Flavonoids and chlorogenic acid from Eriobotrya japonica scavenge peroxynitrite

  • Peroxynitrite is a cytotoxic intermediate produced by the reaction between the superoxide anion radical and nitric oxide. Flavonoids (afzelin, quercitrin and quercetin 3-O-sambubioside), and chlorogenic acid and its methyl ester obtained from leaves of loquat (Eriobotrya japonica) have recently been shown to scavenge 1,1-diphenyl-2-picrylhydrazyl radical and to inhibit lipid peroxidation in mouse liver homogenate. The aim of this study is to investigate the inhibitory effects of the above components on peroxynitrite produced stimulated by 3-morpholinosydnonimine (SIN-1) to produce superoxide anion radical and nitric oxide at the same time. In addition, the present study tests whether or not the components directly scavenge peroxynitrite itself. The results showed that the components with the aromatic ortho-dihydroxyl groups (catechol) were more potent inhibitors of peroxynitrite formation by SIN-1. In particular, the methyl ester form of chlorogenic acid showed the most potent inhibition. At $5\;{\mu}M$ concentration, the order of minimizing peroxynitrite formation were : methyl chlorogenic acid > quercitrin > quercetin 3-O-sambubioside > chlorogenic acid > afzelin. Authentic peroxynitrite was directly scavenged by the components in a manner similar to peroxynitrite formation by SIN-1. In particular, when compared with penicillamine as a positive control, methyl chlorogenate was as effective in inhibiting peroxynitrite formation and approximately 2 times more effective in scavenging an authentic peroxynitrite. These results demonstrate therefore, that components extracted from the leaves of Eriobotrya japonica effectively scavenged peroxynitrite.

Neuroprotective and Anti-Neuroinflammatory Activities of Anthraquinones Isolated from Photorhabdus temperata Culture Broth

  • Photorhabdus temperata (PT), a gram-negative bacterium, lives symbiotically within entomopathogenic nematodes. The insecticidal compounds derived from Photorhabdus are used as biopesticides in agriculture. However, the physiological properties are not well characterized. In the course of our screening for neuroprotective and anti-neuroinflammatory substances from natural products, the culture broth of PT showed considerable activities. By activity-guided purification, five anthraquinones, namely, 3-methoxychrysazine (1), 1,3-dimethoxy-8-hydroxy-9,10-anthraquinone (2), 1,3,8-trihydroxy-9,10-anthraquinone (3), 3,8-dihydroxy-1-methoxy-9,10-anthraquinone (4), and 1,3,4-trimethoxy-8-hydroxy-9,10-anthraquinone (5), were isolated from the ethyl acetate fraction of the PT culture broth. Among the isolated compounds, $75{\mu}M$ 3 significantly protected mouse hippocampal neuronal cells (HT22) against 5 mM glutamate-induced cell death via the inhibition of reactive oxygen species production, $Ca^{2+}$ influx, and lipid peroxidation. Additionally, 3 and 4 effectively suppressed the interferon-${\gamma}$-induced neuroinflammation of mouse-derived microglial cells (BV2) at 10 ng/ml, via the reduction of nitric oxide, interleukin-6, and tumor necrosis factor-${\alpha}$. Anthraquinones 3 and 4 derived from the PT culture broth are a potential starting point to discover neuroprotective and anti-neuroinflammatory drug leads. The novel compound 5 is reported for the first time in this study.

Metronidazole Reduced Ammonia Toxicity in Human Hep G2 cell and Rat Hepatocytes (Hep G2 세포와 rat 간세포에서 Metronidazole에 의한 암모니아 독성 감소)

  • Lipophilic ammonia is toxic gas and can easily diffuse across cell membranes. Excess ammonia is implicated in the pathogenesis of several metabolic disorders including hepatic encephalopathy and may result in the death. The purpose of this study was to clarify the inhibition effect of metronidazole on liver cell damage due to ammonia in human Hep G2 cell and rat hepatocytes. The effects of metronidazole were studied in ammonium chloride treated human Hep G2 cell (75 mM) and rat hepatocyte (100 mM) following $0.1{\mu}M$ metronidazole treatment. In MTZ+AC group, cell viabilities increased prominently and LDH activities decreased over 25% than AC group. Furthermore, ammonia level according to ammonium chloride treatment reduced over 30% and lipid peroxidation as an index of cell membrane damage decreased more than twice. By comparison with control, catalase activity showed more than 30% reduction in AC group while less than 10% reduction in MTZ+AC group, respectively. In addition, MTZ+AC group showed the similar cell structure as control in cell morphology study by using light microscope, and represented fluorescent intensity decrement compared with AC group in fluorescent microscopic study with avidin-TRITC fluorescent dye. And cleaved PARP expression due to ammonia reduced twofold or more in MTZ+AC group. As the results suggest, metronidazole may protect the liver cell by inhibiting cell damages due to ammonia and be used for an effective antagonist of ammonia in hyperammonemia.

