녹용중(鹿茸中)의 아미노산(酸) 함량(含量)에 관(關)하여 (제 1 편)(第 1編) (Study on Amino Acid Contents in Deer Horn)

  • Free amino acids in water extracts and total amino acids in hydrolystates of Deer Horn were analyzed by amino acid autoanalyzer (Technicon PNC-1 Type). The results obtained from this study are as follows ; 1) 17 kinds of amino acid, including 7 kinds of essential amino acid in human nutrition except tryptophan were identified and quantified. 2) Of all free amino acid contained in water extract, glutamic acid is the richest, and then comes Ala, Gly, Leu, Lys, valin in that order. Of all total amino acid which are closely related with nutritional valuation glycin is the richest, and then comes Glu, Lys, Ala, Leu, Ala, Pro, in that order. 3) Besides 17 kinds of amino acid, one kinds of unknown amino acid are found in water extracts and hydrolysates.

Gamakamide-E, a Strongly Bitter Tasting Cyclic Peptide with a Hydantoin Structure from Cultured Oysters Crassostrea gigas

  • A new cyclic peptide (six-membered amino acid), gamakamide-E (L-Leu-L-Met (SO)-L-Me-Phe-L-Leu-D-Lys-L-Phe), was isolated as a strongly bitter tasting compound from cultured oysters, Crassostrea gigas. The molecular formula of $C_{43}H_{61}N_7O_8S$ was deduced from high resolution fast atom bombardment mass spectrometry (HR FAB-MS) ($[M+H]^+$ m/z 836.4356 ${\Delta}$= -2.4 mmu). Its unique structure including a hydantoin structure was firstly elucidated by nuclear magnetic resonance (NMR) analysis. Stereochemistries of constituent amino acids were determined by chiral high performanced liquid chromatography analysis of natural and synthesized peptides.

  • The effects of enzyme inhibitors and penetration enhancers on the permeation of leucine enkephalin (Leu-Enk) and its synthetic analog, [${D-ala}^2$]-leucine enkephalinamide (YAGFL) across the nasal, rectal and vaginal mucosae were evaluated. Enzyme inhibitors and penetration enhancers employed for Leu-Enk permeation study were amastatin(AM), thimerosal(TM) and ethylenediaminetetraacetic acid disodium salt(EDTA), and sodium taurodihydrofusidate (STDHF). Those for YAGFL permeation study were TM, benzalkonium chloride(BC) and EDTA, and STDHF, sodium deoxycholate(SDC), sodium glycholate(SGC), glycyrrhizic acid ammonium salt (GAA), L-$\alpha$-Iysophosphatidylcholine(LPC) and mixed micelle (MM, STDHF: linoleic acid = 15 mM : 5 mM). The addition of TM alone on the donor and receptor solutions for Leu-Enk permeation study across all the three kinds of mucosae failed to inhibit the degradation; it completely degraded in 6 hrs, and no permeation occurred. However, with addition of three kinds of inhibitors together, the fluxes across nasal, rectal and vaginal mucosae were $\20.7{pm}2.5$>/TEX>,$\0.3{pm}0.05$>/TEX> and $\1.4{pm}0.5$ $\mu$\mid$textrm{m}$/$\textrm{cm}^2$/hr, respectively. Moreover, the addition of STDHF in the presence of the above three inhibitors enhanced permeation across nasal, rectal and vaginal mucosae 1.3, 15 and 1.3 times, respectively. YhGFL also degraded in the donor and receptor solutions rapidly as time went. With mixed inhibitors of TM and EDTA, the percents of YAGFL remaining in the donor solutions facing nasal, rectal and vaginal mucosae were 69.7, 69.8 and 79.8%, respectively; the percent permeated increased to 10, 2.1 and 5.7%, respectively. The addition of STDHF in the presence of either BC/EDTA or TW/EDTA increased the permeation 2.2, 11.0 and 2.9 times, and 2.21, 14.0 and 2.7 times for nasal, rectal and vaginal mucosae, respectively. With SDC, SGC, GAA, LPC ud MM in the presence of TM/EDTA increased permeation; especially, they increased permeation across vaginal mucosae effectively, and the enhancement factors were 12.5, 7.6, 8.7, 5.7 and 5.5, respectively. The degradation extent of YAGFL was correlated with protein concentrations in the epidermal and serosal extracts. The flux of YAGFL across nasal mucosa increased dose-dependently.

