Thyrotropin-Binding Inhibiting Immunoglobulin(TBII) in Patients with Autoimmune Thyroid Diseases (자가면역성 갑상선질환에서의 혈청 Thyrotropin-Binding Inhibiting Immunoglobulin치)

  • In order to evaluate the significance of thyrotropin-binding inhibiting immunoglobulin (TBII) in the patients with autoimmune thyroid diseases, the authors investigated 402 cases of Graves' disease and 230 cases of Hashimoto's thyroiditis comparing 30 cases of normal healthy adult at Kyung Pook University Hospital from February 1993 to August 1994. The TBII was tested by radioimmunoassay and assesed on the dynamic change with the disease course, thyroid functional parameters, and other thyroid autoantibodies : antithyroglobulin antibody (ATAb) and antimicrosomal antibody (AMAb) including thyroglobulin. The serum level of TBII was $40.82{\pm}21.651(mean{\pm}SD)%$ in hyperthyroid Graves' disease and $8.89{\pm}14.522%$ in Hashimoto's thyroiditis and both were significant different from normal control of which was $3.21{\pm}2.571%$. The frequency of abnormally increased TBII level was 92.2% in hyperthyroid Graves' disease, 46.7% in euthyroid Graves' disease or remission state of hyperthyroidism, and 23.9% in Hashimoto's thyroiditis. The serum levels of increased TBII in Graves' disease were positively correlated with RAIU, serum T3, T4, and FT4, but negatively correlated with serum TSH(each P<0.001). The TBII in Graves' disease had significant positive correlation with serum thyroglobulin and AMAb, but no significant correlation with ATAb. In the Hashimoto's thyroiditis, the serum levels of TBII were positively correlated with RAIU, serum T3, TSH and AMAb, but not significantly correlated with serum T4, FT4, thyroglobulin and ATAb. Therefore serum level of TBII seemed to be a useful mean of assessing the degree of hyperthyroidism in Graves' disease and correlated well with thyroidal stimulation. The serum level of TBII in Hashimoto's thyroiditis is meaningful for the degree of both functional abnormality reflecting either hyperfunction or hypofunction and the immune logic abnormality.

The Effects of Tryptophan and Tyrosine-Enriched Diet on the Serum Cortisol, Glucose and Free Fatty Acid Levels of Stressed Rats (Tryptophan 및 Tyrosine 보강 식이가 Stress로 인한 혈청 Cortisol, Glucose 및 Free Fatty Acid의 농도 변화에 미치는 영향)

  • We made S.D male rats eat 3.6% tryptophan(trp) or tyrosine(thr)-enriched diet for 4 days and measured the trp or tyr use in serum of stressed rats as well as cortisol, glucose and free fatty acid(ffa) changes in serum. When control group had received stress treatment, their tyr level has dropped significantly but no changes in trp level and their cortisol, glucose, ffa levels in serum were increased significantly all together. Trp-enrichment alone can't change serum cortisol and ffa levels but trp pools in serum and brain enlarged by dietary enrichment. Trp-enriched group's serum glucose concentration was significantly lower than control-dieted group. When trp-enriched group received stress treatment, their responses to stress were different from control-dieted group. Serum trp concentration of trp-enriched-with-stress group has dropped significantly and cortisol level was increased significantly but not as much as control-dieted-with-stress group. Glucose and ffa levels of trp-enriched-with-stress group did not increase at all. Tyr-enriched group has also larger serum pool of tyr and lowest basal cortisol level in all three diet group. tyr-enriched group's and ffa levels were in normal range and those responses to stress were same pattern with control diet group. Most importantly in tyr-enriched-with-stress group, only slight but not significant increase of cortisol level was shown.

Change of Serum Steroid Level in the Whole-body Irradiated Rat (방사선 처리를 받은 흰쥐의 혈청내 스테로이드호르몬의 변화)

  • The effect of radiation on the steroidogenesis of rat ovary was investigated. For this purpose, female rats aged 7-8 weeks were exposed to a single dose of ${\gamma}$ radiation(320 rad or 800 rad) by the cobalt-60. At fourth(day of the first estrus) and eighth(day of the second estrus) days after irradiation, the concentrations of serum steroid hormones were determined by radioimmunoassays (RIA). The correlation between survival rate(Y) and radiation dose(X) was Y=-0.06X+100(r=0.89, n=10). Lethal dose$(LD)_{50(30)}$ and $LD_{20(30)}$ were 833.33rad and 333.33rad respectively. The weights of body and ovary was decreased significantly by the $LD_{50(30)}$ irradiation during the 4 days, but both weights were recovered at day 8. The serum levels of 17a-hydroxyprogesterone(170HP) and estradiol($E_2$) in the irradiated rats were not generally different from those of control. However, the levels of testosterone(T) and 19-norterstosterone(19NT) in the irradiated rat at $LD_{50(30)}$ and $LD_{20(30)}$ radiation doses were decreased, while progesterone levels in serum of the irradiated rats were slightly increased. These results suggest that irradiation may inhibit the first step of steroidogenesis, especially the conversion of P to T, in the rat ovary.