Allium Jesdianum Extract Improve AcetaminophenInduced Hepatic Failure through Inhibition of Oxidative/Nitrosative Stress

  • Objectives: Allium jesdianum (Aj) is a medicinal plant that has highlighted pharmacological features. In this study, the effects of Aj extract were examined on acetaminophen (APAP)-induced hepatic failure in rats. Methods: Methanolic fraction of hydro-alcoholic extract of Aj was obtained by silica gel column chromatography method. Animals were randomly divided into four groups each containing six rats and treated by gavage as follows: the first and second groups received normal saline, the third and fourth groups were received with 50 and 100 mg/kg of Aj extract, respectively. After two consecutive weeks, the groups 2-4 were given a single dose of APAP (2 g/kg). After 48 hours, blood and liver samples were collected for biochemical and histological examinations. Results: The findings of the study demonstrated that APAP caused a significant increase in ALT (P < 0.001), AST (P < 0.001), LDH (P < 0.001), ALP (P < 0.001) serum levels, hepatic lipid peroxidation (LPO; P < 0.001) and nitric oxide (NO; P < 0.001). In this regard, APAP led to the depletion of the total antioxidant capacity (TAC; P < 0.001), glutathione and total thiol groups (TTGs; P < 0.001), and structural change in the liver. In the Aj extract groups, a considerable improvement was found in the hepatic function alongside the histopathologic changes. Conclusion: This investigation indicated that the influential effects of Aj extract in APAP-induced hepatic failure might depend on its effect on improving oxidant/antioxidant balance in hepatic tissue.

Nephroprotective Effects of Taeksa-san Aqueous Extracts on Cisplatin-induced Rat Acute Renal Failure (택사산(澤瀉散) 추출물(抽出物)이 Cisplatin으로 유발(誘發)된 흰쥐의 급성신부전(急性腎不全)에 미치는 영향(影響))

TSS has generally been used for treating various renal diseases, including renal failure. Methods : Three different dosages of TSS extract were orally administered once a day for 28 days, At the 23rd day after TSS extract treatment, cisplatin was also treated. Then, 5 days after cisplatin treatment, all the rats (6 groups of 8 rats each) were sacrificed. Changes in the body weight, kidney weight, serum BUN and creatinine levels were observed with changes on the kidney MDA and GSH contents. The results were compared with captopril 100mg/kg, of which the effects on cisplatin-induced acute renal failures are already confirmed. Results : Dramatic decrease of body weight, increase of kidney weight, increase of serum BUN and creatinine levels were detected in the cisplatin control as compared with the intact control. In addition, marked increase of kidney MDA contents and decrease of kidney GSH contents were also detected in the cisplatin control as compared with the intact control. This means that cisplatin-induced ARF is induced by oxidative stress and related lipid peroxidation in the present study. However, these ARFs and inhibition of antioxidant effects induced by cisplatin were dose-dependently reduced by treatment with all three different dosages of TSS extracts. Conclusion : This study suggests that TSS extracts show favorable effects on cisplatin-induced rat ARF.

Methods : At first, three different dosages of MJMTS extract were orally administered once a day for 28 days. On the 23rd day after MJMTS extract treatment, cisplatin was treated. Then, 5 days after cisplatin treatment, all the rats (6 groups of 8 rats each) were checked in the present study. Nephroprotective Effects of Opi-san Aqueous Extracts on Cisplatin-induced Rat Acute Renal Failure (오피산(五皮散)이 Cisplatin으로 유발(誘發)된 흰쥐의 급성신부전(急性腎不全)에 미치는 영향(影響))

On the 23rd day after OPS extract treatment, cisplatin was treated. All the rats (6 groups of 8 rats each) were checked as follows. Changes of body weight, kidney weight, serum BUN and creatinine levels were observed, as well as changes to the kidney MDA and GSH contents. The results were compared with captopril 100 mg/kg, of which the effects on cisplatin-induced acute renal failures were already confirmed. Results : Cisplatin-induced ARF were induced by oxidative stress and related lipid peroxidation in the present study. However, these ARF and inhibition of antioxidant effects induced by cisplatin were dose-dependently reduced by treatment of OPS extracts except in the 100 mg/kg treated group. The effects of OPS extracts at 500 mg/kg were similar to those of 100 mg/kg of captopril. Conclusion : This study suggests that OPS extracts 500 mg/kg showed favorable effects on the cisplatin-induced rat's ARF as similar to 100 mg/kg of captopril.

