Transforming Growth Factor-Beta Stimulates Osteoclastic Bone Resorption in vitro (파골세포에 대한 Transforming Growth Factor-$\beta$의 활성화 작용)

  • Osteoclast has been known as a primary responsible cell for the bone resorption. The activation of osteoclast, therefore, may be the key event in the regulation of bone growth and remodeling. Various factors were reported to have influence on the resorbing activity of osteoclast in organ culture. Among those factors, transforming growth factor-$\beta$ (TGF-$\beta$) has been known to have a profound effect on bone metabolism. Since a large amount of TGF-$\beta$ presents in bone tissue, it may be important for the understanding the regulatory mechanism of bone resorption to elucidate the effect of TGF-$\beta$ on the osteoclast. We have reported the dlsaggregated chick embryonic osteoclast culture as an useful assay method for determining the resorption activity of osteoclast. In this culture, we found that TGF-$\beta$ significantly enhaced the osteoclastic bone resorption activity. We also found that the timulatory effect seemed to be an indirect one that is mediated by other cells. As nordihydroguaiaretic acid significantly inhibited the TGF-$\beta$1-induced osteoclastic bone resorption, we suggest that the lipoxygenase derivative of arachidonic acid may participate in the action of TGF-$\beta$ as a paracrine or an autocrine mediator.

Antioxidant and Anti-inflammatory Activities of Ethanol Extracts from Medicinal Herb Mixtures (단일 생약재 및 생약복합제 에탄올추출물의 항산화 및 항염증 활성)

  • In this study, antioxidant and anti-inflammatory effects of ethanol extracts from medicinal herb mixtures were investigated in vitro. Contents of total polyphenols in ethanol extracts from Houttuynia cordata Thunb. (HC), Carthamus tinctorius L. (CT), and MIX-2 (Houttuynia cordata Thunb., Acanthopanax gracilistylus Smith, Carthamus tinctorius L., Kalopanax pictus) were 58.98, 60.79 and 57.74 mg/g respectively. Radical scavenging activities of the ethanol extracts were examined using ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl radicals and 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) assay. Anti-inflammatory effects of ethanol extracts from medicinal herbs composites were examined using 5-lipoxygenase (5-LO), cyclooxygenase-2 (COX-2) and nitric oxide (NO) inhibition assays. The ethanol extracts from medicinal herb mixtures showed higher 5-LO and COX-2 inhibition activities than those from single medicinal herbs. The ethanol extracts from medicinal herb mixtures also showed strong anti-inflammatory activity through inhibition of NO production.

Anti-asthmatic Activities of the Extract of Lonicera japonica (인동 추출물의 항천식 효과)

  • The anti-asthmatic activities of the extract of Lonicera japonica (BuOH fraction) and its mode of action were investigated using several in vitro and in vivo models. Lonicera japonica was extracted with 30% ethanol (v/v) and successively partitioned into BuOH. The BuOH fraction reduced antigen-induced contraction of isolated trachea from sensitized guinea pigs in a concentration-dependent manner. The BuOH fraction also inhibited histamine release from rat peritoneal mast cells induced by antigen or calcium ionophore A23187 ($IC_{50}=0.26$ and 0.32mg/ml, respectively). Eosinophil infiltration into bronchoalveolar lavage fluids induced by aeroallergen challenge in passively sensitized guinea pigs was inhibited by the BuOH fraction at a dose of 800mg/kg (51.7%). In addition, the BuOH fraction inhibited leukotriene $B_4$ prodution in rat basophilic leukemia cells ($IC_{50}=0.42\;mg/ml$) as well as phosphodiesterase 4 (PDE4) isolated from rat brain ($IC_{50}=0.015\;mg/ml$). All results from this study strongly suggest that the BuOH fraction of Lonicera japonica may be useful in the treatment of asthma and its mode of action may be related with inhibition of both 5-lipoxygenase and PDE4 enzyme.

Improved Storage Stability of Brown Rice by Coating with Rice Bran Protein (쌀겨 단백질 코팅에 의한 현미의 저장성 향상)

  • Brown rice grains were coated by spraying aqueous protein solution extracted from rice bran using 10% ethanol, and dried at room temperature. Coating procedure was repeated 1-5 times to determine effect of repeated coating, Quality changes in coated brown rice grains were observed during 8 weeks storage. Most coated rice grains gave lower peroxide and acid values, which indicate antioxidative effects of coating. Lipase and lipoxygenase activities generally decreased in grains coated more than three times. Microscopic images of whole kernel and longitudinal section revealed cracking on all brown rice grains including control, and hydration rate constants were not significantly different among treated grains. Compared to non-coated brown rice grains, those coated more than three times, after 8 week storage, showed better quality retention observed in (meaning not clear) higher water-binding capacity, lower gel consistency decrease, less browning, and better textural properties in cooked rice, resulting in better sensory quality.

