Production and Properties of Tannase from Lenzites betulina (Lenzites betulina에 의한 Tannase 생산 및 성질에 관한 연구)

  • Six species under the basidiomycetes were screened for extracellular tannase (tannin acyl hydrolase EC 3.1. 1.20) production in submerged culture and Lenzites betulina was found to be most effective for the production of tannase. The optimum cultural conditions for tannase production were $25^{\circ}C$, pH 6.0 and 21 days of culture period, The efficient composition of culture medium for the production of tannase was performed in synthetic medium containing tannic acid, 2g; sucrose, 5g; bacto-peptone, 2g; ,$ KH_2PO_4, \;2g,\; MgSO_4.7H_2O \;0.5g,\; CuS0_4.5H_2O$, 2 mg; thiamine HCl, 100 ug and distilled water 100 ml, The tannase produced from Lenzites bdulin*r was 223.3 unit (umole of gaUic acidiml of brothlmin). The tannase had an optimal reaction conditions ofpH 6.0 and temperature of $40^{\circ}C$. The enzyme was stable at temperature below $40^{\circ}C$ and lost its activity by 50% above $60^{\circ}C$. And the stable pH range was 5.5 to 6.0.

  • Cellulosic substance which plays an important role in carbon cycle is most abundant in the nature world. Some higher fungi are able to digest cellulose directly to satisfy their carbohydrate requirement. Then, in order to investigate the enzymatic components of Lenzites betulina (Fr.) being wood rot, that fungus was collected in Kwangneung area. The carpophore of the fungus was smashed with cool distilled water, extracted and salted out by ammonium sulfate. And then the precipitate was purified by dialysing with visking tube and dissolved with pH 7.8 ammonia water, and the extract was filtrated. The fraction of filtrate was obtained as light brown powder after lyophilization, and determined cellulolytic activity. Cellulolytic potency of Lenzites betulina (Fr.) was 1. 65 unit/ml. The cellulase of Lenzites betulina (Fr.) was stable at below $45^{\circ}C$ and range of pH $4.5{\sim}6.0$ and is completely inactivated at $60^{\circ}C$ for 15 minutes. The optimum condition for the enzymatic reaction was $40^{\circ}C$ and pH 4.0. The enzyme activity was not influenced by the presence of $Ca^{2+}$ and $Fe^{2+}$.

  • To investigate the hemolytic toxicities of the Korean basidiomycetes, the cold-water extracts of 22 wild mushrooms were tested for their hemolytic activities on the erythrocytes of a mouse and a rat. As a result, three species including Collybia confluence, Laccaria vinaceoavellanea and Lenzites betulina showed strong hemolytic activities. Of these, L. vinaceoavellanea retained more than 80% of its hemolytic activity even after it was boiled for 30 minutes, while the other two were inactivited in five minutes.

  • Fruit bodies of the wood rotting basidiomycetes naturally grown in middle part of Korea were collected from August 1989 to May 1992 and identified to 24 genera and 31 species. Species diversity was differed little from areas, but index of species diversity was higher in Mt. Songni and Mt. Duckyu than that in other areas. The dominant species were Coriolus versicolor, Daedalea dickinsii, Daedaleopsis styracina, Lenzites betulina and Schizophyllum commune.

  • For screening of biological activities of culture broth of some wood rotting basidiomycetes, antimicrobial activity, plant growth regulating activity, antitumor activity, and various enzyme activities were checked. Coriolus versicolor 5129 and C. pubescens 5131 strains showed inhibition activity against gram-positive bacteria and Lenzites betulina 8029 strain showed the activity against gram-negative bacteria. L. betulina 8085 inhibited the growth of both bacteria and plant pathogenic fungi. All of tested basidiomycetes inhibited the germination and growth of radish and cabbage at concentration of 0.8ml/ml. Especially, Fomitopsis pinicolor 8059 and Fomitella fraxinea 8084 showed strong inhibition activity. In contrast, Bjerkandera adusta 8054 stimulated the growth of cabbage and radish at concentration of 0.4 and 0.2ml/ml. All polysaccharides from tested basidiomycetes showed anti-tumor activity against sarcoma 180 and the stronger antitumor activity was observed in L. betulina 8029 and unidentified 8058 strain. All tested basidiomycetes had also an ability to degrade cellulose and lignin.