Effects of a Stair-step Growth Pattern on Improvements in Meat Quality and Growth in Hanwoo Steers

  • The present study was conducted to examine the effect of a stair-stepped feed intake pattern on growth, feed efficiency, and meat quality of Hanwoo steers. Twenty-seven 11-month-old Hanwoo steers were randomly divided into three groups. The control group was fed according to the Korean steer feeding program, and the other two groups were fed according to an alternated feeding schedule of 3-2-4-2 months. During the first three months of the experiment, treatment group 1 (T1) and treatment group 2 (T2) were fed 20% and 30% less than the control group, respectively. For the following two months, the T1 group was fed 20% more than the control group while the T2 group was fed 20% less than the control group. In the third step, T1 and T2 groups were fed 20% and 10% less, respectively, than the control group for four months. In the last two months, T1 and T2 groups were fed 20% more than the control group. After the stair-step feeding trial, steers were fed concentrated feed ad libitum for five months. The altered feed intake pattern did not affect daily body weight gain. However, daily feed intake tended to decrease and growth efficiency tended to increase in the two treatment groups compared to the control group. Altered feed intake also affected blood metabolite levels. The serum glucose and BUN level of the T1 group increased in the first re-fed period compared to the T2 and control groups. The serum cholesterol level of the T2 group decreased in the first restricted-re-fed growth period compared to the T1 and control groups. The serum NEFA levels of the two treatment groups increased from the first restricted period compared to the controls. The serum insulin level of the T2 group increased in the last period compared to the T1 and control groups. Regarding meat yield index, the control group was significantly higher than the T2 group (p<0.05). Regarding meat yield grade, the carcass back fat thickness of the T2 group was significantly higher than the control group (p<0.05). In marbling score, the T1 group was the highest (4.9), followed by the control group (4.1) and the T2 group (4.0). These results indicate that using a stair-stepped growth pattern (T1) can contribute to improvements in growth efficiency and muscle marbling.

  • The Hexane Extract (HE) and Ethyl Acetate Soluble Fraction of the Methanolic Extract (EASFME) of the fruit pulp of Momordica dioica Roxb. (Cucurbitaceae) was evaluated for its anti-hepatotoxic activity in rats. Acute hepatotoxicity was induced by administering paracetamol (2 g/kg, p.o.) for 3 days. The extracts, at a dose of 400 mg/kg (p.o.) administered for 7 days exhibited a significant therapeutic effect by lowering Serum Glutamate Oxaloacetate Transaminase (SGOT), Serum Glutamate Pyruvate Transaminase (SGPT), Serum Alkaline Phosphatase (ALP) and Serum bilirubin and increasing the serum protein levels. These biochemical observations were supplemented by histopathological examination of the liver sections. The activity of extract was also comparable to the standard drug Silymarin, which is a well-known natural anti-hepatotoxic drug.

  • This study aimed to determine the effective dose of intravenous administration of L-tryptophan (L-T) on gastrointestinal hormones (GIH) secretions and melatonin using Hanwoo cattle. Three steers ($362{\pm}23kg$) fitted with indwelling jugular vein catheters were assigned in a $3{\times}3$ Latin square design. Treatments were intravenous administration of saline (control), 28.9 mg L-T/kg body weight (BW; low) and 57.8 mg L-T/kg BW (high) L-T for 1 day with 7 days of adaptation. Samples were collected after adaptation period at -60, 0, 30, 60, 90, 120, 150, 180, 240, and 300 min of sampling day. The levels of serum cholecystokinin (CCK) and secretin were higher (p < 0.05) in the high L-T group than those in the other groups. Serum Melatonin (MEL) levels were increased upon L-T administration (p < 0.05) in the high L-T group. Taken together, the effective dose of L-T administration was defined at 57.8 mg L-T/kg BW in order to stimulate increase of GIH and MEL.

An Evaluation on the Thyroid Function Tests of the Patients with Autoimmune Thyroiditis (자가면역성 갑상선염환자의 갑상선기능 검사소견)

