백호탕 추출물의 지방장벽 손상 개선을 통한 상피 내 Th2 분화 조절 효과 (The Effects of Baekho-tang Extracts on Regulating Th2 Differentiation through Improving Skin Fat Barrier Damage)

  • Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Baekho-tang extract may produce to improves skin lipid barrier damages. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DxT, n=10), and Baekho-tang extract treatment group after lipid barrier elimination group (BHTT, n=10). Baeko-tang extract was administered for 3 days after removal of the skin fat barrier in BHTT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin (FLG), transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7) and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in BHTT group than in LBE group and DxT group. Also, pH and TEWL were significantly decreased with BHTT group. However, FLG was significantly increased in BHTT group compared to LBE, DxT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in BHTT compared to the LBE and DxT. Conclusions As a result of this study, BHT administration decreased pH, TEWL, and increased FLG, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, BHT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.

Optimized cultivation of Ettlia sp. YC001 in eutrophic pond water for nutrient removal and biomass production

  • Ettlia sp. YC001, a highly settleable and productive microalga, was shown to be effective in removing nutrients and capturing suspended solids from eutrophic pond water. The optimum conditions for the Ettlia sp. YC001 cultivation were investigated using water from a landscape pond. The pond water was supplemented with different N : P ratios by weight, and the biomass production and nutrient removal compared in batch cultures. The maximum removal rate of N and P was with an N : P ratio of 16 : 1. Plus, the turbidity dropped to near zero within 4 days. Meanwhile, chemostat cultivation showed that the biomass productivity and nutrient removal rate increased when increasing the dilution rate, where a dilution rate of $0.9d^{-1}$ showed the highest N and P removal rate at $32.4mg\;L^{-1}\;d^{-1}$ and $1.83mg\;L^{-1}\;d^{-1}$, respectively, and highest biomass and lipid productivity at $0.432g\;L^{-1}\;d^{-1}$ and $67.8mg\;L^{-1}\;d^{-1}$, respectively. The turbidity was also reduced by 98% in the chemostat cultivation. Moreover, auto-flocculation and pH were closely connected to the turbidity removal. As a result, this study identified the optimal N : P ratio for small pond water treatment using an Ettlia sp. YC001, while also establishing the optimal conditions for nutrient removal, turbidity reduction, and biomass production.

A Cost Analysis of Microalgal Biomass and Biodiesel Production in Open Raceways Treating Municipal Wastewater and under Optimum Light Wavelength

  • Open raceway ponds are cost-efficient for mass cultivation of microalgae compared with photobioreactors. Although low-cost options like wastewater as nutrient source is studied to overcome the commercialization threshold for biodiesel production from microalgae, a cost analysis on the use of wastewater and other incremental increases in productivity has not been elucidated. We determined the effect of using wastewater and wavelength filters on microalgal productivity. Experimental results were then fitted into a model, and cost analysis was performed in comparison with control raceways. Three different microalgal strains, Chlorella vulgaris AG10032, Chlorella sp. JK2, and Scenedesmus sp. JK10, were tested for nutrient removal under different light wavelengths (blue, green, red, and white) using filters in batch cultivation. Blue wavelength showed an average of 27% higher nutrient removal and at least 42% higher chemical oxygen demand removal compared with white light. Naturally, the specific growth rate of microalgae cultivated under blue wavelength was on average 10.8% higher than white wavelength. Similarly, lipid productivity was highest in blue wavelength, at least 46.8% higher than white wavelength, whereas FAME composition revealed a mild increase in oleic and palmitic acid levels. Cost analysis reveals that raceways treating wastewater and using monochromatic wavelength would decrease costs from 2.71 to 0.73 $/kg biomass. We prove that increasing both biomass and lipid productivity is possible through cost-effective approaches, thereby accelerating the commercialization of low-value products from microalgae, like biodiesel.

흡연인들에서 증가된 혈장지질 농도가 비타민 E 영양상태와 글루타티온 과산화효 활성에 미치는 영향 (The Relation of the Elevated Plasma Lipid Levels to Plasma Vitamin E Status and Activities of Erythrocyte Glutathione Perosicase in Smokers)

  • This study has done to investigate the relationship between the icreased lipid oncentration caused by smoking and plama levels of vitamin A and vitamin E, antiodative enzyme activity, and lipid peroxidation , in 52 male smokers and 32 non-smokers, Dietary vitamin A and vitamin E intake was imilar in both smokers and non-smokers. Absolute plasma concentrations of vitamin A and vitamin E were not significantly different between two groups, whereas vitamin E/cholesterol ration in plasma was low or in smokers than in that of non-smokers(p<0.05). It was considered that this lowered effect was due to the elevated plasma lipid concentration rather than oxidant stress derived from smoking, in view of the fact that smokers had higher cholesterol (15.2%) adn LDL-C(26.6%) levels than non-smokers. In non-smokers, plasma thiobarbiturin acid reactive substances(TBARS) conrrelated positively with total cholesterol(r=0.63466, p<0.001), LDL-C level(r=0.57166, p<0.01) , and LDL-C/HDL-C ratio(r=0.45926, p<0.05) . Activities of glutathione perosidase(GSH-Px) , superoside dismutase(SOD), and catalse made no difference in both groups. However, it was observed in non-smokers that GSH-Px activity had negative correlations with total cholesterol(r=-0.67293, p<0.001), LDL-C level(r=-0.62878, p<0.001), and LDL-C/HDL-C ratio (r=-0.58824, p<0.01), indicating that there was a dependent relationship between lipid perosidation and plasma lipid level. The smokers also showed negative correlations for GSH-Px activity with total cholesterol (r=-0.29946, p<0.05) and LDL-c level (r=0.45914, p<0.001), and LDL-C/HDL-c ratio(r=-0.35438, p<0.05). It seemed that the lipid that the lipid level elevated by sustaines smoking resulted in reducing vitamin E/cholesterol ratio and proportion of antioxidant to oxidant load, and then GSH-Px activity, with insufficient removal of free radicals(TBARS 2.43$\pm$0.51 and 1.81$\pm$0.15nmol/ml in smokers and non-smokers, respectively). These findings suggest that higher plasma lipid levels may play a more important role in perturbing the antioxidant defense system including vitamin E status and GSH-Px activity, at least in circumstances that increase lipid concentration . In addition, in exposure to free radicals like those in cigarette smoke. In those cases the ratio of vitamin E/lipid in plasma can be a more indicator of vitamin E status than plasma levels of vitamin E alone.

