본 연구에서는 방사선 조사식품의 규정을 확립하고 안전성을 입증하기 위해 1 kGy 감마선 조사 오렌지를 ICR 마우스에 투여 또는 섭취시켜 급성 및 아만성 독성시험을 실시하였다. 급성독성의 경우 시험물질을 0, 1,000, 2,000 mg/kg의 용량으로 설정하여 단회 투여한 후 14일간 사망동물, 일반 증상, 체중 변화, 부검소견, 혈액학적 및 혈액생화학적 검사를 수행한 결과 어떠한 이상도 관찰되지 않았으며, 통계학적으로도 유의적인 차이를 나타내지 않았다. 따라서 1 kGy 감마선 조사 오렌지의 대략의 치사량(approximate lethal dose)은 암수 ICR 마우스 모두 2,000 mg/kg을 상회하는 것으로 판명되었다. 아만성독성은 시험물질을 식이에 함유하도록 하여 3개월간 섭취시킨 후 사망동물, 일반증상, 체중 변화, 사료 섭취량, 장기 무게, 조직학적 검사, 혈액학적 및 혈액생화학적 검사를 수행하였다. 시험기간 동안 암수 ICR 마우스 모두에서 사망동물이나 이상증상은 발견되지 않았으며, 체중 변화, 사료 섭취량 및 장기 무게에서도 통계학적으로 유의적인 차이를 보이지 않았다. 혈액학적 및 혈액생화학적 검사에서도 암수 ICR 마우스 모두 정상적인 수치를 나타냈다. 또한 조직학적 검사에서 간, 신장 조직은 모두 정상적인 구조를 유지하고 있었으며, 염증, 괴사 등의 유의할만한 병적 변화도 관찰되지 않았다. 따라서 1 kGy 감마선 조사 오렌지를 ICR 마우스에 3개월간 섭취시켜도 본 시험조건에서는 독성이 없는 것으로 판명되었다.
국내 자생 수생식물인 노랑꽃창포와 창포를 수질 내 카드뮴(Cd) 제거용 식물재료로 이용하고자 Cd 축적과 내성을 조사하였다. Cd 농도에 따른 치사율을 근거로 Cd에 대한 반수치사량($LD_{50}$)을 계산한 결과, 노랑꽃창포는 $78.5{\mu}M$, 창포는 $47.6{\mu}M$로 나타났다. Cd 스트레스에 대해 노랑꽃창포에서는 POD와 SOD, 창포에서는 CAT와 SOD, 폴리페놀이 활성화되었다. 노랑꽃창포에서는 폴리페놀이 측정되지 않았다. 노랑꽃창포와 창포 모두 Cd 처리 농도가 증가하고 기간이 길어질수록 식물체 내 Cd 축적량이 증가하였고, 두 종 모두 지상부보다는 지하부 축적량이 현저하게 많았다. 지상부에 축적된 Cd 함량은 노랑꽃창포 $548.1mg{\cdot}kg^{-1}$(지하부 축적 대비 82.1%) 창포 $121.4mg{\cdot}kg^{-1}$(지하부 축적 대비 13.7%)로 두 종 모두 Cd 고축적식물로 확인되었으며, 특히 노랑꽃 창포가 Cd 축적능력과 내성이 우수하였다.
신선한 왕대(Phyllostachys bambuoides sieb. et zucc.)를 이용하여 기계식 죽초액과 재래식 죽초액을 제조하고 이들의 농도에 따른 항균활성을 조사하였으며 정체 기계식 죽초액이 대두유 저장 과정 중 산패억제 효과를 조사하였다. 각각의 죽초액은 10-50 ${\mu}L/8$ mm paper disc 농도범위에서 공시 모든 공시균주에 대하여 활성을 나타내었으며 재래식 죽초액의 편이 기계식 죽초액에 비하여 항균활성이 더 크게 나타났다. 동일한 조건하에서 죽초액의 항균활성은 그람양성균 > 그람음성균 > 젖산균 > 효모의 순이었다. 그리고 모든 공시균주에 대하여 3.0%의 acetic acid 50 ${\mu}L/8$ mm paper disc에 상당한 죽초액의 항균활성 농도는 10-50 ${\mu}L/8$ mm paper disc 범위이며, 각각의 균주에 대한 활성농도는 서로 차이를 나타내었다. MIC는 대체로 1.0-7.3 ${\mu}L/mL$ 범위였으며 최대치사농도(MLC)는 26.7-116.7 ${\mu}L/mL$ 범위였다. 대두유 저장기간 동안 죽초액 첨가 농도 0.1-1.0% 범위에서 농도가 증가할수록 산패억제 효과도 컸으며, 죽초액 0.1% 첨가하였을 때 효과가 BHT 0.02% 첨가하였을 때와 유사하였으며 ${\alpha}-tocopherol$ 0.02% 첨가한 것보다 높은 산화억제 효과를 보였다. 죽초액 0.1, 0.5 및 1.0% 첨가 대두유의 유도기간은 각각 3.75, 4.57 및 12.06일 이었으며, 무첨가구의 2.86일에 비하여 모두 길었고, 상대적 항산화 효과는 각각 130.85, 159.60 및 421.30%로 죽초액 0.5%를 첨가할 때가 BHT 0.02% 첨가하였을 때의 168.45%와 비슷하게 나타났다. 죽초액은 일부 식중독균 및 식품부패균에 대한 억제효과와 유지 산패억제 효과가 인정되므로 이를 이용한 식품첨가제의 개발이 기대된다.
