초임계 이산화탄소에서 소르비탄 모노팔미테이트 계면활성제의 상거동에 관한 연구 (Phase Behavior of Sorbitan Monopalmitate Surfactant in Supercritical Carbon Dioxide)

  • 가변부피 투시셀이 장착된 고압 상평형 장치를 사용하여 이산화탄소 + 계면활성제 이성분계와 이산화탄소 + 계면활성제 + 물 삼성분계의 상평형 거동을 318 K부터 348 K의 온도범위에서 관찰하였다. 계면활성제로는 소르비탄 모노팔미테이트를 사용하였다. 이산화탄소 + 소르비탄 모노팔미테이트 이성분계에서 관측된 운점압력은 온도가 상승함에 따라 증가하며, 소르비탄 모노팔미테이트의 조성이 0.226 wt%에서 최대 운점압력이 측정되었다. 반면에 계면활성제의 농도를 0.1 wt%로 고정한 삼성분계에서 관측된 운점압력은 온도와 물의 조성이 증가함에 따라 커지는 것을 확인할 수 있었다. 물의 농도를 0.2 wt%로 고정한 삼성분계에서의 압력-조성의 곡선은 상대적으로 수평의 형태를 나타냈다. 이 때, 운점압력은 온도와 계면활성제의 조성이 증가함에 따라 상승함을 확인할 수 있다.

Biotransformation of Pueraria lobata Extract with Lactobacillus rhamnosus vitaP1 Enhances Anti-Melanogenic Activity

  • Isoflavone itself is less available in the body without the aid of intestinal bacteria. In this study, we searched for isoflavone-transforming bacteria from human fecal specimens (n = 14) using differential selection media. Isoflavone-transforming activity as the production of dihydrogenistein and dihydrodaidzein was assessed by high-performance liquid chromatography and we found Lactobacillus rhamnosus, named L. rhamnosus vitaP1, through 16S rDNA sequence analysis. Extract from Pueraria lobata (EPL) and soy hypocotyl extract were fermented with L. rhamnosus vitaP1 for 24 and 48 h at $37^{\circ}C$. Fermented EPL (FEPL) showed enhanced anti-tyrosinase activity and antioxidant capacities, important suppressors of the pigmentation process, compared with that of EPL (p < 0.05). At up to $500{\mu}g/ml$ of FEPL, there were no significant cell cytotoxicity and proliferation on B16-F10 melanoma cells. FEPL ($100{\mu}g/ml$) could highly suppress the content of melanin and melanosome formation in B16-F10 cells. In summary, Lactobacillus rhamnosus vitaP1 was found to be able to biotransform isoflavones in EPL. FEPL showed augmented anti-melanogenic potential.

에멀전 방식의 고분자 분산형 액정의 전기 광학 특성에 미치는 액적 크기 분포의 영향 (Droplet Size Distribution Effect on the Electro-Optical Properties of Emulsion Type Polymer Dispersed Liquid Crystal)

  • We established the emulsion method using membrane filter with precise control of LC droplet distribution in PDLC. PDLC cells with various LC droplet size distributions such as single droplet sizes of $1.0{\mu}m$, $1.9{\mu}m$ and $3.5{\mu}m$, the mixture of two different LC droplet sizes and the mixture of three different LC droplet sizes were fabricated and the electro-optical properties of the emulsion type PDLC cells with various droplet size distribution were investigated. In the appropriate droplet size range, the PDLCs with the single droplet sizes distributions have good electro optical properties than those with the mixture of three different LC droplet sizes. In addition, the PDLC cells with the mixture of two different LC droplet sizes have the better electro optical properties than those with single droplet sizes distribution. The PDLC cell with dual droplet size distribution of $1.0+1.9{\mu}m$ shown the best electro optical properties than the PDLC cells with other size distributions. This method enabled us to find the proper LC droplet size distribution for achieving both high transmittance and contrast ratio.

Neuroprotective Effects of Phlorotannin-Rich Extract from Brown Seaweed Ecklonia cava on Neuronal PC-12 and SH-SY5Y Cells with Oxidative Stress

