Effects of Dimilin on Larval Development of the Portinind Crab Liocarcinus corrugatus (주름꽃게의 유생발생에 미치는 Dimilin의 영향)

  • laboratory experiments were conducted to determine the effects of Dimilin, an insect growth regulator which acts to inhibit chitin synthesis, during the larval development of liocarcinus corrugatus Pennant. The larvae was exposed to control (10 ppb acetone sea water and untreated sea water solution) and five concentrations 0.1, 1.0, 5.0, 10.0 and 25.0 ppb of both TG and WP-25 formulations of Dimilin from the hatching to the megalopal stage, and the effect of Dimilin on development of the larvae were determined. Two formulations (TG and WP-25) had different effect on the different stages in L. corrugatus. and early stage larvae of L. corrugatus were more sensitive to TG than to WP-25. Concentrations of diflubenzuron >5.0 ppb are lethal to L. corrugatus larvae upon chronic exposure. Lethal concentrations are defined here as those in which less than 10% of the larvae survived to the megalopal stage. However, Dimilin (TG and WP-25) showed no significant effects on developmental time of L. corrugatus larvae.

Drought Resistance Assessment of Ground Cover Plants for Low Management and Light Weight Green Roof System (저관리·경량형 옥상녹화를 위한 지피식물의 내건성 평가)

  • This study was carried out to suggest an experimental base in selecting the drought resistance of plants. Adopting the natural drought method, this paper studies the drought resistance of 12 kinds of ground cover plants. focusing on analyzing the changes of relative water content on leaf, relative electric conductivity and chlorophyll content in 12 kinds of plants, and and the relation between soil water content under drought stress. The drought resistance of the plants were subject to laboratory and rooftop drought resistance treatments. The Logistic model of nonlinear regression analysis was used to evaluate the lethal time that were predicted with the range of 10.4~30.1d on roof top, and 19.5~39.0d on hothouse. The result shows that with the increase of stress time, relative water content and chlorophyll content on leaf were in a downward trend; the relative electric conductivity was upward tendency. Among 12 species of ground cover plants, exclude Pulsatilla koreana, Ainsliaea acerifolia were selected for rooftop plants because they showed resist drought strongly and took adaptive ability.

Acute Toxicity of Synthetic Detergent on the Larvae of Loach, Misgurnus angillicaudatus (미꾸리 자어에 대한 합성세제의 급성독성)

  • Short-term acute toxicity of synthetic detergent(LAS) to larvae of loach, Misgurnus angillicaudatus was examined by static bioassay. The larvae were exposed to 15 different concentration of synthetic detergent for 16, 48, 72, 96 and 120 hours in order to determine median lethal concentration($LC_{50}$). The $100\%$ mortarlity of larvae was showed within 120, 96, 48 and 16 hours for 6, 18, 30 and 38 ppm, respectively. The median lethal concentration values of the larvae were 12.59 ppm for 48 hours, 4.00 ppm for 96 hours and 1.02 ppm for 120 hours. The permissible toxicant concentration of acute toxicity to larvae was $0.37{\sim}0.43$ ppm, and application factor of the synthetec detergent was $0.093{\sim}0.108$. The median lethal time($LT_{50}$) for different concentration also was determined. The $LT_{50}$ of 0.2 ppm was found within 165.1 hours and 2 ppm was 106.2 hours, while the $LT_{50}$ of 8 ppm was 60.3 hours and that of 38 ppm was 23.5 hours.

Cloning and Nucleotide Sequence Analysis of the asd Gene from Shigella sonnei KNIH104S (Shigella sonnei KNIH104S로부터 asd 유전자의 클로닝 및 염기서열 분석)

  • Shigella sonnez is important causes of human enleric infcctions. S. sonnei KNIH104S was isolated from patient of shigellosis in Korea and previously reported. We cloned 1.7 kb BamHI fragment containing the asd gene encoding an aspartate $\beta$-semialdehyde dehydrogenase from chromosomal DNA of S. sonnei KNIH104S. This recombinant plasmid was named as pSAB17. E. coli $\chi$6097, an a d mutant, cannol grow on the LB medium without DL-$\alpha$, $\varepsilon$-diaminopimclic acid (50 pgiml) but E. coli x 6097(pSAB17) can grow on the same medium. We sequenccd the asd gene ol Shigella for the first time. The asd gcne was composed of 1,104 base pairs with ATG initiation codon and TAA termination codon. Sequence comparison of the asd gene exhibited 99.9% nucleolide sequence hornology with that of E. coli. Also, We constructed the balanced-lethal vector using pBluescrip SK(+) and asd gene of S. sonnei KNIH104S.