Evaluation and Comparison of Antioxidant Activities of Nineteen Commercial Carbonated Vinegar Liquors (19종 시판 탄화초액의 항산화활성의 평가 및 비교)

  • Biological functionality of nineteen commercially available carbonated vinegar liquors including wood vinegar liquor (WVL), bamboo vinegar liquor (BVL) and chaff vinegar liquor (CVL) were evaluated, focusing mainly on electron donating ability to DPPH radical, reducing power against ferricyanide ($Fe^{3+}$), blockading ability to linoleic acid autoxidation and NO production from LPS-stimulated RAW264.7 cells plus cytotoxicity to RAW264.7 cells. The results showed that crude carbonated vinegar liquors, regardless of their source materials, have high capacity of antioxidation such as electron donating ability, reducing power, blockading ability to lipid peroxidation and NO production, as well as cell cytotoxicity. Refined carbonated vinegar liquors for skin care or bath showed significantly low cell cytotoxicity, however, overall antioxidant potencies were also low. Especially, these carbonated vinegar liquors revealed low levels of inhibition for NO production deeply involved in inflammation. Among nineteen carbonated vinegar liquors examined, chaff vinegar liquor was observed to be the most potent carbonated vinegar liquor with high antioxidant activities together with low cytotoxicity to mammalian cells.

Anti-proliferative and Antioxidant Activities of 1-methoxy-3-methyl-8-hydroxy-anthraquinone, a Hydroxyanthraquinoid Extrolite Produced by Amycolatopsis thermoflava strain SFMA-103

  • Actinobacteria are prolific producers of a large number of natural products with diverse biological activities. In the present study, an actinobacterium isolated from sunflower rhizosphere soil sample collected from Medak, Andhra Pradesh, South India was identified as Amycolatopsis thermoflava strain SFMA-103. A pigmented secondary metabolite in culture broth was extracted by using methanol and it was further purified by silica gel column chromatography with methanol-chloroform solvent system. Structural elucidation studies based on UV-visible, 1D and 2D-NMR, FT-IR, and mass spectroscopic analyses confirmed the structure as 1-methoxy-3-methyl-8-hydroxy-anthraquinone. It showed significant in vitro anticancer activity against lung cancer and lymphoblastic leukemia cells with $IC_{50}$ values of 10.3 and $16.98{\mu}M$, respectively. In addition, 1-methoxy-3-methyl-8-hydroxy-anthraquinone showed good free radical scavenging activity by DPPH method with an $EC_{50}$ of $18.2{\mu}g/ml$. It also showed other promising superoxide radical scavenging, nitric oxide radical scavenging and inhibition of lipid peroxidation activities. This is a first report of anti-proliferative and antioxidant activities of 1-methoxy-3-methyl-8-hydroxy-anthraquinone isolated from A. thermoflava strain SFMA-103 which may find potential application in biotechnological and pharmaceutical fields.

  • The purpose of this study was to investigate the antioxidant activities of water soluble browning reaction products (WS-BRPs) isolated 5\ulcorneron korea red ginseng. Antioxidant activities of WS-BRPs were examined with the various systems. All three WS-BRPs (L, S-1 and S-2) were found to have an ability to linoleic acid, Ox-brain autoxidant, Fe$^{2+}$ ADP/NAD system and cumene hydroperoxide system. Especially, S-2 was had the strongest activity of theses three WS-BRPs to scavenge free radicals such as more effective than S-1, L. MDA determination showed the antioxidant effect on linoleic acid oxidation inhibition ratio of 22.5%, 31.7%, 31.9% and 33.5%, respectivity Especially; Ox-brain autoxidant was strong inhibited activity by 49.52%,62,44,97.54% by addition of various concentration. But three WS-BRPs showed weak inhibitory activity on lipid peroxidation in rat hepatic microsomes induced enzymatically and nonenzymaticallyly
