Lipid accumulation product (LAP) is a new index reflecting central lipid accumulation and is known to be a strong independent indicator for identifying the risk of cardiovascular disease (CVD) or diabetes mellitus (DM). This study was conducted to assess the relationship between the homeostasis model assessment of insulin resistance (HOMA-IR) and LAP according to the presence or absence of obesity in Korean adults. The study was carried out using data from the 2019 Korean National Health and Nutrition Examination Survey (KNHANES) and included 6,090 adults aged 20 years or older. There were several key findings. First, after adjusting for related variables, the mean of the HOMA-IR levels (M±SE, 95% confidence interval) was positively associated with the quartiles of LAP in the overall population (P<0.001), non-obese (P<0.001) or obese groups (P<0.001). Second, in all the groups (overall population, non-obese and obese groups), the mean value of the fasting blood glucose (all, P<0.001), insulin (all, P<0.001), and the metabolic syndrome score (all, P<0.001) increased with the increasing quartiles of LAP. Insulin resistance was thus positively associated with an increase in the LAP in Korean adults with or without obesity.

In relation to the global increase trend of obesity population, there is a demand for the development of foods having high functional activity by mass-extracting anti-obesity active substances using mulberry product such as leaf and fruit. Therefore, we evaluated the antiobesity efficacy according to varieties by using the mulberry leaves and fruit extracts. At the same time, high active varieties were selected. For this purpose, the effects of the extracts of the mulberry leaf and fruit on 3T3-L1 adipocyte differentiation were examined. As a result, in the case of mulberry leaves, the lipid accumulation inhibitory rate of 'Cheongolppong' was higher than that of the control at $500{\mu}g/ml$ treatment. And in the case of the extract of mulberry fruit, 'Daesim' showed the highest lipid accumulation inhibitory rate compared with the control at 50 times of diluted extract.

Obesity increases the risk of mortality and morbidity because it results in hypertension, heart disease, and type 2 diabetes. Therefore, there is an urgent need for pharmacotherapeutic drugs to treat obesity. We performed a screening assay using natural products with anti-adipogenic properties in 3T3-L1 cells and determined that tschimganidine, a terpenoid from the Umbelliferae family, inhibited adipogenesis. To evaluate the anti-obesity effects of tschimganidine in vivo. Mice were fed either a normal chow diet (NFD) or a high-fat chow diet (HFD) with or without tschimganidine for 12 weeks. Treatment with tschimganidine decreased lipid accumulation and adipogenesis, accompanied by reduced expression of adipogenesis and lipid accumulation-related factors. Tschimganidine significantly increased the phosphorylation of AMP-activated protein kinase (AMPK) and decreased that of AKT. Depletion of AMPK relieved the reduction in lipid accumulation resulting from tschimganidine treatment. Moreover, tschimganidine administration drastically reduced the weight and size of both gonadal white adipose tissue (WAT) and blood glucose levels in high-fat diet-induced obese mice. We suggest that tschimganidine is a potent anti-obesity agent, which impedes adipogenesis and improves glucose homeostasis. Tschimganidine can then be evaluated for clinical application as a therapeutic agent.
The LAP (lipid accumulation product) index is an indicator of excessive fat accumulation in combination with the fasting triglyceride concentration and waist circumference. This study examined the relationship between the LAP index and ALT (alanine aminotransferase) level and the effect of the LAP index on the ALT level. Cross-sectional studies were conducted on healthy adult males without a history of liver disease. From January 2015 to June 2017, 13,854 adults between 20 and 70 years of age, who underwent health screening at a general hospital in Gyeonggi-do, were enrolled in this study. The LAP index was calculated as [waist circumference (cm)-65]${\times}$[triglyceride (mmol/L)] for males. Serum ALT abnormalities were set at 40 IU/L or more in males. All subjects underwent measurements of the anthropometric indicators and physiological examinations. A significant increase in ALT with increasing LAP quintiles was observed (p<0.001). The LAP index correlated with ALT (r=0.238, p<0.001). Insulin (r=0.449, p<0.001) and HDL-cholesterol (r=-0.369, p<0.001) were strongly correlated with LAP. In addition, the LAP index is a factor affecting the ALT level (p<0.001). As the LAP index increased, the risk of an elevated ALT was higher (p<0.001). In conclusion, the LAP index was a predictor of an ALT elevation in Korean men.

