열대식물 (파푸아 뉴기니아산)의 항암작용연구

  • 해를 거듭할수록 증가추세에 있는 악성종양은 우리나라의 3대 사망원인 질환의 하나이다. 그러나 현재 암의 화학적 치료법은 40여종의 암치료제가 주종을 이루고 있으나 그 효과면에서 한계점에 도달하고 있어 식물추출물, 해양식물, 미생물 등으로부터 얻어지는 천연물 성분에서 강력하고 부작용이 적은 항암제를 발굴하기 위한 연구가 국내외에서 많이 수행되고 있다. 본 연구는 파푸아뉴기니아산 열대식물 13과 23종의 항암작용을 검색할 목적으로 실시하였는데, 실험동물은 ICR계의 암컷생쥐를 이용하였으며 Sarcoma 180 동물복수암세포에 대한 열대 식물 엑스 투여에 의한 수명 연장 효과를 측정함으로써 항암작용의 지표로 삼았다. 항암작용의 지표는 ILS(Increased Life Span)를 사용하였으며 ILS가 20%이상인 것을 항암성이 있는 것으로 간주하였다. 실험의 결과 Apocynaceae에 속하는 Plumiera rubra의 줄기껍질 및 잎 추출물 Leguminosae과의 Pterocarpus indicus의 잎 추출물과, 같은 과의 Derris trifoliata, Moraceae과의 Artocarpus altilis의 뿌리 및 줄기 껍질, 잎 추출물과 Sterculiaceae과의 Kleinhovia hospita의 잎 추출물, Verbenaceae과의 Vitex trifolia의 5과 5종 8시료가 의의있는 항암작용이 있는 것으로 나타났다.

Inhibitory Effect of Astragaloside I and IV on Passive Cutaneous Anaphylaxis Reaction and Scratching Behaviors in Mice

  • To evaluate the antiallergic effect of the dried root of Astragalus membranaceus Bunge (AM) (Leguminosae), which inhibited the mouse passive cutaneous anaphylaxis (PCA) reaction in a preliminary experiment, its main constituents, astragalosides I and IV, were isolated and their antiallergic effects were investigated. Astragalosides I and IV inhibited the PCA reaction induced by the IgE-antigen complex, and the scratching behaviors induced by compound 48/80. These constituents reduced the protein expressions of $TNF-{\alpha}$ and IL-4 in IgE-induced RBL-2H3 cells. These findings suggest that astragalosides I and IV as well as AM can improve IgE-induced anaphylaxis and scratching behaviors.

아까시아나무 모자익병에 관한 연구 2 (Mosaic Disease of Black Locust on Rodinia pseudo-acacja L.(Par. 2))

  • In order to investigate the host range of the mosaic disease of black locust in the Chunchon area, the sap of the mosaic-diseased leaves of black locust itself and the cowpea leaves infected with the above mentioned sap, were inoculated to 53 species of plants belong to 12 families. As to the result, no difference in infection was found as related to the virus sources, and the infection was recognized in 4 species of the family Chenopodiaceae and 8 species of the family Leguminosae. The plants recognized as hosts are as follows: the plants which showed local infection are Chenopodium album, Ch. ambrosioides, Ch. quinoa; the plants which showed systemic infection are Chenopodium amaranticolor, Phaseolus vulgaris, Robinia pseudo-acacia, Vigna sinensis; and Astragalus sinicus, Melilotus indicus, Phaseolus angularis, Pisum sativum and Vicia faba were recognized as carriers. Through investigating its host ranges and symptoms, this mosaic virus of black locust seems not to be regarded as the group of the black locust mosaic virus in southeastern Europe reported by Milinko et al (1961). And, too, it is thought hardly to exist in combination with the cowpea mosaic virus. It appears, therefore, that this mosaic virus was confined to that of black locust.

  • The cytotoxicity-guided fractionation of the roots of Sophora flavescens (Leguminosae) extracts led to the isolation of fifteen active principles 1~15, responsible for the cytotoxicity against five kinds of cultured human tumor cell lines, i.e., A549(non small cell lung), SK-OV-3(ovary), SK-MEL-2(skin), XF498(central nerve system) and HCT-15(colon), evaluated by SRB method in vitro. Compounds 2~14 were classified as unusual flavonoid occurred exclusively in this species and the proliferation of each examined tumor cells were significantly inhibited during the continuous exposure to compounds 1~15 for 48 hours, respectively.

