It is well known that multipoint and computerized intraoperative mapping systems improve the results of surgery for Wolff-Parkinson-White syndrome and show tremendous potential for opening an entirely new era of surgical intervention for the more common and lethal types of supraventricular tachyarrhythmias such as atrial flutter and atrial fibrillation. In addition, the ability to map and ablate the sometimes fleeting automatic atrial tachycardia is greatly enhanced by computerized mapping systems. In this study, we have developed 64 channel computerized data analysis system using microcomputer (Macintosh ${II}_{x}$) for basic research of electrophysiology and electrical propagation. The bipolar electrogram information is acquired from 64 cardiac sites simultaneously at a sampling rate of 1 ksampls/sec with continuous and total data storage of up to 30 seconds. When the reference electrogram is selected and reference point is picked up, delay time from the reference point is displayed on two dimensional diagram of the heart. System design permits easy expansion to almost 256 simultaneous sites. this system is expected to enable us to study pathophysiology of cardiac arrhythmia and to improve the result of diagnosis and surgical treatment for cardiac arrhythmia.
Pharmaceutically active compounds (PhACs) have become an environmental havoc in last few decades with reported cases of antibiotic resistant bacteria (ARB) and antibiotic resistant genes (ARGs), lethal effects over aquatic organisms, interference in natural decomposition of organic matter, reduced diversity of microbial communities in different environmental compartments, inhibition of growth of microbes resulting in reduced rate of nutrient cycling, hormonal imbalance in exposed organisms etc. Owing to their potential towards bioaccumulation and persistent nature, these compounds have longer residence time and activity in environment. The conventional technologies of wastewater treatment have got poor efficiency towards removal/degradation of PhACs and therefore, modern techniques with efficient, cost-effective and environment-friendly operation need to be explored. Advanced oxidation processes (AOPs) like Photocatalysis, Fenton oxidation, Ozonation etc. are some of the promising, viable and sustainable options for degradation of PhACs. Although energy/chemical or both are essentially required for AOPs, these methods target complete degradation/mineralization of persistent pollutants resulting in no residual toxicity. Considering the high efficiency towards degradation, non-toxic nature, universal viability and acceptability, AOPs have become a promising option for effective treatment of chemicals with persistent nature.
Organic pollution causes eutrophication and dystrophication, which occur when excessive amounts of organic matter enters seawater. Eutrophication can contaminate sediment and harm aquaculture. Polychaeta species have been shown to restore eutrophic sediment. In this study, we used peptone to simulate a eutrophic environment and detect the levels at which eutrophication became toxic to the polychaete Perinereis aibuhitensis. Peptone concentrations were 0, 100, 200, and 500 mg/L. The median lethal concentrations were 950.35 mg/L at 48 h, 340.34 mg/L at 72 h, and 120.22 mg/L at 96 h, which are much higher than those of other aquatic species. Polychaeta species are highly tolerant of eutrophication. During the 15-day long-term experiment, sediment loss on ignition, as well as seawater total organic carbon and total nitrogen all decreased significantly (P<0.05). However, $NH_4^+$ concentration increased with time. Perinereis aibuhitensis slowed the increment of $NH_4^+$ but could not prevent its increase. Our results indicate that this polychaete is helpful in the recovery of seawater and sediment from eutrophication.
Single non-lethal doses of chloroform $(CHCL_3)$ dichlorobromomethane $(CHCL_2Br)$, dibromochloromethane $(CHCIBr_2)$, or bromoform $(CHBr_3)$ were administered to male rats. Routes of exposure including single intraperitional (ip) and subcutaneous (sc) injection were used in order to permit comparison of severity of THM effects and renal toxicity was assessed at varied times following treatment. On an equimolar basis, sc administration of $CHBr_3$ (either 12 or 3 mmoles/kg) is more effective at increasing KW/BW than ip $CHCI_3$ treatment. Plasma urea nitrogen (BUN) following ip THM injections are markedly increased with all four THM at 24 hours post treatment. BUN response to $CHCL_2Br$ and $CHCIBr_3$-effected BUN levels have essentially returned to those of vehicle control. THM sc treatment results in a BUN response similar to that seen following ip treatment, with only the time course being different. With the exception of $CHCL_3$, sc and ip-treatments appear to be equally effective in evoking absolute BUN elevations. These results suggest that THM administration induce renal toxicity dependent upon the route or exposure.
Aquatic ecotoxicity of bisphenol A, a well-known endocrine disrupter in mammals, was studied using laboratory-reared Daphnia magna as a test organism. The static acute 48 h $EC_{50}$ of bisphenol A for daphnid neonates(<24 hold) was 12.9 mg/l and 110 h $LC_{50}$ values of bisphenol A for daphnid embryos of different ages after deposition into the brood chamber increase with ages in the range of 1.55 mg/l-8.91 mg/l. Also, 48 h $EC_{50}s$ generally increase with daphnid's ages in the range of 12.9 mg/l-19.8 mg/l. In the acute toxicity tests using mature daphnids, the lethal response and immobility all showed good concentration-response relationship with exposure concentration and exposure time, showing little difference between lethality and immobility. These results clarify that acute toxicity tests, using daphnid and its embryo, could also be useful tools easily available for the assessment of ecotoxicity of various harmful chemicals.
