Optimum Feeding Rate for Sub-adult Olive Flounder (370 g) Paralichthys olivaceus Fed Practical Extruded Pellets at Low Water Temperature (12-14℃) (저수온(12-14℃)에서 사육한 미성어기(370 g) 넙치(Paralichthys olivaceus)의 배합사료 적정 공급률)

  • We investigated the effects of feeding rate on the growth performance and blood components of sub-adult olive flounder Paralichthys olivaceus. Optimum feeding rate (initial fish mean weight : $370{\pm}5.72g$) was determined under the low water temperature. Two replicated groups of fish were fed a commercial diet at rates of 0%, 0.3%, 0.4%, 0.5%, and 0.6% body weight (BW) per day, and to satiation (0.75%). Feeding trial was conducted under a flow-through system with 12 1.2-metric ton aquaria receiving filtered seawater at $12-14^{\circ}C$ for 4 weeks. Weight gain (WG) and specific growth rate (SGR) for fish fed at 0.6% BW per day was significantly higher than that of unfed fish (0%) and fish fed at 0.3% and 0.4% BW per day. There were no significant differences in WG and SGR among fish fed at 0.5%, 0.6%, and 0.75%. These parameters were negative and significantly lower in the starved fish than in fish fed the experimental diet at all feeding rates. Survival for unfed fish (0%) was significantly lower than that of fish fed at 0.4%, 0.5%, and 0.6%. Hematocrit and hemoglobin content of fish fed at 0% and 0.75% (satiation) were significantly lower than that of fish fed at 0.4% BW per day. Total protein content in unfed fish was significantly lower than those in other treatments. Broken-line regression analysis of weight gain showed that the optimum feeding rate of olive flounder weighing 370 g was 0.51% BW per day at the low water temperature.

Effects of Dietary Protein and Lipid Levels on the Growth Performance, Feed Utilization and Innate Immunity of Juvenile Red Seabream Pagrus major (사료 내 단백질과 지방 수준이 참돔(Pagrus major) 치어의 성장, 사료효율 및 비특이적 면역력에 미치는 영향)

  • A $3{\times}3$ factorial study was conducted to investigate the effects of dietary protein and lipid levels on the growth, feed utilization and innate immunity of red seabream Pagrus major. Nine diets consisting of three protein levels (42%, 46% and 50% crude protein) and three lipid levels (10%, 14% and 18% crude lipid) were formulated. Triplicate groups of red seabream were fed the experimental diets to apparent satiation (5-6 times a day, from 08:00 to 18:00 h at 2-h intervals) for 10 weeks. At the end of the feeding trial, the weight gain and specific growth rate of fish fed P46L14 (46% protein and 14% lipid), P50L10 (50% protein and 10% lipid) and P50L14 (50% protein and 14% lipid) were significantly (P<0.05) higher than those of fish fed P42L18 (42% protein and 18% lipid). The feed conversion ratios (FCR) of the fish were affected by dietary lipid levels (P<0.039), but not dietary protein levels. The FCR tended to increase with increasing dietary lipid levels from 10% to 18% with the 46% and 50% protein levels. The weight gain, protein efficiency ratio, specific growth rate, feed intake and survival of fish were not affected by either dietary protein or lipid levels. Myeloperoxidase activity in the group fed P50L14 (50% protein and 14% lipid) was significantly higher than that in the group fed P42L10 (42% protein and 10% lipid) or P50L18 (50% protein and 18% lipid). However, the myeloperoxidase activity of fish was not affected by either dietary protein or lipid level. The fish fed P46L14 (46% protein and 14% lipid) and P46L18 (46% protein and 18% lipid) showed significantly higher superoxide dismutase activity than did the fish fed P46L10 (46% protein and 10% lipid), P50L10 (50% protein and 10% lipid) of P50L18 (50% protein and 18% lipid). In conclusion, the optimum protein and lipid levels for the growth and feed utilization of juvenile red seabream were 46% and 14%, respectively, and the optimum dietary protein to energy ratio was 27.4 g/MJ.