  • The study was taken to analyze the laboratory findings of the 161 patients with autoimmune thyroiditis treated at Kyungpook University Hospital from January 1992 to July 1993. They were all female and mean age was 33 years ranging from 10 to 73 years. Mean radioactive iodine uptake(RAIU) of the thyroid was $30.90{\pm}21.80(mean{\pm}SD)%$ at 6 hours and $37.97{\pm}23.25%$ at 24 hours. Mean serum levels of thyroid hormones were $1.41{\pm}0.48$(ng/ml) of T3, $7.26{\pm}3.23$(ug/dl) of T4, and $1.11{\pm}0.66$(ng/dl) of free T4, while mean serum level of TSH was $17.99{\pm}30.72$(uIU/ml). Mean levels of serum autoantibodies were 24. $43{\pm}31.91$(U/ml) of antithyroglobulin antibody and $55.32{\pm}41.97$(U/ml) of antimicrosomal antibody. The correlation between RAIU and serum thyroid hormone levels was significantly negative, but the positive correlation between RAIU and serum TSH was noted. The correlation between thyroid hormones and TSH was significantly negative, but the positive correlation between RAIU and serum TSH was noted. The correlation between thyroid hormones and TSH was significantly negative, while antimicrosomal antibody titer revealed significantly positive correlation with TSH. The RAIU and free T4 showed negatively correlated with the increasing age. The Initial clinical findings of the patients with autoimmune thyroiditis revealed euthyroidism in 83.2%, hypothyroidism in 14.9%, and hyperthyroidism in 1.9%. The incidence of abnormally increased serum thyroglobulin, antithyroglobulin antibody, and antimicrosomal antibody were 21.3%, 97.5%, and 87.6%, respectively and these abnormalities were more frequent in the patients with documented clinical thyroid functional disturbances.

EFFECTS OF DIETARY LEVELS OF CHROMIUM PICOLINATE ON GROWTH PERFORMANCE, CARCASS QUALITY AND SERUM TRAITS IN BROILER CHICKS

  • An experiment was conducted to evaluate the effects of dietary levels of chromium in the form of chromium picolinate on growth performance, nutrient utilizability, carcass composition, serum traits, and in vitro lipolysis and lipogenesis in adipose tissues of Arbor Acre broiler chicks. Experimental diets containing six different levels of chromium (0, 100, 200, 400, 600 and 800 ppb) were fed for 6 weeks. Individual treatment had six replicates of eight birds each and their average initial weight was 59.2 g. Dietary addition of chromium did not affect growth performance and nutrient utilizability. However, mortality appeared to be reduced with addition of chromium to the diet. It was obvious that chromium supplementation significantly decreased serum cholesterol and increased serum HDL cholesterol (p < 0.05), but serum insulin, glucose, triglyceride and non-esterified fatty acid concentrations were inconsistent among dietary supplementation levels of chromium. The in vitro lipolysis and lipogenesis in adipose tissues were significantly influenced by dietary addition of chromium (p < 0.05). Chicks fed diets containing 200 or 400 ppb chromium showed the highest protein content and the lowest fat content in their carcass.

  • In 123 examples of admission patients at Dongshin university oriental medical hospital circulatory internal medicine, we sequently investigated serum lipid, at admission state, 2th and 4th weeks. The results were as follows; 1. In the case of 123 stroke patients, only 12cases were related to Hyperlipemia. So stroke was not related to Hyperlipemia significantly. 2. According to sex, woman has more significant results in the T-cholesterol and LDL-cholesterol. 3. According to the living Zone, Triglyceride of the city zone group washigher than that of the contryside zone significantly. 4. According to the history of drinking and smoke, the patients who have the history of drinking and smoke had a significant remark in T-cholesterol and LDL-cholesterol. 5. In the case of patients like eating meat, it was significant gain of HDL-cholesterol. 6. In the patients who had past history of Hypertension, the value of Triglyceride is significantly low, and sequently as blood pressure decreased, serum lipid value decreased too. 7. In the patients who had past history of D.M., the value of Triglyceride is significantly high, and sequently as blood sugar value decreased, serum lipid value decreased too. 8. In the case of patients treated He-Ne laser, Triglyceride was decreased significantly. 9. In the case of treating the stroke patients, serum lipid had non-significant decrease value.

Detection of Serum IgE Specific to Mite Allergens by Immuno-PCR

  • Background: Although a skin test is the primary option for detecting allergen-specific IgE in clinics, the serum IgE immunoassay is also important because it allows for the diagnosis of allergy without any accompanying adverse effect on the patient. However, the low detection limit of IgE levels by immunoassay may restrict the use of the method in some occasions, and improving its sensitivity would thus have a significant implication in allergy-immunology clinics. Methods: In this study, we attempted to detect specific serum IgE by using immuno-polymerase chain reaction (IPCR) which combines the antigen-antibody specificity of enzyme-linked immunosorbent assays (ELISAs) with the amplification power of PCR. Results: Our results demonstrated that Blo t5-specific serum IgE can be detected by IPCR with a 100-fold higher sensitivity than ELISA, and cross-reactivity of serum IgE to other mite allergens is able to be analyzed by using only $0.3{\mu}l$ of serum sample. Use of real-time IPCR seemed to permit more convenient determination of specific serum IgE as well. Conclusion: We believe that IPCR can serve as a valuable tool in determining specific serum IgE, especially when the amount of serum sample is limited.
