Biochemical Characterization of Serine Proteases with Fibrinolytic Activity from Tenodera sinensis (Praying Mantis)

  • Three types of proteases (MEF-1, MEF-2 and MEF-3) were purified from the egg cases of Ten-odera sinensis using ammonium sulfate fractionation, gel filtration on Bio-Gel P-60 and affinity chromatography on DEAE Affi-Gel blue gel. The proteases were assessed homogeneous by SDS-polyacrylamide gel electrophoresis and have molecular weight of 31,500, 32,900 and 35,600 Da, respectively. The N-terminal regions of the primary structure were compared and they were found to be different each other. MEFs readily digested the $A\alpha$ - and B$\beta$-chains of fibrinogen and more slowly the ${\gamma}$-chain. The action of the enzymes resulted in extensive hydrolysis of fibrinogen and fibrin, releasing a variety of fibrinopeptides. MEF-1 was inactivated by Cu$^{2+}$ and Zn$^{2+}$ and inhibited by PMSF and chymostatin. MEF-2 was inhibited by PMSF, TLCK. soybean trypsin inhibitor. MEF-3 was only inhibited by PMSF and chymostatin. Antiplasmin was not sensitive to MEF-1 but antithrombin III inhibited the enzymatic activity qf MEF-1. MEF-2 specifically bound to anti plasmin Among the chromogenic protease substrates, the most sensitive one to the hydrolysis of MEFs was benzoyl-Phe-Val-Arg-p-nitroanilide with maximal activity at pH 7.0 and 3$0^{\circ}C$. MEF-1 preferentially cleaved the oxidized B-chain of insulin between Leu15 and Tyr16. In contrast, MEF-2 specifically cleaved the peptide bond between Arg23 and Gly24. D-dimer concentrations increased on incubation of cross-linked fibrin with MEF-1, indicating the enzyme has a strong fibrinolytic activity.ity.

Purification and Characterization of a Cytochrome P-450 from Pravastatin-Producing Streptomyces sp. Y-110.

  • Streptomyces sp. Y-110 cytochrome P-450, induced by the addition of compactin -Na into the culture medium, was purified from the cell extract to apparent homogeniety, mainly by DEAE-Sepharose, hydroxyapatite, and Mono Q column chromatyography. The sepcific activity of purified enzyme on its substrate, compactin-Na, was determined to be 15 nmol of pravastatin per mg protein. The molecular mass of this enzyme on SDS-PAGE was $37{\pm}0.5$ kDa, pI was 4.5, and its CO difference spectrum showed maximum absorption peaks at 452 and 550nm, respectively. The N-terminal amino acid sequence was determined to be Met>Thr>Cys>Thr>Pro>Val>Thr>Val>The>Gly>Ala>Ala>Gly>Gln>Ile>Gly>Tyr>Ala>Leu. Its apparent $K_m$ on compactin-Na was $1.294{\mu}M{\cdot}min^-1,\;and\;V_{max}\;was\;1.028{\mu}M{\cdot}min^-1$. The maximum substrate concentration ($K_s$) for reaction was $270 {\mu}M$and thus $1/[K_s]$ was $3.7{\mu}M$. These physicochemical characteristics and kinetic behavior of this enzyme were compared and shown to be different from those of Streptomyces cytochrome P-450 enzymes reported, suggesting that this enzyme may be an additional member of the Streptomyces cytochrome P-450 family.