  • Neurodegenerative disorders in the elderly are characterized by gradual loss of memory and cognitive function. Oxidative stress caused by reactive oxygen species is associated with progressive neuronal cell damage and death in Alzheimer's disease, one of the most common neurodegenerative disorders. An edible brown seaweed, Ecklonia cava, contains a variety of biologically active compounds such as phlorotannins. In this study, we comparatively evaluated the total phenolic content, antioxidant capacity, and neuroprotective effects of the phlorotannin-rich extract from E. cava (PEEC). The total phenolic content of PEEC and dieckol was 810.8 mg gallic acid equivalents (GAE)/g and 996.6 mg GAE/g, respectively. Antioxidant capacity of PEEC was 1,233.8 mg vitamin C equivalents (VCE)/g and 392.1 mg VCE/g determined using ABTS and DPPH assays, respectively, while those of dieckol were 2,238.4 mg VCE/g and 817.7 mg VCE/g. High-performance liquid chromatography results revealed 48.08 ± 0.67 mg dieckol/g of PEEC. PEEC had neuroprotective effects in pheochromocytoma (PC-12) and human neuroblastoma (SH-SY5Y) cells against H2O2- and AAPH-induced oxidative damage, partly due to reduced intracellular oxidative stress. PEEC treatment inhibited acetylcholinesterase and butyrylcholinesterase in a dose-dependent manner. Taken together, these findings suggest that PEEC is a good source of antioxidants and neuroprotective materials.

  • Electro-Optical (EO) performances for the ion beam (IB) aligned twisted-nematic (TN)-liquid crystal display (LCD) with ion beam exposure on the new of diamond like carbon (DLC) thin film surface were investigated. Voltage-transmittance (V-T) curve and response time without backflow bounce in the ion beam aligned TN-LCD with ion beam exposure for 0.5 and 1min on the DLC thin film was observed. Also. the fast response time of ion beam aligned TN-LCD with ion beam exposure for 1min on the DLC thin film surface can be achieved. The residual DC voltage of the ion beam aligned TN-LCD on the DLC thin film surface was almost the same as that of the rubbing aligned TN-LCD on the polyimide(PI) surface.

고율 조류 생세포체 배양지에서 조사 조건으로 본 조류 배양 특성 (Algae Culture Characteristics Viewed with Continuous and Cyclic Irradiation in High Rate Algae Biomass Culture Pond)

  • The utilization methods of algae biomass have been studied constantly in whole world. These are $\circled1$the wastewater treatment if waste stabilization pond and oxidation ditch etc. and $\circled2$the biosorption of heavy metals and recovery of strategic' precious metals and $\circled3$the single-celled protein production and the production of chemicals like coloring agent and $\circled4$the production of electric energy through methane gasification. The culture system also has been developed constantly in relation with such utilization method developments. In the result of experimental operation under continuous and cyclic irradiation of light, using high rate algae biomass culture pond(HRABCP), which had been made so as to be an association system with the various items which had been managed to have high efficiency for algae culture, the algae production of the 12 hours-irradiance pond was 41.48 Chlorophyll-a ${\mu}g/L$ only in spite of having the more chance of $CO_2$ synthesis to algae cell than the 24 hours-irradiance pond. This means that the energy supply required for dark-reaction of photosynthesis is very important like this. The difference of algae production between continuous and cyclc irradiation explains that the dark-reaction of photosynthesis acts on algae production as the biggest primary factor. The continuous irradiance on HRABCP made the good algae-production($1403.97{\;}{\mu}g$ Chlorophyll-a/mg) and the good oxygen-production(5.8 mg $O_2/L$) and the good solid-liquid seperation. especially, DO concentration through the oxygen-production was enough to fishes' survival.

Glycerol, 난황 및 Trehalose를 이용한 돼지유정란의 동결 (Cryopreservation of Porcine Embryos using Glycerol, Egg Yolk and Trehalose)

  • The purpose of this study was to examine the suvival rates of cryopreserved porcine embryos collected on Day 6 (Day 0=onset of estrus) with various cryprotectants. Eighty two embryos at different stages, ranged from expanded blastocyst to hatched blastocyst, were allocated to 6 experimental groups in different combinations of cryoprotectants glycerol, egg yolk and/or trehalose. Porcine embryos were cryopreserved using conventinal slow freezing precedures. The embryos were equilibrated with one of the freezing solutions, cooled from 25 to -7$^{\circ}C$ at 1$^{\circ}C$/min, seeded at -7$^{\circ}C$ frozen to -36$^{\circ}C$ at 0.5$^{\circ}C$/min, and then plunged into liquid nitrogen. The frozen embryos were thawed by immersion in 37$^{\circ}C$ water and the cryoprotectants were removed by dilution with 0.5M sucrose solution. Embryonic survival was estimated from the normal development of embryos for 12, 24 and 48hrs culture. Then the embryos were stained and their cell nuclei was counted. The survival rates of morphological embryos were significantly higher in group I (10% glycerol) or group IV (10% glycerol+10% egg yolk+0.5M trehalose) than those in other groups, although the nuclei number was quite higher in embryos treated with 10% glycerol and 0.25M trehalose at expended blastocyst. However, hatched blastocysts showed higher viability and nuclei number treated with either egg york or trehalose, but the survival rates after 48hrs of culture were quite low. These results indicate that egg yolk and trehalos as a supplement to freezing solutions can be useful to the cryopreservation ofn porcine embryos.