Comparative Evaluation of Extruded and Moist Pellets for Development of High Efficiency Extruded Pellets in Olive Flounder Paralichthys olivaceus (넙치(Paralichthys olivaceus)용 고효율 배합사료 개발을 위한 실험 배합사료 및 습사료 비교 평가)

  • This study was conducted to compare an experimental extruded pellet (EP) and raw fish-based moist pellet (MP) on growth performance, whole-body and dorsal muscle composition, and textural properties of dorsal muscle for olive flounder Paralichthys olivaceus. Four groups of 8,000 fish (initial mean weight $27.1{\pm}0.05g$) per concrete tank ($10{\times}10m$) were fed the experimental diets for 16 months. After the feeding trial, no significant differences were observed in final mean body weight, weight gain, specific growth rate, feed intake, survival, and condition factor between EP and MP groups. However, feed efficiency of fish fed the EP was significantly higher than that of fish fed the MP. Moisture content in the whole-body of fish fed the MP was significantly higher than that of fish fed the EP. Significantly higher contents of polyunsaturated fatty acid (PUFA) in whole-body and dorsal muscle were observed in fish fed the EP compared with fish fed the MP. Any notable differences in amino acid contents or textural properties of the dorsal muscle were not observed in fish fed either the EP or the MP. The results of this study indicated that the EP could be developed to replace MP for the market size of production in olive flounder without the adverse effects on growth performance. Formulation of the EP could be used as a practical feed for olive flounder.

Case Study of the Heavy Asian Dust Observed in Late February 2015 (2015년 2월 관측된 고농도 황사 사례 연구)

  • Asian dust is a seasonal meteorological phenomenon influencing most East Asia, irregularly occurring during spring. Unusual heavy Asian dust event in winter was observed in Seoul, Korea, with up to $1,044{\mu}g\;m^{-3}$ of hourly mean $PM_{10}$, in 22~23 February 2015. Causes of such infrequent event has been studied using both ground based and spaceborne observations, as well as numerical simulations including ECMWF ERA Interim reanalysis, NOAA HYSPLIT backward trajectory analysis, and ADAM2-Haze simulation. Analysis showed that southern Mongolia and northern China, one of the areas for dust origins, had been warm and dry condition, i.e. no snow depth, soil temperature of ${\sim}0^{\circ}C$, and cumulative rainfall of 1 mm in February, along with strong surface winds higher than critical wind speed of $6{\sim}7.5m\;s^{-1}$ during 20~21 February. While Jurihe, China, ($42^{\circ}23^{\prime}56^{{\prime}{\prime}}N$, $112^{\circ}53^{\prime}58^{{\prime}{\prime}}E$) experienced $9,308{\mu}g\;m^{-3}$ of hourly mean surface $PM_{10}$ during the period, the Asian dust had affected the Korean Peninsula within 24 hours traveling through strong north-westerly wind at ~2 km altitude. KMA issued Asian dust alert from 1100 KST on 22nd to 2200 KST on 23rd since above $400{\mu}g\;m^{-3}$ of hourly mean surface $PM_{10}$. It is also important to note that, previously to arrival of the Asian dust, the Korean Peninsula was affected by anthropogenic air pollutants ($NO_3^-$, $SO_4^{2-}$, and $NH_4^+$) originated from the megacities and large industrial areas in northeast China. In addition, this study suggests using various data sets from modeling and observations as well as improving predictability of the ADAM2-Haze model itself, in order to more accurately predict the occurrence and impacts of the Asian dust over the Korean peninsula.