각종 white rot fungi에 의한 분산염료의 색제거 비교

  • Batch culture system and continuous culture system were used to investigate the removal of disperse dye using several white rot fungi. White rot fungi used in the study were Coriolus hirsutus IFO 4917, Lenzites betulina IFO 6266, Coriolus versicolor IFO 30340 and Phanerochaete chrysosporium IFO 31249. The results of the batch culture experiment showed that white rot fungi used in this study had excellent dye removal abilities. Phanerochaete chrysosporium IFO 31249 was especially effective on the removal of disperse dyes. And continuous treatment of disperse red-60 was studied under bioreactor with vertical matrix using Phanerochaete chrysosporium IFO 31249. The removal efficiency of disperse red-60 were more than 95% in 0.20 ${\sim} 1.50 $hr^{-1}$ dilution rate and 90% in $1.83h^{-1}$ dilution rate.

Color Removal from Disperse Dye Solution Using White Rot Fungi (백색부후균을 이용한 분산염료용액의 색 제거)

  • Batch culture system and continuous culture systems were used to investigate the removal of disperse dye using white rot fungi. White rot fungi used in the study were Coriolus hirsutus IFO 4917, Lenzites betulina IFO 6266, Coriolus versicolor IFO 30340 and Phanerochaete chrysosporium IFO 31249. The results of the batch culture experiment showed that white rot fungi used in this study had excellent dye removal abilities. Phnerochete chrysosporium IFO 31249 was especially effective on the removal of disperse dyes. And continuous treatment of disperse red 60 was studied under two type of reactor using Phanerochaete chrysosporium IFO 31249. The removal efficiency of disperse red 60 for immobilized Phanerochaete chrysosporium IFO 31249 in continuous reactor with vertical matrix was increased 1.3 fold in $1.4\;hr^{-1}$ dilution rate when compared with continuous reactor without vertical matrix.

Studies on the Relative Durability of Useful Woods Grown in Korea to the Parasitic Wood Destroying Fungi (목재부후균의(木材腐朽菌)의 기생(寄生)에 의한 한국산(韓國産) 유용목재(有用木材)의 비교내후성(比較耐朽性) 연구(硏究))

  • This experiment was undertaken for the purpose of investigation of relative durability about thirty sorts of the korean native wood and nine sorts of the wood destroying fungi Polystictus versicolor, Irpex consors. Poria vaporaria, Merulius lacrymans, Schizophyllum commune, Trametes pini, Lenzites betulina, Paxillus panuoides by means of a laboratory methods. The results obtained are; (from the table 2 to 10) 1. By measuring the percentage of loss in weight with regard to Polystictus versicolor, the sap wood contained in Junipeus chinesis was calculated at 18.2%, therefore it is much higher durability than any others and the sap wood contained in Pinus densiflora was measured at 44.10%, therefore it is much lower durability than any others. 2. By measuring the percentage of loss in weight with regard to Irpex consors, the heart wood contained in Butula schmidtii was calculated at 1.23%, therefore durability is very high and the heart wood contained in Quercus dentata was calculated at 26.79%, therefore durability is very low. 3. By measuring the percentage of lass in weight with regard to Lenzites betulina, the heart wood contained in Betula schmidtii was calculated at 5.33%, therefore it is much higher durability than any others and the sap wood with regard to Abies holophylla was measured at 45.48%, therefore durability is very low. 4. By measuring the percentage of loss in weight with regard to Poria vaporaria, the sap wood contained in Fraxinus rhynchophylla was calculated at 5.57%, therefore durability is very high and the sap wood contained in Paulownia coreana was calculated at 35.6%, therefore it is much lower durability than any others. 5. By measuring the percentage of loss in weight with regard to Merulius lacrymans, the heart wood contained in Castanea crenata was measured at 0.09% and in the next place, the heart wood contained in Abies holophylla calculated at 0.15% did not decay almost and the sap wood contained in Cornus controversa was measured at 32.88%, therefore it is lower durability than any others. 6. By measuring the percentage of loss in weight with regard to Schizophyllum commune, the sap wood cantained in Junipensis chinensis was calculated at 1.09%, therefore durability is very high and the sap wood contained in Populus deltoides was calculated at 36.87%, it is much lower durability than any others. 7. By measuring the percentage of loss ill weight with regard to Trametes pini, the sap wood contained in Robinia pseudo-acacia was calculated at 1.33%, therefore durability is very high and the sap wood contained in Comus controversa was calculated at 33.19%, it is much lower durability than any others. 8. By measuring the percentage of loss ill weight with regard to Paxillus panuoides. the heart wood contained in Zelkova serrata was calculated at 1.73%, therefore durability is very high and the sap wood contained in Populus deltoides was calculated at 42.30%, therefore durability in very low. 9. By measuring the percentage of loss in weight with regard to Dardalea quercina, the heart wood contained in Castanea crenata was calculated at 0.92%, therefore durability is very high and the sap wood contained in Cornus controversa was calculated at 32.00% therefore durability is very low. The above results are summarized as follows 1. Durability between the heart wood and the soft wood find that it is nonsignificant, while durability is higher heart wood than sap wood. 2. Special sorts including in the element like Castanea cenata, Robinia pseudo-acacia, Juniperus chinensis and heart wood like Betula schmiditii and Styrax obasia are highly durable and soft wood like Cornus controversa, Populus davidiana, Salix koreensis, Celtis jessensis are lowly durable in general. 3. Among the wood destroying fungi Paxillus panuoides is highly durable and as follows. Poria vaporaria, Lenzites betulina and the Daedalea quercina is less in general and Irpex consors, Mersulius lacrymans are a middle degree.