Effect of temperature on fractional precipitation for paclitaxel purification

  • 분별침전 공정에서 순도 및 수율에 미치는 온도의 영향을 확인하였다. 분별침전을 위해 일정 온도에서 보관할 경우와 단계적으로 보관 온도를 하강하는 경우에 대한 분별침전 효율을 확인하였다. 일정온도$(0^{\circ}C)$에서 보관할 경우 높은 수율$({\sim}84%)$을 얻을 수 있는 반면 단계적으로 보관 온도를 하강할 경우 상대적으로 높은 순도$({\sim}79%)$를 얻을 수 있었다. 또한 단계적으로, 큰 폭으로 보관 온도를 하강 시킬 경우 더 높은 순도를 얻을 수 있었다. 저장온도$(-20^{\circ}C{\sim}12^{\circ}C)$를 일정하게 유지할 경우 paclitaxel 순도 면에서 $0^{\circ}C$가 가장 효과적이었으며 온도변화에 따른 수율은 거의 영향이 없었다. 따라서 최종 정제를 위한 전 처리 공정으로 분별침전 공정이 간단하고 매우 효과적임을 알 수 있었다.

아미노산액 처리에 의한 인산가용화균주의 생육 및 가용화 효과 (Effect of Amino Acid Solution for Cell Growth and MPS Activity of Mineral Phosphate Microorganisms.)

  • 인산가용화능이 우수한 균주인 Burkholderia sp. DA23과 Klebsiella sp. DA7l-1 두 균주를 사용하여 농도별 아미노산 첨가에 따른 성장 양상과 유리인산 방출량을 조사한 결과, 두 균주의 생육과 인산가용화에 있어 아미노산이 도움이 되는 것으로 나타났다. DA23과 DA7l-1 모두 0.05%와 0.1%의 아미노산을 SM배지에 첨가하여 성장 양상을 확인한 결과 아미노산을 첨가한 배지의 균주가 아미노산을 처리하지 않은 배지의 균주에 비해 높은 세포 밀도 성장을 보여주었다. 인산가용화 균주의 아미노산 첨가의 경우 무처리보다 높은 pH 저하를 보였으며, 또한 아미노산 첨가가 아미노산 무처리구에 비해 2배이상 높은 인산가용화 효과를 보였다. DA7l-1의 아미노산 첨가의 경우는 DA23의 아미노산 첨가보다 아미노산 첨가에 의한 인산가용화 효과가 더 있는 것으로 나타났다. hydroxyapatite와 tri-calcium phosphate 의 인산염 차이에 따른 아미노산 처리 효과는 없었으며, 0.05%의 미량의 농도에서도 인산가용화로의 세포밀도 성장 및 가용화능에 효과를 나타내었다.

고안전성 리튬이차전지 구현을 위한 나노 세라믹 코팅 분리막 제조 및 전기화학특성 분석 (Nano Ceramic Coating on Polypropylene Separator for Safety-Enhanced Lithium Secondary Battery)

  • 폴리올레핀 분리막의 내열성을 향상시키면서도 전기화학특성 개선을 위해 RF Magnetron Sputter기반으로 수십 나노미터 수준의 세라믹 층이 코팅된 내열 분리막을 제조하였다. 분리막 원단의 열적 손상없이 코팅 시간을 최소화하기 위한 증착 조건을 최적화 하였고, 이를 기반으로 제조된 내열 분리막의 물리적, 전기화학적 평가를 진행하였다. 약 20 nm의 $Al_2O_3$가 코팅된 Polypropylene(PP) 분리막은 원단 분리막 대비 통기 특성 (원단: 211.3 sec/100 mL, 코팅 분리막: 250.8 sec/100 mL)은 떨어졌으나, 열 수축율 (원단: 19.4%, 코팅 분리막: 0.0% @ $140^{\circ}C$ & 30 min), 전해액 Uptake(원단: 176%, 코팅 분리막: 190%) 및 이온전도도 (원단: 0.700 mS/cm, 코팅 분리막: 0.877 mS/cm)는 모두 향상되었다. 그 결과, 2032-type Half-cell($LiMn_2O_4/Li$)을 이용한 전기화학적 평가에서도, 향상된 율별 특성과 유사한 수명 특성을 나타내었다.
