A unique genetic lineage at the southern coast of China in the agar-producing Gracilaria vermiculophylla (Gracilariales, Florideophyceae)

  • Ocean warming can have significant negative impacts on population genetic diversity, local endemism and geographical distribution of a wide range of marine organisms. Thus, the identification of conservation units with high risk of extinction becomes an imperative task to assess, monitor, and manage marine biodiversity for policy-makers. Here, we surveyed population structure and genetic variation of the red seaweed Gracilaria vermiculophylla along the coast of China using genome-based amplified fragment length polymorphism (AFLP) scanning. Regardless of analysis methods used, AFLP consistently revealed a south to north genetic isolation. Populations at the southern coast of China showed unique genetic variation and much greater allelic richness, heterozygosity, and average genetic diversity than the northern. In particular, we identified a geographical barrier that may hinder genetic exchange between the two lineages. Consequently, the characterized genetic lineage at the southern coast of China likely resulted from the interplay of post-glacial persistence of ancestral diversity, geographical isolation and local adaptation. In particular, the southern populations are indispensable components to explore evolutionary genetics and historical biogeography of G. vermiculophylla in the northwestern Pacific, and the unique diversity also has important conservation value in terms of projected climate warming.

De novo assembly, annotation and gene expression profiles of gonads of Cytorace-3, a hybrid lineage of Drosophila nasuta nasuta and D. n. albomicans

  • Cytorace-3 is a laboratory evolved hybrid lineage of Drosophila nasuta nasuta males and Drosophila nasuta albomicans females currently passing ~850 generations. To assess interracial hybridization effects on gene expression in Cytorace-3 we profiled the transcriptomes of mature ovaries and testes by employing Illumina sequencing technology and de novo transcriptome assembling strategies. We found 26% of the ovarian, and 14% of testis genes to be differentially expressed in Cytorace-3 relative to the expressed genes in the parental gonadal transcriptomes. About 5% of genes exhibited additive gene expression pattern in the ovary and 3% in the testis, while the remaining genes were misexpressed in Cytorace-3. Nearly 772 of these misexpressed genes in the ovary and 413 in the testis were either over-or under-dominant. Genes following D. n. nasuta dominance was twice (270 genes) than D. n. albomicans dominance (133 genes) in the ovary. In contrast, only 105 genes showed D. n. nasuta dominance and 207 showed D. n. albomicans dominance in testis transcriptome. Of the six expression inheritance patterns, conserved inheritance pattern was predominant for both ovary (73%) and testis (85%) in Cytorace-3. This study is the first to provide an overview of the expression divergence and inheritance patterns of the transcriptomes in an independently evolving distinct hybrid lineage of Drosophila. This recorded expression divergence in Cytorace-3 surpasses that between parental lineages illustrating the strong impact of hybridization driving rapid gene expression changes.

Mitochondrial energy metabolic transcriptome profiles during cardiac differentiation from mouse and human pluripotent stem cells

  • Simultaneous myofibril and mitochondrial development is crucial for the cardiac differentiation of pluripotent stem cells (PSCs). Specifically, mitochondrial energy metabolism (MEM) development in cardiomyocytes is essential for the beating function. Although previous studies have reported that MEM is correlated with cardiac differentiation, the process and timing of MEM regulation for cardiac differentiation remain poorly understood. Here, we performed transcriptome analysis of cells at specific stages of cardiac differentiation from mouse embryonic stem cells (mESCs) and human induced PSCs (hiPSCs). We selected MEM genes strongly upregulated at cardiac lineage commitment and in a time-dependent manner during cardiac maturation and identified the protein-protein interaction networks. Notably, MEM proteins were found to interact closely with cardiac maturation-related proteins rather than with cardiac lineage commitment-related proteins. Furthermore, MEM proteins were found to primarily interact with cardiac muscle contractile proteins rather than with cardiac transcription factors. We identified several candidate MEM regulatory genes involved in cardiac lineage commitment (Cck, Bdnf, Fabp4, Cebpα, and Cdkn2a in mESC-derived cells, and CCK and NOS3 in hiPSC-derived cells) and cardiac maturation (Ppargc1α, Pgam2, Cox6a2, and Fabp3 in mESC-derived cells, and PGAM2 and SLC25A4 in hiPSC-derived cells). Therefore, our findings show the importance of MEM in cardiac maturation.