Objectives : Traditional medicine Oryeong-san (ORS) has been prescribed for a long time to treat light fever, thirst, dysuria, and accompanying edema. However, the acute toxicity and safety were not reported. In this study, we evaluated the potent acute toxicity and safety of ORS. Methods : ICR mice were used to evaluate acute toxicity and safety by oral administration of 0, 500, 1,000, 2,000mg/kg of ORS. Mortality, body weight, and clinical symptoms were observed, and organ weight and blood biochemical parameters were analyzed after necropsy. Results : We found no mortality and no toxic or abnormal clinical symptoms by administration of ORS. Comparing with control group, no significant alterations in organ weight and blood biochemical parameters were observed. Conclusions : ORS recognized as safe and non-toxic medicinal material, and median lethal dose considered to be over 2,000 mg/kg in both male and female ICR mice.
Altered mRNA expression of two metallothionein isoforms (MT-IA and MT-IB) in response to acute heavy metal exposure was examined in mud loach, Misgurnus mizolepis, fry using a real-time RTPCR assay. Sublethal exposure (1 or 5 ${\mu}M$) to Cd, Cr, Fe, Mn, Ni, and Zn resulted in highly variable transcriptional responses of the two MT isoforms to the heavy metal ions, including upregulation, a steady state, and downregulation. Overall, the most potent inducer of both MT isoforms was Cd (up to 6-fold). Another exposure experiment using a series of doses of Cu revealed that the stimulation patterns of the two MT isoforms differed: MT-IA transcription was soon saturated at higher concentrations (about 2-fold at 1-4 ${\mu}M$ of Cu), whereas the activation of MT-IB was more dependent on the treatment dose (increased up to 5-fold at 3 ${\mu}M$). The isoform-specific allotment of constitutive and inducible functions was not as clear in fry as in adult tissues. Coordinated interaction between the MT-IA and MT-IB isoforms was hypothesized based on the finding that MT-IA represented a primary action under 'less stressful' or 'sublethal' conditions, whereas the activation of MT-IB became important under 'more stressful' or 'lethal' circumstances in this species.
A cytoplasmic polyhedrosis virus (CPV) was isolated from the larvae of Thaumetopoea pityocampa and shown to cause an infection of midgut cells. This viral infection revealed several important diagnostic symptoms, including discoloration of the posterior midgut, reduced feeding, and extended development time of the larvae. The virus infection is lethal to Thaumetopoea pityocampa, and with the increasing doses kills the larvae within 4-5 days post infection. Electron microscopy studies showed typical cytoplasmic polyhedral inclusion bodies that are icosahedral, and ranged from 2.4 to $5.3{\mu}m$ in diameter. Electrophoretic analysis of the RNA genome showed that the virus has a genome composed of 10 equimolar RNA segments with the sizes of 3,907, 3,716, 3,628, 3,249, 2,726, 1,914, 1,815, 1,256, 1,058, and 899 bp, respectively. Based on morphology and nucleic acid analysis, this virus was named Thaumetopoea pityocampa cytoplasmic polyhedrosis virus (TpCPV), and belongs to the genus Cypovirus, family Reoviridae.
Toxoplasma gondii is the causative agent of toxoplasmosis with symptoms of congenital neurological and ocular diseases and acquired lymphadenitis, retinochoroiditis, and meningoencephalitis. Small molecules which block the activity of protein kinases were tested in in vitro culture of T. gondii to find new therapeutic drugs of safer and more effective than the combined administration of pyrimethamine and sulfadoxine that sometimes provoke lethal Stevens-Johnson syndrome. Among them, Gefitinib and Crizotinib inhibited intracellular growth of T. gondii in HeLa cells by counting the number of T. gondii per parasitophorous vacuolar membrane whereas Sunitinib did not. Gefitinib inhibited the growth of T. gondii in a dose-dependent manner over $5{\mu}M$ up to the tolerable concentration of HeLa cells and halted the division of the parasite immediately from the time point of treatment. Gefitinib inhibition suggests that tyrosine kinases of EGFR family or other homologous kinases of the parasite itself may be the target to cause the block of T. gondii growth.
The effects of chlorine dioxide on the oxidation of phenol and disinfection were studied in the various test water conditions. With the 0.3mg/l of chlorine dioxide dose, the spiked phenol(initial concentration: 0.1mg/l) was completely oxidized within 10 minute. The removal rate of phenol was much faster in distilled water than in ground water and filtered water. The applied dose of chlorine dioxide concentrations higher than 0.2mg/l was sufficiently enough for the complete oxidation of phenol. However, with 0.1mg/l of dose, chlorine dioxide can oxidize only 20% of the spiked phenol. The reactive substances present in test water may influence the chlorine dioxide demand in water. pH effect of oxidation rate was also investigated. Increasing the pH, the removal rate of phenol was found to be increased. The disinfection test of chlorine and chlorine dioxide were conducted and compared. The lethal effect for the both disinfectants are similarly powerful. The time for 99% inactivation of E. coli was obtained within 120 sec with the 0.2mg/l of each dose.
