Selection and Characteristics of Fermented Salted Seafood (jeotgal)-Originated Strains with Excellent S-adenosyl-L-methionine (SAM) Production and Probiotics Efficacy

  • This study is executed to develop probiotics which produce S-adenosyl-L-methionine (SAM), a methyl group donor of the 5-methyltetrahydrofolate methylation reaction within the animal cell. SAM is an essential substance for the synthesis, activation, and metabolism of hormones, neurotransmitters, nucleic acids, phospholipids, and cell membranes of animals. The SAM is also known as a nutritional supplement to improve brain functions of the human. In this study, the SAM-producing strains are identified in 18 types of salted fish, and then, the strains with excellent SAM productions are being identified, with 1 strain in the Enterococcus genus and 9 strains in the Bacillus genus. Strains with a large amount of SAM production include the lactic acid bacteria such as En. faecium and En. durans, En. sanguinicola, as well as various strains in the Bacillus genus. The SAM-overproducing strains show antibacterial activities with certain harmful microbes in addition to the weak acid resistances and strong bile resistances, indicating characteristics of probiotics. It is possible that the jeotgal-originated beneficial strains with overproducing SAM can be commercially utilized in order to manufacture SAM enriched foods.

김치의 재료별 첨가량에 따른 SAM 함량의 변화 (Change of S-Adenosyl-L-Methionine(SAM) Content in Material by Addition of the Kimchi)

  • 본 연구에서는 발효기간 중 SAM 생성량 증진김치의 최적조건을 개발하기 위해 배추김치의 재료별 및 제조법에 따른 발효기간 중의 SAM 함량 변화를 측정하였다. 발효가 진행됨에 따라 모든 김치군의 pH는 발효초기에는 급격히 감소하다가 후기에는 pH $4.2{\sim}4.3$으로 비슷하게 나타났다. 산도는 pH와 비슷한 경향을 보이면서 발효초기에는 급격히 감소하다가 후기에는 서서히 증가하는 경향을 보였다. SAM 함량은 가장 기본적인 재료로만 제조한 표준(A)김치는 가장 낮게 나타났고, SAM이 검출되는 원 부재료에 SAM 함량이 많은 김치가 SAM 함량도 높게 나타났다. 모든 김치군 중 I김치가 가장 높은 SAM 함량을 나타냈으며, 김치별 SAM 함량이 가장 높은 발효시점은 A, K김치는 pH $4.6{\sim}4.9$, 산도 $0.38{\sim}0.57$0.57, D, H김치는 pH 6.3, 산도 $0.24{\sim}0.27$, E, G김치는 pH $6.1{\sim}6.6$, 산도 $0.05{\sim}0.14$0.14, B, C, F, I, J김치는 pH $4.3{\sim}5.3$, 산도 $0.45{\sim}1.02$에서 나타났다. 따라서 대부분의 김치는 적숙기시점이 되었을 때 B, E, G, H김치는 초기발효 때부터 SAM 함량이 높게 나타났다. 또한 발효가 진행됨에 따라 SAM 함량은 감소하는 경향을 보였다. 따라서 연구결과 주재료인 채소류와 젓갈의 함량비를 적절히 제조하여 최적의 SAM 생산능력의 발효시점과 김치맛에 영향을 끼치지 않는 관능적으로 적합하고 SAM 생성이 보다 효과적으로 유도되는 최적의 조건에 대한 실험들이 추가적으로 필요하다고 판단된다.

Effect of S-Adenosylmethionine on Hepatic Injury from Sequential Cold and Warm Ischemia

  • We investigated whether S-adenosylmethionine (SAM) treatment improved ischemic injury using perfused rat liver after sequential periods of 24 h cold and 20 min re-warming ischemia. SAM (100 $\mu\textrm{mol/L}$) was added to University of Wisconsin (UW) solution and Ringers lactate solution. After cold and sequential warm ischemia, releases of lactate dehydrogenase (LDH) and purine nucleoside phosphorylase (PNP) markedly increased during repefusion. The increase in PNP was significantly reduced by SAM treatment. While the concentration of reduced glutathione (GSH) in ischemic livers significantly decreased, the concentration of glutathione disulfide (GSSG) increased. This decrease in GSH and increase in GSSG were suppressed by SAM treatment. Lipid peroxidation was elevated in cold and warm ischemic and reperfused livers, but this elevation was also prevented by SAM treatment. Hepatic ATP levels were decreased in the ischemic and reperfused livers to 42% of the control levels. However, treatment with SAM resulted in significantly higher ATP levels and preserved the concentration of AMP in ischemic livers. Our findings suggest that SAM prevents oxidative stress and lipid peroxidation and helps preserve hepatic energy metabolism.

Age-related Increase of Sister Chromatid Exchange Frequency in Bone Marrow Cells of Senescence Accelerated Mouse and Its Inhibition by Chronic Treatment of Ginseng

  • Age-related change in the frequency of spontaneous sister chromatid exchange (SCE) and chromosornal aberrations were investigated in bone marrow cells of accelerated senescence-resistant mice (SAM R1) and senescence accelerated ones (SAM P1). And the effect of chronic treatment of ginseng extract (Panax ginseng C.A. Meyer) on these chromosomal abnormalities was tested in SAM P1. SCE frequency in the cells was progressively increased with age in both mice, but it was consistently higher in SAM P1 than in SAM R1 at all corresponding age. Chromosomal aberrations were, however, not significantly changed with age except that it was slightly increased in only aged SAM P1. Interestingly, the rate of these genetic instabilities in SAM P1 was remarkably retarded by long-term administration of ginseng water extract (0.05% in drinking water). These results suggest that frequency of spontaneous SCE in bone marrow cells increase in parallel with senescence of the mice, and SAM P1 is in the condition of being more exposed than SAM R1 to DNA damaging factors. These also indicate that long-term treatment of ginseng may reduce the genetic damage.

