Characterization of the high mannose asparagine-linked oligosaccharides synthesized by microfilariae of Dirofilaria immitis (심장사상충 자충이 합성한 high mannose asparagine-linked oligosaccharides의 분자화학적 분석)

  • This report describes the structures of high-mannose-type N-linked oligosaccharides in glycoproteins synthesized by the microfilariae of Diroflcrio immitis. Microfilariae of D. immitis were incubated in vitro in media containing 2-(3H) mannose to allow metabolic radiolabeling of the oligosaccharide moieties of newly synthesized glycoproteins. Glycopeptides were prepared from the radiolabeled glycoproteins by digestion with pronase and fractionation by chromatography on concanavalin A Sepharose. Thirty eight percent of 2- (3H) mannose incorporated into the microalariae of D. immitis glycopeptides was recovered in high mannose-type asparagine-linked oligosaccharides which were bound to the immobilized lectin. Upon treatment of 2-(3H) mannose labeled glycopeptides with endo - β- N- acetylglu co saminidase H , the high mannose type chains were released and their structures were determined by high performance liquid chromatography and exoglycosidase digestion. The major species of high mannose-type chains synthesized by microfilariae of D. immitis have the composition Man5GlcNAc2, Man6ClcNAc2, Man7GlcNAc2, and Man8GlcNAc2. Structural analyses indicate that these oligosaccharides are similar to high mannose-type chains synthesized by vertebrates.

Identification of Molecular Markers Linked to Ti Locus in Soybean

  • Soybean is a major source of protein meal in the world. Kunitz trypsin inhibitor (KTI) protein is responsible for the inferior nutritional quality of unheated or incompletely heated soybean meal. The objective of this research was to identify RAPD markers linked to KTI protein allele using bulked segregant analysis. Cultivar Jinpumkong2 (TiTi) was crossed with C242 (titi, absence of KTI protein) and F. seeds were planted. The $F_1$. plants were grown in the greenhouse to produce $F_2$ seeds. Each $F_2$ seed from $F_1$. plants was analysed electrophoretically to determine the presence of the KTI protein band. The present and absent bulks contained twenty individuals each, which were selected on the basis of the KTI protein electrophoresis, respectively. Total 94 $F_2$ individuals were constructed and 1,000 Operon random primers were used to identify RAPD primers linked to the Ti locus. The presence of KTI protein is dominant to the lack of a KTI protein and Kunitz trypsin inhibit protein band is controlled by a single locus. Four RAPD primers (OPAC12, OPAR15, OPO12, and OPC08) were linked to the Ti locus. RAPD primer OPO12 was linked to Ti locus, controlling kunitz trypsin inhibitor protein at a distance of 16.0 cM. This results may assist in study of developing fine map including Ti locus in soybean.

An Enzyme-Linked Immunosorbent Assay for Carp (Cyprinus carpio) Vitellogenin and Assay for Oestrogenic Chemicals

  • The egg-yolk precursor vitellogenin(VTG) is secreted by the liver of female and male fish, in response to estrogenic compound. Carp(Cryprinus carpio) vitellogenin of one major protein is 160kDa, two minor proteins is 110kDa and 170kDa. We were induced vitellogenin by inject of 17-estradiol and purified in a two step procedure by separating it from plasma protein precipitated by 35% saturated ${(NH_4)}_2SO_4$ and then from the remainder by Mono-Q chromatography. We found major carp(Cyprinus carpio) vitellogenin band at 160kDa. Effect of different concentration of oestrogen on vitellogenin synthesis in carp(Cyprinus carpio) exposed for 4 weeks. Show differential effects on vitellogenin synthesis 7 days after treatment. Plasma vitellogenin was measured 3 times for each by an enzyme linked immunosorbent assay(ELISA). ELISA was developed for the detection of the egg yolk precursor vitellogenin in plasma of carp(Cyprinus carpio). The ELISAS performance was optimized and characterized.