Characterization of New Antihypertensive Angiotensin I-Converting Enzyme Inhibitory Peptides from Korean Traditional Rice Wine

  • This study describes the characterization of a new angiotensin I-converting enzyme (ACE) inhibitory peptide from a Korean traditional rice wine. After purification of the ACE inhibitor peptides with ultrafiltration, Sephadex G-25 column chromatography, and successively $C_{18}$ and SCX solid-phase extraction, reverse-phase HPLC, and size exculsion chromatography, two types of the purified ACE inhibitors with $IC_{50}$ values of 0.34 mg/ml and 1.23 mg/ml were finally obtained. The two purified ACE inhibitors (F-1 and F-2) were found to have two kinds of novel oligopeptides, showing very little similarity to other ACE inhibitory peptide sequences. The amino acid sequences of the two purified oligopeptides were found to be Gln-Phe-Tyr-Ala-Val (F-1) and Ala-Gly-Pro-Val-Leu-Leu (F-2), and their molecular masses were estimated to be 468.7 Da (F-1) and 357.7 Da (F-2), respectively. They all showed a clear antihypertensive effect on spontaneously hypertensive rats at a dosage of 500 mg/kg.

멸치를 이용한 식품가공용 중간소재의 가공 (Processing of the Intermediate Product (Frozen Seasoned Anchovy Meat) Derived from Anchovy)

  • 멸치를 보다 효율적으로 이용하기 위한 일련의 연구로 각종 조리재료나 식품가공용 중간소재로 이용할 수 있는 냉동멸치조미육의 가공조건을 구명하고 본 시제품의 화학성분을 분석하였다. 냉동멸치조미육제품을 가공하기 위하여 채육, 세절한 멸치육에 대해 분리대두 단백 : 물 : 대두유(1 : 5 : 2)로 만든 유화커어드 12.8%, 식염 0.5%, 설탕 2.0%, sodium bicarbonate 0.4%, 중합인산염 0.2%, MSG 0.2%, 분리대두단백 3.0%, 양파가루 0.3%, 생강가루 0.1%, 후추가루 0.1% 항산화제로서 sodium erythrobate를 0.2% 첨가하여 멸치육 알맹이가 붕괴되지 않도록 혼합한 후$-35^{\circ}C$에서 급속동결시켜 carton box로 포장하여 $-25{\pm}2^{\circ}C$에 동결 저장하는 것이 가장 좋았다. 냉동멸치조미육제품의 수분함량은 73.0%. 조단백질 19.1%, 조지방은 5.0%였으며 생균수는 $1.5{\sim}1.6{\times}10^5/g$, histamine함량은 70.6mg/100g이었다. 유화커어드를 첨가함으로써 제품의 지방함량 조정 및 조직감을 개선 시킬 수 있었다. 제품의 주요구성 지방산은 linoleic, oleic, palmitic, docosahexaenoic, linolenic, palmitoleic, eicosapentaenoic acid 등으로 불포화지방산이 많이 함유되어 있었고, Glu, Asp, Leu, Lvs, Ala 등이 주요 구성아미노산이었다. 멸치조미육제품의 정미성분은 IMP(160.9mg/100g)와 같은 핵산관련물질 및 Glu(179.4mg/100g), His(84.7mg/100g), Ala(54.4mg/100g), Leu(37.9mg/100g) 둥 유리아미노산이 정미성 및 양적으로 보아 주체를 이루고 있었고, 이 외에 total creatinine(158.3mg/100g), 미량의 TMAO, betaine 등으로 이루어져 있었다.