With the COVID-19 pandemic, there is increasing interest in anti-obesity strategies. According to the National Statistical Office, the obesity rate in Korea was 38.3% in 2020 and 37.1% in 2021. Obesity is a risk factor for several severe diseases, including stroke, heart disease, type 2 diabetes, and certain types of cancer. Pinus rigida × Pinus taeda is a hybrid of Pinus rigida Mill and Pinus taeda Linn, and its cones are considered a by-product. Although previous studies have investigated their pharmacological effects on antioxidant activity and protection against oxidative DNA damage, few researchers have explored their potential as functional natural materials. Therefore, we evaluated the anti-obesity effects of the cone of ethyl acetate fraction of P. rigida × P. taeda (ERT), specifically its ability to inhibit lipid accumulation. Our analysis showed that ERT contains phytochemicals (catechin and caffeic acid) which are known to improve immune function and inhibit cell damage. ERT inhibited lipid droplet accumulation at the cellular levels through Oil Red O staining. Furthermore, ERT suppressed the expression of adipogenic transcription factors (PPARγ and CEBP/α) as well as downstream lipogenic target genes (FAS and SREBP-1) thereby inhibiting adipogenesis. ERT also down-regulated key adipogenic markers, including aP2α, while inducing the phosphorylation of AMPK. It has been reported that PPARγ and CEBP/α are expressed in the early stages of adipose differentiation, while SREBP-1 is expressed in the late stage. Therefore, our findings suggest that ERT activates AMPK signaling pathways, which inhibits adipogenic transcription factors (PPARγ, C/EBPα, and SREBP1) and lipogenic genes (FAS and aP2α), thereby blocking lipid accumulation and preventing obesity and related disorders. ERT showed potential as a new resource for developing a functional material for anti-obesity agents.
BACKGROUND/OBJECTIVES: Abeliophyllum distichum is a plant endemic to Korea, containing several beneficial natural compounds. This study investigated the effect of A. distichum leaf extract (ALE) on adipocyte differentiation. MATERIALS/METHODS: The cytotoxic effect of ALE was analyzed using cell viability assay. 3T3-L1 preadipocytes were differentiated using induction media in the presence or absence of ALE. Lipid accumulation was confirmed using Oil Red O staining. The mRNA expression of adipogenic markers was measured using RT-PCR, and the protein expressions of mitogen-activated protein kinase (MAPK) and peroxisome proliferator-activated receptor gamma (PPAR𝛾) were measured using western blot. Cell proliferation was measured by calculating the incorporation of Bromodeoxyuridine (BrdU) into DNA. RESULTS: ALE reduced lipid accumulation in differentiated adipocytes, as indicated by Oil Red O staining and triglyceride assays. Treatment with ALE decreased the gene expression of adipogenic markers such as Ppar𝛾, CCAAT/enhancer binding protein alpha (C/ebp𝛼), lipoprotein lipase, adipocyte protein-2, acetyl-CoA carboxylase, and fatty acid synthase. Also, the protein expression of PPAR𝛄 was reduced by ALE. Treating the cells with ALE at different time points revealed that the inhibitory effect of ALE on adipogenesis is higher in the early period treatment than in the terminal period. Furthermore, ALE inhibited adipocyte differentiation by reducing the early phase of adipogenesis and mitotic clonal expansion. This was indicated by the lower number of cells in the Synthesis phase of the cell cycle (labeled using BrdU assay) and a decrease in the expression of early adipogenic transcription factors such as C/ebp𝛽 and C/ebp𝛿. ALE suppressed the phosphorylation of MAPK, confirming that the effect of ALE was through the suppression of early phase of adipogenesis. CONCLUSIONS: Altogether, the results of the present study revealed that ALE inhibits lipid accumulation and may be a potential agent for managing obesity.
This study investigated the effects of fermented product from Bacillus subtilis (FPBS) on feed conversion efficiency, fat accumulation and ammonia production in broiler chicks. Sixty female broilers (strain Chunky, 7-day old) were divided into four groups and raised in individual cages. One group was fed a commercial diet without supplementation of FPBS as the control and the other three groups were fed commercial diets containing FPBS, either 0.5, 1.0 or 2.0%, for 21 days from 7 to 28 days of age. Water and feed were given ad libitum. Feed conversion efficiency was significantly improved in chicks supplemented with 0.5 or 1.0% of FPBS as compared with the control (p<0.05). The activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase, and contents of triglyceride and cholesterol in the liver were significantly decreased in treatment groups (p<0.05) as compared with the control group. FPBS had no effect on the concentration of plasma triglyceride, phospholipids and cholesterol. Feeding FPBS at 1 % or 2% levels reduced ammonia gas release (p<0.05). The inclusion of FPBS at 1 % level may be recommended both to improve production efficiency and to reduce air pollution caused by ammonia gas release. For production efficiency to reach maximal profit, the inclusion of FPBS at 0.5% level can be recommended. Feeding FPBS reduced fat accumulation in the liver.
