컴퓨터 정액자동분석에 의한 동결융해 한우 정액의 운동특성 연구 (Motional kinematics of Frozen-thawed Korean native cattle semen use of computer aided semen analysis(CASA) system)

  • The aim of this experiments were to assess the time-interval change of motional characteristics in frozen-thawed semen of Korean native cattle (KNC) by using computer aided semen analysis (CASA) technology. Twenty-six KNC frozen semen straws were obtained from Korean KNC improvement department, livestock improvement main division, national livestock cooperatives federation in Korea. Specimens were allowed to thaw at $37^{\circ}C$ for 30 sec in water bath. Semen analysis was performed on semen image analysis system (SIAS, Medical supply, Korea) adjusted to the gate settings and used the semen droplet ($5{\mu}l$) placed on Makler counting chamber (Sefi medical instrument, Israel) prewarmed at $37^{\circ}C$. The same person used the same micropipette to fill the Makler counting chamber. A total of 150 or more of sperms were analysed in each specimen by a single trained person by scanning at least 5 to 10 fields. The measurement parameters in SIAS were as follows ; frame rate = 30 frames per sec, image capture = 1 sec, minimum motile speed = $10{\mu}m/s$, maximum countable sperm number = 400. Statistical analysis was done by Student t-test with use of the Sigma plot program on a IBM personal computer. The dancemean(DNM) and hyperactivated sperm(HYP) of frozen-thawed KNC semen kinematics were significantly decreased(p < 0.05) after 10 min of incubation at $37^{\circ}C$ water bath. But, wobble(WOB) of same sample semen was significantly increased(p < 0.05) after 10 min of incubation and significantly decrease(p < 0.05) after 60 min of same incubation. And, after 30 mim of incubation, significantly differences were found most of motion kinematics, motifity(MOT), curvilinear velocity(VCL), straight line velocity(VSL), average path velocity(VAP), amplitude of lateral head displacement(ALH), beat cross frequency(BCF), mean angular displacement(MAD), dance(DNC), on same sample semen. The DNM of KNC semen sample was variable kinematics after 30 min of incubation. Also, the linearity(LIN) and straightness(STR) was significantly decreased(p < 0.05) from 60 min of incubation. In conclusion, the AI within 30 min after thawing of frozen semen can be an effective method for obtaining high fertility rate in KNC reproductive program.

Brain CT에서 발생하는 선속경화현상 감소방안에 관한 연구 (A study of beam hardening effect reduction occur in brain CT)

  • 본 연구는 Brain CT검사 시 영상에서 발생하는 선속경화현상의 원인과 감소방법을 알아보기 위하여 선속경화현상에 영향을 미치는 관전압, 관전류, 단면두께, 갠트리 각도, 기준선에 변화를 주었다. 사용한 장비로는 Somatom Sensation 16장비로 Bone opaque head phantom을 이용 영상영향인자에 변화를 주어 스캔하였고 획득한 영상 데이터를 이용하여, CT값 분석을 이용한 정량적 분석과 CT영상평가표를 이용한 정성적 평가를 시행하였다. 정량적 분석결과 관전압은 140kVp일 때 $31.56{\pm}2.89HU$로 측정되었고, 관전류의 경우 150mA에서 $-3.87{\pm}0.12HU$, 절편두께는 3mm에서 $2.29{\pm}0.78HU$로 측정되었으며 갠트리 각도에서 IOML이 $13.31{\pm}1.03HU$로 선속경화현상이 가장 적었다. 정성적 분석결과 대부분의 평가자들이 140kVp, 150mA, 3mm, IOML 또는 OML에서 스캔한 영상을 선속경화현상이 적게 발생한 영상으로 평가하였으며 모든 조건에서 변화 인자와 비교 시 통계학적으로 유의한 차이가 있었다.(P<0.05) 따라서, 임상적용 시 허용선량한도 범위 내에서 관전압은 높여주고 관전류는 영상 화질 저하에 영향을 미치지 않는 범위 내에서 낮게 설정하며, 절편두께는 해상도 저하를 고려하여 얇은 절편두께를 사용하고, 갠트리 각도는 IOML 또는 OML을 이용한다면 선속경화현상에 의해 발생되어지는 인공물을 최소화시켜 영상 판독자와 환자에게 보다 정확한 양질의 영상을 제공할 수 있을 것으로 사료된다.