Development of fission 99Mo production process using HANARO

  • The widely used medical isotope technetium-99 m (99mTc) is a daughter of Molybdenum-99 (99Mo), which is mainly produced using dedicated research reactors from the nuclear fission of uranium-235 (235U). 99mTc has been used for several decades, which covers about 80% of the all the nuclear diagnostics procedures. Recently, the instability of the supply has become an important topic throughout the international radioisotope communities. The aging of major 99Mo production reactors has also caused frequent shutdowns. It has triggered movements to establish new research reactors for 99Mo production, as well as the development of various 99Mo production technologies. In this context, a new research reactor project was launched in 2012 in Korea. At the same time, the development of fission-based 99Mo production process was initiated by Korea Atomic Energy Research Institute (KAERI) in 2012 in order to be implemented by the new research reactor. The KAERI process is based on the caustic dissolution of plate-type LEU (low enriched uranium) dispersion targets, followed by the separation and purification using a series of columns. The development of proper waste treatment technologies for the gaseous, liquid, and solid radioactive wastes also took place. The first stage of this process development was completed in 2018. In this paper, the results of the hot test production of fission 99Mo using HANARO, KAERI's 30 MW research reactor, was described.

Mutation analyses in Korean patients with MELAS (mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes)

  • The mitochondrial myopathy, encephalopathy, lactic acidosis, and stroke-like episodes (MELAS) is inherited maternally, in which the MTTL1*MELAS 3243 mutation has been most commonly found as a heteroplasmy of A to G point mutation in the $tRNA^{Leu(UUR)}$ gene. The MTTL1*MELAS 3271 mutation is known to be the second common mutation, though clinical features of both mutations are not remarkably different. Recently, a variety of minor mutations have been reported in patients with MELAS. In this study, major efforts have been made to investigate the allele frequency of major three mutations including MTTL1*MELAS 3243, 3252, 3271 in 10 Korean families with MELAS probands. The PCR and subsequent direct sequencing of the PCR product in the regions spanning these three mutation sites were employed to identify the mutation in each proband. All family members have been screened for the presence of these three mutations by PCR-RFLP assay using Apa I, Acc I and Bfr I restriction enzymes. The MTTL1*MELAS 3243 mutation was most commonly found (7 out of 10 families tested) followed by the MTTL1*MELAS 3271 which was identified in 1 out of 10 families. In the remaining 2 families none of three mutations were found, indicating the presence of either nuclear mutation or yet unidentified mitochondrial DNA mutation in these families.

  • X-linked adrenoleukodystrophy (X-ALD) is a rare peroxisomal disorder, that is rapidly progressive, neurodegenerative, and recessive, and characteristically primary affects the central nervous system white matter and the adrenal cortex. X-ALD is diagnosed basaed on clinical, radiological, and serological parameters, including elevated plasma levels of very long chain fatty acids (VLCFA), such as C24:0 and C26:0, and high C24:0/C22:0 and C26:0/C22:0 ratios. These tests are complemented with genetic analyses. A 7.5-year-old boy was admitted to Department of Pediatrics, Chungnam National University Hospital with progressive weakness of the bilateral lower extremities. Brain magnetic resonance imaging confirmed clinically suspected ALD. A low dose adrenocorticotropic hormone stimulation test revealed parital adrenal insufficiency. His fasting plasma levels of VLCFA showed that his C24:0/C22:0 and C26:0/C22:0 ratios were significantly elevated to 1.609 (normal, 0-1.390) and 0.075 (normal, 0-0.023), respectively. Genomic DNA was extracted from peripheral whole blood samples collected from the patient and his family. All exons of ABCD1 gene were amplified by polymerase chain reaction (PCR) using specific primers. Amplified PCR products were sequenced using the same primer pairs according to the manufacturer's instructions. We identified a missense mutation (p.Arg163Leu) in the ABCD1 gene of the proband caused by the nucleotide change 488G>T in exon 1. His asymptomatic mother carried the same mutation. We have reported an unpublished mutation in the ABCD1 gene in a patient with X-ALD, who showed increased ratio of C24:0/C22:0 and C26:0/C22:0, despite a normal VLCFA concentrations.