A tuber lectin from Arisaema jacquemontii Blume belonging to family Araceae was purified by employing a single step affinity chromatography using column of asialofetuin-linked amino activated silica beads and the bound lectin was eluted with 100 mM glycine-HCl buffer pH 2.5. The purified A. jacquemontii lectin (AJL) showed a single protein band with an apparent molecular mass of 13.4 kDa when submitted to SDS-polyacrylamide gel electrophoresis under reducing as well as non-reducing conditions. The native molecular mass of AJL determined by gel filtration on a Biogel P-200 column was 52 kDa and its carbohydrate content was estimated to be 3.40%. Thus AJL is a tetrameric glycoprotein. The purified lectin agglutinated erythrocytes from rabbit but not from human. Its activity was not inhibited by any of the mono- and disaccharides tested except N-acetyl-D-lactosamine having minimal inhibitory sugar concentration (MIC) 25 mM. Among the glycoproteins tested only asialofetuin was found to be inhibitory (MIC $125\;{\mu}g/mL$). A single band was obtained in native PAGE at pH 4.5 while PAGE at pH 8.3 showed two bands. Isoelectric focusing of AJL gave multiple bands in the pI range of 4.6-5.5. When incorporated in artificial diet AJL significantly affected the development of Bactrocera cucurbitae (Coquillett) larvae indicating the possibility of using this lectin in a biotechnological strategy for insect management of cucurbits. Larvae fed on artificial diet containing sub-lethal dose of AJL showed a significant decrease in acid phosphatase and alkaline phosphatase activity while esterase activity markedly increased as compared to larvae fed on diet without lectin. Out of various human cancer cell lines employed in sulphorhodamine B (SRB) assay, this lectin was found to have appreciable inhibitory effect on the in vitro proliferation of HCT-15, HOP-62, SW-620, HT-29, IMR-32, SKOV-3, Colo-205, PC-3, HEP-2 and A-549 cancer cell lines by 82, 77, 73, 70, 41, 41, 37, 29, 21 and 21% respectively.
Single and 28-day repeated dose toxicity studies of botulimnn toxin type A were carried out in ICR mice and Sprague-Dawley rats, respectively. In the single dose toxicity study, botulinwn toxin was injected intraperitoneally to male and female mice at a single dose of 40, 59, 89 133 and 200 ng/10 ml saline/kg. All animals died from 59 ng/kg group. Some clinical signs, such as decrease in locomotor activity, dyspnea, prone position and ptosis, were observed in most of both sexes from 59 ng/kg group, but no signs were seen in all animals at 40 ng/kg group. The results showed that the median lethal dose of botulinum toxin might be in the range of 40-59 ng/kg in both sexes. In the repeated dose toxicity study, the test material was administered intradermally for 28 days at doses of 0 (vehicle-treated control), 1.25, 2.5, 5.0 and $10.0ng/head/50{\mu}{\ell}$ saline in male and female rats. No test material-related changes were noted in survivals, clinical signs, food and water consumptions and gross finding in any group. Botulinum toxin treatment significantly decreased the body weight gain rate in male of 5.0 ng/head group and over and in female of 10.0 ng/head group compared to vehicle-treated control. One or more relative organ weights (i.e., spleen, thymus, liver and kidney) were increased significantly from 5.0 ng/head group compared to vehicle-treated control in both sexes. Serum biochemistry revealed increases in aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatine phosphokinase, total protein and albumin in male, and increases in AST and ALT and decreases in $K^+$ and $Cl^-$ in female without dose-pendent manners. In the histopathological study, physical stimulation by needle caused slight inflammations of dennis. In addition, botulinum toxin treatment induced denervation of nerve cell and disuse of muscle, resulting in atrophy of skeletal muscle in both sexes from 2.5 ng/head group. When the antibodies to toxin were determined in all animals, a significant increase in serum antibodies was observed from 5.0 ng/head group. The results showed that the NOAEL of botulinum toxin might be 1.25 ng/head for 28-day repeated dose toxicity in rats.