Studies on Quantitative Analysis of Salivary Gland using Computed Tomography (전산화단층사진을 이용한 타액선의 정량분석에 관한 연구)

  • Purpose: The purpose of this study was to calculate the size and CT number of both normal parotid and submandibular gland. and evaluate their relation to sex, age and obesity using computed tomography. Materials and Methods: The computed tomography was performed parallel to the Frankfurt plane in 46 subjects with healthy salivary gland. The subjects were divided into the three groups (young, middle. old) according to their ages. The size of salivary gland was determined as maximum cross-sectional area and the CT number of salivary gland was determined as the mean CT number of three ROI's. The body mass index was calculated from weight and height. Results: The mean maximum cross-sectional area was 7.79(±1.25)cm² on parotid gland and 4.12(±0.83) cm² on submandibular gland. The mean CT number was -4.43(±23.87) HU on parotid gland and 50.01(±15.63) HU on submandibular gland. There was decreasing pattern of the maximum cross-sectional area of submandibular gland and the CT number of both parotid and submandibular gland according to age(p<0.05). As the body mass index increased. the maximum cross-sectional area of parotid gland increased and CT number of both parotid and submandibular gland decreased(p<0.05). The maximum cross-sectional area of submandibular gland in male was larger than that in female(p<0.05). As the maximum cross-sectional area and CT number of left salivary gland increased. those of right gland increased(p<0.05). Conclusion : Intra-individual differences in salivary gland size and CT number is considered in the age and individual obesity.

Screening of GLA (γ-Linolenic Acid) from Fungi by Gas Chromatography and Mass Spectroscopy (Gas Chromatograph-Mass Spectrometer를 이용한 γ-Linolenic Acid(GLA) 생성 균주탐색 및 확인)

  • In order to select ${\gamma}-Linolenic$ acid (GLA)-producing fungi, a total of forty-four strains of 4 genera such as Phytophthora, Pythium, Mucor and Rhizopus were obtained from Koran Agricultural Culture Collection (KACC) and then analysed by using GC-FID and GC-MS. GLA was detected on 39 fungal strains, and the highest rate of GLA was found as 24.8% of total fatty acids on Mucor hiemalis f. sp. hiemalis KACC 40264. Total GLA content of Zygomycota was comparatively high - Mucor (14.2%) and Rhizopus (14.3%), whereas that of Oomycetes was low - Phytophthora (3.3%) and Pythium (3.0%). Moreover, total fatty acids of the Zygomycota fungi such as Mucor (15.4 mg/100 ml) and Rhizopus (7.1 mg/100 ml) were higher compared with the Oomycetes such as Phytophthora (2.6 mg/100 ml) and Pythium (4.5 mg/100 ml). Thus, two genera such as Mucor and Rhizopus have higher potential as an useful microbial resource. The total fatty acid content varies even within the strains of the same genus e.g. Mucor. M. blumbeus KACC 40935 showed the highest values on productivity (18.2%) of GLA and total fatty acid contents (50.8 mg/100 ml liquid medium).

Development of natural fermented seasoning with Flammulina velutipes powder fortified with γ-aminobutyric acid (GABA) by lactic acid fermentation (팽이버섯(Flammulina velutipes) 분말의 젖산발효를 통한 고농도 γ-aminobutyric acid 함유 천연 발효조미료 개발)

  • Lactic acid fermentation of Flammulina velutipes (FV) powder was optimized to produce higher content of ${\gamma}-aminobutyric$ acid (GABA). FV powder (10%) was fermented with 0.5% yeast extract, 1% glucose, 5% mono sodium-L-glutamate (MSG) by Lactobacillus plantarum EJ2014 for 5 days at $30^{\circ}C$. The pH decreased from 6.1 to 4.4 for first 2 days after then increased to 6.2 for following 5 days. While the acidity increased from 0.5% to 1.3% for 2 days, after then decreased to 0.4% for 5 days. Viable cell count showed higher value of $2.2{\times}10^9CFU/mL$ after fermentation for 5 days. In particular, 3.54% MSG as a substrate was completely utilized during lactic acid fermentation, indicating higher 2.31% GABA content. The fermented FV powder showed higher antioxidant properties than that of un-fermented FV power. $IC_{50}$ values of DPPH radical scavenging and ABTS radical scavenging activities were 1.11 mg/mL and 2.58 mg/mL, respectively. Conclusively, natural fermented seasoning from the lactic acid fermentation of 30g of FV powder and 1 g of roasted wheat bran could provide the functional ingredients with 17% GABA, probiotics and dietary fiber, which is used for health food and functional seasoning.