  • 박테리오 파이지 K11 lysozyme은 최근에 우리 실험실에서 클로닝 되었으며, 숙주균주의 세포벽을 분해하는 고유의 lysozyme활성과 박테리오 파아지 K11 RNA 중합효소의 전사반응을 억제하는 활성을 가지고 있는 것으로 확인되었다. 이미 잘 연구된 박테리오 파아지 T7 lysozyme의 경우 클로닝되고 분리 정제된 T7 lysozyme 단백질의 3차 구조 및 T7 RNA 중합효소와의 결합양상에 대하여 밝혀졌다. 따라서 우리 실험실에서는 K11 lysozyme과 K11 RNA 중합효소와의 결합 정도 및 그 특성을 파악할 목적으로 yeast two hybrid 시스템을 통하여 K11 RNA 중합효소와 K11 lysozyme의 단백질-단백질 상호작용을 알아보고자 하였다. LexA 시스템을 이용하여 LexA DNA 결합 부위를 갖고 있는 pLexA에 K11 lysozyme 유전자를 삽입하여 prey로 하였따. 활성 부위로는 B42 융합 단백질을 만드는 pJG4-5에 K11 RNA 중합효소의 결합은 생체 밖에서 reporter 유전자인 lacZ와 leu2의 발현으로 확인되었으며 이들의 결합정도와 결합부위에 대한 연구들은 진행중에 있다.

Effect of In ovo Injection of Critical Amino Acids on Pre- and Post-hatch Growth, Immunocompetence and Development of Digestive Organs in Broiler Chickens

  • Two experiments were conducted to standardize in ovo injection of amino acids (AA) and to evaluate the effect of in ovo injection of limiting AA(s) on pre and post hatch growth performance, immune response and development of digestive organs. Combinations of essential and non-essential amino acids (Lys+Arg, Lys+Met+Cys, Thr+Gly+Ser, Ile+Leu+Val and Gly+Pro) were injected into 50 eggs in each treatment group at 14 d of embryonic age. Standardization of injection site, needle length and embryonic age revealed that when AA were injected in to the broad end of the egg with a 11 mm needle and at the narrow end with a 24 mm needle both at the 7$^{th}$ and 14$^{th}$ d of incubation there was poor hatchability. However, better hatchability was recorded when the AA were injected in the narrow end of the egg with a 11 mm needle and in the broad end with a 24 mm needle on the 14$^{th}$ d of incubation. The chick to egg weight ratio was higher (p<0.018) when AA were injected on the 14$^{th}$ d of incubation. When a combination of amino acids were injected a 63.6 or 63.2 g difference in body weight of bird at 21 d was recorded between uninjected control and Ile+Leu+Val or Gly+Pro group, respectively. Higher feed intake (p<0.047) was recorded in the AA injected groups and feed conversion ratio (FCR) was numerically better in Gly+Pro, Lys+Met+Cys AA injected groups than in the uninjected control. Significantly higher immune response to cell mediated (p<0.033) and humoral (p<0.002) immunity was observed in in ovo amino acid injected birds, especially in Lys+Met+Cys, Thr+Gly+Ser or Ile+leu+Val groups. The digestive organ weights at 21 d did not differ between specific AA injected groups and the uninjected control. In ovo injected amino acids may act as immunomodulators and their role in gastrointestinal development needs further research.

Neurofibromatosis type 1: a single center's experience in Korea

  • Purpose: Neurofibromatosis 1 (NF1) is an autosomal dominant condition caused by an NF1 gene mutation. NF1 is also a multisystem disorder that primarily affects the skin and nervous system. The goal of this study was to delineate the phenotypic characterization and assess the NF1 mutational spectrum in patients with NF1. Methods: A total of 42 patients, 14 females and 28 males, were enrolled in this study. Clinical manifestations and results of the genetic study were retrospectively reviewed. Results: Age of the patients at the time of NF1 diagnosis was $15.8{\pm}14.6$ years (range, 1-62 years). Twelve patients (28.6%) had a family history of NF1. Among the 42 patients, $Caf\acute{e}$-au-lait spots were shown in 42 (100%), neurofibroma in 31 (73.8%), freckling in 22 (52.4%), and Lisch nodules in seven (16.7%). The most common abnormal finding in the brain was hamartoma (20%). Mental retardation was observed in five patients (11.9%), seizures in one patient (2.4%), and plexiform neurofibromas (PNFs) in four patients (9.5%). One patient with PNFs died due to a malignant peripheral nerve sheath tumor in the chest cavity. Genetic analysis of seven patients identified six single base substitutions (three missense and three nonsense) and one small deletion. Among these mutations, five (71.4%) were novel (two missense mutations: p.Leu1773Pro, p.His1170Leu; two nonsense mutations: $p.Arg2517^*$, $p.Cys2371^*$; one small deletion: $p.Leu1944Phefs^*6$). Conclusion: The clinical characteristics of 42 Korean patients with NF1 were extremely variable and the mutations of the NF1 gene were genetically heterogeneous with a high mutation-detection rate.