Effect of trans-10, cis-12 Conjugated Linoleic Acid on Production of Prostaglandin E2, Cyclooxygenase-2 and 5-lipoxygenase in Lipopolysaccharide-Stimulated Porcine Peripheral Blood Mononuclear Cells

  • The objective of this study was to examine the effect of trans-10, cis-12 conjugated linoleic acid (t10c12-CLA) on the expression of cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) pathway in lipopolysaccharide (LPS)-stimulated porcine peripheral blood mononuclear cells (PBMCs). t10c12-CLA was treated with different concentrations in culture medium of LPS$na{\ddot{i}}ve$ and LPS-stimulated PBMCs. The mRNA expressions of prostaglandin $E_2$ ($PGE_2$)-synthase, COX-2 and 5-LOX were measured using quantitative real-time PCR. In addition, the production levels of $PGE_2$ and 5-LOX in culture supernatant from PBMCs with or without LPS were assessed by ELISA. In LPS$na{\ddot{i}}ve$ PBMCs, treatment of t10c12-CLA significantly (p < 0.05) increased the mRNA expressions of PGE2 synthase and 5-LOX compared to vehicle control. Expression of COX-2 mRNA did not show significant difference compared to vehicle control by t10c12-CLA treatment in LPS$na{\ddot{i}}ve$ PBMCs. However, the addition of LPS in PBMCs markedly (p < 0.05) increased the mRNA expression of COX-2, $PGE_2$ synthase and 5-LOX, and also significantly (p < 0.05) enhanced the production of $PGE_2$ and 5-LOX relative to LPS$na{\ddot{i}}ve$ PBMCs, respectively. However, the addition of t10c12-CLA significantly (p < 0.01) suppressed the LPS-induced excessive expression of COX-2, $PGE_2$ synthase, and 5-LOX compared to those of PBMCs treated with LPS alone. The production levels of $PGE_2$ and 5-LOX in culture supernatant from LPS-stimulated PBMCs were also significantly (p < 0.05) inhibited by the treatment of t10c12-CLA compared to LPS alone. These results suggested that t10c12-CLA has an anti-inflammatory effect via dual inhibition of COX-2 and 5-LOX with gene expression and production level in LPS-stimulated porcine PBMCs. Therefore, it was thought that t10c12-CLA can attenuate the inflammatory response by down-regulation of eicosanoids production.

Anti-oxidant, Anti-inflammatory and Anti-allergic effects of mixed herb medicines extracts by steaming process (증숙과정에 따른 생약 혼합추출물의 항산화, 항염 및 항알러지 활성)

  • 본 연구는 국내 생약중 도라지를 포함, 인삼, 당귀, 생강, 마 5가지를 선별하여 각 원물별로 항산화 활성 가장 높은 기준으로 증숙시킨 조건의 흑도라지, 흑삼, 흑당귀, 흑생강, 흑마를 확보하였다. 증숙시킨 5가지 생약은 다시 혼합 추출(BMHE; Black Mixed Herbal Extract)하여 증숙하지 않은 생약 혼합 추출물(MHE; Mixed Herbal extract)과 항산화, 항염 및 항알러지 활성을 비교하여 증숙공정이 생약의 생리활성에 미치는 긍정적인 영향을 바탕으로 피부향장 소재로 활용가능성을 확인하고자 연구하였다. 실험은 MHE와 BMHE를 각각 50% Ethyl alcohol로 추출하여 감압농축하여 생리활성을 평가하였으며, MHE와 BMHE 추출물에 대한 세포생존률을 확인한 결과 $1,000{\mu}g/ml$의 농도에서 각각 89.4%와 95.3%의 세포생존률을 보임에 따라 $1,000{\mu}g/ml$ 이하의 농도에서 HME와 BMHE 모두 세포독성이 거의 적은 것을 확인하였다. DPPH radical scavenging effects 항산화 활성에 대하여 MHE와 BMHE을 비교한 결과 농도 의존적으로 항산화 활성을 보였으며 $1,000{\mu}g/ml$의 농도에서는 각각 66.8%와 74.2%의 라디칼 소거능을 확인하였다. NO assay로부터 항염활성을 비교한 결과에서도 MHE와 BMHE 모두 농도 의존적으로 NO의 함량 감소시켰으며 $1,000{\mu}g/ml$로 처리한 농도에서는 59.5%와 64.7%의 억제율을 각각 보임에 따라 항염 효과도 우수한 것을 확인하였다. 항알레르기 효능 실험 중 하나인 5-LO 억제효능을 측정하여 ME와 BME의 항알러지 효능을 측정한 결과 상기의 결과와 마찬가지로 농도 의존적으로 5-Lipoxygenase를 억제하였으며, $1,000{\mu}g/ml$의 농도로 MHE와 BMHE를 처리하였을 시 5-Lipoxygenase 효소활성에 대하여 각각 76.2%와 83.5%의 높은 억제율을 확인 하였다. 이는 양성대조군인 EGCG를 $200{\mu}g/ml$의 농도로 처리한 9.10%에 근접한 억제율을 보임에 따라 항알러지 효과가 우수한을 확인할 수 있었다. 실험 결과를 바탕으로 BMHE가 MHE보다 항산화, 항염, 항알러지 활성이 우수한 것을 확인 할 수 있었으며, 이는 증숙기법이 생약의 생리활성을 높여주는 것은 물론 더 나아가 본 연구를 통해 확보된 5가지 흑도라지, 흑삼, 흑당귀, 흑생강, 흑마는 향후 피부향장 소재로 활용이 용이할 것으로 사료된다.