수체 추출을 위한 Geo-SAM 기법의 응용: 국토위성영상 적용 실험 (Application of Geo-Segment Anything Model (SAM) Scheme to Water Body Segmentation: An Experiment Study Using CAS500-1 Images)

  • Meta에서 신속한 영상 분할 기능을 제공하는 대규모 컴퓨터 비전 생성 모델을 발표한 이후, 여러 활용 분야에서 이를 적용하려는 연구가 이루어지고 있다. 이 연구에서는 위성 영상 자료에 Segment Anything Model (SAM)을 사용할 수 있는 QGIS 플러그인 Geo-SAM을 사용하여 수체 객체 탐지와 추출에 대한 SAM의 적용성을 조사해 보고자 하였다. 실험 대상 자료는 국토위성(Compact Advanced Satellite 500, CAS500-1) 영상을 사용하였다. 이 자료를 가지고 SAM을 적용하여 얻은 결과는 같은 입력 영상으로부터 수작업으로 제작한 수체 객체 자료, Open Street Map (OSM)의 수체 자료, 국토지리정보원의 수계 수치지도와 비교하였다. SAM 처리 결과와 비교 대상 자료를 이용하여 추출된 모든 객체를 대상으로 계산한 경계사각형의 교집합/합집합의 평균값을 나타내는 mean Intersection over Union (mIoU)은 각각 0.7490, 0.5905, 0.4921로 나타났고, 각 자료에서 공통으로 나타나거나 추출된 객체에 대해 계산한 결과는 차례대로 0.9189, 0.8779, 0.7715로 나타났다. SAM을 적용한 결과와 다른 비교 자료와의 공간적 일치도를 분석한 결과, SAM에서는 한 개의 수체 객체를 여러 개의 분할 요소로 나타내므로 수체 객체 분류를 지원하는 의미 있는 결과를 보이고 있음을 알 수 있다.

Commercial MgB2 superconducting wires at Sam Dong

  • Since 2014, Sam Dong Co., Ltd. has successfully developed high-performance MgB2 superconducting wires with a kilometer-scale. Herein, we studied performances of various MgB2 wires fabricated by the Sam Dong with different Cu fractions and diameters for practical applications. Critical current densities of our commercial wire, 18+'1'Cu multifilamentary MgB2 wire, are estimated to be 270,000 A/㎠ at 3 T and 4.2 K and 100,000 A/㎠ at 2 T and 20 K, respectively. We further discuss research progress of various MgB2 superconducting wires at Sam Dong Co., Ltd and make an effort to align with customers' requirements.

  • In Korean peninsular, Air Defense with SAM(Surface-to-Air Missile) is very important, because of threatening by North Korea's theater ballistic missiles installed with nuclear or biochemistry. Effective and successful defense operation largely depends on two factors, SAM's location and the number of SAM for each target based on missile's availability in each SAM's location. However, most previous papers have handled only the former. In this paper, we developed Multi-heuristic algorithm which can handle both factors simultaneously for solving allocation problem of the batteries and missile assignment problem in each battery. To solve allocation problem, genetic algorithm is used to decide location of the batteries. To solve missile assignment problem, a heuristic algorithm is applied to determine the number of SAM for each target. If the proposed model is applied to allocation of SAM, it will improve the effectiveness of missile defense operations.

노화 방지를 위한 한약재의 효능 연구 (I) (Age-related Changes in blood Chemistry and Thyroid hormones in Senescence Accelerated Mice(SAM R1 and SAM P6))

  • Aging process can be explained by many factors. In this study, we counted complete Blood Cells (CBC) such as WBC, Lymphocytes, Monocytes, Granulocytes, RBC, HGB, and HCT of both SAM P6 and SAM P1 during the aging process. Plasma albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea concentrations were also measured at the same time. In addition to these, plasma concentrations of cortisol, total T3, and total T4 were analyzed by Chemiluminescent Immunoassay. There were no change in CBC counts of SAM R1 and SAM P6 during this study. Plasma concentratins of albumin and glucose decreased significantly in SAM R1. However, plasma alkaline phosphatases and creatinine concentration in SAM P6 decreased significantly at 16 week after birth comparing to the control. Total T4 levels were siginificantly increased although cortisol and total T3 concentrations were the same in SAM R1 groups. Especially, the after birth of creatinine, alkaline phosphatase, T4 of SAM P6 at 16 week were significantly different form those of SAM R1. At 12 week after birth, pilose antler extract was given 5g/kg/day p.o. for 0, 7, 14, 21, and 30 days each in both SAM R1 and SAM P6. The RBC, HGB, and HCT levels started to increase significantly from 7 days after the dose at SAM P6 only. Total T4 conectrations were elevated gradually during the study although the antler extract administration did not prevent or inhibit the increase in total T4 concentration during the study. Therefore, the elevation of erythrocytes after administration of the extract needs to be studied in future.

In this paper, we developed Set covering model which can handle both factors simultaneously and Multi-heuristic algorithm for solving allocation problem of the batteries and missile assignment problem in each battery. Genetic algorithm is used to decide optimal location of the batteries. To determine the number of SAM, a heuristic algorithm is applied for solving missile assignment problem. If the proposed model is applied to allocation of SAM, it will improve the effectiveness of air defense operations.