법랑기질유도체를 도포한 타이태늄 표면에서 조골세포의 증식 및 분화 (Effects of enamel matrix derivative and titanium on the proliferation and differentiation of osteoblasts)

  • Among objectives of periodontal therapy. the principal one is the morphological and functional reconstruction of lost periodontal supporting tissues. This includes de novo formation of connective tissue attachment and the regrowth of alveolar bone. The use of enamel matrix derivative(EMD) may be a suitable means of regeneration new periodontal attachment in the infrabony defects. Implant used to replace lost tooth but, implantitis occurred after installation. The purpose of this study was to investigate the effects of EMD on differentiation and growth of osteoblast in titanium disc. Twentyfive millimeter diameter and 1mm thick Ti disc which was coated 25, 50, 100, 200${\mu}g$/ml of EMD(Emdogain(R)) used as experimental group, 25, 50, 100, 200ng/d of rhBMP-2 as positive control group, and no coat as negative control group. A human osteosarcoma cell line Saos-2 was cultured in Ti disc and cell proliferation and Alkaline phosphatase (ALP) activity were measured at 1 and 6 days. PCR was performed at 2 and 8 hours. Semi-quantitative RT-PCR for mRNA expressions of various osteoblastic differentiation markers -type I collagen, ALP, osteopontin, and bone sialoprotein - were performed at appropriate concentrations based upon the results of MTT and ALP assay. Cultured cell-disc complexes were prepared for scanning electron microscopy (SEM) at 2 hour. Data were analyzed using Mann-Whitney and repeated- measures 1-way analysis of variance(SPSS software version 10,SPSS. Chicago. IL). After culture, there was more osteoblast in EMD100${\mu}g$/ml than in EMD50, 200${\mu}g$/ml on day 6. There was significant difference in experimental and positive control group compared control group, as times go by(1 and 6 days). Alkaline phosphatase activity was different significantly in EMD100, 200${\mu}g$/ml and BMP100, 200${\mu}g$/ml on day 6. The results of reverse transcriptase-polymerase chain reaction (RT-PCR) showed that expression of mRNA for ALPase, collagen type I, osteopontin. hone sialoprotein and BMP-2 was detected at 2 hour and 8 hour in EMI 200${\mu}g$/ml subgroup and BMP100ng/ml subgroup. The results of this study suggest that application of enamel matrix derivative on osteoblast attached to titanium surface facilitate the expression of bone specific protein and the differentiation and growth of osteoblast.

광섬유를 이용한 광영상단층촬영기 제작에 관한 연구 (A Study on Optical Coherence Tomography System by Using the Optical Fiber)

  • 본 연구에서는 인체에 무해하며, 고분해능 측정과 저가격화 및 소형으로 제작이 가능한 광섬유를 이용한 광영상단층촬영기 제작에 관한 것이다. 시스템의 기본원리는 마이켈슨 간섭계를 이용한 것으로서 광섬유를 이용하여 간섭계를 구성하였다. 시스템의 구성으로는 광원은 분해능 및 측정범위를 고려하여 1,300(nm) 중심파장을 가지며 대역폭이 35(nm)인 상용제품의 SLD(Superluminersent diode)를 사용하였으며, 샘플내부의 영상을 검출하기 위한 간섭신호 검출방법은 기준거울이 선형적으로 이동하여 광경로를 일치하는 광지연선로를 구성하였다. 그리고 간섭계는 단일모드 광섬유를 이용하여 마이켈슨 간섭계를 구성하였으며, 스캐너는 시준기를 이용하여 샘플에 대한 초점을 고정하였으며, 스텝모터를 이용하여 샘플에 대한 횡단방향의 이동을 통해 샘플의 2차원 단층영상을 측정하도록 하였다. 수광부는 소신호인 간섭신호를 검출하기 위하여 감도가 뛰어나면 잡음특성이 우수한 800-1,700(nm) 측정범위의 광검출기를 사용하였다. 신호처리부에서 간섭신호의 포락선 신호만을 검출하기 위하여 증폭 및 필터링 하여 A/D 변환을 거친 후 영상검출 프로그램을 통해 실시간으로 단층영상을 나타내도록 하였다. 측정결과 분해능은 약 30($\mu\textrm{m}$)로서 이론식과 일치함을 확인하였으며, 샘플을 이용한 단층촬영에서 실시간으로 양파조직의 조직형태를 측정하였다.