Expression of lipid metabolism genes provides new insights into intramuscular fat deposition in Laiwu pigs

  • Objective: The objective of this study was to measure the special expression pattern of lipid metabolism genes and investigate the molecular mechanisms underlying intramuscular fat (IMF) deposition in Longissimus dorsi muscle of Laiwu pigs. Methods: Thirty-six pigs (Laiwu n = 18; Duroc×Landrace×Yorkshire n = 18) were used for the measurement of the backfat thickness, marbling score, IMF content, and expression of lipid metabolism genes. Results: Significant correlations were found between IMF content and the mRNA expression of lipid metabolism genes. Of the 14 fat deposition genes measured, fatty acid synthase (FASN) showed the strongest correlation (r = 0.75, p = 0.001) with IMF content, and of the 6 fat removal genes, carnitine palmitoyl transferase 1B (CPT1B) exhibited the greatest negative correlation (r = -0.66, p = 0.003) with IMF content in Laiwu pig. Multiple regression analysis showed that CPT1B, FASN, solute carrier family 27 member 1 (SLC27A1), and fatty acid binding protein 3 (FABP3) contributed 38% of the prediction value for IMF content in Laiwu pigs. Of these four variables, CPT1B had the greatest contribution to IMF content (14%) followed by FASN (11%), SLC27A1 (9%), and FABP3 (4%). Conclusion: Our results indicate that the combined effects of an upregulation in fat deposition genes and downregulation in fat removal genes promotes IMF deposition in Laiwu pigs.

  • Maltol(2-methyl-3-hydroxy-r-pyrone), a component known to be present in Korean Ginseng root showed an antioxidant action but its potency as an antioxidant was low; about 1150th that of other antioxidants such as p-phenylenediamine , BHA and BHT. However, maltol was able to protect the oxidation adamants in biological systems such as adriamycin-induced membrane damage in isolated cardiomyocytes, parquet-induced toxicities in isolated hepatocytes and repercussion injury in isolated hearts. The antioxidant action of maltol was also shown to be effective in vivo. The antioxidant action of this compound was probably due to the removal of hydroxyl radicals. In view of the roles of oxygen radical in various pathological processes, Korean Ginseng root, which contains several antioxidants including maltol, is expected to have beneficial efforts on the oxygen radical-involved processes. The Novel Angiotensin I Converting Enzyme Inhibitory Peptide from Rainbow Trout Muscle Hydrolysate

  • The purpose of this study was the purification and characterization of an angiotensin I converting enzyme (ACE) inhibitory peptide purified from enzymatic hydrolysates of rainbow trout Oncorhynchus mykiss muscle. After removal of lipid, the approximate composition analysis of the rainbow trout revealed 24.4%, 1.7%, and 68.3% for protein, lipid, and moisture, respectively. Among six hydrolysates, the peptic hydrolysate exhibited the highest ACE inhibitory activity. We attempted to purify ACE inhibitory peptides from peptic hydrolysate using high performance liquid chromatography on an ODS column. The $IC_{50}$ value of purified ACE inhibitory peptide was $63.9{\mu}M$. The amino acid sequence of the peptide was identified as Lys-Val-Asn-Gly-Pro-Ala-Met-Ser-Pro-Asn-Ala-Asn, with a molecular weight of 1,220 Da, and the Lineweaver-Burk plots suggested that they act as a competitive inhibitor against ACE. Our study suggested that novel ACE inhibitory peptides purified from rainbow trout muscle protein may be beneficial as anti-hypertension compounds in functional foods.

Metastatic lipid-rich mammary carcinoma in a dog

  • An adult female dog was presented for evaluation of mammary gland masses. Complete blood count and serum chemistry data were within normal limits. Fine-needle aspiration cytology of the mammary masses revealed clusters of malignant epithelial cells with clear cytoplasmic vacuoles. Based on histopathological findings, a diagnosis of lipid-rich mammary carcinoma was made. Approximately 5 weeks after surgical removal, the tumor recurred at the surgery site and metastasis to the tibia was detected. Due to the poor prognosis and deterioration of the condition, the dog was euthanized.

  • To compare seed components of plus trees, seed powder ground after seedcoat removal was analyzed for two oak species, i. e., Quercus monglica (white oak) and Quercus variabilis (red oak), which are typical oak trees in Korea but have different fruiting characteristics. Thus we aimed at analyzing and comparing many ingredients including minerals, sugars, etc. Two species were similar to each other in the content of water, crude ash, crude protein and carbohydrates, but crude lipid content in Q. variabilis was 2.5 times higher than that in Q. mongolica. Crude proteins of Clone 124 was 1.5 times higher than that of Clone 75 in Q. mongolica. Crude lipid content showed the highest value in Clone 0511 of Q. variabilis, and more phosphate and iron was found in Q. monglica than in Q. variabilis. Glucose showed 85.4% and 88.3% on average of the total monosacchrides in two species, and galactose and arabinose were also found. In the content of phosphate, iron, and crude lipid, differences were found between two species and among clones of two species.
