Development of Competitive Direct Enzyme-linked Immunosorbent Assay for the Detection of Gentamicin Residues in the Plasma of Live Animals

  • Competitive direct ELISA was developed to detect gentamicin residues. Mice immunized with gentamicin-keyhole limpet hemocyanin (KLH) conjugate developed good antiserum titers, which gradually increased with booster injections, indicating immunization was successfully processed. Monoclonal antibody against gentamicin was prepared using hybridoma cells cloned by limit dilution of fused cells. IgG was purified from ascites fluid of hybridoma cell-injected mice through ammonium sulfate precipitation and Sephadex G-25 gel filtration. After the gel filtration, fractions of high antibody titer were further purified through affinity chromatography on protein A/G column. Monoclonal antibody against gentamicin was confirmed as IgG1, which has kappa light chain. Cross-reactivities ($CR_{50}$) of gentamicin monoclonal antibody to other aminoglycosides (kanamycin, neomycin, and streptomycin) were less than 0.005%, indicating the monoclonal antibody was highly specific for gentamicin. Standard curve constructed through competitive direct ELISA showed measurement range (from 80 to 20% of B/$B_0$ ratio) of gentamicin was between 1 and 40 ng/ml, and 50% of B/$B_0$ ratio was about 4 ng/ml. The gentamicin concentration rapidly increased to 1,300 ng/ml after the intramuscular administration up to 2 h, then sharply decreased to less than 300 ng/ml after 4 h of withdrawal, during which the elimination half-life ($t_{1/2}$) of gentamicin in the rabbit plasma was estimated to be 1.8 h. Competitive direct ELISA method developed in this study using the prepared monoclonal antibody is highly sensitive for gentamicin, and could be useful for detecting gentamicin residues in plasma of live animals.

Obtaining Mechanical Properties of Fe Powder Using a Combined Nanoindentation and the Finite Element Method (나노인덴테이션과 유한요소법을 결합한 철 분말의 기계적 물성 취득)

  • Stress-strain curves are fundamental properties to study characteristics of materials. Flow stress curves of the powder materials are obtained by indirect testing methods, such as tensile test with the bulk materials and powder compaction test, because it is hard to measure the stress-strain curves of the powder materials using conventional uniaxial tensile test due to the limitation of the size and shape of the specimen. Instrumented nanoindentation can measure mechanical properties of very small region from several nanometers to several micrometers, so nanoindentation technique is suitable to obtain the stress-strain curve of the powder materials. In this study, a novel technique to obtain the stress-strain curves using the combination of instrumented nanoindentation and finite element method was introduced and the flow stress curves of Fe powder were measured. Then obtained stress-strain curves were verified by the comparison of the experimental results and the FEA results for powder compaction test.

Classification of Tumor cells in Phase-contrast Microscopy Image using Fourier Descriptor (위상차 현미경 영상 내 푸리에 묘사자를 이용한 암세포 형태별 분류)

  • Tumor cell morphology is closely related to its migratory behaviors. An active tumor cell has a highly irregular shape, whereas a spherical cell is inactive. Thus, quantitative analysis of cell features is crucial to determine tumor malignancy or to test the efficacy of anticancer treatment. We use 3D time-lapse phase-contrast microscopy to analyze single cell morphology because it enables to observe long-term activity of living cells without photobleaching and phototoxicity, which is common in other fluorescence-labeled microscopy. Despite this advantage, there are image-level drawbacks to phase-contrast microscopy, such as local light effect and contrast interference ring. Therefore, we first corrected for non-uniform illumination artifacts and then we use intensity distribution information to detect cell boundary. In phase contrast microscopy image, cell is normally appeared as dark region surrounded by bright halo ring. Due to halo artifact is minimal around the cell body and has non-symmetric diffusion pattern, we calculate cross sectional plane which intersects center of each cell and orthogonal to first principal axis. Then, we extract dark cell region by analyzing intensity profile curve considering local bright peak as halo area. Finally, we calculated the Fourier descriptor that morphological characteristics of cell to classify tumor cells into active and inactive groups. We validated classification accuracy by comparing our findings with manually obtained results.