Inhibitory Activities of Basidiomycetes on Prolyl Endopeptidase, Acetylcholine Esterase and Coagulation (담자균 추출물의 Prolyl Endopeptidase, Acetylcholine Esterase 저해 및 항혈전 응고활성)

  • Fifty six mycelial cultured Basidiomycetes were screened for their inhibitory effects against prolyl endopeptidase(PEP), acetylcholine esterase(AChE) and thrombus coagulation. Out of them, methanolic extract of mycelium and/or ethylacetate(EtOAc) soluble fraction from culture broth of Peniophora quercina, Amanita aspera, Phellinus chrysoloma, Grifola frondosa, Wolfiporia extensa, Clavicorona pyxidata and Phanerochaete sordida inhibited more than 90% of PEP activity at 40 ppm. The extracts of Lenzites betulina, Phellinus chrysoloma, Wolfiporia extensa, Phanerochaete sorrlida, Hypocrea nigricans, Coriolus azureus, Flammulina velutipes, Phlebiopsis gigantea and Bondarzewia montana exhibited about 40% of inhibitory activity against AChE at 40 ppm. In thrombin times assay, the extracts of Amanita aspera, Oxyporus latemarginata, Peniophora quercina, Fomes fomenfarius, Trametes versicolor, and Phlebiopsis gigantea delayed coagulation of thrombus about two to three times over control at ca 550 ppm. In activated partial thromboplastin times assay, none of the tested Basidiomycetes showed significant effect.

Selection of Newly Isolated Mushroom Strains for Tolerance and Biosorption of Zinc In Vitro

  • Nine newly isolated mushroom strains were tested to assess both their zinc tolerance and potential for zinc removal from an aqueous solution. Four strains of ectomycorrhizal fungi, namely Clavariadelphus truncatus (T 192), Rhizopogon roseolus (T 21), Lepista nuda (T 373), and Tricholoma equestre (T 174), along with five strains of white rot fungi, Lenzites betulina (S 2), Trametes hirsuta (T 587), Ganoderma spp. (T 99), Polyporus arcularius (T 438), and Ganoderma carnosum (M 88), were investigated using zinc-amended solid and liquid media. Their biosorption properties were also determined. The colony diameter and dry weight were used as tolerance indices for fungal growth. C. truncatus and T. equestre were not strongly inhibited at the highest concentrations of (225 mg/l) zinc in solid media. The most tolerant four strains with solid media, C. truncatus, G carnosum, T. hirsuta, and T. equestre, were then chosen for tolerance tests in liquid media. An ectomycorrhizal strain, C. truncatus, was also detected as the most tolerant strain in liquid media. However, the metal-tolerant strains demonstrated weak activity in the biosorption studies. In contrast, the highest biosorption activity was presented by a more sensitive strain, G. carnosum. In addition, seven different biosorbent types from G. carnosum (M 88) were compared for their Zn (II) biosorption in batch experiments.