Physiological Genetic Studies on the Erects of Methyl methanesulfonate in Drosophila melanogaster (Drosophila melanogaster에 있어서 Methyl methane sulfonate의 영향에 대한 생리유전학적 연구)

  • Methyl methanesulfonate (MMS) was fed to Drosophila melnogaster in order to investigate its toxic capability at developmental and adult stages, and the hereditary effect of toxicity and the potency for induction of sex-linked lethal mutation during the slyer-matogenesis by the means of an attached-X method. In the control group, the egg to adult viability of D. melnogaster was 95.2%, while 3. 5mM and 5.0mM treated groups were 90.0% and 84.1%, respectively. In the case of their progenies (Fl), the viability was 96.9% in the control group, while 3.5mM and 5.0mM treated groups were 54.5% and 1.6%, respectively. Therefore, these differences between two generations show significant physiological toxic effects in the next generation. In the parental generation, the developmental time was calculated 11.05 days in the control group, 12.43 days In 3.5%mM treated group, and 13.23 days in 5.0mM. In the case of Fl it was estimated 10.35 days in the control group, and 11.43 days In 3.5mM treated group. Compared with the control groups In two generations, the developmental time generally delayed as the dose of MMS increased. As to the sex-ratio, there was no differences between the control and MMS treated groups. The toxic values of adult stage showed which increased the frequency of mortality with MMS concentrations. The mortality at 120hr In the control group was 1.67% and it in 0.5mM MMS treated group 3.33%. In 2.5mM MMS treated group, it was 33.3% at 72hr, and it 95% at 120hr The increase of the morality was shown from 72hr in 4.0mM treated group which was 100% at 96hr. There was the concentration-dependent induction of sex-linked lethal mutation during the spermatogenesis by means of an attached-X method, MMS had more pronounced effect in sperm and spermaid stages in D. melnogaster.

Morphological, Phylogenetic and Biological Characteristics of Ectropis obliqua Single-Nucleocapsid Nucleopolyhedrovirus

  • The tea looper caterpillar, Ectropis obliqua, is one of the major pests of tea bushes. E. obliqua single-nucleocapsid nucleopolyhedrovirus (EcobSNPV) has been used as a commercial pesticide for biocontrol of this insect. However only limited genetic analysis for this important virus has been done up to now. EcobSNPV was characterized in this study. Electron microscopy analysis of the occlusion body showed polyhedra of 0.7 to $1.7\;{\mu}m$ in diameter containing a single nucleocapsid per envelope of the virion. A 15.5 kb genomic fragment containing EcoRI-L, EcoRI-N and HindIII-F fragments, was sequenced. Analysis of the sequence revealed that the fragment contained eleven potential open reading frames (ORFs): lef-1, egt, 38.7k, rrl, polyhedrin, orfl629, pk-1, hoar and homologues to Spodoptera exigua multicapsid NPV (SeMNPV) ORFs 15, 28, and 29. Gene arrangement and phylogeny analysis suggest that EcobSNPV is closely related to the previously described Group II NPV. Bioassays on lethal concentration $(LC_{50}\;and\;LC_{90})$ and lethal time $(LT_{50}\;and\;LT({90})$ were conducted to test the susceptibility of E. obliqua larvae to the virus.

Bioassays On marine Organisms: Acute Toxicity Test Of mercury, Cadmium And Copper To Arkshell, Anadara broughtonii, From Jin-Dong Bay, And To Oyster, Crassostrea gigas, From Kwang-Do Bay, South Coast Of Korea

  • Short-term acute toxicity of mercury, cadmium and copper to arkshell, Anadara broughtonii, and to oyster, Crassostrea gigas, was determinedby static bioassays from 20 May to 27 June in 1977. During the observations of the opening rate of the shell mercury was the most sensitive toxicant of the three toxic substances to the test animals and caused them to close their shellvalves together after being exposed to a mercury solution for an hour during the test. Opening rate to cadmium and copper increased gradually at the higher concentration. the 96hr-LC50 values for the test animals are 4.84mg/l for mercury and 1.86mg/l for cadmium, while the 72hr-LC50 value for copper is 0.31mg/l. the death rate of oysters for cadmium showed lower than that of the mercury and copper test solutions. The 96hr-LC50 values of mercury, copper and cadmium were 1.1mg/l, 2.54mg/l and 19.5mg/l, respectively. For oysters mercury was the most toxic substance, and cadmium was the least toxic one. the medium lethal time (LT 50) value decreased gradually at higher concentration of heavy metals. The LT 50 of 2mg/l was found within 96 hours ofr copper, 104 hours for mercury and 121 hours for cadmium. The lethal threshold concentrations for 7 days were found to be about 1mg/l for mercury and copper, and 2mg/l for cadmium.

