Formation of Dictyosome and Spherosome in Endosperm Cells of Panax ginseng during seed Formation (인삼(人蔘)(Panax ginseng)의 종자형성(種子形成)에 따른 배유세포(胚乳細胞)의 딕티오좀 및 Spherosome 형성)

  • This study has been carried out to investigate the development of dictyosome, and roles of dictyosome about the formation of spherosome in the endosperm cell during seed formation of Panax ginseng with electron microscope. The result is as follows; In the endosperm cells of early stage during seed formation of Panax ginseng, plastid, mitochondria, endoplasmic reticulum, dictyosome and ribosomes are evenly distributed in cytoplasm. Electron lucent vesicles derived from dictyosome are observed in endosperm cells. Vesicles that contain low electron density are derived from forming face of dictyosome and releases into the cytosol. This vesicles formed multi vesicular body or fused with the plasma membrane. The spherical spherosomes are formed from dictyosome containing the lipid materials of even electron density and are gradually increased in size and number. Dictyosome is located in between vacuole and spherosome and it's cisternae form a semicircle and a circle. Some membrane of the protein body that accumulate the storage protein are originate from the spherical vacuole which interfused between vesicles and vacuoles derived from dictyosome.

  • The identification of eicosapentaenoic acid (EPA) by Pseudomonas sp. CH-414 were investigated under the optimal culture conditions. The maximum dry cell weight and phospholipid production showed about 10 mg/ml and 0.6 mg/ml, respectively, at the 48 hr cultivation. The phospholipid form produced by Pseudomonas sp. CH-414 was elucidated as a phosphatidyt ethanolamine by thin layer chromatography. EPA was contained about 15% in the. extractable lipid, and the amount of EPA was about 83 $\mu $g/ml from the culture broth. Also myristic, palmitic, palmitoleic, stearic and oleic acids were identified with gas chromatography.

  • Persistent cellular oxidative stress and enhanced lipid peroxidation (LPO) of PUFAs, leading to macromolecular damage and disruption of signaling pathways, are implicated in the development of human malignancies and other chronic degenerative diseases. LPO generates by oxidation of linoleic acid (LA) or arachidonic acid ($\omega$ -6 PUPAs) reactive aldehydes, such as trans-4-hydroxy-2-nonenal, which form etheno $\varepsilon$ -DNA adducts in a variety of human tissues and thus can contribute to diet-related cancers.(omitted)

  • The cellular fatty acids and quinones of streptomycetes isolated from volcanic soils were analysed. The strains contained fatty acids of 14 to 17 carbon chains, and 12-methyltetradecanoic acid and 14 methylpentadecanoic acid were dominant in most strains. The total profiles consisted of 74% branched fatty acid family, 16.8% linear family and 8.2% unsaturated family. The largest cluster of grey spore meases defined by numerical classification was separated from the remainders in the principal component analysis, but the other clusters were overlapped with one another. In the analysis of respiratory quinones, all of the strains contained either the menaquinone of 9 isoprene units with 6 hydrogenations of 8 hydrogenations as the major species. The distribution of menaquinones among the clusters could provide an important key in the chemotaxonomy of streptomycetes.

  • Yeast-form cells of cadmium ion-tolerant Hansenula anomala B-7 were changed to mycelial cells in medium containing more than $400\mu$g/ml of cadmium. Moreover, the mycelial cells were exchanged into clumped cells in a medium containing more than $1,000\mu$g/ml of cadmium. Optimal conditions of mycelial cell formation were achieved in the presence of .$1,000\mu$g/ml of cadmium with shaking cultivation for 7 days. Glucan and mannan contents of the yeast cell wall frown with $1,000\mu$g/ml of cadmium decreased by 10% compared with those grown without cadmium. However, protein and lipid contents increased about 20% respectively. By cadmium, no significant findings in specific amino acid contents were discovered.

  • Anatomical and histochemical changes taking place in Piper nigrum berries during their ripening are described. The important observations on the pericarp are the development of sclereids in the exocarp, a continuous band of oil cells in mesocarp and the wall thickening of the endocarpic cells. The mature seed with a single layer of seed coat, representing the innermost tegment layer, encloses abundant perisperm. The endosperm and embryo are situated laterally at the terminal part of the seed. The perisperm is distinguished into an outer protein-rich zone and inner starch-filled zone. Starch and protein are also deposited in the mature pericarpic tissue. Lipid bodies are seem in the form of oil globules in oil cells.

  • Because of the ready availability of aspirin, salicylate poisoning remains a common problem in many countries. Another potential source of salicylate poisoning is medicated oil containing methyl salicylate (oil of wintergreen). Methyl salicylate poses a much greater safety threat than aspirin tablets because of its liquid, concentrated form and high lipid solubility. Because of this danger, the toxic potential of medicated oil containing methyl salicylate should be fully appreciated both by physicians and by the general public. We encountered two cases of salicylate poisoning resulting from accidental ingestion of Chinese medicated oil. We report these cases along with a review of the literature.

  • Diacetylene lipid monomers possess the capability to self-assemble into vesicles via polymerization under ultraviolet irradiation, resulting in the formation of polydiacetylene (PDA) liposomes. Exposure of the polymerized vesicles to external stimuli is known to induce a unique blue-to-red color transition. The cyclic oligosaccharide ${\alpha}$-cyclodextrin known for its use in many applications, such as drug delivery, purification, and stimulus sensing, is able to form an inclusion complex with poly(ethylene glycol) (PEG) in aqueous solution. In this study, we prepared polymeric liposomes with PEG (PEG-PDA) with the aim of improving the stability of the vesicles and colorimetric response toward ${\alpha}$-cyclodextrin. We demonstrated that PEG-PDA liposome displays unique characteristics compared with native PDA liposome and it also shows apparent chromic properties of the inclusion complex formation with ${\alpha}$-cyclodextrin.

  • In order to evaluate the anticancer activity of 1,4-naphthoquinone derivatives, several 1,4-naphthoquinone derivatives were newly synthesized and subjected to mouse leukemia p-388 cell line by MTT cytotoxicity assay. Among the several 1,4-naphtho-quinone derivatives, YC-001 has showed the most potent anticancer activity. To determine the safety of YC-001, hematotoxicity was tested. YC-001 induced hemolysis increased with both concentration and time dependent manner. The mechanism of hemolysis considered to be the generation of oxygen free radicals and lipid peroxydation of erythrocyte membrane which composed of phospholipids. Also methemoglobin, oxidized form of hemoglobin, was formed by YC-001.

  • It is reasonably well known that there is a relationship between myelin formation and methylation of myelin basic protein in nerve cells. One of the suggestions is that arginine methylation of myelin basic protein could be of aid in the conjugation of myelin protein with the nonpolar lipid to form myelin. Abnormality in methylation of myclin basic protein might induce the neurological diseases in experimental animals as well as in human being. In the biological system, the methylation reaction is catalyzed by protein methaylse I using S-adenosyl-L-methionine as methyl donor. In this study, we examined the changes of S-adenosyl-L-methionine concentration and protein methylase I activity in developing rat brain tissues. The results are sumraerized as followings: (1) In brain tissues of fetus rat, the concentration of S-adenosyl-L-methionine was gradually decreased until to birth. However, the concentration in brain tissues of infant rat was suddenly increased at 7th day(just before myelination occur) birth. (2) Protein methylase I activity was decreased until to birth in brain of fetus rat and increased temporally just after birth, However, the enzyme activity showed no changes around 7th day after birth.