Dyeability and functionality of Catechu(Part I) -Characteristics of Catechu and Dyeing properties of Cotton- (아선약의 염색성과 기능성(제1보) -아선약 색소의 특성과 면섬유에 대한 염색성-)

  • Catechu is widely used as in natural dyeing as well as in various medicines and tannage. This study investigated the dyeing properties and functionality of Catechu through a series of dyeing experiments using cotton fabric. It was shown that the absorbance of Catechu solution has two peaks at 220nm and 280nm. Thus, it can be concluded that the color constituent of Catechu is a catechol tannin and color tone is YR. The dyeability of Catechu increases as the concentration of the dye is increased. Its variation was shown to be similar to the isothermal absorption curve of the Freundlich. As dyeing time was increases, the dyeability did not change significantly after the 30 minute mark was passed. At high temperatures the dyeability was shown to increase. As the dyeing temperature was increased the color of dyeing fabrics changes from Y and YR to R. Experiments with pre-mordanting method showed that various colors can be obtained using Catechu. Cotton is effective in using K, Cu, Fe mordants, and the dyeability was shown to be improved with low pH. Both washing fastness and light fastness were shown to be low. However, the fabric color gradually changed to red was due to mailard reaction of catechol tannin causing by repeated washing and sunlight. The ultraviolet-cut ability was improved for cotton fabric dyed with Catechu. Also, dyed fabric with Catechu showed very good antimicrobial abilities at 99.9%.

  • Crystlline silicon solar cells markets are increasing at rapid pace. now, crystlline silicon solar cells markets screen-printing solar cell is occupying. screen-printing solar cells manufacturing process are very quick, there is a strong point which is a low cost. but silicon and metal contact, uses Ag & Al pates. because of, high contact resistance, high series resistance and sintering inside process the electric conductivity decreases with 1/3. and In pastes ingredients uses Ag where $80{\sim}90%$ is metal of high cost. because of low cost solar cells descriptions is difficult. therefore BCSC(Buried Contact Solar Cell) is developed. and uses light-induced plating, ln-line galvanization developed equipments. Ni/Cu matel contact solar cells researches. in Germany Fraunhofer ISE. In order to manufacture high-efficiency solar cells, metal selections are important. metal materials get in metal resistance does small, to be electric conductivity does highly. efficiency must raise an increase with rise of the curve factor where the contact resistance of the silicon substrate and is caused by few with decrement of series resistance. Ni metal materials the price is cheap, Ti comes similar resistance. Cu and Ag has the electric conductivity which is similar. and Cu price is cheap. In this paper, Ni/Cu/Ag metal contact cell with screen printing manufactured, silicon metal contact comparison and analysis.

Face Detection for Automatic Avatar Creation by using Deformable Template and GA (Deformable Template과 GA를 이용한 얼굴 인식 및 아바타 자동 생성)

  • This paper proposes the method to detect contours of a face, eyes and a mouth in a color image for making an avatar automatically. First, we use the HSI color model to exclude the effect of various light condition, and we find skin regions in an input image by using the skin color is defined on HS-plane. And then, we use deformable templates and Genetic Algorithm(GA) to detect contours of a face, eyes and a mouth. Deformable templates consist of B-spline curves and control point vectors. Those can represent various shape of a face, eyes and a mouth. And GA is very useful search procedure based on the mechanics of natural selection and natural genetics. Second, an avatar is created automatically by using contours and Fuzzy C-means clustering(FCM). FCM is used to reduce the number of face color As a result, we could create avatars like handmade caricatures which can represent the user's identity, differing from ones generated by the existing methods.

Growth and optic characteristics of AgGaS$_2$/GaAs single crystal thin film by hot wall epitaxy (HWE 방법에 의한 AgGaS$_2$/GaAs 단결정 박막 성장과 광학적 특성)