RSSI 판독 라이브러리 함수 및 옥내 측위 모듈 구현 (Implementation of a Library Function of Scanning RSSI and Indoor Positioning Modules)

  • IEEE 802.11 기술 덕분에 학교와 대형 쇼핑몰을 비롯한 사무실, 병원, 역 등지에서도 무선 LAN을 통한 인터넷 접속이 가능하다. 본 논문은 무선 LAN에 현재 가장 많이 사용되는 2.4GHz 대역의 802.11b와 802.11g 프로토콜이 탑재된 액세스포인트(AP: Access Point)로부터 수신한 신호의 세기(RSSI: Received Signal Strength Indicator)를 판독할 수 있는 C# 라이브러리 함수를 제안한다. 위치기반서비스는 사용자의 현재 위치를 실시간으로 측정하여 현재 위치를 기반으로 길을 안내하거나, 현재 위치와 관련한 콘텐츠를 제공하는 등의 유용한 서비스를 제공한다. 옥내에서 위치기반서비스를 제공하려면 옥내에 있는 사용자의 위치를 판정하는 옥내측위가 반드시 선결되어야 한다. 옥내측위 기술로 적외선, 초음파, UDP 패킷의 신호세기 등을 이용하는 방법들이 소개된 바 있다. 이러한 방법들은 측위를 위한 특수 장비를 설비해야만 한다는 단점이 있다. 본 논문은 RSSI를 판독하는 라이브러리 함수를 제공할 뿐만 아니라 제공하는 함수를 이용한 옥내 측위 구현 예도 소개한다. 구현에 적용된 방법들은 이미 널리 알려진 K-NN(K Nearest Neighbors), 베이시안 방법 그리고 삼각측량법이다. K-NN 방법과 베이시안 방법은 일종의 지문방식인데, 지문방식은 준비단계와 실시간단계로 구성되며, 실시간 단계의 처리 과정은 처리속도가 빨라야만 한다. 본 논문은 실시간 단계의 속도를 개선하는 방법으로 판단나무 방법(Decision Tree Method)을 제안하고, 이러한 방법들의 성능을 실험적으로 평가한 결과를 소개한다.

Effects of Tiam 1 on Invasive Capacity of Gastric Cancer Cells in vitro and Underlying Mechanisms

  • Objective: To investigate changes in the invasive capacity of gastric cancer cells in vitro after expression inhibition of T lymphoma invasion and metastasis inducing factor 1 (Tiam 1) and underlying mechanisms. Methods: Using adhesion selection, two subpopulations with high ($M_H$) or low ($M_L$) invasive capacity were separated from the human gastric cancer cell line MKN-45 ($M_0$). Tiam 1 antisense oligodeoxynucleotide (ASODN) was transfected into $M_H$ cells with liposomes, and expression of Tiam 1 mRNA and protein was determined by RT-PCR and quantitative cellular-ELISA. Changes in the cytoskeleton, invasive capacity in vitro and expression of ras-related $C_3$ botulinum toxin substrate 1 (Rac 1), integrin ${\beta}1$ and matrix metalloproteinase 2 (MMP 2) between Tiam 1 ASODN transfected $M_H$ cells and non-transfected cells were observed by HE staining, cytoskeletal protein staining, scanning electron microscopy, Boyden chamber tests and cyto-immunohistochemistry. Results: A positive correlation existed between the expression level of Tiam l mRNA or protein and the invasion capacity of gastric cancer cells. After ASODN treatment ($0.43{\mu}M$ for 48 h), Tiam 1 mRNA transcription and protein expression in $M_H$ cells were decreased by 80% and 24% respectively (P < 0.05), compared with untreated controls, while invasive capacity in vitro was suppressed by 60% (P < 0.05). Morphologic and ultrastructural observation also showed that ASODN-treated $M_H$ cells exhibited smooth surfaces with obviously reduced filopodia and microspikes, which resembled $M_0$ and $M_L$ cells. Additionally, cytoskeletal distribution dramatically altered from disorder to regularity with reduced long filament-like structure, projections, pseudopodia on cell surface, and with decreased acitn-bodies in cytoplasm. After Tiam 1 ASODN treatment, the expression of Rac 1 and Integrin ${\beta}1$ in $M_H$ cells was not affected (P > 0.05), but that of MMP 2 in $M_H$ cells was significantly inhibited compared with untreated cells (P < 0.05). Conclusion: Over-expression of Tiam-1 contributes to the invasive phenotype of gastric cancer cells. Inhibition of Tiam 1 expression could impair the invasive capacity of gastric cancer cells through modulating reconstruction of the cytoskeleton and regulating expression of MMP 2.