Activin-A 처리에 의해 분화 촉진된 인간 배아 줄기세포 유래 내배엽성 세포의 효과적인 정제 (Effective Isolation of Endodermal Lineage Cells Derived from Human Embryonic Stem Cells Post Activin-A Treatment)

  • Embryoid bodies (EBs) generated from human embryonic stem cells (hESCs) include spontaneously induced endodermal lineage cells (ELCs). Activin-A plays important roles in the endoderm differentiation of hESCs. Despite studies on the generation of ELCs from hESCs with treatment of Actvin-A, it was unclear for localization and pattern of ELCs by Activin-A during differentiation of hESCs. Accordingly in this study, we knew that Actvin-A increased the cystic EBs formation, including the highly enriched AFP (endoderm lineage specific marker)-expressing cells in the surface of cystic EBs. To induce the EBs formation from undifferentiated hESCs, cells were transferred onto petri-dish and cultured in suspension condition without bFGF removed hESC media (EB media) for 3 days. Next to investigate the effect of Activin-A, EBs were subsequently cultured in EB media supplement with 100 ng/ml Activin-A for 3 days. After 5~7 days of Activin-A treatment, cystic EBs began to appear which increased in numbers reaching ~60% of initially formed EBs over 5 days. Endoderm lineage marker, AFP were highly expressed and specifically localized at the surface region of cystic EBs comparison with normal EBs. We next attached the cystic EBs onto gelatin-coated plates and cultured for 5 days. In the results of real-time PCR and immunocytochemistry analysis, AFP-expressing cells migrated and localized at the outgrowth region of attached cystic EBs. To obtain the AFP-expressing cells of the outgrowth region, we manually isolated by using micro-dissection and cultured them. These cells strongly express AFP over 70% of isolated cells post re-plating. Here, we first showed an expression pattern of specifically localized ELCs by Activin-A during differentiation of hESCs. From this observation, we could highly purified ELCs from undifferentiated hESCs. Taken together, our system will provide a novel and efficient option to generate ELCs from hESCs.

상품으로서의 온라인 게임 세계와 역할 놀이자로서의 온라인 게임 소비자의 행동특성 (The Online Game World as a Product and the Behavioral Characteristics of Online Game Consumers as Role Player)

  • 본 연구에서는 현재 한국 사회에서 가장 대표적인 디지털 컨텐츠 제품으로 등장한 온라인 게임 중 물리적으로 유사한 특성을 지니고 있지만 전혀 다른 형태의 세계로 발전해간 두 개의 제품, 리니지와 에버퀘스트를 대상으로 소비자의 게임 인식과 소비 행위에 따라 게임의 성격이 어떻게 다르게 규정되고, 그리고 소비자의 행동 특성이 특정 제품의 성격을 어떻게 변화시키는지를 탐색하였다. 첫째로 각각의 게임에 대한 소비자의 인식의 차이를 알아보기 위해, 게임의 매력과 만족을 주는 주된 활동, 친구관계, 캐릭터의 선택과 육성의 측면에서 비교해 본 결과, 리니지 게이머들은 리니지 세계를 현실과 유사한 가치와 의미를 지닌 새로운 생활공간으로 인식하고 있는 반면, 에버퀘스트 게이머들은 에버퀘스트 세계를 게임 자치로서의 기능에 초점을 둔 환타지 세계로 인식하고 있었다. 또한 게임 인식에 따른 게임 소비 행동을 게임 속 공동체 활동과 연애와 같은 사적인 관계의 측면에서 비교해본 결과, 구체적인 게임 소비 행동에서 차이가 나는 것으로 나타났다. 이러한 결과는 온라인 게임과 같은 사이버 공간의 가상의 디지털 이미지 또는 컨텐츠 제품들의 경우, 소비자가 지닌 경험이나 가치에 따라 제품에 대한 인식이 달라질 수 있으며, 이런 인식의 차이는 각각의 소비자들이 동일한 제품도 다르게 소비할 수 있음을 시사한다. 즉 디지털 이미지 또는 컨텐츠 제품들의 가치와 의미는 소비자에 의해 결정된다는 것을 시사한다고 할 수 있다.

Comparative Analysis for In Vitro Differentiation Potential of Induced Pluripotent Stem Cells, Embryonic Stem Cells, and Multipotent Spermatogonial Stem Cells into Germ-lineage Cells

  • In the present study, embryoid bodies (EBs) obtained from induced pluripotent stem cells (iPSCs) were induced to differentiate into germ lineage cells by treatment with bone morphogenetic protein 4 (BMP4) and retinoic acid (RA). The results were compared to the results for embryonic stem cells (ESCs) and multipotent spermatogonial stem cells (mSSCs) and quantified using immunocytochemical analysis of germ cell-specific markers (integrin-${\alpha}6$, GFR-${\alpha}1$, CD90/Thy1), fluorescence activating cell sorting (FACS), and real time-RT-PCR. We show that the highest levels of germ cell marker-expressing cells were obtained from groups treated with 10 ng/$m{\ell}$ BMP4 or 0.01 ${\mu}M$ RA. In the BMP4-treated group, GFR-${\alpha}1$ and CD90/Thy-1 were highly expressed in the EBs of iPSCs and ESCs compared to EBs of mSSCs. The expression of Nanog was much lower in iPSCs compared to ESCs and mSSCs. In the RA treated group, the level of GFR-${\alpha}1$ and CD90/Thy-1 expression in the EBs of mSSCs Induced pluripotent stem cells, Mouse embryonic stem cells, Multipotent spermatogonial stem cells, Germ cell lineage, Differentiation potential. was much higher than the levels found in the EBs of iPSCs and similar to the levels found in the EBs of ESCs. FACS analysis using integrin-${\alpha}6$, GFR-${\alpha}1$, CD90/Thy1 and immunocytochemistry using GFR-${\alpha}1$ antibody showed similar gene expression results. Therefore our results show that iPSC has the potential to differentiate into germ cells and suggest that a protocol optimizing germ cell induction from iPSC should be developed because of their potential usefulness in clinical applications requiring patient-specific cells.