Effects of nitrite exposure on survival and physiology of white leg shrimp, Litopenaeus vannamei (아질산 노출이 흰다리새우, Litopenaeus vannamei의 생존율 및 독성 생리에 미치는 영향)

  • Juveniles of the white leg shrimp Litopenaeus vannamei (Weight 0.18±0.08 g) were exposed to nitrite-N at 0, 25, 50, 100, 200 and 400 mg/L for 72 hours, and the lethal concentration, heamolymph and genes regulation were evaluated. The lethal concentration 50 (LC50) of L. vannamei exposed to nitrite-N was 141.2 mg/L at 25℃ and 33 psu. In Total protein, total cholesterol, and BUN in heamolymph temporarily increased after the start of the experiment and then stabilized, but glucose, an indicator of stress, decreased over time in the entire experimental group, and creatines, an indicator of tissue damage, decreased with nitrite concentration until the first 12 hours. The genes of immune-related showed that masquerade-like serine proteinase(Mas) increased at 50 and 400 ppm for 24 hours, and then gradually decreased depending on concentration. In the case of prophenoloxidase, it was highest at 400 ppm for 40 hours, and other genes(Ras-related nuclear protein, Masquerade-like serine proteinase, proPO-activating enzyme) showed a response for 48 hours and then gradually decreased. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of L. vannamei.

Insecticidal Activity of Metarhizium anisopliae FT83 against the Different Stages of Beet Armyworm, Spodoptera exigua (곤충병원성 곰팡이 Metarhizium anisopliae FT83의 파밤나방 생육단계별 살충활성)

  • The beet armyworm, Spodoptera exigua is pest which is difficult to control. For eco-friendly beet armyworm managements, we isolated entomopathogenic fungi from soil samples by insect-bait method using Tenebrio molitor and conducted bioassay to larvae of beet armyworm. The result of bioassay, a selected strain Metarhizium anisopliae FT83 caused 100% mortality against first ~ third instar larva of S. exigua at $1{\times}10^8conidia/ml$ and medial lethal time ($LT_{50}$) were 0.5 days, 2.6 days and 2.5 days respectively. Mortality against fourth and fifth larvae were $83.3{\pm}6.2%$ and $86.0{\pm}5.7%$ and medial lethal time ($LT_{50}$) were 4.2 days and 3.6 days respectively. Mortality against pupae and eggs of S. exigua were 100%. M. anisopliae FT83 showed high virulence at all developmental stages of S. exigua.

Effect of Water Temperature Condition on Growth and Survival of Juvenile Geoduck (Panopea japonica A. Adams, 1850) (사육 수온이 코끼리조개 Panopea japonica (A. Adams, 1850) 치패의 성장과 생존에 미치는 영향)

  • This study was performed to determine the effect of water temperature condition on survival rate and growth of juvenile geoduck (Panopea japonica). Panopea japonica juveniles were bred for 6 weeks at 12, 15, 18, 21, 24 and $27^{\circ}C$ on incubator. Juveniles median lethal times (LT50) were 29 hours in $27^{\circ}C$, 14.5 day in $24^{\circ}C$, 37.4 day in $21^{\circ}C$. Survival rate of water temperature 12, 15 and $18^{\circ}C$ showed a high survival rate in 95.0%, 95.0% and 93.3% at 6 weeks. As a result of culturing for 6 weeks at 12, 15, 18, 21 and $24^{\circ}C$, a daily growth of shell length and total weight was 0.10 mm, 0.12 mm, 0.13 mm, 0.16 mm and 0.14 mm, and 2.21 mg, 2.65 mg, 2.84 mg, 3.13 mg and 2.93 mg. Juvenile shell length and total weight have significantly increased at $15-21^{\circ}C$. However, mortality rate has significantly increased at more than $21^{\circ}C$. As a result, appropriate water temperature for juveniles culturing considered $15-18^{\circ}C$.