  • YEp 13 plasmid에 B. amyloliquefaciens의 $\alpha$-amylase gene을 cloning시켜서 얻은 hybrid plasmid를 E. coli C 600으로 형질전환시켜서 amylase 활성을 나타내는 균주를 선별하였다. 선별된 E. coli C 600균주를 plasmid추출하여 전기영동해 본 결과 plasmid가 매우 불안정하였으며, 그중 가장 단순한 plasmid band를 지니고 있으며 amylase활성이 강한 E. coli균주를 선별하였다. 선별된 균주의 균체내에 있는 2개의 plasmid DNA를 분리하여 각각의 plasmid를 pTG 17-1, pTG 17-2로 명명하였으며 S. cerevisiae MC 16에서 형질전환이 가능한 pTG 17-2 DNA를 제한효소 EcoRI과 Pst I으로 restriction한 결과 EcoRI으로 처리한 경우는 7.3, 4.8, 2.4 kb인 3개의 분획으로 나타났으며 Pst I으로 처리한 경우는 linear로 14.5kb임을 알았으며 이로써 pTG 17-2 plasmid의 size가 약 14kb임을 알았다. 또한 E.coli균체내에서의 ampicillin sensitive로써 이 plasmid의 ampicillin resistance site가 결실되었음을 알았고 효모의 형진전환체로 부터의 $\alpha$-amylase는 균체외로 분비되지 않았고 효모균체내의 $\alpha$-amylase는 Somogyi-Nelson방법과 Agar diffusion 방법으로 확인하였다.

Purification and Characterization of Anticoagulant Protein from the Tabanus, Tabanus bivittatus

  • Tabanus anticoagulant protein (TAP) was isolated from the whole body of the tabanus, Tabanus bivittatus, using three purification steps (ammonium sulfate fractionation, gel filtration on Bio-Gel P-60, and ion exchange chromatography on DEAE Sephadex gel). The purified TAP, with a molecular weight of 65 kDa, was assessed to be homogeneous by SDS-polyacrylamide gel electrophoresis, and an isoelectric point of 7.9 was determined by isoelectric focusing. The internal amino acid sequence of the purified protein was composed of Ser-Leu-Asn-Asn-Gln-Phe-Ala-Ser-Phe-lle-Asp-Lys-Val-Arg. The protein was activated by $Cu^{2+}\;and\;Zn^{2+}$, and the optimal conditions were found to be at pH $3\sim6\;and\;40\sim70^{\circ}C$. Standard coagulation screen assays were used to determine thrombin time and activated partial thromboplastin time. Chromogenic substrate assays were performed for thrombin and factor Xa activity. TAP considerably prolonged human plasma clotting time, especially activated partial thromboplastin time in a dose-dependent manner; it showed potent and specific antithrombin activity in the chromogenic substrate assay. Specific anti-factor Xa activity in TAP was not detected. Overall, this result suggested that TAP has significant anticoagulant activity on blood coagulation system.

약용식물 추출물에 대한 항미생물 활성 검색과 폴리페놀 함량 (Antimicrobial Effect of Methanol Extracts from Some Medicinal Herbs and the Content of Phenolic Compounds)

  • 다양한 기능성을 갖는 생약재 32종을 대상으로 천연보존료 개발의 일환으로 메탄올로 추출한 항균활성 물질을 몇 종의 병원균과 식중독균, 식품과 관련이 있는 세균, 젖산균 및 효모 등 12종의 균주 B. subtilis, M. luteus, Sta aureus, Sta. epidermidis, Strep. mutans, E. coli, Pse. aeruginosa, Sal. typhimurium, V. vulnificus, Lac. brevis, Leu. mesenteroides, Sacch. cerevisiae에 대하여 항균활성을 검색하였으며, 항균력이 강한 생약재 추출물은 최소저해농도(MIC)를 측정하였다. 검색한 결과 계피, 목단, 적양, 정향, 팔각향 등의 메탄올 추출물은 12균주 중 세균에 대하여 항균활성이 강하였으나, 젖산균 및 효모에 대해서는 낮은 항균활성을 보였다. 최소저해농도는 B. subtilis에 대해 강한 증식억제효과를 보였고, Sta. epidermidis나 Pse. aeruginosa에 대해서도 거의 모든 추출물이 저해활성을 나타내었다. 하지만 항균활성 측정시 거의 저해가 없었던 Lue. mesenteroides도 대부분의 추출물에서 억제 효과를 나타내 항균성과는 약간 다른 결과를 보였다. 생약재 중 강한 항균력을 보인 녹차, 정향, 적양, 인진호 등은 폴리페놀 함량이 약 7 mg/g 이상으로 높아 이들 페놀성 물질들이 항균활성에 많은 영향을 준 것으로 사료된다. 따라서 여러 미생물에 의해 변패될 수 있는 식품에서 천연보존료로의 이용이 가능할 것으로 생각된다.