표면처리방법에 따른 Electroforming Gold와 레진과의 전단결합강도 (The Shear Bond Strength of Resin to Electroforming Gold according to the Surface Treatment)

  • Statement of problem. The success of the bonding between electroforming gold and resin is dependent on the surface-conditioning technique but its effective technique has net been studied widely. Purpose. The purpose of the study was to evaluate the bond strength between the electroforming gold and resin with varying the surface-conditioning technique. Materials and methods. Sixty rectangular shaped metal specimens were made and one side of each specimen were gold hard plated. The sand-blasted specimens were divided into four experimental groups with fifteen specimens in each group and were treated as follows. Group 1: Silicoating (Rocatec, 3M ESPE)+ Sinfony (3M ESPE), Group 2: SR Link+ SR Adoro (Ivoclar Vivadent), Group 3: Tin plating (Microtin, Danville Engineering)+ SR Link+ SR Adoro, Group 4: Tin plating (Micro tin, Danville Engineering)+ Silicoating (Rocatec)+ Sinfony. Shear bond strength at metal-resin interface were measured using universal testing machine. Energy Dispersive x-ray analysis was done and scanning electron microscope images were taken and observed. Results and Conclusion. The following conclusions were drawn. 1. The mean shear bond strength values in order were 11.69MPa (Group 2), 22.35MPa (Group 3), 22.40MPa (Group 1) and 27.71MPa (Group 4). There was no significant difference in Group 1, Group 3 and Group 4(P>0.05). 2. In the EDX line analysis, the Au was detected on the surface of all specimen. $SnO_2$ showed on the surface of Group 2 and $SiO_2$ was detected on the surface of Group 1. 3. Increasing of roughness by sandblasting(Group 2), formation of micro-irregularities and tin crystals by electrolytic tin plating(Group 3) and formation of surface irregularities and $SiO_2$ layer(Group 1,4) were observed in SEM photo. 4. Tin plating(Group 3) and Rocatec treatment(Group 1) showed clinically effective shear bond strength(>20MPa), but when the two surface conditioning method were used together higher bond strength were achieved.

  • High-k materials have been paid much more attention for their characteristics with high permittivity to reduce the leakage current through the scaled gate oxide. Among the high-k materials, $ZrO_2$ is one of the most attractive ones combing such favorable properties as a high dielectric constant (k= 20 ~ 25), wide band gap (5 ~ 7 eV) as well as a close thermal expansion coefficient with Si that results in good thermal stability of the $ZrO_2$/Si structure. During the etching process, plasma etching has been widely used to define fine-line patterns, selectively remove materials over topography, planarize surfaces, and trip photoresist. About the high-k materials etching, the relation between the etch characteristics of high-k dielectric materials and plasma properties is required to be studied more to match standard processing procedure with low damaged removal process. Among several etching techniques, we chose the inductively coupled plasma (ICP) for high-density plasma, easy control of ion energy and flux, low ownership and simple structure. And the $BCl_3$ was included in the gas due to the effective extraction of oxygen in the form of $BCl_xO_y$ compounds. During the etching process, the wafer surface temperature is an important parameter, until now, there is less study on temperature parameter. In this study, the etch mechanism of $ZrO_2$ thin film was investigated in function of $Cl_2$ addition to $BCl_3$/Ar gas mixture ratio, RF power and DC-bias power based on substrate temperature increased from $10^{\circ}C$ to $80^{\circ}C$. The variations of relative volume densities for the particles were measured with optical emission spectroscopy (OES). The surface imagination was measured by scanning emission spectroscope (SEM). The chemical state of film was investigated using energy dispersive X-ray (EDX).