Thirty-day-old seedlings of tomato (Lycopersicon esculentum) were treated with different concentrations of HgCl$_2$(0. 10 and 50 $\mu$M) for up to 20 days. and the detailed distribution of Hg absorbed and its toxicity in different plant parts (roots, stems and leaves) were investigated. The accumulation of Hg in plants increased with external Hg concentrations. and Hg is strongly retained by roots. Further. Hg content in leaves was various. showing more accumulation in older leaves. Seedlings exposed to toxic levels of Hg showed not only the reduction of dry weight and length of both shoot and root. and chlorophyll levels in leaves but also the enhancement of malondialdehyde (a lipid peroxidation product) formation in all plant parts investigated. These results suggest that physiological impairment of a plant exposed to Hg may be achieved by internal distribution of Hg absorbed and Hg-induced oxidative stress in different plant parts.
The effect of salicylic acid(SA) on antioxidant system and protective mechanisms against UV-B induced oxidative stress was investigated in cucumber(Cucumis sativus L.) leaves. UV-B radiation and SA were applied separately or in combination to first leaves of cucumber seedlings, and dry matter accumulation, lipid peroxidation and activities of antioxidant enzymes were measured in both dose and time-dependant manner. UV-B exposure showed reduced levels of fresh weight and dry matter production, whereas SA treatment significantly increased them. SA noticeably recovered the UV-B induced inhibition of biomass production. UV-B stress also affected lipid peroxidation and antioxidant enzyme defense system. Malondialdehyde(MDA), a product of lipid peroxidation, was greatly increased under UV-B stress, showing a significant enhancement of a secondary metabolites, which may have antioxidative properties in cucumber leaves exposed to UV-B radiation. Combined application of UV-B and SA caused a moderate increase in lipid peroxidation. These results suggest that SA may mediate protection against oxidative stress. UV-B exposure significantly increased SOD, APX, and GR activity compared with untreated control plants. Those plants treated with 1.0 mM SA showed a similar pattern of changes in activities of antioxidant enzymes. SA-mediated induction of antioxidant enzyme activity may involve a protective accumulation of $H_2O_2$ against UV-B stress. Moreover, their activities were stimulated with a greater increase by UV-B+SA treatment. The UV-B+SA plants always presented higher values than UV-B and SA plants, considering the adverse effects of UV-B on the antioxidant cell system. ABA and JA, second messengers in signaling in response to stresses, showed similar mode of action in UV-B stress, supporting that they may be important in acquired stress tolerance. Based on these results, it can be suggested that SA may participates in the induction of protective mechanisms involved in tolerance to UV-B induced oxidative stress.
Background: Lipid accumulation product (LAP) is associated with the presence and severity of nonalcoholic fatty liver disease (NAFLD) in adults. Purpose: Here we evaluated the ability of LAP to predict NAFLD in obese children. Methods: Eighty obese children (38 girls; age 6-18 years) were included. Anthropometric measurements and biochemical values were obtained from the patients' medical records. LAP was calculated as [waist circumference (WC) (cm) - 58]×triglycerides (mmol/L) in girls; [WC (cm) - 65]×triglycerides (mmol/L) in boys. The minLAP and adjLAP were described (3% and 50% of WC values, respectively) and the total/high-density lipoprotein cholesterol index (TC/HDL-C) was calculated. NAFLD was observed on ultrasound, and patients were divided into 3 groups by steatosis grade (normal, grade 0; mild, grade 1; moderate-severe, grade 2-3). The area under the curve (AUC) and appropriate index cutoff points were calculated by receiver operator characteristic analysis. Results: LAP was positively correlated with puberty stage (rho=0.409; P<0.001), fasting insulin (rho= 0.507; P<0.001), homeostasis model assessment of insulin resistance (rho=0.470; P<0.001), uric acid (rho=0.522; P<0.001), and TC/HDL-C (rho=0.494; P<0.001) and negatively correlated with HDL-C (rho=-3.833; P<0.001). LAP values could be used to diagnose hepatosteatosis (AUC=0.698; P=0.002). The LAP, adjLAP, and minLAP cutoff values were 42.7 (P=0.002), 40.05 (P=0.003), and 53.47 (P= 0.08), respectively. For LAP, the differences between the normal and mild groups (P=0.035) and the normal and moderate-severe groups were statistically significant (P=0.037), whereas the difference between the mild and moderate-severe groups was not (P>0.005). There was a statistically significant difference between the normal and mild groups for adjLAP (P=0.043) but not between the other groups (P>0.005). There was no significant intergroup difference in minLAP (P>0.005). Conclusion: LAP is a powerful and easy tool to predict NAFLD in childhood. If LAP is ≥42.7, NAFLD should be suspected. This is the first study to assess LAP diagnostic accuracy for childhood obesity.