We investigated the effects of temperature changes on the oxygen consumption rhythm in Japanese eels, Anguilla japonica, using an automatic intermittent flow respirometer (AIFR). The endogenous rhythm of the oxygen consumption rate (OCR) in the eels (n = 18; 44-74 cm, 145-690 g), freshly collected by bag net from estuaries, was nearly synchronous with the tidal pattern of the estuarine collection site. The magnitude of mean OCR (mOCR) of eels showed variable range of 82.2 - 116.5 ml $O_2\;kg^{-1}ww \;h^{-1}$ under constant conditions. In case of increasing temperature from 25 to $38^{\circ}C$, the OCR of eels exhibited a gradually increasing trend with a rhythmic pattern until $36^{\circ}C$. Above $36^{\circ}C$ the rhythms of the OCR dampened and the OCR decreased rapidly at around $36-37^{\circ}C$. The OCR of the eels exhibited the maximum value at $38^{\circ}C$, and then it sharply decreased. The results suggested that the critical thermal maximum (CTM) regarding the endogenous rhythms of the eels was at around $36-37^{\circ}C$ when water temperature increased at $0.5^{\circ}C$/14 h following the acclimation at $25^{\circ}C$. In case of decreasing temperature ($0.5^{\circ}C$/14 h) from 25 to $0^{\circ}C$, the OCR of the eels displayed a abrupt decrease up to $23^{\circ}C$, and between at 23 and $20^{\circ}C$, there was an agitation which showed a slight increase in the OCR with a duration of 1-2 days. Below $9^{\circ}C$, the OCR rhythm of the eels showed a constant state regardless of temperature decreasing. These results suggest that the Japanese eel has an upper incipient lethal temperature at $36^{\circ}C$, with a lower thermal limit at $9^{\circ}C$. The biochemical aspects of the eels influenced by water temperature need to be further studied.
Objectives To evaluate Shinbaro Pharmacopuncture safety through analysis of potential single-dose intramuscular toxicity of Sinbaro Pharmacopucture in SD rats and Beagle dogs. Methods Single-dose intramuscular toxicity of Shinbaro Pharmacopuncture was assessed in accordance with Korea Food and Drug Administration Guidelines for toxicity testing of Medicinal Products. The SD rats were treated intramuscularly with Shinbaro Pharmacopuncture at doses of 0, 4.6, 9.2, and 18.5 mg/kg, respectively. The Beagle dogs were treated intramuscularly with Shinbaro Pharmacopuncture at doses of 2.3, and 4.6 mg/kg, respectively, and after 3 days, the procedure was repeated a second time at doses of 0.6, and 1.2 mg/kg, respectively, for toxicity testing. Mortality, change in body weight, and necropsy findings were examined for the study period. Results There were no mortalities, general symptoms, or body weight changes in the SD rats. While pyelectasis of the left kidney was observed in a male rat in the 4.6 mg/kg administration group, natural occurrence is common, and does not appear to be related with the test substance. No mortalities were observed in the Beagle dogs. In assessment of general symptoms, a female dog in the 9.2 mg/kg group displayed body weight decrease due to leftover food, but the change in body weight was within the normal range seen at 6~7 months, and the necropsy findings were not significant. The toxicity of the test substance appears to be minimal. Conclusions The results suggest that the lethal dose 50 ($LD_{50}$) and approximate lethaldose (ALD) value in single intramuscular administration of Shinbaro Pharmacopuncture in SD rats and Beagle dogs are higher than 18.5 mg/kg.