Mapping QTL for ratooning ability in advanced backcross lines from an Oryza sativa × O. rufipogon cross

  • Ratooning ability is one of the major different traits from perennial to cultivated rice and annual type. We developed a set of 126 introgression lines derived from a cross between Hwayeong and W1944 (O. rufipogon) to gain an insight into the genetic factors underlying differences between common wild rice and cultivated rice. One IL, CR6 among the 126 ILs of $BC_3F_4$ showed a significant difference in rationing ability compared with Hwayeong. To further characterize the rationing ability, CR6 was selected and crossed to Hwayeong to produce three secondary populations, $BC_4F_2$, $BC_4F_3$ and $BC_5F_2$. In the Hwayeong background, the W1944 allele was associated with an increase in rationing ability. QTL analysis showed that the qRAT5 for rationing ability was linked to RM194 ($R^2$=6.6%, 19.6%, and 44.5% in the $BC_4F_2$, $BC_5F_2$, and $BC_5F_3$, respectively). The putative qRAT5 was also tightly linked to QTLs for spikelets per panicle and grain weight indicating that this region harbors a QTL cluster related to domestication. To our knowledge, this is the first report to map the major QTL for ratooning ability in rice. The SSR markers linked to qRAT5 would be useful in marker-assisted selection for breeding lines with enhanced ratooning ability.

Assessing Factors Linked with Ozone Exceedances in Seoul, Korea through a Decision Tree Algorithm

  • Since prolonged exposure to elevated ozone ($O_3$) concentrations is known to be harmful to human health, appropriate control strategies for ozone are needed for the non-attainment area such as Seoul, Korea. The goal of this research is to assess factors linked with the 1-hour ozone exceedance through a decision tree model. Since ozone is a secondary pollutant, lag times between ozone and explanatory variables for ozone formation are taken into account in the model to improve the accuracy of the simulation. Results show that while ozone concentrations of the previous day and $NO_2$ concentrations in the morning are major drivers for ozone exceedances in the early afternoon, meteorology plays more important role for ozone exceedances in the late afternoon. Results also show that a selection of lag times between ozone and explanatory variables affect the accuracy of predicting 1-hour ozone exceedances. The result analyzed in this study can be used for developing control strategies of ozone in Seoul, Korea.

Redistribution/Dehydrocoupling of Tertiary Alkylstannane $n-Bu_3 SnH$ Catalyzed by Group 4 and 6 transition Metal Complexes

  • The catalytic transformation of sterically bulky tertiary stannane n-Bu3SnH by the Cp2MCl2/Red-Al (M=Ti, Zr, Hf) and M(CO)6 (M=Cr, Mo, W) catalysts yielded two kinds of catenated products: one is a cross-linked polystannane as minor product, and the other is hexabutyldistannane (n-Bu3Sn)2 as major product. The distannane was produced by simple dehydrocoupling of n-Bu3SnH, whereas the cross-linked polystannane could be obtained via redistribution/dehydrocoupling combination process of n-Bu3SnH. The redistribution/dehydrocoupling combination process may initially produce a low-molecular-weight oligostannane with partial backbone Sn-H bonds which could then undergo an extensive cross-linking reaction of backbone Sn-H bonds, resulting in the formation of an insoluble polystannane.

A Study on Faculty's Perceptions on Convergence Education in N-University (N-대학의 융합교육에 대한 교수의 인식조사 연구)

  • The purpose of this study was to investigate the perceptions of faculty as a foundation for establishing a convergence education model at N-University. In September 2019, a survey was conducted using an online survey system, and a total of 80 responses were obtained. SPSS 23 was used for analysis, and a t-test, ANOVA, and multiple regression analysis were performed. First, the female faculty were significantly more positive about linked-major expansion than were the male faculty. Second, assistant faculty were significantly more positive about convergence curriculum establishment than were associate faculty. Third, as a result of the multiple regression analysis, the preference for multimajor expansion was the most significant and positively influencing factor on the convergence curriculum establishment. Universities must provide various incentives to form a consensus among university faculty. The Ministry of Education must support individual universities through the establishment of clearer guidelines and a kind of convergence education support center.

