hairy root under the dark condition, whereas IAA and CH did not affect on the growth of ginseng hairy root. In the MS liquid culture, the growth was maintained regularly by the treatments of IAA and NAA. IBA and CH restrained the growth of ginseng hairy root.

Styrene Degradation in a Polyurethane Biofilter Inoculated with Pseudomonas sp. IS-3

  • In a search for bacteria capable of degrading styrene better than previously isolated strains, bacterium IS-3 was isolated from activated sludge and found to be most closely related to Pseudomonas sp. Styrene degradation by this strain was tested in liquid cultures and polyurethane-packed biofilters. In liquid cultures, the rate of styrene degradation by this bacterium increased from 24.93 to $76.53\;{\mu}mol\;g^{-1}\;DCW\;H^{-1}$ for an initial mass range from 8.7 to $34.8{\mu}mol$. The maximum styrene elimination capacity was 580-635 $g/m^{3}\cdot$h at a space velocity (SV) of 50-200/h. The critical elimination capacities guaranteeing $95\%$ removal of the input styrene were determined to be 635, 170, and 38 $g/m^{3}\cdot$h, respectively, at SVs of 50, 100, and 200/h. Kinetic analysis revealed that the maximum styrene elimination velocity ($V_{m}$) for this biofilter was 1,000 g/m$\cdot$h, and the saturation constant ($K_{m}$) was 454 ppmv. Together, these results suggest that a polyurethane biofilter containing Pseudomonas sp. IS-3 could have potential practical applications for the effective removal of styrene gas.

Mycobacterium intracellulare Pleurisy Identified on Liquid Cultures of the Pleural Fluid and Pleural Biopsy

  • Pleural effusion is a rare complication in non-tuberculous mycobacterial infection. We report a case of Mycobacterium intracellulare pleuritis with idiopathic pulmonary fibrosis in a 69-year-old man presenting with dyspnea. Pleural effusion revealed lymphocyte dominant exudate. M. intracellulare was identified using a polymerase chain reaction-restriction fragment length polymorphism method and liquid cultures of pleural effusion and pleural biopsy. After combination therapy for M. intracellulare pulmonary disease, the patient was clinically well at a 1-month follow-up.

  • Cultures of hairy root induced from ginseng(Panax C.A. Meyer) transformed with Agrobacterium rhizogenes (strain A4, ATCC 15834) were established and morphologically two different hairy root strains (HB1, HB2) were obtained. To determine the optimum growth rate, the hairy root (HB2) was cultured in various liquid medium supplemented with or without plant growth hormone. The growth rate of hairy root cultured on MS medium was 1.3-3.1 times higher than those cultured on other media, and the optimum sucrose concentration and pH were 3-6%, 5.5-6.5, respectively. Also, the growth rate of hairy root was increased when 0.02 M ammonium nitrate, 1.2 mM potassium phosphate (monobasic) and 0.5 mg/l IBA were supplied to liquid medium. The saponin patterns and contents of hairy root (HB2) were determined by TLC and HPLC. The crude saponin contents were 4.67% and the total saponin contents were 1.0%, on dry weight basis.