황기의 지질과산화 억제작용 (Antilipidperoxidative activity of Astragalus membranaceus)

  • The root of Astragalus membranaceus (Leguminosae) has been used in traditional chinese prescriptions for strengthning the superficial resistance, promoting pus discharge and tissue regeneration, diuretis and alleviating edema. Lipid peroxidation has been suggested as a major cause of atherosclerosis, cancer, liver disease, and the aging process. In order to investigate the anti-lipid peroxidation activity, Astragali Radix was extracted with $80\%$ MeOH and fractionated with hexane, $CH_{2}Cl_2$, EtOAc, BuOH and Water. The antilipid peroxidation activities of them were determined by human erythrocyte ghost and $CCl_4$-induced lipid peroxidation. $CH_{2}Cl_2$ and EtOAc fractions, especially, isoflavonoids, 7,2'-dihydroxy-3',4'-dimethoxyisoflavan-7-O-${\beta}$-D-glucoside and calyco sin-7-O-${\beta}$-D-glucoside from EtOAc fraction showed anti-lipid peroxidation activities.

소목 추출물의 항산화 및 간보호작용 (Antioxidant and Liver-protective Effects of Caesalpinia sappan)

  • The heartwood of Caeralpinia sappan L. (Leguminosae) has been used to activate blood flow, remove blood stasis, reduce swelling and relieve pain in Korean folk medicine. In this study. the antioxidant effects of this crude drug and its hepatoprotective activity on CCl$_4$-induced liver injury in rats were evaluated. The levels of antioxidant enzymes, superoxide dismutase (SOD) and catalase (CAT) reduced by CCl$_4$treatment, were recovered by this crude drug. It is suggested that Caesalpinia sappan L. have antioxidant effect. The increased levels of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) by CCl$_4$were also recovered by treatment with this crude drug. These results can be attributed to the agent's antioxidant and membrane - stabilizing actions.

  • Reactive oxygen species are capable of damaging biomolecules such as lipids, proteins, and DNA, which can not only lead to various diseases, but also oxidative damage resulting aging. In our previous study, Cercis chinensis (Leguminosae) showed a potent antioxidant activity. Twenty compounds including a new flavonol glycoside were isolated through antioxidant activity-guided fractionation. C. chinensis and some of the constituents exhibited a potent antioxidant activity on the free radicals and lipid peroxidation and a notable protective effect on the t-BuOOH induced oxidative damage.   • Isoflavones have been a central subject in natural phytoestrogens found in Leguminosae. Their effects on bone formation and remodeling are an important turning point in that they can act like estrogen by binding on estrogen receptors on target cell surface. We, therefore, believed that isoflavones may be applied in estrogen deficiency disease such as osteoporosis in terms of estrogen replacement therapy (ERT). As commonly known, osteoporosis is one of hormonal deficiency diseases, especially in menopausal women. (omitted)

  • This study was carried out to examine the constituents of Amorpha fruticosa (Leguminosae), a shrub originated from North Africa. Dried and ground fruit of A. fruticosa were extracted with methanol and then concentrated to give the crude extracts. The crude extracts was successively fractioned with organic sovents, such as n-hexane, CH$_2$Cl$_2$ and EtOAc. Seven compounds were isolated from the fruits of A. fruticosa. On the basis of spectrosopic data, the structures of these compounds were determined as: kaempferol 7-O-${\alpha}$-L-rhamnopyranoside (I), methyl 3,4,5- trihydroxybenzoate (methyl gallate, II), tephrosin (III), dalbinol (IV), gallic acid (V), 2",4",5",7-tetramethoxyisofavone (Ⅵ) and Dalbinol 2"-O-${\beta}$-D-Glucopyranoside (Ⅶ) respectively.   • Two phenolic flavonoids were isolated from the traditional medicine of Eastern Asia, Caesalpinia sappan L (Leguminosae). Brazilin (1) showed a significant inhibitory activity against inducible Nitric Oxide Synthase (iNOS) in lipopolysaccharides (LPS)-induced macrophage RAW 264.7 cells with an IC$\sub$50/ value of 1.68 mg/ml, which is more potent than the positive control, L-N$\^$6/-(1-iminoethyl)lysine (IC$\sub$50/ 3.49 mM). On the other hand, caesalpine J (2) was found to be inactive in the present iNOS assay system despite of their structural similarities. (omitted)