  • The stochiometric composition of AgGaS$_2$ polycrystal source materials for the AgGaS$_2$/GaAs epilayer was prepared from horizontal furnace. From the extrapolation method of X-ray diffraction patterns it was found that the polycrystal AgGaS$_2$ has tetragonal structure of which lattice constant a$\sub$0/ and c$\sub$0/ were 5.756 ${\AA}$ and 10.305 ${\AA}$, respectively. AgGaS$_2$/GaAs epilayer was deposited on throughly etched GaAs(100) substrate from mixed crystal AgGaS$_2$ by the Hot Wall Epitaxy (100) system. The source and substrate temperature were 590$^{\circ}C$ and 440$^{\circ}C$ respectively. The crystallinity of the grown AgGaS$_2$/GaAs epilayer was investigated by the DCRC (double crystal X-ray diffraction rocking curve). The optical energy gaps were found to be 2.61 eV for AgGaS$_2$/GaAs epilayer at room temperature. The temperature dependence of the photocurrent peak energy is well explained by the Varshni equation, then the constants in the Varshni equation are given by ${\alpha}$ : 8.695${\times}$10$\^$-4/ eV/K, and ${\beta}$ = 332 K. From the photocurrent spectra by illumination of polarized light of the AgGaS$_2$/GaAs epilayer, we have found that crystal field splitting ΔCr was 0.28 eV at 20 K. From the PL spectra at 20 K, the peaks corresponding to free and bound excitons and a broad emission band due to D-A pain are identified. The binding energy of the free excitons are determined to be 0.2676 eV and 0.2430 eV and the dissociation energy of the bound excitons to be 0.4695 eV.

  • We demonstrate epitaxial growth of ZnO thin films on 4H-SiC(0001) substrates using pulsed laser deposition (PLD). ZnO and SiC have attracted attention for their special material properties as wide band gap semiconductors. Especially, ZnO could be applied to optoelectronic applications such as light emitting devices and photo detectors due to its direct wide bandgap (Eg) of ~3.37eV and large exciton binding energy of ~60meV. SiC shows a good lattice matching to ZnO compared with other commonly used substrates and in this regard SiC is a good candidate as a substrate for ZnO. In this work, ZnO thin films were grown on 4H-SiC(0001) substrates by PLD using an Nd:YAG laser with a 355nm wavelength. The crystalline properties of the films were evaluated by x-ray diffraction (XRD) $\theta-2\theta$, rocking curve and pole figure measurements using a high-resolution diffractometer. The surface morphology of the films was studied by atomic force microscopy (AFM).

  • A quantitative flow visualization technique was developed to measure velocity and temperature fields simultaneously in a two-dimensional cross section of thermo-fluid flows. Thermochromic liquid crystal(TLC) particles are used as temperature sensor and velocity tracers. Illuminating a thermo-fluid flow with a thin sheet of white light, the reflected colors from the TLC particles in the flow were captured simultaneously by two CCD cameras; a 3-chip CCD color camera for temperature field measurement and a black and white CCD camera for velocity field measurement. Variations of temperature field were measured by using a HSI true color image processing system and TLC solution. The relationship between the hue values of TLC color image and real temperature was obtained and this calibration curve was used to measure the true temperature under the same camera and illumination condition. The velocity field was obtained by using a 2-frame PTV technique using the concept of match-probability to track true velocity vectors from two consecutive image frames. These two techniques were applied at the same time to the unsteady thermal-fluid flow in a Hele-Shaw cell to measure the temperature and velocity field simultaneously and some results are discussed.

  • This study is about the excavated costume representing fundamental "Conservation and Restoration". It's not a report of a specific case, but it is a guideline that contains the costume and textile of museum collection. It is not research based on experiments; however, this paper is basically consists of various reported-documents. Followings are the conclusions of this study 1. These are the factors that we have to know to prevent the causes of fabric's degradation. -Light, -Humidity and temperature, -Microscopic organism, -Insect and rodent animal, -Air pollution, -Ph, -Handling 2. Basic principles of conservation and restoration follows are: -Select the Reversible method, -Represent the easily distinguishable repaired place, -Should be acted by an expert or people with experience, -Before the restoration, accurate and specified records should be completed, -Procedure, treatment method, and materials used should be recorded prior to restoration, -Should be cared minimally, -Be cautious when using the conservation materials, -When caring, make sure nothing is against the principle of aesthetic, historic, and form of preservation 3. The types of restoration are type of straight or curve, type of hole, type of without warp or weft, type of special part damage something like sleeve, collar, type of form that is severely damaged, and type of separated pieces. 4. The method of restoration is sewing, stitching, and the combination of sewing and stitching. 5. The restoration seams are welt seam, plain seam, flat felled seam, french seam etc. And there are kinds of used-sewing, such as, broad stitching, backstitch, half backstitch, basking, hemming, saddle stitching etc.