Changes in the quality of frozen vegetables during storage (저장기간에 따른 동결채소의 품질 변화)

  • This study was conducted to assess the possibility of preserving frozen vegetables (Aster scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion) for a long period and of using them after such storage by measuring changes in quality due to their preservation. Various freshly harvested vegetables were blanched under optimal conditions (that were determined in a preliminary experiment), quick-frozen at $-40^{\circ}C$ for 24 h, and stored at $-20^{\circ}C$. The change in the chromaticity of the frozen A. scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion did not vary. The hardness of the frozen A. scaber, green pumpkin, Chinese cabbage and Welsh onion did not change during the preservation period, whereas the hardness of the cotyledon and hypocotyl of the soybean sprouts significantly increased on the sixth month of their storage. The total bacterial counts of the A. scaber during the preservation period remained constant at $10^3$ CFU/g, whereas those of soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion decreased slightly to $10^2$ CFU/g. Coliform was not detected in any of the samples. The sensory evaluation showed that the preservation period that was used in this study did not significantly affect the marketability of the frozen vegetables. Therefore, it was considered that A. scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion can be safely preserved by freeze-storing them for up to 12 months.

Effects of the Ge Prearmophization Ion Implantation on Titanium Salicide Junctions (게르마늄 Prearmophization 이온주입을 이용한 티타늄 salicide 접합부 특성 개선)

  • We studied the effects of Ge preamorphization (PAM) on 0.25$\mu\textrm{m}$ Ti-salicide junctions using comparative study with As PAM. For each PAM schemes, ion implantations are performed at a dose of 2E14 ion/$\textrm{cm}^2$ and at 20keV energy using $^{75}$ /As+and GeF4 ion sources. Ge PAM showed better sheet resistance and within- wafer uniformity than those of As PAM at 0.257m line width of n +/p-well junctions. This attributes to enhanced C54-silicidation reaction and strong (040) preferred orientation of the C54-silicide due to minimized As presence at n+ junctions. At p+ junctions, comparable performance was obtained in Rs reduction at fine lines from both As and Ge PAM schemes. Junction leakage current (JLC) revels are below ~1E-14 A/$\mu\textrm{m}^{2}$ at area patterns for all process conditions, whereas no degradation in JLC is shown under Ge PAM condition even at edge- intensive patterns. Smooth $TiSi_2$ interface is observed by cross- section TEM (X- TEM), which supports minimized silicide agglomeration due to Ge PAM and low level of JLC. Both junction break- down voltage (JBV) and contact resistances are satisfactory at all process conditions.

Inhibition of Adipocyte Differentiation and Adipogenesis by Supercritical Fluid Extracts and Marc from Cinnamomum verum (초임계 추출 계피오일의 3T3-L1 지방전구세포의 분화 전사인자 억제에 의한 지방대사 조절)

  • This study was performed to evaluate the antiobesity effect of supercritical fluid extracts (SFC) and marc methanol extracts (SFM) from Cinnamomum verum in 3T3-L1 preadipocytes. In inducing the differentiation of 3T3-L1 preadipocytes in the presence of an adipogenic cocktail, iso-butylmethylanthine (IBMX), dexamathasone, and insulin, treatment with fraction residue SFC and SFM. SFC significantly reduced the mRNA expression of the transcription factor peroxisome proliferator-activate-dreceptor-${\gamma}$ ($PPAR{\gamma}$), the sterol regulatory-element-binding protein-1c (SREBP1c), and the CCAAT enhancer-binding-protein ${\alpha}$ ($C/EBP{\alpha}$) in a concentration-dependent manner. Moreover, SFC markedly down-regulated acyl-CoA synthetase-1 (ASC1), fatty acid synthesis (FAS), fatty acid transport-1 (FATP1), fatty acid binding protein 4 (FABP4), and perilipin. These findings suggest that SFC may be a potential therapeutic adjunct for obesity by targeting the differentiation of preadipocytes, as well as their functions.