Studies on the Characteristics of Extractives in Japanese Larch (Larix leptolepis Gordon) Grown in Korea (낙엽송(落葉松) 추출성분(抽出成分) 이용(利用)에 관(關)한 기초연구(基礎硏究))

  • Red pine and Japanese larch (Larix leptolepis Gordon) grown in Korea have been the main species of coniferous resources in Korea. Especially, planting area of Japanese larch has been increased continueously in the recent years due to its superior plant type and rapid growth rate and its stocks reached approximately 4.32 million cubic meters at the present time. Although many research works have been done for the utilization of the larch wood in various ways, still many problems are existed in its chemical applications due to a large proportion of soluble extractives. In this study, chemical composition of larch extractives and chemical structure of its major component were analyzed. In order to identify the basic structure of major component, gas-liquid chromatography for separation of some completely methylated alditols as their acetates on a 3% - ECNSS-M on Gas Chrom Q. column was used. Proportion of extractives of Japanese larch wood was higher than that of other conifers and major component of the soluble extractives was arabinogalactan, a schematic structural formula which was presented in Figure 2. The molar ratio of arabinose and galactose was 1:4.5. The main chain of arabinogalactan was composed of 1,3 linked ${\beta}$-D-galactopyranose residues, each of which carried a side chain, attached to the C-6 positions. The exact nature of all of the side chains is not known, but the majority of these side chain was composed of 1, 6 linked ${\beta}$-D-galactopyranose residues, with 2~3 such units present per average chain. Some of the galactose units in the main chain had a residue of 3 - 0 - ${\beta}$-L-arabinopyranosyl-L-arabinofuranose. In addition, a few terminal residues of D-glucuronic acid also was confirmed, attached to C-6 position of the D-galactopyranose residue. It could concluded that the main structure of highly branched arabinogalactan from Japanese larch extractive was essentially the same as those of the other larch species.

Functional Characterization of the Major Surface Protein of Treponema maltophilum in Human Gingival Fibroblasts

  • Treponema maltophilum, a Group IV oral spirochete, is associated with periodontitis and endodontic infections. In this study we analyzed the functional role of the major surface protein of this organism (MspA) in human gingival fibroblasts (HGFs). The full-length gene encoding MspA was cloned and expressed in Escherichia coli by using the expression vector pQE-30. The recombinant protein (rMspA) was purified by affinity chromatography with nickel-nitrilotriacetic acid agarose and possible contamination of E. coli endotoxin in rMspA was removed by using polymyxin B-agarose. rMspA significantly induced the expression of pro inflammatory cytokines like IL-6 and IL-8 and intercellular adhesion molecule (ICAM)-1 in HGFs, when analyzed by reverse transcription-PCR, flow cytometry, and enzyme-linked immunosorbent assay. Our results indicate that MspA of T. maltophilum may play an important role in amplifying the local immune response by upregulating the expression of proinflammatory cytokines and ICAM-1.

Genetics of Broodiness in Poultry - A Review

  • In poultry, the selection against broodiness took up presumably naturally occurred mutations in the White Leghorn breed and led to an almost complete loss of the avian form of parental behaviour (incubation of eggs). Early studies on the genetics of broodiness demonstrated that the trait is polygenic with a major sex-linked effect. The reassessment of the studies on putative genes located on the Z chromosome, which are implicated in the control of broodiness, has resulted in the denial of this hypothesis. The recent experiments bear witness that incubation behaviour in chickens is not controlled by a major gene (or genes) on Z chromosome and there must, therefore, be major autosomal genes contributing to the expression of the behaviour. If a broody gene does exist on the Z chromosome it is one of at least three genes including two dominant autosomal genes, one causing and other one inhibiting incubation behaviour, with probably equal influence.