Comparison of Fluoroquinolone Resistance Determinants in Uropathogenic Escherichia coli between 2 Time Periods of 1989 and 2010-2014 at Gangwon Province in Korea

  • Fluoroquinolone (FQ) resistant uropathogenic Escherichia coli (UPEC) have become a major problem in urinary tract infections (UTIs). The purpose of this study was to compare the quinolone resistance-determining region (QRDR) and plasmid mediated quinolone resistance (PMQR) determinants of FQ resistant UPEC between 1989 and 2010-2014. A total of 681 strains of UPEC clinical isolates was collected from Korean healthcare facility in 1989 (123 strains) and in 2010-2014 (558 strains). The minimum inhibitory concentrations (MICs) of FQs were determined by agar dilution method. QRDRs (gyrA, gyrB, parC and parE) and PMQR determinants (qnrA, qnrB, qnrS, aac(6')-Ib-cr and qepA) were analyzed polymerase chain reaction and sequencing method. Among 681 isolates, FQ resistant UPEC were 3 strains (2.4%) in 1989 isolates and 220 strains (39.4%) in 2010-2014 isolates. The rate of the FQ resistant UPEC strains in 2010-2014 isolates was increased than that of in 1989 isolates. UPEC isolates from 1989 and 2010-2014 were shown to carry mutations in gyrA (Ser83 and Asp87), gyrB (Ser464 and Thr469), parC (Ser80 and Glu84) and parE (Glu460, Ser458, Ile464 and Leu445). The most common mutations of QRDRs in 1989 isolates were Ser83Leu and Asp87Gly in gyrA and Ser80Ile in parC (2 strains: 66.7%) while those in 2010-2014 isolates were Ser83Leu and Asp87Asn in gyrA and Ser80Il2 and Glu84Val in parC (88 strains: 40.0%). PMQR determinants were detected only in 2010-2014 UPEC strains (47 strains: 21.4%).

Influence of age and type of feed ingredients on apparent and standardized ileal amino acid digestibility in broiler chickens

  • Two experiments were conducted to determine the effects of bird age on apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) for 10-d-old Experiment (Exp. 1) and 22-d-old (Exp. 2) male broilers. This study investigated the effects of different broiler ages and feed ingredients on AID and SID of AA in corn and soybean meal (SBM). Four hundred and eighty (age = 7 d; initial body weight [BW] = 173.4 ± 12.65 g) and 192 (age = 18 d; initial BW = 772.2 ± 62.13 g) birds were allocated to three dietary treatments in a randomized complete block design with eight replicate cages per treatment. Two diets were formulated based on corn or SBM as the sole source of AA in the diet. A nitrogen-free diet was also formulated to measure basal endogenous losses of AA. Experimental diets were given for 3 and 4 days in Exps. 1 and 2, respectively. An interaction was observed (p < 0.05) between the age of birds and the type of ingredient for the AID of most AA, except for methionine, valine, cysteine (Cys), and tyrosine; however, the effects of age and type of ingredients were diminished in the SID of AA, except for histidine, isoleucine, leucine (Leu), phenylalanine, alanine (Ala), and glutamic acid (Glu). The AID of AA, except for Leu and Cys and the SID of AA, except for Leu, Ala, Glu, and Pro in SBM were greater (p < 0.05) than in corn. As the age of birds increased from 10 to 22 d, digestibility of all AA increased (p < 0.05), regardless of the expression of AA digestibility (i.e., AID and SID). In conclusion, the AID and SID of AA in both corn and SBM increased with increasing age, and the AID and SID of AA in SBM were greater than in corn.