Kinetic Studies of Peptidylprolyl cis-trans Isomerase from Porcine Spleen

  • Peptidylprolyl cis-trans isomerase (PPlase) catalyzes the cis-trans isomerization of prolyl peptide and facilitates the folding of cellular proteins and peptides. PPlase consists of two distinct immunophilins, each specifically binding to the immunosupressive drug cyclosporin A (CsA) or FK506, respectively. A PPlase was isolated and partially purified from porcine spleen. The molecular weight of porcine spleen PPlase was determined to be ~14,000 on the basis of SDS-PAGE. The purified enzyme was strongly inhibited by FK506, but not by CsA. The inhibition constant and the true concentration of enzyme preparations were determined by active site titration using the tight binding inhibitor FK506: $K_{i}=18.7$ nM and $E_{t}=172$ nM. The equilibrium ratio of conformer. [cis]/[trans], of prolyl peptide substrates (N-Suc-Ala-Xaa-Pro-Phe-p-NA) in anhydrous trifluoroethanol/LiCl solvent system varied from 0.24 to 0.85 depending on the nature of Xaa. Overall. in this solvent-salt system, the populations of the cis conformer of substrates in equilibrium are higher than in an aqueous solution so that the substantial error caused by high background absorption can be reduced. The reactivities of porcine spleen PPlase are shown to be highly sensitive to changes in the structure of substrates. Thus, $k_{cat}/K_m$ value for the most reactive substrate (Xaa Leu) is $4.007+10^{6}M^{1}s^{1}$ and, is 2,636 fold higher than that for the least reactive peptide substrate tested, Xaa=Glu.

  • Catharsius protease-2 (CPM-2) was isolated from the body of dung beetles, Catharsius molossus, using a three step purification process (ammonium sulfate fractionation, gel filtration on Bio-Gel P-60, and affinity chromatography on DEAE Affi-Gel blue). The purified CPM-2, having a molecular weight of 24 kDa, was assessed homogeneously by SDS-polyacrylamide gel electrophoresis. The N-terminal amino acid sequence of CPM-2 was composed of X Val Gin Asp Phe Val Glu Glu lie Leu. CPM-2 was inactivated by $Cu^{2+}\;and\;Zn^{2+}$ and strongly inhibited by typical serine proteinase inhibitors such as TLCK, soybean trypsin inhibitor, aprotinin, benzamidine, and ${\alpha}_1$-antitrypsin. However, EDTA, EGTA, cysteine, $\beta$-mercaptoethanol, E64, and elastatinal had little effect on enzyme activity. In addition, antiplasmin and antithrombin III were not sensitive to CPM-2. Based on the results of a fibrinolytic activity test, CPM-2 readily cleaved $A{\alpha}-$ and $B{\beta}$-chains of fibrinogen and fibrin, and y-chain of fibrinogen more slowly. The nonspecific action of the enzyme resulted in extensive hydrolysis, releasing a variety of fibrinopeptides of fibrinogen and fibrin. Polyclonal antibodies of CPM-2 were reactive to the native form of antigen. The ELISA was applied to detect quantities, in nanograms, of the antigen in CPM-2 protein.