Evaluation of the Accuracy of Distance Measurements on 3D Volume-rendered Image of Human Skull Using Multi-detector CT: Effects of Acquisition Section Thickness and Reconstruction Section Thickness

  • The image quality of three-dimensional (3D) images has been widely investigated by the qualitative analysis method. A need remains for an objective and quantitative method to assess the image quality of 3D volume-rendered images. The purpose of this study was to evaluate the quantitative accuracy of distance measurements on 3D volume-rendered images of a dry human skull by using multi-detector computed tomography (MDCT). A radiologist measured five times the twenty-one direct measurement line items composed among twelve reference points on the skull surface with a digital vernier caliper. The water filled skull specimen was scanned with a MDCT according to the section thicknesses of 1.25, 2.50, 3.75, and 5.00 mm for helical (high quality; pitch 3:1) scan mode. MDCT data were reconstructed with its acquisition section thickness and with 1.25 mm section thickness for all scans. An observer also measured seven times the corresponding items on 3D volume-rendered images with measuring tools provided by volumetric analysis software. The quantitative accuracy of distance measurements on the 3D volume-rendered images was statistically evaluated (p-value < 0.05) by comparatively analyzing these measurements with the direct distance measurements. The accuracy of distance measurements on the 3D volume-rendered MDCT images acquired with 1.25, 2.50, 3,75 and 5.00 mm section thickness and reconstructed with its section thickness were 48%, 33%, 23%, and 14%, respectively. Meanwhile, there were insignificant statistical differences in accuracy of distance measurements among 3D volume-rendered images reconstructed with 1.25 mm section thickness for the each acquisition section thickness. MDCT images acquired with thick section thickness and reconstructed with thin section thickness in helical scan mode should be effectively used in medical planning of 3D volume-rendered images. The quantitative analysis of distance measurement may be a useful tool for evaluating the quantitative accuracy and the defining optimal parameters of 3D volume-rendered CT images.

아몰퍼스실리콘의 결정화에 따른 복합티타늄실리사이드의 물성변화 (Property of Composite Titanium Silicides on Amorphous and Crystalline Silicon Substrates)

  • 반도체 메모리 소자의 스피드 향상을 위해 저저항 배선층을 채용하는 방안으로 70 nm-두께의 아몰퍼스실리콘과 폴리실리콘 기판부에 $TiSi_2$ 타켓으로 각각 80 nm 두께의 TiSix 복합실리콘을 스퍼터링으로 증착한 후 RTA $800^{\circ}C$-20sec 조건으로 실리사이드화 처리하고 사진식각법으로 선폭 $0.5{\mu}m$의 배선층을 만들었다. 배선층에 대해 다시 각각 $750^{\circ}C-3hr,\;850^{\circ}C-3hr$의 부가적인 안정화 열처리를 실시하였으며, 이때의 면저항의 변화는 four-point probe로 실리사이드층의 미세구조와 수직단면 두께 변화를 주사전자현미경과 투과전자현미경으로 관찰하였다. 아몰퍼스실리콘 기판인 경우 후속열처리에 따른 결정화 진행과 함께 급격한 면저항의 증가가 확인되었고, 이 원인은 결정화 과정에서 실리콘과 복합티타늄실리사이드 층과의 상호확산으로 표면 공공(void)을 형성한 것으로 미세구조 관찰에서 확인되었다. 따라서 복합티타늄실리사이드의 하지층의 종류와 열처리 조건을 바꾸어 저저항 또는 고저항 실리사이드를 조절하여 제작하는 것이 가능하여 복합 $TiSi_2$를 저저항 배선층 재료로 채용할 수 있음을 확인하였다.