한국재래염소의 mtDNA 다양성 및 계통유전학적 분석 (mtDNA Diversity and Phylogenetic Analysis of Korean Native Goats)

  • 한국재래염소는 흑모색의 특징을 나타내며, 유일한 염소 품종으로서 오랫동안 한반도에서 사육되어 왔다. 하지만 이들에 대한 유전적 다양성, 계통유전학적 분석 등을 통한 기원 추정 등에 대한 연구는 미비한 실정이다. 본 연구에서 한국재래염소 5개 집단, 60두를 대상으로 mtDNA D-loop 영역 중 HVI 영역의 서열을 이용하여 유전적 다양성 및 계통유전학적 분석을 실시하였다. 한국재래염소는 다른 나라 염소들에 비해서 haplotype 다양성 지수가 낮게 나타났다. 또한 본 연구에서 분류된 한국재래염소 10개 haplotype 중 현재까지 보고되지 않은 6개의 새로운 haplotype이 확인되었다. 계통유전학적 분석 결과, 분석에 사용된 모든 한국재래염소는 mtDNA 모계혈통 A에 속하였다. 10개의 haplotype 중 8개는 베트남, 일부 중국 염소와 함께 subgroup을 형성하였다. 그러나 나머지 2개 haplotype은 각각 서로 독립적인 계통유전학적 위치를 보였다. 이런 결과들을 토대로 한국재래염소는 상대적으로 높은 근친상황으로 외부 유전자 유입이 적었을 것이라고 추정된다. 한국재래염소의 새로운 mtDNA haplotype의 발견 및 유전자원 보존 및 평가를 위해서 더 많은 분석집단 및 개체를 수집하고, MS 마커를 이용한 추가분석이 필요하다고 사료된다.

본부와 방면 - 대순진리회 종교조직의 특성 - (Bonbu and Bangmyeon: The Lineage Principle in Daesoon Jinrihoe)

  • 대순진리회는 1969년 박우당에 의해 공식적으로 창설되었다. 이 글은 대순진리회가 증산을 신앙하는 다른 종단과 역사와 사상을 공유하고 있지만 조직 측면에서 타종단과 구별됨을 보여주고자 한다. 대순진리회 조직의 양대 기구인 본부와 방면은 대순진리회가 신종교로서 급속한 성장을 이루는 데 서로 기여했다. 본부는 중앙 집권적인 위계질서가 있는 반면, 방면은 친족 같은 강한 충성도와 공동체의식을 보여준다. 두 조직 기구에 공통적인 특성은 계보(lineage) 개념으로 이는 대순진리회 양대 조직의 분절화 원리이다. 미세하게 균형 잡힌 두 조직기구는 대순진리회의 종교적 사명 실현에 크게 기여했다. 균형의 첫 번째 축은 본부이다. 본부는 여주와 여주 외 지역 도장 및 수련센터의 핵심 종단 기구를 포함한다. 이러한 기구들은 모두 박우당 도전에 의해 설립되었다. 박우당 도전은 종단 운영의 제반사항을 총괄하는 종단의 헌법 곧 도헌을 제정하였다. 중앙종의회로부터 포덕, 감사, 기획 등에 이르는 본부의 제반 운영은 박우당 도전의 운영지침에 충실히 부합한다. 균형의 두 번째 축은 방면이다. 비교적 대규모 방면들은 그 자체가 복합적인 조직이다. 이 연구의 결론은 본부와 방면이라는 양대 축에 대한 조직유형적 특성구분이다. 로버트 퀸과 킴 카메론의 분류에 따른다면 대순진리회의 본부는 효율성을 중시하는 전형적인 중앙집권화 조직이다. 이에 반해 방면은 집단정체성과 공동체의식을 중시하는 친족에 가까운 조직이다.

Genetic Diversity and Origin of Chinese Domestic Goats Revealed by Complete mtDNA D-loop Sequence Variation

  • China has numerous native domestic goat breeds, but so far there has been no extensive study on genetic diversity, population demographic history, and origin of Chinese goats. To determine the origin and genetic diversity of Chinese goats, we analyzed the complete mtDNA D-loop sequences of 183 goats from 13 breeds. The haplotype diversity value found in each breed ranged from 0.9333 to 1.0000. The nucleotide diversity value ranged from 0.006337 to 0.025194. Our results showed that there were four mtDNA lineages (A, B, C and D), in which lineage A was predominant, lineage B was moderate, and lineages C and D were at low frequencies. Lineages C and D were observed only in the Tibetan breed. The results revealed multiple maternal origins of Chinese domestic goats. There was weaker geographical structuring in the 13 Chinese goat populations, which suggested that there existed high gene flow among goat populations caused by the extensive transportation of goats in the course of history.
