Influence of Total Saponin from Korean Red Ginseng on Structural Changes in Phospholipid Membranes and Ghost Erythrocytes (고려홍삼의 총사포닌에 의한 인지질막과 적혈구막의 구조적 변화)

  • Total saponin from Korean red ginseng changed thermodynamic parameters of membranes from dipalmitoylphosphatidylcholine (DPPC) and ghost erythrocytes of human. In liposomes from DPPC, temperature of the main transition (Lb'-La) in liquid-crystalline phase increases by 0.2$^{\circ}C$ in average, but enthalpy does not change. Total saponin at a concentration of smaller than $10^5$% "stabilizes" the timid bilayers. At larger than 0.07 of saponin/DPPC ratio, saponin leads to an exclusion of the bound lipid molecules from the main phase transition into lamella liquid crystalline La-phase. Total saponin influences specifically all erythrocyte membrane transitions in a concentration-dependent manner, i.e. on the structures of all the main membrane skeleton proteins. A high structural specificity of saponin with membrane proteins, could be a base of specificity of physiological response of not only erythrocytes, but also other cells.her cells.

  • The effect of vesicular L-$\alpha$-phosphatidylcholine on the rate of formation of all-trans-N-retinylidene-n-butylamine (3) and on the regeneration kinetics of bacteriorhodopsin pigment from retinal and bacterio-opsin have been studied. An estimate of the relative positions of retinal and n-butylamine in the vesicles has been made by fluoresence quenching experiments. Partition coefficient of retinal and microviscosity of the retinal-binding region have also been determined. The results are discussed in terms of the nature of chemical interaction between retinal and amine in a lipid environment.

  • The effects of polyacetylenes on living membrances, rat erythrocyte and murine leukemic L1210 cell as well as artificial lipid bilayer were determined to investigate the cytotoxic mechanism of polyacetylenes against cancer cell lines. As results, panaxydol and panaxynol caused erythrocyte hemolysis dose-dependently while panaxytriol had no lysis. For liposomes composed of phosphatidyl choline (PC) and phosphatidic acid(PA), all three polyacetylenes supressed the osmotic behavior at the same degree.

  • Previously we showed that $Ba^{2+}$ block of large conductance $Ca^{2+}$-activated $K^{+}$ (BK) channel was larger in the planar lipid bilayer formed with negatively-charged phosphatidylserine (PS) than neutral phosphatidylethanolamine (PE).   • Effects of SDS and temperature on the conformational changes of bacteriorhodopsin were studied using a, b, c bands of bacteriorhodopsin. In the SDS denaturation, bacteriorhodopsin in purple membrane was more labile than bacteriorhodopsin reconstituted into PC vesicles. These rather interesting results may be understood by effective SDS concentration in lipid layer.

  • Effects of dimethipin on the senescence of excised barley first leaves were investigated. Dimethipin markedly inhibited chlorophyll and protein loss and reduced peroxidase activity relevant to senescence phenomena in th excised leaves. Dimethipin decreased hydrogen peroxide content, later malondialdehyde content, and increased the activities of superoxide dismutase and catalase. The antisenescence effects of dimethipin may result from the stabilization of membrane structure through inhibiting the peroxidation of unsaturated lipid and the accumulation of free radicals during senescence.