Antioxidative and Antiallergic Effects of Aronia (Aronia melanocarpa) Extract (아로니아(Aronia melanocarpa)로부터 유래한 추출물의 항산화 및 항알레르기 효능)

  • The purpose of this study was to investigate the antioxidative and antiallergic effects of aronia extract. The aronia extract was tested for various antioxidative potentials (scavenging activity of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and superoxide anion radical) and inhibitory effect of 5-lipoxygenase (5-LO) and cyclooxygenase (COX). In aronia extract, polyphenol content was $81.6{\pm}3.3\;mg/g$ and flavonoid content was $5.15{\pm}0.16\;mg/g$. DPPH radical-scavenging activity ($SC_{50}$) of aronia extract was $6.15{\pm}0.343\;ppm$. $SC_{50}$ value of aronia extract against superoxide anion radical was $6.99{\pm}1.28\;ppm$. In addition, $IC\_{50}$ values of aronia extract against 5-LO, COX-1, and COX-2 were $42.07{\pm}0.15\;ppm$, $24{\pm}4.29\;ppm$, and $24.3{\pm}3.5\;ppm$, respectively. These results suggest that aronia extract may be useful for the prevention or treatment of allergic disease.

Physicochemical and Sensory Characteristics of Commercial Soybean Sprouts (시판 콩나물의 물리화학적 특성 및 관능적 특성)

  • The physicochemical and sensory characteristics of commercial film-packed soybean sprouts from domestic cultivars were investigated. The hardness of the cotyledons was higher than that of the hypocotyls in fresh and cooked soybean sprouts (Kongnamulmuchim). Furthermore, the color determination showed that the hypocotyls and cotyledons of the soybean sprouts had a light green color that was not eliminated by cooking. The acceptability of fresh soybean sprouts was negatively correlated with a beany odor, beany taste, and grassy taste, but positively correlated with a nutty odor, nutty taste, and sweet taste. In addition, the acceptability of boiled soybean sprouts was negatively correlated with a beany and grassy taste, and positively correlated with a nutty odor and taste. Furthermore, the acceptability of cooked soybean sprouts was negatively correlated with a beany odor, beany taste, grassy odor, and grassy taste, while it was positively correlated with a nutty odor and taste. Finally, the lipoxygenase activity of the cotyledons was higher than that of the hypocotyls in fresh and boiled soybean sprouts.

Effect of Cooking Methods and Reheating on Texture of Sausage (시판소시지의 조리방법 및 재가열이 조직특성에 미치는 영향)

  • The Purpose of this study was to evaluate the effect of cooking methods and reheating on texture characteristics of sausage. Sausage samples were cooked(microwaving, gas-broiling and frying), reheated on day 3 and 6 after refrigeration and put to evaluation of sensory and texture characteristics. The results of sensory evaluation showed that hardness, salty taste, rancid flavor and after-taste were increased but wetness and chewiness were decreased by reheating after refrigeration. Overall acceptability was the highest when the samples were gas broiled and reheated on day 3 after refrigeration. As for the estimation of texture changes by rheometer, hardness, cohesiveness, elasticity, gum-miness and chewiness tended to increase by reheating after refrigeration. Hardness, cohesiveness, gumminess and chewiness were the highest when the samples were gas-broiled and reheated on day 3 after refrigeration.

Anti-oxidant and Anti-inflammation Activity of Fractions from Aster glehni Fr. Schm. (섬쑥부쟁이(Aster glehni Fr. Schm.) 분획물의 항산화 및 항염증 활성)

  • The Plants and their extracts containing polyphenol have been shown to be associated with decreased the cause of aging and variety of disease such as reaction oxygen species (ROS) and reactive nitrogen species (RNS) in several recent studies. We conducted to investigate whether the extracts and fractionation isolated from Aster glehni Fr. Schm. has an inhibitory effect association with oxidation or inflammation. The Aster glehni Fr. Schm. 70% aq. MeOH was fractioned according to polarity with n-hexane layer, EtOAc layer, n-BuOH layer and water layer. The electron donating ability of EtOAc, n-BuOH solvent fraction from Aster glehni Fr. Schm. was about 58.0%, 46.4% at $50\;{\mu}g/mL$, respectively. The superoxide anion radical inhibitory effect of EtOAc extracts was about 64.65% at $50\;{\mu}g/mL$, and n-BuOH extracts was 35.66% at $50\;{\mu}g/mL$. EtOAc layer to the inhibition activity of hyaluronidase and lipoxygenase were inhibited about 24.37%, 29.5% at $5\;{\mu}g/mL$. In the anti-inflammation effect of EtOAc layer inhibited the generation of nitric oxide. also, these results showed that EtOAc extract inhibited 81.5% at $50\;{\mu}g/mL$ on the expressions of iNOS protein in Raw264.7 cell line.