Hemoflavoenzyme으로서 cellobiose dehydrogenase(CDH)는 셀룰로오스를 분해하는 과정에서 세포 외부로 분비되는 효소로서 amorphous cellulose와 강하게 결합하여 셀룰라아제(cellulase)에 의해 microcrystalline cellulose의 가수분해를 증가시킨다. 따라서 CDH는 바이오 에탄올 생산의 당화공정에서 중요한 역할을 할 것으로 예상된다. 여러 백색부후균으로부터 CDH 생산이 높은 Phanerochaete chrysosporium ATCC 32629 균주를 선정하였으며, 균주로부터 생산된 CDH 효소활성의 최적 온도와 pH는 각각 ${55^{\circ}C}$와 4이었다. CDH 활성을 증가시키기 위하여 P. chrysosporium ATCC 32629 균주를 돌연변이시켰다. 돌연변이는 새로운 시도로써 국부적으로 큰 에너지를 줄 수 있는 특징을 가진 양성자 빔을 이용하였다. 양성자 빔 조사 후 사멸율이 약 99.9%인 1.2 kGy에서 CDH 활성이 증가된 변이주를 얻었다. 선별된 변이주와 모균주를 액체배양했을 때 변이주가 모균주보다 CDH와 $\beta$-glucosidase 활성이 각각 약 1.4배와 20배 증가하였다. 따라서, CDH 뿐만 아니라 $\beta$-glucosidase 활성이 높은 P. chrysosporium 변이주를 확보하였다.
방향족 탄화수소 화학물질들은 자연계에 오염되면 미생물에 의한 분해가 미미하여 장기 축적됨으로써 생명체에 독성을 나타낸다. 이러한 방향족 탄화수소가 준치사 수준의 농도로 미생물에 노출되면 stress-shock 단백질을 합성하거나 세포 구성물질에 생화학적 변화가 일어나 적응현상이 나타나게 된다. 본 연구에서는 Pseudomonas sp. DJ-12를 여러 가지 농도의 catechol 로 처리했을 때 나타나는 stress-shock 단백질의 합성양상과 함께 세포의 형태와 생존을 위한 내성의 변화를 연구하였다. Pseudomonas sp. DJ-12는 0.5~1mM 의 catechol 이 6시간 배양 후 60% 이상이 분해되었으나, 3mM 도는 그 이상의 농도에서는 전혀 분해되지 않앗고 생존 세포수는 30시간 처리 했을 때부터 \10^2$ cell/ml 또는 그 이상이 사멸되었다. DnaK는 1mM 이상의 catechol 로 10분간 처리할 때 유도 생성되었고, GroEL은 0.5 mM 이상에서 생성되었다. 10mM 의 catechol로 처리한 Pseudomonas sp. DJ-12 의 세포는 세포벽에 구멍이 생겼으며 간균의 형태가 일그러지는 변화가 관찰되었다. 준치사 농도인 1mM의 catechol 이나 benzoate 또는 4-chlorobenzoate 로 전처리한 Pseudomonas sp. DJ-12는 stress-shock 단백질이 합성되었을 뿐 아니라, 치사 농도인 10mM 의 catechol에서 tolerance를 나타내었다.
수온 2$0^{\circ}C$에서 넙치가 수용된 실험구에 ClO$_2$를 농도별로 처리하여 10분간 노출시킨 다음 해수를 유수하면서 넙치의 생존과 생리상태에 미치는 영향을 조사하였다. ClO$_2$ 처리직후의 잔류농도 0.51 ppm에서는 90분이 지나면 모든 개체가 사망하였고, 0.43 ppm에서는 3시간 후의 생존율이 13.3%, 0.34 ppm에서는 24시간이 경과하는 동안 평균 92.0%가 생존하였으며, 0.43과 0.51 ppm에서 넙치의 반수치사시간(($LT_{50}$)은 각각 103분과 32분이었다. 0.34~0.51 ppm $CIO_2$에 노출된 넙치의 Ht, Hb 및 혈청 전해질과 삼투질 농도는 $CIO_2$ 농도가 높고 노출 후 시간이 지날수록 대조군에 비해 유의하게 높은 값을 나타내었다. 0.27 ppm의 경우 Ht, Hb및 혈청 전해질과 삼투질 농도는 대조군과 차이는 없었으나 혈청 glucose 농도는 $CIO_2$ 노출 후 시간이 경과할수록 높아지는 현상을 보여 주었다. 또한 SS 농도가 높은 해수는 SS가 낮은 해수에 비해 잔류 $CIO_2$가 빠르게 환원되었다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
